Abstract:The self-assembly of cyclodipeptides composed of natural aromatic amino acids into supramolecular structures of diverse morphologies with intrinsic emissions in the visible light region is demonstrated. The assembly process can be halted at the initial oligomerization by coordination with zinc ions, with the most prominent effect observed for cyclo-dihistidine (cyclo-HH). This process is mediated by attracting and pulling of the metal ions from the solvent into the peptide environment, rather than by direct interaction in the solvent as commonly accepted, thus forming an "environment-switching" doping mechanism. The doping induces a change of cyclo-HH molecular configurations and leads to the formation of pseudo "core/shell" clusters, comprising peptides and zinc ions organized in ordered conformations partially surrounded by relatively amorphous layers, thus significantly enhancing the emissions and allowing the application of the assemblies for ecofriendly color-converted light emitting diodes. These findings shed light into the very initial coordination procedure and elucidate an alternative mechanism of metal ions doping on biomolecules, thus presenting a promising avenue for integration of the bioorganic world and the optoelectronic field.

Abstract:Biosynthesis offers opportunities for cost-effective and sustainable production of semiconductor quantum dots (QDs), but is currently restricted by poor controllability on the synthesis process, resulting from limited knowledge on the assembly mechanisms and the lack of effective control strategies. In this work, we provide molecular-level insights into the formation mechanism of biogenic QDs (Bio-QDs) and its connection with the cellular substrate metabolism in Escherichia coli. Strengthening the substrate metabolism for producing more reducing power was found to stimulate the production of several reduced thiol-containing proteins (including glutaredoxin and thioredoxin) that play key roles in Bio-QDs assembly. This effectively diverted the transformation route of the selenium (Se) and cadmium (Cd) metabolic from Cd3(PO4)2 formation to CdS xSe1- x QDs assembly, yielding fine-sized (2.0 ± 0.4 nm), high-quality Bio-QDs with quantum yield (5.2%) and fluorescence lifetime (99.19 ns) far exceeding the existing counterparts. The underlying mechanisms of Bio-QDs crystallization and development were elucidated by density functional theory calculations and molecular dynamics simulation. The resulting Bio-QDs were successfully used for bioimaging of cancer cells and tumor tissue of mice without extra modification. Our work provides fundamental knowledge on the Bio-QDs assembly mechanisms and proposes an effective, facile regulation strategy, which may inspire advances in controlled synthesis and practical applications of Bio-QDs as well as other bionanomaterials.

Abstract:Quantum confined materials have been extensively studied for photoluminescent applications. Due to intrinsic limitations of low biocompatibility and challenging modulation, the utilization of conventional inorganic quantum confined photoluminescent materials in bio-imaging and bio-machine interface faces critical restrictions. Here, we present aromatic cyclo-dipeptides that dimerize into quantum dots, which serve as building blocks to further self-assemble into quantum confined supramolecular structures with diverse morphologies and photoluminescence properties. Especially, the emission can be tuned from the visible region to the near-infrared region (420 nm to 820 nm) by modulating the self-assembly process. Moreover, no obvious cytotoxic effect is observed for these nanostructures, and their utilization for in vivo imaging and as phosphors for light-emitting diodes is demonstrated. The data reveal that the morphologies and optical properties of the aromatic cyclodipeptide self-assemblies can be tuned, making them potential candidates for supramolecular quantum confined materials providing biocompatible alternatives for broad biomedical and opto-electric applications.

Abstract:The rapid advance and growth of the point-of-care diagnosis industry has provided an impetus for the development of novel signal labels for highly sensitive bio-molecule detections. Colloidal quantum dots (QDs) exhibit superior brightness, facile surface functionalization and exceptional photostability, making them the preferred option for these biological applications. However, the significantly reduced fluorescence intensity and limited photochemical stability in complex biological environments have greatly hampered their further use. Herein, with an innovative alloying engineering strategy, the high-quality water-soluble CdSe/CdxZn1−xS QDs with near-unity PL quantum yield and monoexponential PL decay dynamics are obtained. Notably, for the first time, a record-breaking stability at the single QD level in water with nonblinking behavior persisting for an hour is achieved, which approaching those of state-of-the-art hydrophobic QDs. Furthermore, the nanocomposites formulated with these novel alloyed QDs demonstrate a remarkable PL QY of 96 % and provide an ultrasensitive detection for prostate specific antigen on lateral flow immunoassays. These findings presented here shed new light on the design of high-brightness water-soluble QDs in single-molecule level and QDs-based nanocomposites, significantly pushing ahead toward high-sensitivity biomedical detections and diagnosis.

