Abstract:Enantiocomplementary hydroxylation of alkyl aromatics through a one‐pot photo‐biocatalytic cascade reaction is described. The photoredox process is implemented in aqueous phase with O 2 as oxidant and the subsequent ( R )‐ or ( S )‐selective bioreduction is performed by whole cell system without the addition of the expensive cofactor (NADPH). This mild, operationally simple protocol transforms a wide variety of readily available aromatic compounds into valuable chiral alcohols with high yield (up to 90%) and stereoselectivity (up to 99%), thereby displaying important potentials in organic synthesis.

Abstract:Herein,an organocatalytic enantioselective desymmetrizing hydroalkoxylationof 1,4- and 1,8-diynes is disclosed, which represents a unique chiralBronsted acid-catalyzed desymmetrization of diynes. This protocolprovides a facile and practical access to enantioenriched 1,3-diaminopropanolderivatives and & gamma;-butyrolactones with a wide substrate scopeand generally high enantioselectivities. Besides, the backbones ofthese products constitute the structural core of numerous bioactivemolecules, and they can serve as valuable precursors for the expeditiousassembly of versatile N- and O-heterocycles. Abstract:The development of efficient and sustainable methods for decarboxylative transformations is of great importance due to the ease of availability and nontoxicity of carboxylic acids. Despite tremendous efforts in this area, it remains challenging to develop enantioselective transformations direct from carboxylic acids. Herein we disclose a photoelectrocatalytic method for the direct and enantioselective decarboxylative cyanation. The photoelectrochemical reactions convert carboxylic acids to enantioenriched nitriles by employing cerium/copper relay catalysis with a cerium salt for catalytic decarboxylation and a chiral copper complex for stereoselective C-CN formation.

Abstract:The development of efficient and straightforward method for decarboxylative coupling using common alkyl carboxylic acid is of great value. However, the decarboxylative coupling with nucleophiles always needs stoichiometric chemical oxidants or substrate prefunctionalization. Herein, we report a protocol for Cu-catalyzed enantioselective decarboxylative cyanation via the merger of photocatalysis and electrochemistry. CeCl3 and Cu/BOX were used as co-catalyst to promote the decarboxylation and cyanation, and both catalysts were regenerated via the anodic oxidation. This method establishes the proof of concept enantioselective transformation via photoelectrocatalysis. Studies by DFT calculation provided mechanistic insight on enantioselectivity control.

Abstract:Herein, we reported the enantioconvergent Csp(3)-Csp(2) cross-coupling reaction with 1,4-dihydropyridine (DHP) derivatives via photoredox/nickel dual catalysis to access chiral products with good yield and enantioselectivity. The operationally simple reaction was carried out under mild conditions with good functional group tolerance. Due to the use of a stoichiometric equivalent of aryl/alkenyl halides as coupling partners, the sequential and iterative synthesis could be achieved smoothly in one pot for the synthesis of position isomers and stereoisomers. In the proposed mechanism, kinetic experiments and mechanistic studies indicated that the radical generation, depended on the excited photocatalyst and DHP, was the rate-determining step.

Abstract:The integration of organocatalysis and enzyme catalysis in one-pot cascade processes allows for the efficient construction of complex molecular architectures with high levels of stereocontrol. However, challenges related to reaction compatibility between both processes are often a limitation for the development of efficient synthetic routes. In this study, we describe the combination of an enzymatic aerobic oxidation followed by the squaramide-mediated asymmetric formation of C-P and C-C bonds to access important building blocks such as chiral α-hydroxy phosphonates and β-nitro alcohols in good yields and enantiomeric ratios. This sequential process is conducted in a one-pot fashion within a biphasic system and represents a pioneering example of a chemoenzymatic cascade involving aerobic biooxidation and an organocatalytic step operating under hydrogen-bond activation mode.

Abstract:A one‐pot photobiocatalytic system integrates liposomal photocatalysts with whole‐cell catalysts, enabling efficient light‐driven transformations under mild conditions. This approach combines photo‐ and biocatalytic activities to facilitate complex cascade reactions, streamlining chemical synthesis, enhancing sustainability, and reducing environmental impact, while offering potential to advance green chemical synthesis, providing a versatile platform for sustainable and innovative synthetic applications. Cooperative photobiocatalytic processes have seen extensive potentials for the synthesis of both bulk and fine chemicals owing to their versatility, eco‐friendliness, and cost‐effectiveness. Nevertheless, developing a universal and effective synthetic strategy compatible with both catalytic systems remains challenging. In this study, we explored cationic liposomes as biocompatible photocatalyst encapsulation systems and combined them with bacteria overexpressing enzymes for two‐step and three‐step cascade reactions. Specifically, the water‐soluble photocatalyst anthraquinone‐2‐sulfonate (AQS), which can oxidize benzyl alcohol, is encapsulated within the core of cationic liposomes composed of dioleoyl‐3‐trimethylammonium propane (DOTAP) and the helper lipid cholesterol. The positive charge on the liposome surface enabled electrostatic interactions with the negative charges on the membrane of Escherichia coli cells. Bacterial cells overexpressing various enzymes, such as Candida antarctica lipase B (CalB) and benzaldehyde lyase (BAL), and coated with liposomes enabled the production of added value compounds through cascade reactions with excellent production. These cascades involve CalB‐catalyzed hydrolysis, BAL‐catalyzed condensation, and AQS‐driven photo‐oxidation reactions. Therefore, the strategy offers more possibilities of combining photocatalysis with biocatalysis for recoverability, enhanced mass transfer, and enhanced compatibility in both industrial biotechnology and synthetic chemistry.

