Abstract:Layer-by-layer (LBL) assembly is a simple and effective method for the fabrication of a three-dimensional (3D) scaffold for nanotechnological and biomedical applications. Herein, a novel 3D scaffold based on an alternate LBL assembly of graphene oxide (GO) nanosheets and fibrinogen nanofibers (Fg NFs) on a silicon substrate was fabricated and utilized to create a 3D hydroxyapatite (HA) scaffold by biomimetic mineralization in 1.5x simulated body fluid for different nucleation periods. The obtained 3D (GO-NF)(n)-HA scaffold was characterized using atomic force microscopy, scanning electron microscopy, transmission electron microscopy, X-ray diffraction, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. The results demonstrate that the Fg NFs could promote the nucleation and growth of HA crystals along the axis. The 3D (GO-NF)(10)-HA scaffold composed of 10 layers of GO alternating with 10 layers of NFs was successfully created by LBL assembly and subsequent biomimetic mineralization. The effects of the created 3D (GO-NF)(10)-HA scaffolds on the adhesion, morphology, and proliferation of L-929 cells were investigated. The in vitro cell culture indicates that the 3D (GONF)(10)-HA scaffold has a higher proliferation ability and better cytocompatibility than the other control samples.

Abstract:Biomolecules-mediated biomimetic mineralization has been extensively investigated and applied to fabricate nano-assemblies with unique hierarchical architectures and salient properties. The confined-source ion diffusion plays a key role in the biomineralization process, but little investigative efforts have focused on it. Here, we developed a simple method to mimic the in vivo condition by a confined diffusion method, and hydroxyapatite nanoflower assemblies (HNAs) with exquisite hierarchical architectures were obtained. The HNAs were assembled from needle-like hybrid nanocrystals of hydroxyapatite and hyaluronan. The results revealed that the strong interactions between ions and hyaluronan led to the nucleation of hydroxyapatite and the following aggregation. The combination of the external diffusion field and the internal multiple interactions induced the self-assembling processes. Additionally, HNAs with colloid stability and excellent biocompatibility were proved to be a promising cargo carrier for intranuclear delivery. This work presents a novel biomimetic mineralization strategy based on confined diffusion system for fabricating delicate hydroxyapatite, which offers a new perspective for the development of biomimetic strategies.

Abstract:Up to now, effectively assembling micro biomimetic CaCO3 particles to form a functional material is still an important research topic. In our study, bacterial cellulose is used to in-situ assemble micro biomimetic lamellar CaCO3 particles induced by egg white and a new hybrid is obtained. Experimental data suggests that the hybrid has a rough surface and an elaborate three-dimensional structure with controllable porosity and the cycle stress–strain curves show that it has fairly good elasticity. Its hardness is adjustable allowing it to become comparable to the middle hardness plastic during a 72h mineralization procedure. The analysis on its application prospects shows that it is very suitable to be used as a multifunctional scaffold material in different fields of tissue engineering.

Abstract:Hydroxyapatite (HA) coatings were introduced onto Poly L-lactic Acid (PLLA) polymer in a controlled manner by immobilized urease method with a shortened precipitation time. Osteoblastic-like cellular responses to the composite were examined in terms of cell proliferation, differentiation and cell morphology, as well as the expression of bone-associated genes. The cells exhibited higher cellular proliferation at 2 and 4 days on the HA/PLLA composite compared to PLLA scaffold, while no significant difference was observed later at 6 days. The alkaline phosphatase (ALP) activity by cells at 7 days was statistically higher on HA/PLLA scaffold than on PLLA. Moreover, the gene expression of ALP and osteocalcin (OC) was up regulated on HA/PLLA composite by RT-PCR analysis. The preliminary study suggested that the use of the controlled modification of hydroxyapatite coating on PLLA scaffold to produce HA/PLLA composite might enhance cellular activity, indicating the potential use for bone substitute in tissue engineering.
Preparation and Characterization of Bionic Bone Structure Chitosan/hydroxyapatite Scaffold for Bone Tissue Engineering.