Abstract:Heavy-metal-free I-III-VI2 quantum dots (QDs) have recently emerged as favorable alternatives to toxic II-VI QDs for optoelectronic and biological applications, but low fluorescence efficiency is a key issue related to the promising nanocrystals. In this work, we demonstrate big enhancement of two-photon fluorescence efficiency and low threshold two-photon pumped amplification of spontaneous emission (ASE) of Zn-processed CuInS2 (CIS) QDs for the first time. The change of two-photon process caused by Zn2+ cation exchange in CIS QDs is systematically investigated. Two-photon ASE in Zn-processed CIS QDs films is achieved with a record of low threshold fluence of 5.7 mu J/cm(2). With high two-photon fluorescence quantum yields and low threshold ASE, the Zn-processed CIS QDs could become a promising candidate for two-photon bioimaging, nonlinear optical and novel QDs laser devices.

Abstract:N-rich metal-free and metal-doped carbon quantum dots (CQDs) have been prepared through one-step hydrothermal method using tetraphenylporphyrin or its transition metal (Pd or Pt) complex as precursor. The structures and morphology of the as-prepared nanoparticles were analyzed by X-ray diffraction, high-resolution transmission electron microscopy, X-ray photoelectron spectroscopy, and Fourier transform infrared spectra. Three kinds of nanocomposites show similar structures except for the presence of metal ions in Pd-CQDs and Pt-CQDs indicated by X-ray photoelectron spectroscopy. All of them display bright blue emission upon exposure to ultraviolet irradiation. The CQDs exhibit typical excitation-dependent emission behavior, with the emission quantum yield of 10.1%, 17.8%, and 15.2% for CQDs, Pd-CQDs, and Pt-CQDs, respectively. Moreover, the CQDs, Pd-CQDs, and Pt-CQDs could serve as fluorescent probes for the specific and sensitive detection of Fe3+ ions in aqueous solution. The low cytotoxicity of CQDs is demonstrated by MTT assay against HeLa cells. Therefore, the CQDs can be used as efficient probes for cellular multicolor imaging and fluorescence sensors for the detection of Fe3+ ions due to their low toxicity, excellent biocompatibility, and low detection limits. This work provides a new route to synthesize highly luminescent N-rich metal-free or metal-doped CQDs for multifunctional applications.

Abstract:A facile method is developed for the preparation of high-quality, water- soluble, and near-infrared (NIR)-emitting CdSeTe alloyed quantum dots (AQdots) with L-cysteine as the capping agent. By changing the size and the composition of AQdots the photoluminescent quantum yield (QY) can reach as high as 53% and the emission color can be tuned between visible and NIR regions (580-814 nm). Furthermore, the prepared NIR-emitting AQdots have been successfully applied for HL-60 cell imaging and glucose and cholesterol assay, which demonstrates the great potential of the AQdots for biological applications.

In our work, a facile method was developed for the preparation of high quality, water-soluble, and NIR-emitting CdSeTe alloyed Qdots (A-Qdots) with L-cysteine (L-cys) as capping agent. By changing the size and the composition of the A-Qdots, the photoluminescent quantum yields (QYs) can reach as high as 53% and the emission color can be tuned between visible and NIR regions. Based on the fluorescence of the A-Qdots selectively quenched in the presence of Cu2+, the NIR-emitting CdSeTe A-Qdots were applied in ultrasensitive Cu2+ sensing. Furthermore, the prepared CdSeTe A-Qdots have been successfully applied for cell imaging, glucose and cholesterol assay, which demonstrates the great potential of the Qdots for biological applications. In order to improve the biocompatibility of the CdSeTe A-Qdots, new water-soluble CdSeTe/ZnS core-shell Qdots (CS-Qdots) with excellent NIR emission were synthesized in aqueous solution. The prepared CS-Qdots not only possessed high QYs but also exhibited excellent photobstability and favorable biocompatibility. Moreover, the CS-Qdots showed high electrogenerated chemiluminescence (ECL) signal. These characteristics showed their potential applications in cell imaging and biosensing with high sensitivity.

