Abstract:We herein report the functionalization of alpha-C-H in alcohols through cross-dehydrogenative coupling reactions. Selectfluor was used as a mild oxidant. In situ-generated HF participated in the reaction and no external strong acid was necessary. A variety of heteroaryl-substituted alcohols were achieved with good yields and with good functional group tolerance.

Abstract:The daunting challenges in converting alcoholic O-H bonds with high bond-dissociation energy (BDE) to alkoxy radicals and harnessing those unruly reaction species largely limit exploiting free alcohols in C(sp3)-H functionalization. Herein we describe a novel radical alkynylation and allylation of unactivated C(sp3)-H bonds with unmodified aliphatic alcohols. The use of phenyliodine bis(trifluoroacetate) (PIFA) enables the formation of alkoxy radicals under mild photochemical conditions. α-Methyl styrene serves as a sacrificial-reagent that significantly improves the reaction outcomes. This transition-metal free protocol further features broad substrate scope, exclusive site-selectivity, high product diversity, and simple operation, supplying a robust manifold for C(sp3)-H functionalization using easily available aliphatic alcohols as feedstock.

Abstract:Oxygen‐containing heterocycles are an important class of compounds with diverse biological activities. In recent years, direct α‐C–H functionalization of inexpensive and abundant readily available ethers and alcohols by oxidative radical cross‐coupling with different nucleophilic partners (C–H, N–H, O–H and S–S), leading to the construction of C–C and C–X (X=C, N, O, S) bonds, has emerged as one of the vital strategies among oxidative cross‐coupling reactions. Owing to the features of being atom‐economic, environmentally benign, having a simple operation and biologically properties, a series of ether α‐C(sp3)–H bond activation reactions have been developed by metal or metal‐free systems via the radical oxidative coupling pathway, since the radical oxidative coupling reactions have wide range of applications. This review aims to highlight the recent advances in direct α‐C(sp3)–H bond functionalization of ethers and alcohols via radical oxidative process.

Abstract:Aliphatic alcohols and amides are highly valued, ubiquitous chemicals in synthetic chemistry. Radical-promoted regioselective functionalization of unactivated C(sp3)-H bonds offers an atom and step economic manner for direct transformation of aliphatic alcohols and amides, especially structural elaboration of complex natural products and bioactive molecules without de novo synthesis. Despite the rapid growth of hydrogen atom transfer (HAT) processes mediated by O- or N-centered radicals generated from prefunctionalized precursors of alcohols and amides in the past few years, exploration of remote C(sp(3))-H functionalization via HAT mediated by unprotected alcohols or amides lags behind, due to the difficult homolysis of O-H and N-H bonds with high bond-dissociation energies (BDEs). In this minireview, the recent advances in regioselective C(sp(3))-H functionalization mediated by unprotected alcohols and amides are summarized.

Abstract:Propargylic alcohols are regarded as a class of readily available and versatile synthons in organic synthesis. Due to their unique bifunctional character, the transformation of propargylic alcohols has been widely utilized as a powerful strategy for the construction of various functionalized frameworks over the past few decades. Recently, the radical transformation of propargylic alcohols has attracted considerable attention in synthetic chemistry. This review summarises recent progress in the radical transformation of propargylic alcohols. Based on the different mechanisms of radical formation underlying these transformations, this review has been divided into four parts: (1) transition‐metal catalyzed radical transformation; (2) photo‐induced radical transformation; (3) electroc‐induced radical transformation; and (4) metal‐free or oxidant‐mediated radical transformation of propargylic alcohols.

Abstract:The C-O bond cleavage for activation of alcohols is synthetically useful and practically challenging. This work describes carbazate as a new type of electrochemically activated alkylating agent derived from ubiquitous alcohols for direct functionalization of heteroarenes under mild electrolytic conditions. The simple undivided cell at low oxidative potentials with carbon/platinum electrode set-ups offers excellent substrate tolerance, affording a variety of primary, secondary and tertiary alkyl-decorated heterocycles in good chemical yields. Furthermore, the mechanism for this electrochemical deoxyalkylation reaction has been investigated.

Abstract:Construction of C-C bonds via alkoxy radical-mediated remote C(sp(3))-H functionalization is largely unexplored, as it is a formidable challenge to directly generate alkoxy radicals from alcohols due to the high bond dissociation energy (BDE) of O-H bonds. Disclosed herein is a practical and elusive metal-free alcohol-directed heteroarylation of remote unactivated C(sp(3))-H bonds. Phenyliodine bis(trifluoroacetate) (PIFA) is used as the only reagent to enable the coupling of alcohols and heteroaryls. Alkoxy radicals are readily generated from free alcohols under the irradiation of visible light, which trigger the regioselective hydrogenatom transfer (HAT). A wide range of functional groups are compatible with the mild reaction conditions. Two unactivated C-H bonds are cleaved and one new C-C bond is constructed during the reaction. This protocol provides an efficient strategy for the late-stage functionalization of alcohols and heteroaryls.

