Abstract:In this report, a mitochondria-targeted probe based on an efficient platform constructed by pyridinium-coumarin and 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) with high selectivity and sensitivity toward SO2 derivatives was presented. As far as we know, pyridinium-coumarin is first used as a donor moiety to construct a FRET system. Compared with our previous work, it showed better solubility, higher biocompatibility and faster response speed. It also presented low detection limit (80.5 nM), excellent energy transfer efficiency (98.2%) and low cytotoxicity. Moreover, the probe could detect exogenous and endogenous bisulfite in living cells as well as in mitochondria successfully.

Abstract:We developed a new mitochondria-targeted ratiometric fluorescent probe (named PPA) for SO2 derivatives (HSO3-/SO32-) with a large (pseudo) Stokes shift (209 nm) based on FRET platform in which water-soluble (E)-4-(4-(dimethylamino)styryl)-1-alkylpyridin-1-ium was chosen as acceptor moiety and phenothiazine oxide as donor moiety. Probe PPA showed specific response toward HSO3-/SO32- over H2S, GSH, Cys and Hcy. The recognition group underwent a nucleophilic addition reaction with HSO3-/SO(3)(2- )to give rise to clear dual-emission signal changes in enhanced blue channel and subdued red channel under a single wavelength excitation. Moreover, probe PPA was successfully used for the detection of endogenous HSO3-/SO32- and bioimaging of mitochondria-targeted in living cells.

Abstract:Intracellular reactive oxygen species is involved in a wide variety of physiological and pathological processes. In this work, we have developed a new mitochondria-targeting probe (DPP-S) for superoxide anion detection with ratiometric fluorescence response. DPP-S exhibited an obvious color change from violet to orange along with a distinct fluorescence change with maximum emission peak from 652 to 545 nm in response to superoxide anion. The limit of detection of DPP-S for superoxide anion was calculated to be 20.5 nM. Imaging studies taken in MCF-7 and RAW264.7 cells showed that DPP-S could be employed as a ratiometric fluorescent probe for endogenous superoxide anion detection and imaging in living cells with a large emission shift. Furthermore, the colocalization study indicated that DPP-S can localize in mitochondria specifically. Finally, the fluorescent probe was successfully applied for superoxide anion imaging in mice.
A dicyanoisophorone-quinolinium-based near-infrared-emission fluorescent probe for ratiometric sensing of bisulfite/sulfite in living cells

Abstract:Sulfur dioxide (SO 2 ), as a known important sulfur oxide in RSS, has an essential role in achieving the homeostasis of organisms. The development of a highly sensitive and selective fluorescent probe for monitoring SO 2 is of great interest in understanding its pathophysiology. Herein, we developed a new near-infrared FRET-based fluorescent probe QD , in which quinolinium fluorophore was selected as the donor and the dicyanoisophorone fluorophore was selected as the acceptor. Probe QD had dual colorimetric and ratiometric fluorescence, larger Stokes shift (277 nm) and longer emission wavelength (677 nm). The fluorescence spectrum showed that QD was highly selective and sensitive with excellent linear response concentration range of 2.5 to 80 μM. Importantly, probe QD allowed detection of SO 2 derivatives and bioimaging of lysosome-targeted in live cells.
A multi-signal mitochondria-targetable fluorescent probe for simultaneously discriminating Cys/Hcy/H2S, GSH, and SO2 and visualizing the endogenous generation of SO2 in living cells

Abstract:Reactive sulfur species (RSS) include sulfur dioxide (SO2) and various biothiols, such as hydrogen sulfide (H2S), glutathione (GSH), cysteine (Cys), and homocysteine (Hcy). These RSS are closely related to each other through complicated symbiotic networks, and play indispensable roles in multiple pathophysiological processes. Due to their similarity in chemical properties, it is challenging for developing a single fluorescent probe to differentiate them simultaneously. Herein we report the first mitochondria-targetable fluorescent probe NIR-NBD for simultaneous discrimination of Cys/Hcy/H2S, GSH, and SO2 from each other. The probe is characterized of the presence of three different types of electrophilic sites, and displays multiple sets of signal pattern in response to Cys/Hcy/ H2S, GSH, and SO2, including red-green for Cys/Hcy/H2S, red for GSH, and green for SO2. Notably, the probe exhibits excellent selectivity, outstanding sensitivity (detection limit = 3.5 nM) and fast response (within 4 min) toward SO2. Furthermore, the probe has been successfully applied for monitoring mitochondrial SO2 and various biothiols, visualizing the endogenous generation of SO2, and probing into the role of biothiols during apoptosis process.