Abstract:Three-dimensional oriented chitosan (CS)/hydroxyapatite (HA) scaffolds were prepared via in situ precipitation method in this research. Scanning electron microscopy (SEM) images indicated that the scaffolds with acicular nano-HA had the spoke-like, multilayer and porous structure. The SEM of osteoblasts which were polygonal or spindle-shaped on the composite scaffolds after seven-day cell culture showed that the cells grew, adhered, and spread well. The results of X-ray powder diffractometer and Fourier transform infrared spectrometer showed that the mineral particles deposited in the scaffold had phase structure similar to natural bone and confirmed that particles were exactly HA. In vitro biocompatibility evaluation indicated the composite scaffolds showed a higher degree of proliferation of MC3T3-E1 cell compared with the pure CS scaffolds and the CS/HA10 scaffold was the highest one. The CS/HA scaffold also had a higher ratio of adhesion and alkaline phosphate activity value of osteoblasts compared with the pure CS scaffold, and the ratio increased with the increase of HA content. The ALP activity value of composite scaffolds was at least six times of the pure CS scaffolds. The results suggested that the composite scaffolds possessed good biocompatibility. The compressive strength of CS/HA15 increased by 33.07% compared with the pure CS scaffold. This novel porous scaffold with three-dimensional oriented structure might have a potential application in bone tissue engineering.

Abstract:Biomineralization generates hierarchically structured minerals with vital biological functions in organisms. This strategy has been adopted to construct complex architectures to achieve similar functionalities, mostly under chemical environments mimicking biological components. The molecular origin of the biofacilitated mineralization process is elusive. Herein, we describe the mineralization of hydroxyapatite (HAp) accompanying the biological secretion of nanocellulose by Acetobacter xylinum. In comparison with mature cellulose, the newly biosynthesized cellulose molecules greatly accelerate the nucleation rate and facilitate the uniform distribution of HAp crystals, thereby generating composites with a higher Young modulus. Both simulations and experiments indicate that the biological metabolism condition allows the easier capture of calcium ions by the more abundant hydroxyl groups on the glucan chain before the formation of hydrogen bonding, for the subsequent growth of HAp crystals. Our work provides more insights into the biologically accelerated mineralization process and presents a different methodology for the generation of biomimetic nanocomposites.
Biomimetic 3D Hydroxyapatite Architectures with Interconnected Pores Based on Electrospun Biaxially Orientated PCL Nanofibers

Abstract:We report here a facile strategy to fabricate three-dimensional (3D) hydroxyapatite (HA) architectures with well-defined long continuous interconnected pores by using electrospinning and biomimetic mineralization. To this end, a polymeric nanofiber (NF) scaffold with well-defined architecture was fabricated by electrospinning, and bone morphogenetic protein 2 (BMP2) was then adsorbed onto the chemically modified NFs through bio-conjugation. The 3D nanoporous HA architecture was finally fabricated by biomimetic mineralization of the NF–BMP2 hybrid in simulated body fluids and subsequent dissolution of NFs in hexafluoroisopropanol. The formation of NF–BMP2 hybrid was identified by confocal laser scanning microscopy analysis. The crystal structure of HA crystals formed on NFs was examined by X-ray diffraction. The chemical composition and interconnected porous structure of the created 3D HA architectures were measured by X-ray photoelectron spectroscopy, focused ion beam scanning electron microscopy, and transmission electron microscopy, respectively. This bottom-up strategy based on electrospinning and biomimetic mineralization opens up a new way to prepare diverse porous HA-based hybrid materials and shows great potential in drug delivery, gene transfer and tissue engineering.
Template-assisted, Sol-gel Fabrication of Biocompatible, Hierarchically Porous Hydroxyapatite Scaffolds

Abstract:Hierarchically porous hydroxyapatite (HHA) scaffolds were synthesized by template-assisted sol-gel chemistry. Polyurethane foam and a block copolymer were used as templates for inducing hierarchically porous structures. The HHA scaffolds exhibited open porous structures with large pores of 400-600 µm and nanoscale pores of ~75 nm. In comparison with conventional hydroxyapatite (CHA), HHA scaffolds exhibited significantly higher surface areas and increased protein adsorption for bovine serum albumin and vitronectin. Both the HHA and CHA scaffolds exhibited well in vitro biocompatibility. After 1 day, Saos-2 osteoblast-like cells bound equally well to both HHA and CHA scaffolds, but after 7 days in culture, cell proliferation was significantly greater on the HHA scaffolds (p < 0.01). High surface area and hierarchical porous structure contributed to the selective enhancement of osteoblast proliferation on the HHA scaffolds.
Preparation of collagen/hydroxyapatite/alendronate hybrid hydrogels as potential scaffolds for bone regeneration.