Abstract:QDs that emit in the infrared (IR) range are of special interest at the moment because of their potential as tissue imaging reagents. Due to autofluorescence from tissues, QDs that emit in the visible range fail to produce good signal to noise ratios. Here we report the production of CdxPb1-xTe tertiary-alloyed QDs that emit in the 1100-1300 nm wavelength range, capped with the hydrophilic ligands mercaptopropionic acid (MPA) or glutathione (GSH), together with DNA, as specific surface tags. We observed an interesting dependence of the QD emission peaks on the species of capping ligand used. ICP-MS analysis confirmed that changing the identity of the surface ligand in the reaction mixture shifted the elemental composition of the particles and resulted in different Cd/Pb ratios. Further, DNA directed assembly of the particles onto DNA nanostructures ensures that the particle remains stable in high salt conditions, which is crucial to biological applications.

Abstract:Quantum dots (QDs), owning to their unique optical and electronic properties, are increasingly used for bio-imaging/biosensing in medical and energy fields. Biosynthesis of QDs offers several advantages over the conventional chemical synthesis route, such as low cost, environmental-benignity, and no need for further modification of the products. However, information about the regulation and mechanism of synthesis of QDs with great fluorescence properties is still limited. In this work, we prepared cadmium selenium (CdSe) QDs in vivo using Candida utilis WSH02-08. We regulated the fluorescence properties of the bio-QDs through different precursor concentrations, and found that the bio-QDs with high fluorescence intensity and long photostable lifetime were favoured at an elevated Cd content over Se. The synthesis of the high-quality bio-QDs was attributed to the less formation of Se(0) and the increased formation of CdSe and CdS. Furthermore, such synthesized QDs were directly used in live-cell imaging without further modification. These findings are expected to enable a further design of biosynthesized QDs with great fluorescence properties and expand our understanding of the regulation mechanism of nanoparticles fabrication in microorganisms. Abstract:A novel luminescent nanocomposite assembly was prepared in aqueous solutions, via the electrostatic interaction between poly(ethylene glycol)-b-poly(N,N-dimethylaminoethyl methacrylate) (PEG-b-PDMAEMA) double hydrophilic block copolymer (DHBC) and thioglycolic acid (TGA)-stabilized CdTe quantum dots (QDs). By tuning the molar ratios of QDs to DHBC, the nanocomposite assemblies could be well controlled and the dimension of these regular spherical QDs/PEG-b-PDMAEMA assemblies ranged from 40 to 75 nm in aqueous media. As the periphery of QDs/PEG-b-PDMAEMA nanocomposite assembly was composed of PEG segments when the PDMAEMA blocks and QDs were immobilized inside the assembly, the cytotoxicity of the assembly was significantly reduced, compared with that of pure PEG-b-PDMAEMA and QDs. The nanocomposite assembly also exhibited superior stability in salt solutions and remained strongly photoluminescent even when free radicals existed. Thus, the QDs/PEG-b-PDMAEMA nanocomposite assembly is potentially useful for a number of applications in biolabeling and imaging.

Abstract:The hollow spherical CdSe QD assemblies were synthesized via a sonochemical approach that utilizes β-cyclodextrin as a template reagent in aqueous solution. The hollow nanospheres have an average diameter of 70nm and are found to consist of an assembly of monodispersed 5nm sized CdSe quantum dots. Following an electrochemical reaction with persulfate ions, strong electrogenerated chemiluminescence (ECL) was observed from the CdSe nanoassemblies suspended in an aqueous solution of pH⩽7.95. The study indicates that the morphology of the 70nm nanoassembly plays an important role in generating the stable ECL since individually dispersed quantum dots did not exhibit any significant ECL. The unique ECL intensity and stability of the synthesized spherical nanoassemblies could allow for potential sensor applications of CdSe quantum dots in water.
pH-dependent network formation of quantum dots and fluorescent quenching by Au nanoparticle embedding

Abstract:A simple approach to the creation of colloidal assemblies is in high demand for the development of functional devices. Here, we present the preparation of CdTe-QD (quantum dot) networks in as little as 1 day simply by pH modification without the use of oxidants. The QD network was tractable in water and casting from a droplet produced a porous networked film on both hydrophobic and hydrophilic solid substrates. Further, we found that citrate-protected gold nanoparticles (AuNPs, d = 5 nm) could be incorporated into the QD networks to afford a QD/Au composite network, and that the fluorescence from the QDs was largely decreased by the addition of a small proportion of AuNPs (QD:AuNP = 99.4:0.6), probably due to the efficient charge transfer through the network. These data indicate that our method is suitable for application to the creation of metal/QD hybrid materials that can be integrated into wet-based processes.