Abstract:Based on 4-bromo-1,8-naphthalic anhydride,one novel ratiometric fluorescence H 2 S-probe（IDNA） was designed and synthesized.Further studies indicate that IDNA can sensitively recognize H 2 S（detection limit of 7 μmol/L） with good selectivity and anti-interference ability.In addition,IDNA has satisfactory photostability in HeLa cells,ability of mitochondrial co-localization,and can be utilized in fluorescence imaging of H 2 S.

Abstract:The level of sulfur dioxide (SO2) and viscosity in mitochondria play vital roles in various of physiological and pathological processes. Abnormalities in mitochondrial SO2 and viscosity are closely associated with numerous biological diseases. It is of great significance to develop novel fluorescence probe for simultaneous detection of SO2 and viscosity within mitochondria. Herein, we have developed a water-soluble, mitochondrial-targeted and near-infrared fluorescent probe, CMBT, for the simultaneous detection of SO2 and viscosity. The probe CMBT incorporates benzothiazolium salt as a mitochondrial targeting moiety and 7-diethylaminocoumarin as a rotor for viscosity detection, respectively. Based on the prompt reaction between nucleophilic HSO3−/SO32− and the backbone of benzothiazolium salt derivative, probe CMBT displayed high sensitivity and selectivity toward SO2 with a limit of detection as low as 0.17 μM. As viscosity increased, the twisted intramolecular charge transfer (TICT) process was restricted, resulting in fluorescence emission enhancement at 690 nm. Moreover, probe CMBT demonstrated exceptional mitochondrial targeting ability and was successfully employed to image variation of SO2 and viscosity in living cells and mice. The work highlights the great potential of the probe as a convenient tool for revealing the relationship of SO2 and viscosity in biological systems.

Abstract:Stimulated Raman Scattering (SRS) coupled with alkyne tags has been an emerging imaging technique to visualize small-molecule species with high sensitivity and specificity. Here we describe the development of a ratiometric Raman probe for visualizing hydrogen sulfide (H2S) species in living cells as the first alkyne-based sensor for SRS microscopy. This probe uses an azide unit as a selective reactive site, and it targets mitochondria with high specificity. The SRS ratiometric images show a strong response to H2S level changes in living cells.

Abstract:Similar to nitric oxide (NO) molecule, carbon monoxide (CO) molecule and hydrogen sulfide (H 2 S) molecule, sulfur dioxide (SO 2 ) is also considered to be another important endogenous signaling molecule. SO 2 derivatives are widely used as food antioxidants and antimicrobial agents. However, SO 2 is also a toxic environmental pollutant and intake of elevated levels of sulfites can lead to diarrhea, low blood pressure and allergic and asthmatic reactions in susceptible people. The specific physiological effects and transformation processes of SO 2 and its derivatives are not well understood due to the limitations of existing detection methods. Therefore, it is an urgent need to develop highly sensitive, high-resolution, real-time in situ assays. Herein, we specifically designed a ratiometric near-infrared fluorescent probe CQT for detecting SO 2 derivatives. After the reaction of CQT with HSO 3 − /SO 3 2− , a gradual decrease in red fluorescence at 662 nm accompanied by an enhanced blue fluorescence at 489 nm, reaching a detection range of 2.8–80 μM. In addition, we found that CQT had high energy transfer efficiency, large Stokes shift and low detection limit, which meant that CQT could sensitively monitor small changes of SO 2 concentration in cells and accurately target lysosomes.

