Abstract:Sensitive detection of telomerase and its inhibitor are of great significance to early diagnosis and treatment of malignant tumors. A cascade signal amplification strategy based on cyclic strand displacement reaction was designed to detect telomerase activity in cell lysates and living tumor cells. The primer can be elongated with repeated sequences of (TTAGGG)n due to the presence of telomerase. The telomerase extension product (TEP) competes C-DNA with G-DNA by forming more stable duplex. Thus, G-DNA was released and trigger the cyclic strand displacement reaction. Two molecular beacons (MB1 and MB2) were applied to fulfill the cyclic amplification reaction and report the fluorescence signal. Compared with unamplified OMB strategy, the limit of detection for telomerase activity with amplified TMB strategy was enhanced by two orders of magnitude. It provides a new enzyme-free and PCR-free amplification method for quantification and monitoring of telomerase activity in living tumor cells. It can also be utilized for distinguishing tumor cells from normal cells by in situ imaging. The monitoring of telomerase activity with inhibitor demonstrated its potential applications in the discovery of anti-cancer drugs.

Abstract:A new electrochemical method for telomerase activity assay was developed on the basis of hybridization chain reaction (HCR). assisted multiple signal amplification, aiming at improving the sensitivity and specificity of telomerase assay. The experiments utilized HeLa cells as original source of the telomerase in the electrochemical studies. The telomerase primer was firstly self-assembled on the surface of gold electrode. The telomerase catalyzed the elongation of the primer, producing the complementary sequences of hairpin probe H1. In this case, HCR was then initiated by interacting with two hairpin probes H1 and H2. Because both H1 and H2 were modified by biotin, horseradish peroxidase could be captured on the electrode surface through the high. affinity interaction between biotin and streptavidin, catalyzing the oxidation of o. phenylenediamine to produce 2,3-diaminophenazine. Therefore, the telomerase assay was realized by tracing the electrochemical signals with differential pulse voltammetry. This electrochemical method was of high efficiency and feasibility for detecting telomerase activity, and could trace the telomerase activity down to 10 cells/mL HeLa cells with a wide linear range. Besides, it could also easily distinguish the target enzyme from the control proteins with high specificity.
Enzymatic DNA Repair Cascade-Driven Fluorophore Encoding for Sensitively Sensing Telomerase Activity in Cancer Cells

Abstract:Telomerase is an important reverse transcriptase that is in charge of maintaining telomeres length, and the dysregulation of telomerase activity is closely associated with cancers. Accurate measurement of telomerase activity is critical to clinical diagnosis and therapeutics. Herein, by taking advantage of in vivo repair mechanisms and single-molecule detection, we demonstrate the enzymatic DNA repair cascade-driven fluorophore encoding for sensitively sensing telomerase activity in cancer cells. The presence of telomerase enables repetitive addition of (TTAGGG)(n) to the 3'-OH end of telomerase substrate (TS) primer to generate long telomeric products. Telomeric product then hybridizes with the apurinic (AP) probe to induce the cleavage of AP site in dsDNA by APE1, yielding a ssDNA with a biotin and an OH group at 5' and 3' ends and simultaneously releasing the telomeric product which in turn hybridizes with new AP probes to yield numerous ssDNAs. By adding streptavidin-coated MBs, the ssDNAs are captured and subsequently functioned as primers to initiate template-free TdT-catalyzed random incorporation of dATPs and Cy5-dATPs into 3'-OH ends, producing long poly-A chains with multiple Cy5 molecules encoding and finally forming Cy5-ssDNA-MB complexes. After magnetic separation, Cy5-ssDNA-MBs are digested by Exo I, releasing large numbers of Cy5 molecules which can be measured by single-molecule detection. This strategy exhibits excellent specificity and high sensitivity with a limit of detection (LOD) of 1 cancer cell. Moreover, it can be employed to screen inhibitors and discriminate different cancer cell lines from normal cell lines, holding enormous potential in clinical diagnosis and therapeutics.

