Abstract:The interactions between nanoparticles and biomembrane have important influences on nanoparticle applications in cell imaging,biosensor design,drug delivery,disease diagnoses and therapy.Here,the morphological deformation of the biomembranes and the change of the free energy of the system during the translocation of a rodlike nanoparticle across the biomembranes under the conditions of different interactions is investigated with the self-consistent field theory.The results show that,during the translocation of nanoparticles across the biomembrane,the morphology of the biomembranes is deformed differently with varying the interactions between the nanoparticles and the biomembranes;furthermore,through analyzing the change of the free energy of the system,it is found that the interactions between nanoparticles and biomembranes can obviously influence the translocation of the nanoparticles across the biomembranes.These results may yield some theoretical insights into the relevant biological applications of nanoparticles.
The dual effect of natural organic matter on the two-step internalization process of Au@Sio2 in freshwater

Abstract:Dissolved organic matter (DOM) in aquatic ecosystems reshapes the surface of nanoparticles (NPs) greatly. Understanding how these changes influence the bioavailability of NPs is critical for accurately predicting the ecological risks of NPs. A quantitative model based on the two-step internalization process enabled the differentiation between the adhesion ability of NPs to membranes and the internalization capacity. Using protozoa Tetrahymena thermophila as the test organism, fluorescein isothiocyanate-modified silica NPs (FITC-SiO2) and silica-coated gold NPs (2) were prepared to validate the model and study the influence of DOM on uptake. DOM reduced the ability of 2 to adhere onto cell membranes and the inhibitory effect of bovine serum albumin (BSA) and β-lactoglobulin was higher due to their higher molecular weights and the weaker interaction. Moreover, DOM increased the internalization capacity. 80% 2 was internalized in the presence of humic acid (HA), over 90% 2 was internalized in the presence of the two proteins, whereas only 60% were internalized by the control group. Next, the specific recognition of the cell internalization in the presence of DOM was confirmed. We concluded that the traditional "accumulation" may misestimate the true biological effect caused by NPs coated with DOM. NPs coated with highly bioavailable DOM pose a greater risk to aquatic ecosystems because they are more likely to be internalized by living organisms.

Abstract:The environmental behavior and toxicity of nanomaterials can be significantly affected by the ubiquitous natural organic matter (NOM) of aquatic environments. However, general mechanisms governing the effects of different organic matter samples on nanomaterials remain elusive at present. Thus, the environmental behavior and toxicity of organic matter with different components or conformation, such as humic acid (HA), bovine serum albumin (BSA) and denatured BSA (DB), on five nanomaterials (i.e., Ag nanoparticles (NPs), polystyrene nanoparticles (PS NPs), TiO2 NPs, nano-zerovalent iron (NZVI), and multiwall carbon nanotubes (MWCNTs)) to Tetrahymena thermophila (T. thermophila) were studied. BSA and DB were found to significantly increase (up to 99%) the bioaccumulation of nanomaterials (Ag NPs and PS NPs) in T. thermophila by specific recognition. This increase was lower in DB-coated nanomaterials, compared to BSA-coated nanomaterials, revealing the importance of protein conformation in the specific recognition process. The toxicity results revealed that different organic matter samples mostly affected Ag NPs and NZVI (toxicity by dissolved metal ions) by altering their biological uptake. In the case of nanomaterials whose toxicity was dominated by suspended particles, the organic matter could alter the toxicity of these materials by changing their surface properties and their state in environmental media, and by scavenging free radicals, instead of altering the bioaccumulation process. In general, the toxicity of HA-coated nanomaterials was significantly lower than those of DB- or BSA-coated nanomaterials. Our results suggest that it is essential to consider the type and characteristics of the organic matter when evaluating the toxicity of NPs, since this represents a more realistic state of exposure.