Abstract:Development of biomimetic scaffolds represents a promising direction in bone tissue engineering. In this study, we designed a two-step process to prepare a type of biomimetic hybrid hydrogels that were composed of collagen, hydroxyapatite (HAP) and alendronate (ALN), an anti-osteoporosis drug. First, water-soluble ALN-conjugated HAP (HAP-ALN) containing 4.0wt.% of ALN was synthesized by treating HAP particles with ALN. Hydrogels were then formed from HAP-ALN conjugate and collagen under physiological conditions using genipin (GNP) as the crosslinker. Depending on the ALN/collagen molar ratio and GNP concentration, the gelation time of hydrogels ranged from 5 to 37min. Notably, these hybrid hydrogels exhibited markedly improved mechanical property (storage modulus G'=38-187kPa), higher gel contents, and lower swelling ratios compared to the hydrogels prepared from collagen alone under similar conditions. Moreover, they showed tunable degradation behaviors against collagenase. The collagen/HAP-ALN hybrid hydrogels supported the adhesion and growth of murine MC3T3-E1 osteoblastic cells well. Such tough yet enzymatically degradable hybrid hydrogels hold potential as scaffolds for bone tissue engineering.
A Highly Interweaved HA-SS-nHAp/collagen Hybrid Fibering Hydrogel Enhances Osteoinductivity and Mineralization

Abstract:The combination of bioactive hydroxyapatite (HAp) with biomimetic bone matrix biomaterials as bone filling scaffolds is a promising strategy for bone regeneration, but the undesirable dispersion of HAp and its interfacial interaction result in inefficient mineralization, mechanical instability, incomplete osteointegration, and even repair failure. Herein, the size dispersion and stabilization of nano-hydroxyapatite (nHAp) in aqueous media were obviously improved by hydrophilic solubilisation and strong negatively charged thiolated hyaluronic acid (HA-SH). Furthermore, the highly interweaved HA-SS-nHAp/collagen hybrid fibering hydrogel exhibited significantly improved mechanical properties and structural stability due to its thickened and densified interweaved fiber network, which ensured the homogeneous dispersion of nHAp in the matrix materials and its integration with the hydrogel network structure completely by covalent self-crosslinking among the sulfhydryl groups derived from the free HA-SH polymer and the mercapto functional groups on the surface of nHAp. Compared with the physically combined micro-hydroxyapatite (μHAp) (d≤25 μm) and nHAp (∼530 nm) with injectable bionic HA-SH and collagen type I biopolymers, HA-SS-nHAp/collagen achieved the maximum efficiency in facilitating rabbit bone marrow stromal cell (rBMSC) adhesion, proliferation and osteogenic differentiation in vitro. The in vivo murine dorsal subcutaneous implantation results further confirmed that the interweaved fiber network structure in HA-SS-nHAp/collagen significantly promoted osteoinductivity and mineralization. This work provides novel insights for the development of new low invasive bone filling biomaterials.

Abstract:Hydrogels have been widely used to mimic the biochemical and mechanical environments of native extracellular matrices for cell culture and tissue engineering. Among them, self-assembling peptide hydrogels are of special interest thanks to their great biocompatibility, designability and convenient preparation procedures. In pioneering studies, self-assembling peptide hydrogels have been used for the culture of bone marrow cells. However, the low mechanical stability of peptide hydrogels seems to be a drawback for these applications, as bone marrow cells prefer hard substrates for osteogenic differentiation. In this work, we explored the use of hydroxyapatite (HAP)-peptide hybrid hydrogels for three-dimensional (3D) culture and differentiation of osteogenic MC3T3-E1 cells. We used HAP nanoparticles as crosslinkers to increase the mechanical stability of peptide hydrogels. Meanwhile, HAP provided unique chemical cues to promote the differentiation of osteoblasts. A phosphate group was introduced to the self-assembling peptide so that the peptide fibers could bind to HAP nanoparticles specifically and strongly. Rheological characterization indicated that the hybrid hydrogels were mechanically more stable than the hydrogels containing only peptides and can be used for long term cell culture. Moreover, the hydrogels were biocompatible and showed very low cytotoxicity. The favorable mechanical properties of the hybrid hydrogels and the chemical properties of HAP synergistically supported the differentiation of MC3T3-E1 cells. Based on these characterizations, we believe that these hybrid hydrogels can potentially be used as scaffolds for cartilage and bone regeneration in the future.