Abstract:Hydrogen sulfide (H2S) is a versatile modulator in mitochondria and involved in numerous diseases caused by mitochondrial dysfunction. Therefore, many efforts have been made to develop fluorescent probes for mitochondrial H2S detection. However, these cationic small molecule probes are inapplicable for in vivo imaging because of the shallow tissue penetration and poor biostability. Herein, a ratiometric upconversion luminescence nanoprobe with the acid-activated targeting strategy is developed for detecting and bioimaging of mitochondrial H2S. The merocyanine TPAMC modified upconversion nanophosphors, acting as the targeting and response component, are encapsulated into a pH-sensitive husk, composed of DSPE-PEG and poly(L-histidine)-b-PEG, which improved the nanoprobe's stability during transport in vivo. Under lysosomal pH, the PEG shell is interrupted and the targeting sites are exposed to further attach to mitochondria. Taking advantage of the LRET process between TPAMC and UCNPs, ratiometric detection of mitochondrial H2S can be achieved with high selectivity and sensitivity. Cellular testing reveals the precise targeting to mitochondria via a lysosme delivery process. Importantly, the nanoprobe can be used for monitoring of mitochondrial H2S levels in living cells and colon cancer mouse models.

Abstract:The sulfur dioxide (SO 2 )-associated dynamic interplay between mitochondria (Mito) and endoplasmic reticulum (ER) is essential for intracellular homeostasis regulation. Visualizing such organelles interactions can help elucidate basic pathogenesis. However, it is hampered by challenges in developing quick response, remarkable sensitivity and high selectivity within a single fluorescent probe for simultaneous and distinct tracking of their crosstalk. Herein, we engineered a SO 2 -responsive Mito-ER dual-targeted fluorescent probe (MER-SO 2 ) for dual-color visualization of Mito (red fluorescence) and ER (blue fluorescence) in living cells and in vivo. Under physiological conditions, the benzopyranium cationic structure in MER-SO 2 could target Mito, following fast response to SO 2 (5 s), which converted to a neutral structure, targeting to ER emitting blue fluorescence via functional fluorescence conversion. Probe MER-SO 2 also displayed high sensitivity, favourable specificity (against 24 types of non-targeting analytes). Importantly, such a high-performance probe allowed in-situ and dynamic tracking of Mito-ER interactions in Mito-dysfunction living cells and zebrafish via co-localization. Furthermore, the probe enabled accurate detection of sulphite in food with recoveries rate (96–100%), and incorporation into an agarose-based portable device for SO 2 assay. Taken together, this initial SO 2 -responsive probe represents a promising tool for discriminative tracking of functional Mito-ER interactions in living systems and holds great potential in many other fields such as food engineering.

Abstract:A novel chromenylium-based fluorescent probe was exploited for sulphur dioxide (SO₂) detecting. The probe displayed a remarkable fluorescence turn-on response towards SO₂ based on the nucleophilic addition reaction to the carbon-carbon double bond with 105 nm Stock shift. The probe was successfully applied for the quantification of SO₂.The linear detection range was from 0-160 μM with the detection limit as low as 99.27 nM. It also exhibited high selectivity for SO₂ than other reactive species and amino acids. Furthermore, cell staining experiments indicated that the probe was cell membrane permeable and could be used for high-performance imaging of SO₂ in living cells. The superior properties of the probe made it highly promising for use in chemical and biological applications.

Abstract:The objective of this study was to review the literature on bronchoalveolar lavage fluid cell profiles in healthy children and adolescents, as well as on the use of BAL as a diagnostic and follow-up tool for lung disease patients in this age bracket. To that end, we used the Medline database, compiling studies published between 1989 and 2009 employing the following MeSH descriptors (with Boolean operators) as search terms: bronchoalveolar lavage AND cytology OR cell AND child. In healthy children, the cell profile includes alveolar macrophages (> 80%), lymphocytes (approximately 10%), neutrophils (approximately 2%) and eosinophils (< 1%). The profile varies depending on the disease under study. The number of neutrophils is greater in wheezing children, especially in non-atopic children, as well as in those with pulmonary infectious and inflammatory profiles, including cystic fibrosis and interstitial lung disease. Eosinophil counts are elevated in children/adolescents with asthma and can reach high levels in those with allergic bronchopulmonary aspergillosis or eosinophilic syndromes. In a heterogeneous group of diseases, the number of lymphocytes can increase. Evaluation of the BAL fluid cell profile, when used in conjunction with clinical and imaging findings, has proven to be an essential tool in the investigation of various lung diseases. Less invasive than transbronchial and open lung biopsies, BAL has great clinical value. Further studies adopting standard international protocols should be carried out. Such studies should involve various age groups and settings in order to obtain reference values for BAL fluid cell profiles, which are necessary for a more accurate interpretation of findings in children and adolescents with lung diseases.
Rapid detection of SO2 in living cells and zebrafish by using an efficient near-infrared ratiometric fluorescent probe with large emission shift