Abstract:With the rapid development of biotechnology and nanomedicine, extensive research has focused on the investigations of delivering large-cargo molecules using nanoparticles through the cell membrane for disease diagnosis and treatment. Various inorganic and polymeric nanoparticles with optimized surface properties have been developed to carry these active cargo molecules such as organic molecules, oligonucleotides and proteins. Phagocytosis and pinocytosis have been suggested as the two major uptake mechanisms for nanoparticles to enter into cellular interior, but such mechanisms are still under debate. In order to enhance the efficiency of cellular uptake of nanoparticles and further understand the physiological process, it is important to investigate detailed interaction mechanisms between nanoparticles and cell membranes. Here, we will review the recent advances of the effect of nanoparticle properties (e.g., nanoparticle shape, size, charge, surface modification, etc.) on cellular uptake mechanisms. These will aid in the future design and development of nanoparticles with improved surface properties for drug and biomolecule delivery. Up to now, novel analytical techniques have been used to examine nanoparticle-cell membrane interactions, but their detailed uptake mechanisms and pathways still need more in-depth research. It is suggested that developing appropriate analytical techniques to study cellular uptake mechanisms of nanoparticles in real time is urgently desired.
Organic matter excreted by the protozoan Tetrahymena thermophila and its effects on the bioaccumulation of nanoparticles

Abstract:Although organic matter (exudate) excreted by aquatic organisms is an important component of dissolved organic matter (DOM) in the natural environment, its potential effects on the bioaccumulation of nanoparticles (NPs) remain unclear. In the present study, we examined the effects of the exudates from the protozoan Tetrahymena thermophila on the bioaccumulation (including uptake and cell surface adsorption) of iron oxide (Fe2O3, polyacrylate coated) and silica (SiO2) NPs in T. thermophila. The exudates were mostly (93.6 %, in carbon) composed of < 1-kDa molecules (e.g., lipids). When the exudates were mixed with the NPs, significant adsorption occurred on SiO2 NPs but not on Fe2O3 NPs. Independent of their adsorption by the NPs, the exudates significantly inhibited the bioaccumulation of both SiO2 NPs and Fe2O3 NPs by T. thermophila. This inhibitory effect was shown to be mainly due to their inhibition of NP adsorption on the cell surface. By contrast, the exudates had negligible effects on the uptake of either NP type, most likely due to their low molecular weight. Since DOM in the aquatic environment is dominated by molecules < 1 kDa, the potential effects of low-molecular-weight DOM, such as exudates from aquatic organisms, on the bioaccumulation of NPs merits greater attention. ENVIRONMENTAL IMPLICATION: Nanoparticles (NPs) are hazardous materials widespread in the natural environment. Previous studies showed that dissolved organic matter (DOM) in aquatic environments determine the environmental behavior and ecological effects of NPs. Although organic matter (exudate) excreted by aquatic organisms is an important component of DOM, its potential effects on the bioaccumulation of NPs remain unclear. In the present study, we found that the exudates inhibited the cell-surface adsorption of NPs but had no effects on NP uptake, as different from the well-known effects of DOM on NP bioaccumulation. This finding merits attention during evaluations of the environmental risks of NPs.

Abstract:AbstractThe interactions of nanoparticles with the soft surfaces of biological systems like cells play key roles in executing their biomedical functions and in toxicity. The discovery or design of new biomedical functions, or the prediction of the toxicological consequences of nanoparticles in vivo, first require knowledge of the interplay processes of the nanoparticles with the target cells. This article focusses on the cellular uptake, location and translocation, and any biological consequences, such as cytotoxicity, of the most widely studied and used nanoparticles, such as carbon‐based nanoparticles, metallic nanoparticles, and quantum dots. The relevance of the size and shape, composition, charge, and surface chemistry of the nanoparticles in cells is considered. The intracellular uptake pathways of the nanoparticles and the cellular responses, with potential signaling pathways activated by nanoparticle interactions, are also discussed.