Abstract:Telomerase, as a significant tumor biomarker, is highly associated with cancers. Highly sensitive determination of telomerase activity is beneficial to accurate diagnosis and treatment of malignant tumors. A new simple and elegant fluorescence sensing strategy based on thioflavin T (ThT) and conjugated polymer nanoparticles was established to determine telomerase activity in living cells. In the presence of telomerase, the primer is prolonged with repeated sequence of (TTAGGG)(n). This long G-riched sequence can form G-quadruplex in the mediation of ThT. The combination of ThT and G-quadruplex allows ThT to emit robust fluorescence. The fluorescence can be further amplified by positively charged conjugated polymer nanoparticles (PPE-PLL). The remarkable amplification is attributed to the disaggregation of PPE-PLL nanoparticles and a good superposition of fluorescence emission from ThT and PPE-PLL. The results demonstrated a new amplification assay for the determination of telomerase activity in cell extracts and living cells with high specificity and sensitivity. The limit of detection is estimated to be 3 HeLa cells in 400 mu L. It is not only more sensitive than most reported biosensors for telomerase detection. Still, it is extremely simple, rapid, and low-cost since it is PCR-free, label-free, and enzyme-free.

Abstract:Telomerase is a well-known cancer biomarker, and the reliable and sensitive detection of telomerase activity is of vital significance for cancer diagnosis and treatment. Despite many attempts, the sensitive methods for telomerase activity assay mainly depend on the various nucleic acid amplification, which requires complex primers/probes design and multiple enzymes. In addition, the multiple experimental procedures may introduce false-positive results from cross-contamination of amplification products challenging the reliability of telomerase-based diagnostics detection. Herein, we present a reliable, amplification-free, and sensitive strategy for the detection of telomerase activity based on the self-extension of telomerase and trans-cleavage of CRISPR/Cas12a. In this work, crRNA/Cas12a complexes can specifically target the self-extending telomere-repetitive DNA and activate the typical CRISPR/Cas12a-based fluorescent output system, which allows the detection of telomerase activity in very few cancer cells (5 HeLa cells) without complex primer design and any nucleic acid amplification. Furthermore, the proposed method holds high reliability and specificity in distinguishing tumor cells from normal cells. Therefore, this work may provide a new strategy for telomerase-related research and broaden the CRISPR-Cas12a-based sensing toolbox.

Abstract:Precise evaluation of telomerase activity is essential for the clinical diagnosis of early tumors. Herein, we have ingeniously designed a tetrahedral DNA nanostructure, with hairpin-shaped DNA probes rich in cytosine bases at four vertices for telomerase detection. The DNA-templated silver nanoclusters can be formed after the addition of Ag. Then the introduction of telomerase adds the single-strand TTAGGG extension, which can “turn on” the fluorescence of silver nanoclusters quickly by the proximity of the resulting guanine-rich sequences to silver nanoclusters and realize accurate detection of telomerase activity. In this study, integration of high stability tetrahedral DNA nanostructure and fluorescence signal amplification of four DNA-templated silver nanoclusters offers the advantage of high sensitivity, with a low detection limit of 1 cell. More than that, this method is low-cost, facile, and feasible for practical clinical applications.

Abstract:As a biomarker for early cancer diagnosis, the quantitative and sensitive detection of telomerase is very hot topic in the field of biological detection. In this work, we reported a novel telomerase assay strategy-based glass nanopipettes (GNs) which were constructed with DNA functionalized metal-organic frameworks (MOFs, PCN-224, C 144 H 112 N 12 O 64 Zr 15 ) as composite probes (DNA-PCN-224, DNA-PCN-224-MB (methylene blue)). The composite probes provided more probe binding sites, so the aggregation behavior of the composite probes would occur obviously in nanopores restricted domain space under the recognition and amplification of telomerase. The relative size of the GNs reduced due to the size effect of MOFs. This size effect exhibits a good signal-to-noise ratio in large pores, resulting in signal amplification. With different modifications of the GNs, on the one hand, the reduction of the relative pore size amplified the ionic current signal; on the other hand, the electrolytic current signal further verified the reliability of this signal amplification, which originated from the transfer of electrons occurring on the gold layer by the electroactive substance MB. The former quantified the assay, allowing a telomerase detection range of 0–10,000 Hela cells/mL and a detection limit of 100 Hela cells/mL. This work provided a detection platform for detecting related telomerase diseases and opened a new detection method of double signals.

