A Three-Dimensional DNAzyme Motor for Sensitive Imaging of Telomerase Activity in Living Cells

Abstract:Telomerase has been recognized as one of the common tumor markers since it has a high level in most tumor cells. Here, we construct a gold nanoparticle (AuNP)-based three-dimensional DNAzyme motor with DNAzyme strands that silenced by a locked strand. In the absence of telomerase, the DNAzyme strand hybridized with locking strand, the catalytic activity of DNAzyme strand was inhibited and the fluorescence of carboxy-fluorescein (FAM) was quenched by AuNP. In the presence of telomerase and Mn2+, TS-primer was extended and experienced the strand displacement reaction with the locking strands on DNAzyme, yielding an "active" DNAzyme. Then, tens or hundreds of substrate strands were cleaved and released by one extended primer. As a result, the fluorescence increased sharply. DNAzyme motor could be taken up by cells autonomously and the primers were delivered to cells by assembling on MnO2 nanosheets. The method was successfully applied to recognize telomerase in living cells by confocal imaging. It has also been applied to assay actual clinical samples with satisfactory, which is significant to clinical diagnosis.
