Abstract:Radical difunctionalization of aliphatic alkynes provides direct access to valuable multi-substituted alkenes, but achieving a high level of chemo- and stereo-control remains a formidable challenge. Herein we report a novel photoredox neutral alkyne difunctionalization through functional group migration followed by radical-polar crossover and energy transfer-enabled stereoconvergent isomerization of alkenes. In this sequence, a hydroxyalkyl and an aryl group are incorporated concomitantly into an alkyne, leading to diversely functionalized E-allyl alcohols. The scope of alkynes is noteworthy, and the reaction tolerates aliphatic alkynes containing hydrogen donating C-H bonds which are prone to intramolecular hydrogen atom transfer. The protocol features broad functional group compatibility, high product diversity and exclusive chemo- and stereoselectivity, thus providing a practical strategy for the elusive radical difunctionalization of unactivated alkynes.
What problem does this paper attempt to address?
The main problem that this paper attempts to solve is to achieve the selective radical difunctionalization of unactivated alkynes, especially the preparation of functional E - allylic alcohols. Due to their inherent reactivity and the difficulty in selectivity control, unactivated alkynes have always been challenging in radical - mediated difunctionalization reactions. Specifically, the paper focuses on the following aspects:
1. **Achievement of high chemical and stereoselectivity**: Through photoredox - neutral alkyne difunctionalization, functional group migration, radical - polarity crossover and energy - transfer - promoted olefin stereoconvergent isomerization are achieved, thus obtaining products with high chemical and stereoselectivity.
2. **Wide substrate applicability**: The reactivity of a variety of unactivated aliphatic alkynes has been studied, including substrates containing C - H bonds that are prone to intramolecular hydrogen atom transfer, and these substrates usually lead to undesired side reactions.
3. **Multifunctional group compatibility**: The wide compatibility of this method with a variety of functional groups has been demonstrated, including unprotected alcohols, iodides, azides, carboxylic acids and silanes, etc., and these functional groups remain stable during the reaction process.
4. **High product diversity**: By changing the reaction conditions and using different sulfonyl reagents, a variety of diverse E - allylic alcohol products can be synthesized, including cyclic compounds and labeled compounds.
5. **Modification of complex structures**: This method can also be used for the structural modification of natural products and drug derivatives, showing good stereocontrol ability.
In general, this paper proposes a novel photocatalytic strategy. Through sequential functional group migration, radical - polarity crossover and stereoconvergent olefin isomerization, the selective radical difunctionalization of unactivated alkynes is achieved, providing new ideas and methods for research in this field.