Abstract:Eliminating colloidal toxicity and enabling its intrinsic fluorescence in aggressive environmental conditions are the key challenges for commercializing hydrophobic cadmium based quantum dots (QD). Polyarylene ether nitriles (PEN) are an example of super-engineering thermoplastics that possess a unique combination of thermal stability, intrinsic fluorescence, biocompatibility and distinct emulsion self-assembly feature. Herein, the co-self-assembly of amphiphilic PEN with hydrophobic CdSe@ZnS QD, confined in the three dimensional (3D) oil-in-water emulsion droplets, has been explored to fabricate fluorescent microparticles (FMP). It was found that these FMP demonstrated good biocompatibility (cell viability above 90%), while exhibiting a fluorescence emission in aqueous solution that was retained (intensity retention ratio above 80%) within the whole pH range of 1–14, as well as, after being subjected to autoclaving at 120 °C for 1 h. Interestingly, it was discovered that introduction of calcium ions in the emulsion self-assembly contributed to in-situ generation of phase changing nanoplates inside the FMP, which led to the photo-thermal modulated solid state fluorescence from drop-casted FMP film. Thanks to their versatile fluorescence, these FMP colloids were exploited as fluorescent probes for macrophages imaging, while micro-patterns with reversible changing of emission color were induced via thermal treatment and direct laser lithography.

Abstract:Organisms served as factories of bio-assembly of nanoparticles attracted a lot of attentions due to the safe, economic and environmental-benignity traits, especially the fabrication of the super fluorescence properties quantum dots (QDs). However, information about the developmental dynamics of QDs in living organisms is still lacking. In this work, we synthesized cadmium-selenium (CdSe) QDs in Candida utilis WSH02-08, and then tracked and quantitatively characterized the developmental dynamics (photoactivation, photostable and photobleaching processes) of bio-QDs by translating fluorescence microscopy movies into visual quantitative curve. These findings shed light on the fluorescence properties of the bio-assembled QDs and are expected to accelerate the applications of the synthesized QDs in vivo. It provided a new way to screen bio-QDs and monitor the quality of QDs in vivo.

Abstract:In comparison with conventional organic dyes, quantum dots (QDs) have unique optical and electronic properties, which provide QDs with a wide scope of prospective application in biology and biomedicine. However, the toxicity of QDs and the fluorescence intensity of labeled bacteria must precede their application in bacterial imaging and tracing in vivo. Here, we show that treatment with CaCl 2 significantly improved bacterial labeling efficiency of CdSe/ZnS QDs with the CdSe core size of ~3.1 nm (relative fluorescence unit (RFU) value and ratio of fluorescent E. coli ) with rising CdSe/ZnS QDs concentration in a concentration-dependent manner. At 12.5 nmol/L CdSe/ZnS QDs concentration, labeled Escherichia coli ( E. coli ) DH5 α appeared as short rod-shaped and luminescent with normal size, and the survival rate and ultrastructure did not change in comparison to the control. But the ratio of fluorescent bacteria and RFU were very low. However, the survival rate of transformed E. coli was significantly inhibited by high CdSe/ZnS QDs concentrations (≥25 nmol/L). Moreover, internalization of CdSe/ZnS QDs resulted in ultrastructure damage of transformed E. coli in a concentration-dependent manner (≥25 nmol/L). Therefore, CdSe/ZnS QDs may not suitable for tracing of bacteria in vivo. Moreover, our study also revealed that colony-forming capability assay and transmission electron microscopy could be used to comprehensively evaluate the toxicity of QDs on labeled bacteria. Our findings do provide a new direction toward the improvement and modification of QDs for use in imaging and tracing studies in vivo.
Aqueous Synthesis and Fluorescence-Imaging Application of Cdte/Znse Core/Shell Quantum Dots with High Stability and Low Cytotoxicity

Abstract:CdTe/ZnSe core/shell quantum dots were directly synthesized in an aqueous condition by heating a mixed solution of ZnCI2, NaHSe and CdTe QDs in the presence of mercaptosuccinic acid as a stabilizer. By controlling the size and composition, the CdTe/ZnSe QDs with emission wavelength ranging from 540 to 630 nm, high quantum yield (44%) and narrow full width at half maximum (FWHM) could be obtained. Characterization results with HRTEM, XRD and EDX have shown that the synthesized CdTe/ZnSe QDs have good monodispersity and a nice crystal structure, and exhibited better stability and less cytotoxicity as compared with CdTe QDs. Furthermore, luminescent QD-IgG bioprobes were produced to detect the breast cancer marker Her2 on the surface of fixed MCF-7 cancer cells for their optical imaging.