Abstract:The merits of nanomedicines are significantly impacted by the surrounding biological environment. Similar to the protein corona generated on the surface of nanoparticles in the circulation system, the intracellular protein corona (IPC) might be formed on nanoparticles when transported inside the cells. However, little is known currently about the formation of IPC and its possible biological influence.Methods: Caco-2 cells, a classical epithelial cell line, were cultured in Transwell plates to form a monolayer. Gold nanoparticles (AuNPs) were prepared as the model nanomedicine due to their excellent stability. Here we focused on identifying IPC formed on the surface of AuNPs during cell transport. The nanoparticles in the basolateral side of the Caco-2 monolayer were collected and analyzed by multiple techniques to verify IPC formation. High-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based proteomics was utilized to analyze the composition of IPC proteins. In particular, we established a dual-filtration strategy to exclude various interference in IPC identification. Based on the subcellular localization of specific IPC proteins, we elicited the nano-trafficking network of AuNPs. The transport pathways of AuNPs identified by proteomic analysis were also verified by various conventional technologies. Finally, we explored the influence of IPC on the uptake and stress response of endothelium.Results: The existence of IPC was demonstrated on the surface of AuNPs, in which 227 proteins were identified. Among them, 40 proteins were finally ascertained as the specific IPC proteins. The subcellular location analysis indicated that these "specific" IPC proteins could back-track the transport pathways of nanoparticles in the epithelial cell monolayer. According to the subcellular distribution of IPC proteins and co-localization, we discovered a new pathway of nanoparticles from endosomes to secretory vesicles which was dominant during the transcytosis. After employing conventional imageology and pharmacology strategies to verify the result of proteomic analysis, we mapped a comprehensive intracellular transport network. Our study also revealed the merits of IPC analysis, which could readily elucidate the molecular mechanisms of transcytosis. Besides, the IPC proteins increased the uptake and stress response of endothelium, which was likely mediated by extracellular matrix and mitochondrion-related IPC proteins.Conclusion: The comprehensive proteomic analysis of IPC enabled tracing of transport pathways in epithelial cells as well as revealing the biological impact of nanoparticles on endothelium.

Meanwhile, the biological effect and safety of nanoparticles have attracted worldwide attention. To ensure the healthy and sustainable development of nanotechnology,, the interaction of nanoparticles with organisms and the biological effect produced by nanoparticles can not be neglected. To understand the biological effects of nanoparticles, investigating how the nanoparticles entrance into organisms and the complicated molecular aspects of nano-bio interactions is crucial. The main routes of nanoparticles entering the organisms are introduced. The interaction of nanoparticles with protein, the influencing factors and characterization of the interaction are reviewed in detail. The effects of nano-protein interaction on the structure and function of protein and the biological impact of nanoparticles are summarized.
TiO2 Nanoparticles Act As a Carrier of Cd Bioaccumulation in the Ciliate Tetrahymena Thermophila.

Abstract:When nanoparticles can enter a unicellular organism directly, how may they affect the bioaccumulation and toxicity of other pollutants already present in the environment? To answer this question, we conducted experiments with a protozoan Tetrahymena thermophila. The well-dispersed polyacrylate-coated TiO2 nanoparticles (PAA-TiO2-NPs) were used as a representative nanomaterial, and Cd as a conventional pollutant. We found that PAA-TiO2-NPs could get into Tetrahymena cells directly. Such internalization was first induced by low concentrations of Cd, but later suppressed when Cd concentrations were higher than 1 μg/L. Considering its significant adsorption on PAA-TiO2-NPs, Cd could be taken up by T. thermophila in the form of free ion or metal-nanoparticle complexes. The latter route accounted for 46.3% of Cd internalization. During the 5 h depuration period, 4.34-22.1% of Cd was excreted out, which was independent of the concentrations of intracellular Cd and PAA-TiO2-NPs. On the other hand, both free and intracellular Cd concentrations only partly predicted its toxicity at different levels of PAA-TiO2-NPs. This may have resulted from PAA-TiO2-NPs' synergistic effects and the distinct subcellular distribution of Cd taken up via the two routes above. Overall, we should pay attention to the carrier effects of nanoparticles when assessing their environmental risks.
Effect of Agricultural Organic Inputs on Nanoplastics Transport in Saturated Goethite-Coated Porous Media: Particle Size Selectivity and Role of Dissolved Organic Matter

