Abstract:The general electrochemical biosensors for telomerase detection require the immobilization of primers on the electrode surface for telomeric extension and hybridization reactions. However, immobilization of primers may suffer from the challenges of hindrance effect and configuration freedom, thus reducing the extension and hybridization efficiency. Herein, we developed a sensitive electrochemical biosensor for telomerase detection by integration of homogeneous extension and hybridization reactions and surface-tethered detection. In the presence of telomerase, the biotinylated primer (bio-primer) was efficiently elongated with telomeric repeats of (TTAGGG) n at the 3' end in solution. Then, the extension product (bio-DNA) was hybridized with the signal probe DNA modified on the surface of ferrocene (Fc)-capped gold nanoparticle (AuNP). The bio-DNA/DNA/Fc-AuNP hybrids were then tethered by streptavidin-modified electrodes through the specific avidin-biotin interactions, thus producing strong electrochemical signals from the oxidation of Fc tags. The biosensor was successfully used to determine telomerase in HeLa cells and monitor the inhibition efficiency of inhibitor. A wide linear range for the detection of telomerase extracted from HeLa cells was attained. This method has great potential in clinical diagnosis and anti-cancer drug development, and should be beneficial for the fabrication of novel biosensors by integration of homogeneous catalysis and hybridization reactions.

Abstract:Telomerase activity detection has attracted much attention concerning its importance for early cancer diagnosis. Here, we established a ratiometric electrochemical biosensor for telomerase detection based on CuS quantum dots (CuS QDs) dependent DNAzyme-regulated dual signals. The telomerase substrate probe was used as the linker to combine the DNA fabricated magnetic beads and CuS QDs. In this way, telomerase extended the substrate probe with repeated sequence to from hairpin structure, releasing CuS QDs as an input to DNAzyme modified electrode. DNAzyme was cleaved with high current of ferrocene (Fc) and low current of methylene blue (MB). On the basis of the obtained ratiometric signals, telomerase activity detection was achieved in the range of 1.0 × 10-12-1.0 × 10-6 IU/L, with the limit of detection down to 2.75 × 10-14 IU/L. Moreover, telomerase activity from HeLa extracts was also tested to verify the clinical application.

Abstract:BACKGROUND The aim of this study was to assess whether the morphological and functional changes of the left atrium (LA) differ after catheter ablation (ABL) from those after electrical cardioversion (ECV) in atrial fibrillation (AF). METHODS AND RESULTS AF patients who had maintained sinus rhythm for 3 months after either ECV (n=30) or ABL (n=33) were studied. Both 2-dimensional and Doppler echocardiography were performed at baseline, 1 week, 1 month, and 3 months after these procedures. LA dimensions, mitral inflow velocity, and tissue Doppler imaging of the mitral annulus were examined. LA dimensions decreased in both groups. The mean late mitral annulus velocity increased from 1 week to 3 months in both groups, but the mean late transmitral peak velocity increased only in the ECV group (42.1+/-14.4 cm/s to 56.7+/-14.8 cm/s, p<0.001 vs 38.7+/-12.5 cm/s to 44.8+/-16.7 cm/s, p=NS). CONCLUSION Reverse morphological remodeling of the LA occurred after successful ABL and ECV for AF. However, because LA function was lower in the ABL group than in the ECV group and did not recover to the baseline levels until 3 months after ABL, a meticulous anticoagulation program should be considered.

Abstract:A new electrochemical method for telomerase activity assay was developed on the basis of hybridization chain reaction (HCR). assisted multiple signal amplification, aiming at improving the sensitivity and specificity of telomerase assay. The experiments utilized HeLa cells as original source of the telomerase in the electrochemical studies. The telomerase primer was firstly self-assembled on the surface of gold electrode. The telomerase catalyzed the elongation of the primer, producing the complementary sequences of hairpin probe H1. In this case, HCR was then initiated by interacting with two hairpin probes H1 and H2. Because both H1 and H2 were modified by biotin, horseradish peroxidase could be captured on the electrode surface through the high. affinity interaction between biotin and streptavidin, catalyzing the oxidation of o. phenylenediamine to produce 2,3-diaminophenazine. Therefore, the telomerase assay was realized by tracing the electrochemical signals with differential pulse voltammetry. This electrochemical method was of high efficiency and feasibility for detecting telomerase activity, and could trace the telomerase activity down to 10 cells/mL HeLa cells with a wide linear range. Besides, it could also easily distinguish the target enzyme from the control proteins with high specificity.
Electrochemical Detection of Telomerase in Cancer Cells Based on the In-Situ Formation of Streptavidin-Biotin-dna-biotin Networks for Signal Amplification