Abstract:By maintaining the telomere lengths, telomerase can make the tumor cells avoid the apoptosis, thus, achieving the cell immortalization. In the past, a series of telomerase detection systems have been developed through utilizing the unique characteristic of telomerase extended primer. However, fluctuation of telomerase activity, along with the cell cycle progression, leads to ambiguous detection results. Here, we reported a dual signal output detection strategy based on cell-cycle synchronization for precisely detecting telomerase activities by using a new AIEgen probe SSNB. Experimental and simulating calculation results demonstrated that positively charged SSNB could interact with DNA through the electrostatic interaction and π–π interaction, as well as the hydrogen bonds. The aggregation of SSNB caused by the extended template strand primer (TP) could be observed in tumor cells, thus, indicating the telomerase activity in various cell lines. Furthermore, after cell cycle synchronization, it was found that the telomerase activity in the S phase was the highest, no matter from the fluorescence intensity or the ROS generation situation. Dual signal outputs of SSNB verified the significance and necessity of cell-cycle synchronization detection for telomerase activity. Abstract:We developed a multiplex single-molecule quantitative assay of intracellular telomerase that used target-triggered signal amplification to enhance sensitivity, substrate reaction to increase signal stability, and quantum dots to enhance signal-to-noise ratio, obtaining an LOD of 5 x 10-14 IU for intracellular telomerase and LOD of 3 cells for multiple cancer cells. An ultrasensitive detection method was developed, which combined the catalytic hairpin assay and the single-molecule assay to analyze intracellular telomerase.

Abstract:An intracellular cascade amplification strategy for ultrasensitive "off-on" imaging of telomerase activity in living cells was designed. The method was based on fabrication of a dual function module-encapsulated liposome nanoprobe, which consisted of a telomerase-targeting responder-transmitter DNA complex (HPT) module and a catalyzed hairpin assembly (CHA) signal amplification module. Upon transfected into living cells, the released HPT could be specifically recognized and extended by telomerase, leading to the release of the transmitter DNA. The transmitter could act as the initiator and catalyzer of CHA amplification, resulting in the lightening up of the reporter complex. The telomerase activity could be monitored in situ by the fluorescence signal without the need for obtaining cell extracts. Because of the recycling use of the transmitter, multiplied enhancement of signal outputs from one extension event was achieved. The proposed strategy could be employed for in situ monitoring of the change of intracellular telomerase activity in response to drugs to detect drug efficacy. Thus the method has great potential in the study of the molecular mechanisms of telomerase-related life processes.
Triple Signal Amplification Strategy for Ultrasensitive in Situ Imaging of Intracellular Telomerase RNA

Abstract:Abnormal upregulation of telomerase RNA (TR) is a hallmark event at various stages of tumor progression, providing a universal marker for early diagnosis of cancer. Here, we have developed a triple signal amplification strategy for in situ visualization of TR in living cells, which sequentially incorporated the target-initiated strand displacement circuit, multidirectional rolling circle amplification (RCA), and Mg2+ DNAzyme-mediated amplification. All oligonucleotide probes and cofactors were transfected into cells in one go, and then escaped from lysosomes successfully. Owing to the specific base pairing, the amplification cascades could only be triggered by TR and performed as programmed, resulting in a satisfactory signal-to-background ratio. Especially, the netlike DNA structure generated by RCA encapsulated high concentrations of DNAzyme and substrates (FQS) in a local region, thereby improving the reaction efficiency and kinetics of the third amplification cycle. Under optimal conditions, the proposed method exhibited ultrasensitive detection of TR mimic with a detection limit at pM level. Most importantly, after transfection with the proposed sensing platform, tumor cells can be easily distinguished from normal cells based on TR abundance-related fluorescence signal, providing a new insight into initial cancer screening.

Abstract:We report an electrochemical sensor for telomerase activity detection based on spherical nucleic acids gold nanoparticles (SNAs AuNPs) triggered mimic-hybridization chain reaction (mimic-HCR) enzyme-free dual signal amplification. In the detection strategy, SNAs AuNPs and two hairpin probes were employed. SNAs AuNPs as the primary amplification element, not only hybridized with the telomeric repeats on the electrode to amplify signal but also initiated the subsequent secondary amplification, mimic-hybridization chain reaction of two hairpin probes. If the cells' extracts were positive for telomerase activity, SNAs AuNPs could be captured on the electrode. The carried initiators could trigger an alternative hybridization reaction of two hairpin probes that yielded nicked double helices. The signal was further amplified enzyme-free by numerous hexaammineruthenium(III) chloride ([Ru(NH3)6](3+), RuHex) inserting into double-helix DNA long chain by electrostatic interaction, each of which could generate an electrochemical signal at appropriate potential. With this method, a detection limit of down to 2 HeLa cells and a dynamic range of 10-10,000 cells were achieved. Telomerase activities of different cell lines were also successfully evaluated.