Abstract:Artificial construction of tissue engineering implants with bone-mimicking structure and osteogenic function is of great significance for bone regeneration, but it remains a formidable challenge. Herein, inspired by natural bone, a biomimetically hierarchical hydrogel scaffold with highly ordered arrangement and multi-layered concentric circular structure is constructed by a convenient free-injection method for osteoimmunomodulation and bone defect regeneration. Ultralong hydroxyapatite nanowires act as skeleton unit to assemble into the hierarchically ordered nanowire bundles and further hybridization with hyaluronic acid methacrylate and loading with layered double hydroxide nanosheets to form the stable biomimetic scaffold. The biomimetic scaffold exhibits anisotropic structure, high water content and porosity, mechanical robustness, and sustained release of bioactive ions. In particular, the excellent biocompatibility together with ordered topography and biochemical cue can guide recruitment and directional migration of stem cells, and effectively promote osteogenic differentiation and vascularization in vitro. Moreover, the biomimetic scaffold induces M2 phenotype polarization of macrophages, thus creating a favorable osteoimmune environment. The in vivo study proves that the biomimetic scaffold can attenuate inflammation, promote vascularized ossification, and further accelerate new bone formation. This study provides an innovative strategy to construct multifunctional anisotropic biomaterials for bone tissue engineering.

Abstract:Hydroxyapatite (HA) micro/nano particles show great promise as artificial bone and dental substitutes, or drug carrier systems. However, the precise regulation of hydroxyapatite micro/nano particles with controllable physicochemical properties (such as hierarchical structure, particle size, potential and crystallinity) is still a challenge. Furthermore, the effects of different hierarchical structures on biological responses have been rarely reported. Herein, the HA particles with a precisely tailored micro/nano hierarchical structure have been developed using an elaborate biomimetic synthesis technology. Three representative particles, namely, micro/nano needle-like HA particles, micro/nano rod-like HA particles, and micro/nano flake-like HA particles, were featured to evaluate their biological responses to stem cells. The pore structure facilitated the adsorption of serum adhesive proteins, which together with the unique hierarchical architecture of micro/nano flake-like HA particles remarkably promoted the endocytosis efficiency in a concentration-dependent manner. The qRT-PCR together with RNA-seq and western blot analyses showed that micro/nano flake-like HA particles more significantly up-regulated the expression of genes and production of proteins related to osteogenic differentiation among the three particles through the activated ERK/MAPK signalling pathway. RNA-seq further revealed a complex mechanism of cell interface events, suggesting that the hierarchical architecture of HA particles is of crucial importance for the regulation of actin cytoskeleton involved in the modulation of cell adhesion which positively stimulated osteogenic differentiation of stem cells. Moreover, the endocytosis of particles into lysosomes resulted in an increase in the intracellular Ca2+ levels, which activated possible intracellular Ca2+-mediated signaling cascades (Ras/cAMP/Rap1/MAPK signaling pathways) related to osteogenic differentiation of stem cells. Our findings shed light on the effects of different hierarchical structures of HA particles on stem cell differentiation and contribute to the optimal design of implant materials.
Biomimetic Mechanically Strong One-Dimensional Hydroxyapatite/Poly( d , l -lactide) Composite Inducing Formation of Anisotropic Collagen Matrix

Abstract:Natural bone is a complex composite, consisting predominantly of collagen and hydroxyapatite (HA), which form a highly organized, hierarchical structure from the nano- to the macroscale. Because of its biphasic, anisotropic, ultrafine structural design, bone tissue possesses excellent mechanical properties. Herein, inspired by the composition and microstructure of natural bone, a biphasic composite consisting of highly aligned strontium/copper-doped one-dimensional hydroxyapatite (Sr/Cu-doped 1D HA) and poly(d,l-lactide) (PDLA) was developed. The presence and alignment of Sr/Cu-doped 1D HA crystals resulted in mechanical reinforcement of the polymer matrix, including compressive and tensile strength and modulus, fracture toughness, swelling resistance, and long-term structural stability. The compressive strength, tensile strength, and Young's modulus of the biomimetic composite were comparable to that of cortical bone. Biologically, the biomimetic composite showed a sustained release of the incorporated Sr and Cu ions, facilitated mineral deposition from simulated body fluid, and supported attachment, proliferation, and alkaline phosphatase activity of human mesenchymal stromal cells (hMSCs). Moreover, the highly aligned Sr/Cu-doped 1D HA crystals in the 3D porous scaffolds induced the alignment of hMSCs and secretion of an anisotropic collagen fiber matrix in 3D. The biomimetic Sr/Cu-doped 1D HA/PDLA composite presented here contributes to the current efforts aiming at the design and development of load-bearing bioactive synthetic bone graft substitutes. Moreover, the biomimetic composite may serve as a 3D platform for studying cell-extracellular matrix interactions in bone tissue.