Abstract:The transport of nanoplastics (NPs) through porous media is influenced by dissolved organic matter (DOM) released from agricultural organic inputs. Here, cotransport of NPs with three types of DOM (biocharDOM (BCDOM), wheat strawDOM (WSDOM), and swine manureDOM (SMDOM)) was investigated in saturated goethite (GT)-coated sand columns. The results showed that codeposition of 50 nm NPs (50NPs) with DOM occurred due to the formation of a GT-DOM-50NPs complex, while DOM loaded on GT-coated sand and 400 nm NPs (400NPs) aided 400NPs transport due to electrostatic repulsion. According to the quantum chemical calculation, humic acid and cellulose played a significant role in 50NPs retardation. Owing to its high concentration, moderate humification index (HIX), and cellulose content, SMDOM exhibited the highest retardation of 50NPs transport and promoting effect on 400NPs transport. Owing to a high HIX, the effect of BCDOM on the mobility of 400NPs was higher than that of WSDOM. However, high cellulose content in WSDOM caused it to exhibit a 50NPs retardation ability that was similar to that of BCDOM. Our results highlight the particle size selectivity and significant influence of DOM type on the transport of NPs and elucidate their quantum and colloidal chemical-interface mechanisms in a typical agricultural environment.
Cellular Internalization of Polypeptide-Based Nanoparticles: Effects of Size, Shape and Surface Morphology

Abstract:Nanoparticles (NPs) can be taken up by cells; however, the effects of the structural characteristics of NPs on their cellular internalization have not been well explored. In this work, cellular internalization performances of various NPs including rods with helical surface (helical rods), spheres with stripe-pattern surface (striped spheres), and spheres with smooth surface (smooth spheres) were investigated by a combination of experiments and theoretical simulations. This study focuses on the effects of the size, shape, and surface morphology on their cellular internalization behaviors. These NPs were self-assembled from mixtures of fluorescein isothiocyanate (FITC)-labelled poly(γ-benzyl-l-glutamate)-block-poly(ethylene glycol) (PBLG(FITC)-b-PEG) block copolymers and PBLG or polystyrene (PS) homopolymers. It was found that the NPs possessing smaller size, rod-like shape, and helical/striped surface morphology exhibit higher cellular internalization efficiency. Such differences in the internalization efficiency for the NPs can be attributed to the differences in both their surface areas and internalization pathways. This study could not only guide the design of nanocarriers with enhanced cellular internalization efficiency, but also deepen our understanding of the internalization behavior of natural NPs with similar structures (e.g., virus).

Abstract:The interactions of nanomaterials such as carbon nanotubes, nanoTiO_2 , nanoFe_2O_3 and nanoFe_3O_4. with biomolecules including amino acids, cells and biological bodies have been studied by the methods of molecular and cellular biologies, respectively. The results obtained show that these nanomaterials can all influence the prolification of the cell and structures or conformations of the biomolecules, while their toxicity is lower. However, the influence of some nanocorpuscles on the hearts of animals is considerable, which should be given serious consideration.