Abstract:Telomerase, a key enzyme involved in telomere homeostasis, is a major player involved in or required for sustained cell proliferation. It is expressed in ∼90% tumor but rarely in normal somatic cells. Therefore, telomerase serves as a diagnostic marker and therapeutic target of cancers. Although many methods are available for measuring telomerase activity, a convenient, fast, sensitive, and reliable method is still lacking for routine use in both clinics and research. Here, we present a single-enzyme sensitivity telomere repeat amplification protocol for quantifying telomerase activity. With multiple optimizations, the protocol pushes the ultimate detection limit down to a single telomerase complex, enabling measurement of telomerase activity of not only multiple cancerous/normal cell samples but also single cancer cells alone or even in the presence of 8000 normal cells. Implemented in a one-step mix-and-run format, the protocol offers a most sensitive, fast, accurate, and reproducible quantification of telomerase activity with linearity ranging from 20,000 HeLa cancer cells to a single telomerase complex. It requires minimal manual operation and experimental skill and is convenient for either low or high throughput of samples. We expect that the protocol should provide practical routine analyses of telomerase in both research and clinical applications. As an example, we demonstrate how telomerase activity evolves at the single-cell level and partitions in cell division in early mouse embryo development.This article has not yet been cited by other publications.

Abstract:A stable and enzyme-free method is described for highly sensitive determination of telomerase activity. It is based on the use of a framework nucleic acid (FNA) nanomachine and doxorubicin-spherical nucleic acid (DSNA) tags. Upon incubation with telomerase, the primer-tetrahedron becomes elongated to form the handed swing arm. The extended swing arm autonomously moves along the predefined track consisting of entropy-tetrahedron by consecutive strand displacement under the aid of fuel-tetrahedron. As a result, many (entropy-tetrahedron)-(fuel-tetrahedron) complexes are assembled for combining the DSNA tags. This results in an amplified electrochemical signal, typically measured at around -0.63 V (Ag/AgCl). The use of an enzyme-free FNA nanomachine and of DSNA tags warrants outstandingly high stability and sensitivity. The method shows a broad dynamic correlation of telomerase activity in cell extracts. The analytical range extends from 10 to 1.0 × 104 HeLa cells mL-1 with a lower detection limit of 2 cells mL-1. The differences in telomerase activity between different cancer cells can be easily evaluated. The method was further verified by quantifying telomerase activity of cancer cells in accumulated normal cells. Therefore, the sensing method has great potential for clinical application. Abstract:This work reports on a simple and sensitive electrochemical strategy for the detection of telomerase extracted from HeLa cells. It was based on the signal amplification of in-situ performed streptavidin-biotin-DNA-biotin (SA-biotin-DNA-biotin) networks. Specifically, the DNA primer immobilized on a gold electrode was elongated by telomerase. After the hybridization of biotin-DNA-biotin to the extended primer, the in-situ formation of SA-biotin-DNA-biotin networks was initiated on the electrode surface within 10 min through the SA-biotin interactions. The resulting networks created an insulating layer, thus hampering the electron transfer and causing the significant increase in the electrochemical impedance. Telomerase extracted from 2 HeLa cells can be readily measured. The method showed a linear relationship in the range of 50 similar to 5000 cells/mL. The signal-amplified strategy can be applied to develop novel sensing platforms for the detection of different biomarkers.

Abstract:The strong correlation between cancer and telomerase activity has inspired the development of new strategies to evaluate telomerase activity. Here, a personal glucose meter (PGM) system that uses DNA-based machine amplification to detect telomerase in cancer cells is reported. In this assay, telomerase elongation products are amplified in the form of another type of product by a DNA-based machine. This process can only be activated by the hybridization of the extended telomerase substrate (TS) probe and the complementary primer in the presence of telomerase. The obtained products are then transformed to glucose-related signals via a three-component assay, which enables the simple use of a PGM to indirectly quantify the telomerase activity. The proposed method realizes sensitive telomerase activity detection down to 20 HeLa cells with a significantly enhanced dynamic range. Additionally, short telomerase elongation products, such as telomerase substrate probes with two repetitive sequences, that cannot be detected using the most widely used telomeric repeat amplification protocol assay were detected.