Abstract:This work reports on a simple and sensitive electrochemical strategy for the detection of telomerase extracted from HeLa cells. It was based on the signal amplification of in-situ performed streptavidin-biotin-DNA-biotin (SA-biotin-DNA-biotin) networks. Specifically, the DNA primer immobilized on a gold electrode was elongated by telomerase. After the hybridization of biotin-DNA-biotin to the extended primer, the in-situ formation of SA-biotin-DNA-biotin networks was initiated on the electrode surface within 10 min through the SA-biotin interactions. The resulting networks created an insulating layer, thus hampering the electron transfer and causing the significant increase in the electrochemical impedance. Telomerase extracted from 2 HeLa cells can be readily measured. The method showed a linear relationship in the range of 50 similar to 5000 cells/mL. The signal-amplified strategy can be applied to develop novel sensing platforms for the detection of different biomarkers.
DNAzyme-Mediated Biodeposition Coupling Adjustable Cascade Electric Fields for Photoelectrochemical Telomerase Activity Monitoring

Abstract:Telomerase, as a specialized reverse transcriptase, plays a vital role in early cancer diagnostics and prognosis; thus, developing efficient sensing technologies is of vital importance. Herein, an innovative "signal-on-off" photoelectrochemical (PEC) sensing platform was developed for ultrasensitive evaluation of telomerase activity based on an electron-transfer tunneling distance regulation strategy and DNAzyme-triggerable biocatalytic precipitation. Concretely, cascade internal electric fields between CuInS2 quantum dots (QDs), graphitic carbon nitride nanosheets (g-C3N4 NSs), and TiO2 nanorod arrays (NRAs) were developed to realize cascade electron extraction and hole transfer. Enabled by such a design, an effective "signal-on" state to gain a progressively enhanced PEC output was designed by suppressing the photogenerated electron-hole pair recombination. With the introduction of hairpin probe H2 and the subsequent extension of the primer sequence driven by the target telomerase, the CuInS2 QDs labeled with hairpin probe H1 were programmatically unfolded, resulting in CuInS2 QDs' close proximity to the working electrode away from the cascade interface, accompanied by the formation of G-quadruplex/hemin complexes. The gradual undermining of tunneling distance and implantation of DNAzyme-initiating biocatalytic precipitation tremendously induced the sluggish migration kinetics of the photoinduced charge, accompanied by the photocurrent intensity decrement, leading to the "signal-off" state. Under optimized conditions, the as-prepared PEC biosensor realizes ultrasensitive detection of telomerase activity from 10 to 105 cell·mL-1 with a detection limitation of 3 cells·mL-1. As a proof of concept, this well-designed method provides new insights into signal amplification for telomerase activity evaluation and also presents promising potential for further development in drug screening, healthcare diagnostics, and biological assays.

Abstract:We report an electrochemical sensor for telomerase activity detection based on spherical nucleic acids gold nanoparticles (SNAs AuNPs) triggered mimic-hybridization chain reaction (mimic-HCR) enzyme-free dual signal amplification. In the detection strategy, SNAs AuNPs and two hairpin probes were employed. SNAs AuNPs as the primary amplification element, not only hybridized with the telomeric repeats on the electrode to amplify signal but also initiated the subsequent secondary amplification, mimic-hybridization chain reaction of two hairpin probes. If the cells' extracts were positive for telomerase activity, SNAs AuNPs could be captured on the electrode. The carried initiators could trigger an alternative hybridization reaction of two hairpin probes that yielded nicked double helices. The signal was further amplified enzyme-free by numerous hexaammineruthenium(III) chloride ([Ru(NH3)6](3+), RuHex) inserting into double-helix DNA long chain by electrostatic interaction, each of which could generate an electrochemical signal at appropriate potential. With this method, a detection limit of down to 2 HeLa cells and a dynamic range of 10-10,000 cells were achieved. Telomerase activities of different cell lines were also successfully evaluated.

Abstract:Telomerase activity has piqued scientists' interest for the reason that it has the potential to be employed for early-stage cancer detection, anticancer therapy and studies related to cancer progression and metastasis. Several approaches have been developed to detect telomerase activity. However, these approaches were lengthy, challenging to quantify, limited sensitivity and prone to polymerase chain reaction (PCR)-related artefacts. We herein developed a novel nanoplasmonic sensing platform for colorimetric detection of telomerase activity relying on the telomere elongation of telomerase at the 3' end, structure-specific cleavage activity of exonuclease III that removes mononucleotides from the 3 ́-hydroxyl termini of double-stranded DNA, and electrostatic interaction of elongated telomeres with plasmonic nanoparticles. Using HeLa cells as a model for colorimetric detection of telomerase activity, this biosensor could detect telomerase activity with detection limit of 100 cells/reaction by visible inspection and 5 cells/reaction by spectroscopic measurement, and analysis time within about three hours. The proposed sensing method provided a novel tool for simple, rapid, and low-cost detection of telomerase activity, eliminating the necessity for thermal cycling, primers in PCR-based assays, and amplification of telomerase extension products. It exhibits significant potential as a label-free, simple, ultrasensitive strategy for on-site detection of telomerase activity in proteomics and clinical diagnostics.