Abstract:Sulfur dioxide (SO2), a possible endogenous gaseous signaling molecule, has been found to be correlated with various diseases in living organisms. Thus, the development of effective methods for detection of SO2 are of great physiological and pathological significance. In this work, we report a ratiometric NIR fluorescent probe Cou-Rh for monitoring bisulfite on basis of Michael addition reaction. This probe is prepared from coumarin by acid-catalyzed cyclization reaction. The addition of bisulfite results in a significant decrease of fluorescence intensity at 695 nm and enhancement at 550 nm. The probe also displays high selectivity over various biologically related analytes with nanomolar limit of detection. What's more, fluorescence imaging further confirms that the probe is capable of detecting bisulfite in living cells and zebrafish larvae. The results demonstrate that probe Cou-Rh can be employed for a useful scaffold to study the physiological function of sulfur dioxide.

A remarkable membrane-permeable fluorescent probe for real-time imaging of mitochondrial SO2 with high fidelity during ferroptosis

Abstract:Mitochondrial sulfur dioxide (SO2) plays a double-edged role in cells, and the real-time and in situ tracing of its dynamic behaviors to elucidate its complicated functions in detail is of great significance. Here, we developed a simple mitochondria-targeted fluorescent probe ZW for tracing SO2 with good membrane permeability. In probe ZW, the 1-phenylpyrrolidine-decorated benzopyrylium unit is employed as the selective response site for SO2. Besides, it also acts as the main fluorophore for signal conversion. The spectral results displayed that ZW could emit near-infrared (NIR) fluorescence (670 nm) and has a highly sensitive and selective response to SO2 (LOD = 0.19 μM). For biological imaging, compared with the control probe ZE, concentration- and time-dependent results verified that probe ZW has remarkable cell delivery with low concentration (200 nM) and fast response time (3 min). Furthermore, the NIR emission of ZW rendered high-fidelity imaging in living cells. Owing to its positive charge, ZW showed favorable mitochondria-targeting properties by colocalization experiments. Probe ZW could detect SO2 in real-time and in situ with high photostability in cells. Significantly, it has the ability to monitor the changes of endogenous SO2 during ferroptosis.
Structure-regulated mitochondrial-targeted fluorescent probe for sensing and imaging SO2 in vivo

Abstract:SO2 and its derivatives play an important role in the antioxidation and anticorrosion of food and medicine. In biological systems, abnormal levels of SO2 lead to the occurrence of many biological diseases. Hence, the development of suitable tools for monitoring SO2 in mitochondria is beneficial for studying the biological effect of SO2 in subcellular organelles. In this research, DHX-1 and DHX-2 are fluorescent probes designed on the basis of dihydroxanthene skeletons. Importantly, DHX-1 (650 nm) and DHX-2 (748 nm) show near-infrared fluorescence response toward endogenous and exogenous SO2, which showed advantages of great selectivity, good sensitivity and low cytotoxicity, and the detection limit is 5.6 μM and 4.08 μM of SO2, respectively. Moreover, DHX-1 and DHX-2 realized SO2 sensing in HeLa cells and zebrafish. Moreover, cell imaging demonstrated that DHX-2 with a thiazole salt structure possesses good mitochondria-targeting ability. Additionally, DHX-2 was perfectly achieved by in situ imaging of SO2 in mice.

Abstract:Sulfite (SO32-) is considered as a monitor of a wide range of physiological processes. However, cells and tissues are adversely affected when the body ingests high level of sulfite. Here, we designed and synthesized a "turn on" fluorescent probe ImiSft-1 with 2-cyano-N-methylacetamide as the specific recognition site of SO32-. This probe predominantly achieved high response intensity to SO32- and desirable properties such as large Stokes shift (similar to 180 nm), fast response time (within 15 s), and high sensitivity (LOD = 0.12 mu M). Importantly, the probe was highly selective for sulfite from other bio-species including biological thiols. Other functional properties included broad pH adaptability (5.0-10.0) and low cytotoxicity. Given these advantages and the fluorescence imaging in living MCF-7 cells, it was demonstrated that probe ImiSft-1 could monitor the changes of sulfite concentration in living cells.