Abstract:Radical difunctionalization of aliphatic alkynes provides direct access to valuable multi-substituted alkenes, but achieving a high level of chemo- and stereo-control remains a formidable challenge. Herein we report a novel photoredox neutral alkyne difunctionalization through functional group migration followed by radical-polar crossover and energy transfer-enabled stereoconvergent isomerization of alkenes. In this sequence, a hydroxyalkyl and an aryl group are incorporated concomitantly into an alkyne, leading to diversely functionalized E-allyl alcohols. The scope of alkynes is noteworthy, and the reaction tolerates aliphatic alkynes containing hydrogen donating C-H bonds which are prone to intramolecular hydrogen atom transfer. The protocol features broad functional group compatibility, high product diversity and exclusive chemo- and stereoselectivity, thus providing a practical strategy for the elusive radical difunctionalization of unactivated alkynes.

Abstract:The C–H and C–C bonds are abundant in organic compounds, yet generally inert in chemical transformations. Therefore, direct functionalization of inert chemical bonds remains challenging. The fluorine-containing compounds are of special interest for their uses in medicinal chemistry. Direct fluorination of C–H and C–C bonds undoubtedly represents one of the most ideal and attractive approaches to incorporate fluorine atom into complex molecules. Herein, we summarize the recent advances in radical-mediated C–H and C–C bond fluorination. Three types of transformations are discussed: (1) direct C–H abstraction/fluorination of alkanes; (2) decarboxylative fluorination of alkyl carboxylic acids; (3) ring-opening fluorination.

Among the various transformations, the radical‐involved alkylation reaction continues to blossom since the alkyl unit is ubiquitous in biological and pharmaceutical compounds. The utilization of aliphatic aldehydes as alkyl radical precursors via peroxides or oxygen‐initiated decarbonylation reactions has attracted great attention over the past decade due to their ready availability and the simple operation. Abstract:The construction of C(sp(3))-heteroatom bonds represents a fundamental research field in synthetic chemistry. We report herein an electrochemical dehydroxymethylative functionalization reaction of aliphatic alcohols under mild and catalyst-free conditions, representing a new paradigm for forging various C(sp(3))-heteroatom bonds between diverse alkanols and N- or O-centered nucleophiles. In addition, detailed mechanistic investigations have been carried out, revealing that these reactions proceed through a beta-scission/anodic oxidation/nucleophilic addition sequence.

Abstract:The polar nature of the C═O bond commonly allows it to undergo direct attack by nucleophiles at the electrophilic carbon atom in which ketones and aldehydes act as alkyl carbocation equivalents. In contrast, transformations in which ketones and aldehydes act as alkyl radical equivalents (generated in carbonyl carbon) are unknown. Here, we describe a new catalytic activation mode that combines proton-coupled electron transfer (PCET) with spin-center shift (SCS) and enables C─H alkylation of heteroarenes using ketones and aldehydes as alkyl radical equivalents. This transformation proceeded via reductive PCET activation of the ketones and aldehydes to form α-oxy radicals, addition of the radicals to the N -heteroarenes to form C─C bonds, and SCS to cleave the C─O bonds of the resulting alcohols. This mild protocol represents a general use of abundant, commercially available, ketones and aldehydes as latent alkyl radical equivalents.
Recent Advances in Ring-Opening Functionalization of Cycloalkanols by C-C σ-Bond Cleavage

Abstract:Cycloalkanols prove to be privileged precursors for the synthesis of distally substituted alkyl ketones and polycyclic aromatic hydrocarbons (PAHs) by virtue of cleavage of their cyclic C-C bonds. Direct functionalization of cyclobutanols to build up other chemical bonds (e. g., C-F, C-Cl, C-Br, C-N, C-S, C-Se, C-C, etc.) has been achieved by using the ring-opening strategy. Mechanistically, the C-C cleavage of cyclobutanols can be involved in two pathways: (a) transition-metal catalyzed β-carbon elimination; (b) radical-mediated 'radical clock'-type ring opening. The recent advances of our group for the ring-opening functionalization of tertiary cycloalkanols are described in this account.

Abstract:Aliphatic alcohols are common and bulk chemicals in organic synthesis. The site-selective functionalization of non-activated aliphatic alcohols is attractive but challenging. Herein, we report a silver-catalyzed δ-selective Csp3-H bond functionalization of abundant and inexpensive aliphatic alcohols. Valuable oximonitrile substituted alcohols are easily obtained by using well-designed sulphonyl reagents under simple and mild conditions. This protocol realizes the challenging δ-selective C-C bond formation of simple alkanols.

Abstract:Phenols are important readily available synthetic building blocks and starting materials for organic synthetic transformations, which are widely found in agrochemicals, pharmaceuticals, and functional materials. The C–H functionalization of free phenols has proven to be an extremely useful tool in organic synthesis, which provides efficient increases in phenol molecular complexity. Therefore, approaches to functionalizing existing C–H bonds of free phenols have continuously attracted the attention of organic chemists. In this review, we summarize the current knowledge and recent advances in ortho-, meta-, and para-selective C–H functionalization of free phenols in the last five years.