Abstract:We developed a multiplex single-molecule quantitative assay of intracellular telomerase that used target-triggered signal amplification to enhance sensitivity, substrate reaction to increase signal stability, and quantum dots to enhance signal-to-noise ratio, obtaining an LOD of 5 x 10-14 IU for intracellular telomerase and LOD of 3 cells for multiple cancer cells. An ultrasensitive detection method was developed, which combined the catalytic hairpin assay and the single-molecule assay to analyze intracellular telomerase.

Abstract:Telomerase, which is regarded as a common biomarker for early cancer diagnostics and a potential target for clinical therapies, has attracted considerable interests concerning its detection and monitoring. Herein, we propose a sensitive method by designing a gold nanoparticle (AuNP) probe for visually intracellular detection of telomerase activity. The AuNPs were functionalized with a telomerase substrate primer (SH‐prime). A 6‐carboxy‐fluorescein (FAM) modified strand (FAM‐probe) was attached to the surface of AuNP through its complementary stand (SH‐attach).In the absence of telomerase, the fluorescence resonance energy transfer (FRET) from FAM to AuNPs results in efficient fluorescence quenching. In the presence of telomerase, SH‐primers on AuNPs were extended with the repeat units (TTAGGG)n. The extension sequence triggered the strand displacement of FAM‐probe to restore the fluorescence signals. It is worth mentioning that the proposed strategy does not need to design complex hairpin structure and allows the measurement of telomerase in crude cell extracts down to 0.5 HeLa cells/μL in 2 h. In addition, the present sensing platform can be applied to the visually intracellular detection of telomerase activity in living cells.

Abstract:In this paper, we report a non-PCR-based electrochemical assay that can detect telomerase activity. Telomerase from HeLa cells may induce telomerization of thiolated primers immobilized on a gold electrode surface. With the telomerization reaction, more and more guanine-rich telomeric repeats are formed, so the electrochemical oxidation signal of guanine at about 1.00V, which is utilized to indicate the elongated guanine-rich telomeric repeats tethered to the primers, will be increased. This assay method can detect the telomerase activity originated from 3000 HeLa cells and thus holds promise as a simple and sensitive approach in clinical diagnosis of cancer.

Abstract:Telomerase is an important reverse transcriptase that is in charge of maintaining telomeres length, and the dysregulation of telomerase activity is closely associated with cancers. Accurate measurement of telomerase activity is critical to clinical diagnosis and therapeutics. Herein, by taking advantage of in vivo repair mechanisms and single-molecule detection, we demonstrate the enzymatic DNA repair cascade-driven fluorophore encoding for sensitively sensing telomerase activity in cancer cells. The presence of telomerase enables repetitive addition of (TTAGGG)(n) to the 3'-OH end of telomerase substrate (TS) primer to generate long telomeric products. Telomeric product then hybridizes with the apurinic (AP) probe to induce the cleavage of AP site in dsDNA by APE1, yielding a ssDNA with a biotin and an OH group at 5' and 3' ends and simultaneously releasing the telomeric product which in turn hybridizes with new AP probes to yield numerous ssDNAs. By adding streptavidin-coated MBs, the ssDNAs are captured and subsequently functioned as primers to initiate template-free TdT-catalyzed random incorporation of dATPs and Cy5-dATPs into 3'-OH ends, producing long poly-A chains with multiple Cy5 molecules encoding and finally forming Cy5-ssDNA-MB complexes. After magnetic separation, Cy5-ssDNA-MBs are digested by Exo I, releasing large numbers of Cy5 molecules which can be measured by single-molecule detection. This strategy exhibits excellent specificity and high sensitivity with a limit of detection (LOD) of 1 cancer cell. Moreover, it can be employed to screen inhibitors and discriminate different cancer cell lines from normal cell lines, holding enormous potential in clinical diagnosis and therapeutics.

Abstract:Telomerase, as a significant tumor biomarker, is highly associated with cancers. Highly sensitive determination of telomerase activity is beneficial to accurate diagnosis and treatment of malignant tumors. A new simple and elegant fluorescence sensing strategy based on thioflavin T (ThT) and conjugated polymer nanoparticles was established to determine telomerase activity in living cells. In the presence of telomerase, the primer is prolonged with repeated sequence of (TTAGGG)(n). This long G-riched sequence can form G-quadruplex in the mediation of ThT. The combination of ThT and G-quadruplex allows ThT to emit robust fluorescence. The fluorescence can be further amplified by positively charged conjugated polymer nanoparticles (PPE-PLL). The remarkable amplification is attributed to the disaggregation of PPE-PLL nanoparticles and a good superposition of fluorescence emission from ThT and PPE-PLL. The results demonstrated a new amplification assay for the determination of telomerase activity in cell extracts and living cells with high specificity and sensitivity. The limit of detection is estimated to be 3 HeLa cells in 400 mu L. It is not only more sensitive than most reported biosensors for telomerase detection. Still, it is extremely simple, rapid, and low-cost since it is PCR-free, label-free, and enzyme-free.