Abstract:Carbonated hydroxyapatite is a main inorganic component of hard tissues in vertebrates, which usually appears as orderly arranged superstructures in tissues and exhibits excellent biological and mechanical properties. Recently, mimicking the structure and composition of organisms in the field of biomaterials and tissue engineering has caused much attention. Herein, a family of orderly arranged textures composed of two-dimensional carbonated hydroxyapatite in nanoscale was prepared in the complex matrix of succinylated gelatin and cetyltrimethyl ammonium bromide, which is an analogue of bone in calcified turkey tendon. The building blocks of carbonated hydroxyapatite nanosheets elongate along the c-axis direction and self-organize into well-aligned bundles, This study may offer an example to design and synthesize laminated materials and may provide new evidence in support of the hypothesis that hierarchical structures in nature result from cooperative interactions between biological membrane and crystal growth.
Hierarchical Self-Assembly of Nano-Fibrils in Mineralized Collagen

Abstract:A designed hierarchical structure was made by self-assembly of nano-fibrils of mineralized collagen resembling extracellular matrix. The collagen fibrils were formed by self-assembly of collagen triple helices. Hydroxyapatite (HA) crystals grew on the surface of these fibrils in such a way that their c-axes were oriented along the longitudinal axes of the fibrils. The mineralized collagen fibrils aligned parallel to each other to form mineralized collagen fibers. For the first time, the new hierarchical self-assembly structure of collagen−hydroxyapatite composite was verified by conventional and high-resolution transmission electron microscopy.
One-step method to construct hydroxyapatite scaffolds with 3-D interconnected structure by a novel hydrogel bead porogen process

Abstract:The different pore sizes of hydroxyapatite (HA) scaffolds with an adjustable network of interconnected pores were constructed by a novel one-step alginate hydrogel bead porogen method. Monodisperse alginate hydrogel beads were manufactured by needle-based droplet method with different sizes of needles and pre-treated with organic solvent exchange to enhance mechanical support and deformability. HA slurry was mixed with alginate hydrogel beads followed by physical squeeze for simultaneous construction of three-dimensional scaffold and interconnected windows. The sizes of interconnected windows of HA scaffolds could be adjusted by changing the compressive force of physical squeeze. Finally, the interconnected HA scaffolds were fabricated after sintering the initial blocks. The advantage of the present approach is that the alginate hydrogel beads could shrink during drying the HA scaffolds and release a small amount of gas during sintering, which avoid cracking of cellular structure.

Abstract:In this research we utilize three polymers, including collagen, diblock PEG-PLGA copolymer and triblock PEG-PLGA-PEG copolymer for biomimetic hydroxyapatite (HAp) preparation. Average crystallite sizes of precipitated HAp from polymer solutions were estimated from XRD patterns to be ca. 23 nm for those in collagen-HAp hybrids and 30 nm for those without polymer addition. Scanning electron micrographs showed that the organic macromolecules induced HAp with regular plate-like shape and nano-sized structure. Thermogravimetric analyses showed 2-3 wt% polymer content in the hybrids. Solid state NMR revealed polymer incorporation in HAp crystal. The hybrids containing these biodegradable polymers can be processed to various morphologies for tissue engineering applications.
Construction of Biomimetic Natural Wood Hierarchical Porous-Structure Bioceramic with Micro/Nanowhisker Coating to Modulate Cellular Behavior and Osteoinductive Activity

Abstract:Scaffolds with a biomimetic hierarchy micro/nanoscale pores play an important role in bone tissue regeneration. In this study, multilevel porous calcium phosphate (CaP) bioceramic orthopedic implants were constructed to mimic the micro/nanostructural hierarchy in natural wood. The biomimetic hierarchical porous scaffolds were fabricated by combining three-dimensional (3D) printing technology and hydrothermal treatment. The first-level macropores (∼100–600 μm) for promoting bone tissue ingrowth were precisely designed using a set of 3D printing parameters. The second-level micro/nanoscale pores (∼100–10,000 nm) in the scaffolds were obtained by hydrothermal treatment to promote nutrient/metabolite transportation. Micro- and nanoscale-sized pores in the scaffolds were recognized as <i>in situ</i> formation of whiskers, where the shape, diameter, and length of whiskers were modulated by adjusting the components of calcium phosphate ceramics and hydrothermal treatment parameters. These biomimetic natural wood-like hierarchical structured scaffolds demonstrated unique physical and biological properties. Hydrophilicity and the protein adsorption rate were characterized in these scaffolds. <i>In vitro</i> studies have identified micro/nanowhisker coating as potent modulators of cellular behavior through the onset of focal adhesion formation. In addition, histological results indicate that biomimetic scaffolds with porous natural wood hierarchical pores exhibited good osteoinductive activity.