Effects of Different Nanoparticles on Microbes

Abstract:Nanoparticles widely exist in nature and may be formed through inorganic or organic pathways, exhibiting unique physical and chemical properties different from those of bulk materials. However, little is known about the potential consequences of nanomaterials on microbes in natural environments. Herein, we investigated the interactions between microbes and nanoparticles by performing experiments on the inhibition effects of gold, ludox and laponite nanoparticles on Escherichia coli in liquid Luria–Bertani (LB) medium at different nanoparticle concentrations. These nanoparticles were shown to be effective bactericides. Scanning electron microscopy (SEM) images revealed the distinct aggregation of cells and nanoparticles. Transmission electron microscopy (TEM) images showed considerable cell membrane disruption due to nanoparticle accumulation on the cell surfaces, resulting in cell death. We hypothesized that this nanoparticle accumulation on the cell surfaces not only disrupted the cell membranes but also physically blocked the microbes from accessing nutrients. An iron-reducing bacterium, Shewanella putrefaciens, was tested for its ability to reduce the Fe (III) in solid ferrihydrite (HFO) or aqueous ferric citrate in the presence of laponite nanoparticles. It was found that the laponite nanoparticles inhibited the reduction of the Fe (III) in solid ferrihydrite. Moreover, direct contact between the cells and solid Fe (III) coated with the laponite nanoparticles was physically blocked, as confirmed by SEM images and particle size measurements. However, the laponite particles had an insignificant effect on the extent of aqueous Fe (III) bioreduction but slightly enhanced the rate of bioreduction of the Fe (III) in aqueous ferric citrate. The slightly increased rate of bioreduction by laponite nanoparticles may be due to the removal of inhibitory Fe (II) from the cell surface by its sorption onto the laponite nanoparticle surface. This result indicates that the scavenging of toxic heavy metals, such as Fe (II), by nanoparticles may be beneficial for microbes in the environment. On the other hand, microbial cells are also capable of detoxifying nanoparticles by coagulating nanoparticles with extracellular polymeric substances or by changing nanoparticle morphologies. Hence, the interactions between microbes and nanoparticles in natural environments should receive more attention.

Abstract:The application and consumption of nanoparticles (NPs) inevitably result in the contamination of environmental water. The internalized NPs in unicellular organisms could travel to human bodies along food chains and raise health concerns. Current research failed to determine the characteristics of cellular uptake of NPs by unicellular organisms at extremely low concentration in the real environment. We here developed a label-free high-throughput mass cytometry method to investigate gold NP (AuNP) uptake in a unicellular organism (Tetrahymena thermophila) at the single-cell level. The limit of detection for Au is as low as to 6.67 × 10–18 g/cell, which equals ∼5.3 5 nm AuNPs. We demonstrated that active engulfment pathways were responsible for the cellular accumulation of AuNPs and T. thermophila could also eliminate the cellular AuNPs rapidly. The interaction between AuNPs and T. thermophila is highly dependent on the sizes of nanoparticles; i.e., the population of T. thermophila containing AuNPs decreased with the increment of the diameters of AuNPs when exposed to the same mass concentration. For each type of AuNP, distinct heterogeneous cellular uptake of AuNPs by T. thermophila was observed. Intriguingly, for 5 nm AuNP, even at 0.001 ng/mL, some T. thermophila cells could concentrate AuNPs, indicating a real environmental concern even when water was contaminated by only trace level of NPs. The uptake and distribution of gold nanoparticles in tetrahymena pyriformis

Abstract:Tetrahymena pyriformis was chosen as the model organism to investigate the uptake and distribution of gold nanoparticles in aquatic animal.Gold nanoparticles with an average size of 13 nm were used in this experiment.The results showed that ingestion and endocytosis were the two most important processes for gold nanoparticle uptake by Tetrahymena pyriformis.Gold nanoparticles distributed in food vacuole,cytoplasm and macronuclear of Tetrahymena pyriformis.Statistical results showed that there was no obvious toxicity to Tetrahymena pyriformis with the exposure concentrations of gold nanoparticles used in the experiment.