Abstract:Precise evaluation of telomerase activity is essential for the clinical diagnosis of early tumors. Herein, we have ingeniously designed a tetrahedral DNA nanostructure, with hairpin-shaped DNA probes rich in cytosine bases at four vertices for telomerase detection. The DNA-templated silver nanoclusters can be formed after the addition of Ag. Then the introduction of telomerase adds the single-strand TTAGGG extension, which can “turn on” the fluorescence of silver nanoclusters quickly by the proximity of the resulting guanine-rich sequences to silver nanoclusters and realize accurate detection of telomerase activity. In this study, integration of high stability tetrahedral DNA nanostructure and fluorescence signal amplification of four DNA-templated silver nanoclusters offers the advantage of high sensitivity, with a low detection limit of 1 cell. More than that, this method is low-cost, facile, and feasible for practical clinical applications.

Abstract:A stable and enzyme-free method is described for highly sensitive determination of telomerase activity. It is based on the use of a framework nucleic acid (FNA) nanomachine and doxorubicin-spherical nucleic acid (DSNA) tags. Upon incubation with telomerase, the primer-tetrahedron becomes elongated to form the handed swing arm. The extended swing arm autonomously moves along the predefined track consisting of entropy-tetrahedron by consecutive strand displacement under the aid of fuel-tetrahedron. As a result, many (entropy-tetrahedron)-(fuel-tetrahedron) complexes are assembled for combining the DSNA tags. This results in an amplified electrochemical signal, typically measured at around -0.63 V (Ag/AgCl). The use of an enzyme-free FNA nanomachine and of DSNA tags warrants outstandingly high stability and sensitivity. The method shows a broad dynamic correlation of telomerase activity in cell extracts. The analytical range extends from 10 to 1.0 × 104 HeLa cells mL-1 with a lower detection limit of 2 cells mL-1. The differences in telomerase activity between different cancer cells can be easily evaluated. The method was further verified by quantifying telomerase activity of cancer cells in accumulated normal cells. Therefore, the sensing method has great potential for clinical application. Extending model checkers for hybrid system verification: the case study of SPIN

Abstract:A hybrid system is a system that evolves following a continuous dynamic, which may instantaneously change when certain internal or external events occur. Because of this combination of discrete and continuous dynamics, the behaviour of a hybrid system is, in general, difficult to model and analyse. Model checking techniques have been proven to be an excellent approach to analyse critical properties of complex systems. This paper presents a new methodology to extend explicit model checkers for hybrid systems analysis. The explicit model checker is integrated, in a non‐intrusive way, with some external structures and existing abstraction libraries, which store and manipulate the abstraction of the continuous behaviour irrespective of the underlying model checker. The methodology is applied to SPIN using Parma Polyhedra Library. In addition, the authors are currently working on the extension of other model checkers. Copyright © 2013 John Wiley & Sons, Ltd.

Abstract:The accurate evaluation of telomerase activity, a typical cancer biomarker, is vital for early cancer screening. In this study, we developed a dark-field microscopy (DFM) visual single-particle detection scheme to detect telomerase activity based on automatic counting gold nanoparticles (AuNPs). This method started with attaching the telomerase substrate (TS) primer to the magnetic beads (MBs) through streptavidin-biotin interaction. In the presence of telomerase and dNTPs, the TS primer was expanded with (TTAGGG)(n) repeat units to form the telomerase extension product (MBs-telomerase extension product), which could be hybridized with the complementary DNA (cDNA) modified with AuNPs through Au-S bonds (AuNPs-SH-cDNA). After magnetic separation and DNA double-strand unwinding, AuNPs were collected from the supernatant, and the telomerase activity was quantitatively measured by visually counting bright spots based on DFM. This strategy achieved a limit of detection as low as 1 HeLa cell and distinguished telomerase activity among different cell lines, thus verifying its excellent sensitivity and specificity. Further, two common telomerase inhibitors (BIBR1532 and curcumin) were screened with the consistent IC50 values with other methods, respectively. It is worth mentioning that this strategy can clearly identify bladder cancer among various urinary diseases. Consequently, the visualized automatic particle counting strategy is potential as a powerful tool in early and noninvasive diagnosis of bladder cancer.