Abstract:We report a new assay for human telomerase activity that relies on polyvalent oligonucleotide nanoparticle conjugates as diagnostic probes and amplification units. Gold nanoparticles functionalized with specific oligonucleotide sequences can efficiently capture telomerase enzymes and subsequently be elongated. Both the elongated and unmodified oligonucleotide sequences are simultaneously measured. The two strands not only serve as internal positive controls for each other but also provide a way of amplifying signal. At high concentrations, both elongated and unmodified strands exhibit measurable responses. At low telomerase concentrations (e.g., from 10 HeLa cells), elongated strands cannot be detected, but the unmodified sequences, which come from the same probe particles, can be detected because their concentration is higher, providing a novel form of amplification. This new assay rivals the sensitivity of the conventional PCR-based method of telomerase detection.
DNAzyme-Mediated Biodeposition Coupling Adjustable Cascade Electric Fields for Photoelectrochemical Telomerase Activity Monitoring

Abstract:Telomerase, as a specialized reverse transcriptase, plays a vital role in early cancer diagnostics and prognosis; thus, developing efficient sensing technologies is of vital importance. Herein, an innovative "signal-on-off" photoelectrochemical (PEC) sensing platform was developed for ultrasensitive evaluation of telomerase activity based on an electron-transfer tunneling distance regulation strategy and DNAzyme-triggerable biocatalytic precipitation. Concretely, cascade internal electric fields between CuInS2 quantum dots (QDs), graphitic carbon nitride nanosheets (g-C3N4 NSs), and TiO2 nanorod arrays (NRAs) were developed to realize cascade electron extraction and hole transfer. Enabled by such a design, an effective "signal-on" state to gain a progressively enhanced PEC output was designed by suppressing the photogenerated electron-hole pair recombination. With the introduction of hairpin probe H2 and the subsequent extension of the primer sequence driven by the target telomerase, the CuInS2 QDs labeled with hairpin probe H1 were programmatically unfolded, resulting in CuInS2 QDs' close proximity to the working electrode away from the cascade interface, accompanied by the formation of G-quadruplex/hemin complexes. The gradual undermining of tunneling distance and implantation of DNAzyme-initiating biocatalytic precipitation tremendously induced the sluggish migration kinetics of the photoinduced charge, accompanied by the photocurrent intensity decrement, leading to the "signal-off" state. Under optimized conditions, the as-prepared PEC biosensor realizes ultrasensitive detection of telomerase activity from 10 to 105 cell·mL-1 with a detection limitation of 3 cells·mL-1. As a proof of concept, this well-designed method provides new insights into signal amplification for telomerase activity evaluation and also presents promising potential for further development in drug screening, healthcare diagnostics, and biological assays.

Abstract:Sensitive and specific sensing of intracellular telomerase is of significance for early cancer diagnosis and treatment. And, efficient signal amplification technology is a powerful tool to achieve low level biomarker detection. However, the delivery and release of amplification reagents in live cells is currently a major challenge to achieve sensitive intracellular detection of biomarkers. Here, we describe the development of a powerful in-cell amplification approach for label-free and highly sensitive detection of telomerase in live cells via zeolitic imidazolate framework-8 (ZIF-8)-mediated DNAzyme/light-up RNA aptamer transcription amplification cascades. The amplification reagents can be readily transported into and released in the target cells by ZIF-8 under intracellular acidic environment. Telomerase then activates DNAzyme to trigger the DNAzyme-aided cleavage reaction and T7 RNA polymerase-initiated light-up RNA aptamer transcription cycles for the yield of many light-up RNA aptamers, which associate with the non-fluorescent dye to recover significant fluorescence for achieving intracellular telomerase imaging with high sensitivity in a label-free approach. Moreover, the proposed strategy can discriminate telomerase level variations in different cells and shows promising potentials for anticancer drug screening, which makes such method a robust bioassay for early tumor-related diagnosis and therapeutic monitoring.