Abstract:Although nanoparticles' (NPs) toxicity has been intensively studied, their effects on bacterial ingestion by protozoans (as an important component of the microbial loop) is unknown. This study investigated the effects of NPs of different chemical composition [hematite (HemNPs), anatase (AnaNPs), and silica (SiNPs) NPs] and size [SiNPs with particle size of 20 (Si-20), 100 (Si-100), and 500 (Si-500) nm] on the ingestion of Escherichia coli by the protozoan Tetrahymena thermophila. Potential differences between pre- vs. co-exposure were also assessed. Pre-exposure to HemNPs had no effects on bacterial ingestion but the other NPs caused a significant inhibition, due to their inhibition of ATP synthesis and the down-regulation of phagocytosis-related genes (ACT1 and CTHB). Contrastively, co-exposure to HemNPs and Si-20 didn't affect bacterial ingestion while co-exposure to AnaNPs (Si-100 and Si-500) induced (inhibited) ingestion. The stimulatory effect of AnaNPs was due to their induction of an increase in the intracellular Ca concentration of T. thermophila whereas the inhibitory effects of Si-100 and Si-500 were attributable to ATP synthesis reduction, enhanced bacterial cell aggregation, and competition between the bacterial cells and the NPs. These findings provide insights into the mechanisms underlying the environmental risks of NPs.

Abstract:The application of nanomedicines in oral drug delivery effectively promotes the drug absorption and transportation through enterocytes.Nevertheless,the absence of mechanism studies on efficacy and safety limits their final translation in humans.Although the vesicular trafficking has been verified as the general character for transport of nanomedicines,the deeper mechanism in molecular mechanism is still unclear.Moreover,the cellular transport of nanomedicines is a dynamic process involved by different organelles and components.However,most of existing studies just pay attention to the static location of nanomedicines,but neglect the dynamic biological effects on cells caused by them.Here,we prepared gold nanoparticles(AuNPs)as the model and cultured epithelial cell monolayer to explore the nano-bio interactions at the molecular level.The traditional pharmacological inhibition strategy and subcellular imaging technology elucidated the macropinocytosis/endosome/MVB/lysosome pathway during the transportation of AuNPs.Proteomics strategy based on mass spectrometry(MS)was utilized to identify and quantify proteins involved in the cellular transport of nanomedicines.Multiple proteins related to subcellular structure,signal transduction,energy transformation and metabolism regulation were demonstrated to be regulated by nanoparticle transport.These alterations of protein expression clarified the effects of intracellular proteins and verified the conventional findings.More importantly,it revealed a feedback mechanism of cells to the nano-trafficking.We believed that these new regulatory mechanisms provided new insights into the efficient transport of nanomedicines through epithelial barriers.

Abstract:Titanium dioxide nanoparticles (TiO2 NPs) are widely used in photodynamic therapy (PDT) of cancer treatment as excellent regenerative photocatalysts. However, there are some challenges because of their poor dispersity. Transferrin (Tf) was tried to modify the surface of TiO2 NPs to reduce the aggregation, which further affected uptake and excretion on SMMC-7721 human liver cancer cells. Initially, TiO2 NPs modified with Tf (TiO2-Tf NPs) entered into the cells faster than the pure TiO2 NPs which remain attaching on the cell membrane after short-term co-incubation. Tf modification increased the rate and amount of cellular endocytosis. Both TiO2 NPs and TiO2 Tf NPs were observed in lysosomes after long-term co-incubation through clathrin-mediated endocytosis pathway. Expulsion of NPs was then observed and it was found that the exocytosis of TiO2-Tf NPs was fast in the first 24 h, and then slowed down gradually from 24 h to 144 h. Totally, existence of Tf decreased the exocytosis of TiO2 NPs. Furthermore, the differences of cytotoxicity and genotoxicity between TiO2 NPs and TiO2-Tf NPs show that surface-adsorbed Tf components provide some protection from the cytotoxic effect by reducing the production of intracellular ROS. TiO2-Tf NPs obviously affected cell cycle, indicating a significant G2/M phase cell cycle arrest. Our results offer a promising application of easily aggregated TiO2 NPs in the nanomedicine field.