Abstract:There is a growing interest in developing cooperative chemoenzymatic reactions to harness the reactivity of chemical catalysts and the selectivity of enzymes for the synthesis of nonracemic chiral compounds. However, existing chemoenzymatic systems with more than one chemical reaction and one enzymatic reaction working cooperatively are rare. Moreover, the application of oxidoreductases in cooperative chemoenzymatic reactions is limited by the necessity of using expensive and unstable redox equivalents such as nicotinamide cofactors. Here, we report a light-driven cooperative chemoenzymatic system comprised of a photoinduced electron transfer reaction (PET) and a photosensitized energy transfer reaction (PEnT) with an enzymatic reduction in one-pot to synthesize chiral building blocks of bioactive compounds. As a proof of concept, ene-reductase was directly regenerated by PET in the absence of external cofactors. Meanwhile, enzymatic reduction worked cooperatively with photocatalyst-catalyzed energy transfer that continuously replenished the reactive isomer from the less reactive one. The whole system stereoconvergently reduced E/Z mixtures of alkenes to the enantiopure products. Additionally, enantioselective enzymatic reduction worked competitively with photocatalyst-catalyzed racemic background reaction and side reactions to channel the overall electron flow to the single enantiopure product. Such a light-driven cooperative chemoenzymatic system holds great potential for asymmetric synthesis using inexpensive petroleum or biomass-derived alkenes.

Abstract:While photochemical deracemization significantly enhances atom economy by eliminating the necessity for additional oxidants or reductants, the laborious presynthesis of substrates from feedstock chemicals is often required, thereby compromising the practicality of this method. In this study, we propose a novel approach known as de novo deracemization synthesis, which involves direct utilization of simple substrates undergoing both photochemical transformation and reversible photochemical transformation. The efficient enantiocontrol of chiral catalysts in the latter process establishes an effective platform for deracemization. This alternative and practical approach to address the challenges of asymmetric photocatalysis has been successfully demonstrated in the photosensitized de novo deracemization synthesis of azaarene-functionalized cyclobutanes featuring three stereocenters, including an all-carbon quaternary center. By exclusively employing a suitable chiral catalyst to enable kinetically controlled [2 + 2] photocycloreversion, we pave a creative path toward achieving more cost-effective photochemical deracemization.

Abstract:High-efficiency photo-enzymatic synthesis of chalcone chemicals was achieved by tandem catalysis based on the photocatalytic oxidation of porphyrin-based metal organic frameworks (MOFs) confined copper nanoclusters and the non-specific catalytic activity of hydrolases. A series of photocatalysts were successfully synthesized and the Cu nanoparticles (NPs) were loaded on the materials by in-situ reduc-tion of Cu2+. Subsequently, in situ oxidation of benzyl alcohol and aldol reaction catalyzed in tandem by Cu@PCN-222(M) and hydrolase were used as template reactions and the reaction conditions were sys-tematically studied. 20 chalcone derivatives were successfully synthesized (yield up to 98 %). Finally, the optical properties difference of photocatalysts were studied by spectroscopic and electrochemical analysis. The reaction system used greener benzyl alcohol as the reaction substrate and was driven by visible light under O2. This method has good yield and recovery performance, which provides a new strat-egy for the construction of MOF-enzyme combined catalytic system.(c) 2022 Published by Elsevier Inc.

Abstract:In a continuous effort to emulate the efficiency of biosynthetic pathways, considerable progress has been made in developing one-pot chemoenzymatic processes that take full advantage of the chemo-, regio-, and stereoselectivity of biocatalysts and the productivity of chemical catalysts. Over the last 20 years, research in this area has provided us with proof of concept examples in which chemical and biological transformations occur in one vessel, sequentially or concurrently. These transformations typically access products with high enantiopurity and chemical diversity. In this perspective, we present some of the most successful reports in this field.

Abstract:Living organisms rely on simultaneous reactions catalysed by mutually compatible and selective enzymes to synthesize complex natural products and other metabolites. To combine the advantages of these biological systems with the reactivity of artificial chemical catalysts, chemists have devised sequential, concurrent, and cooperative chemoenzymatic reactions that combine enzymatic and artificial catalysts1–9. Cooperative chemoenzymatic reactions consist of interconnected processes that generate products in yields and selectivities that cannot be obtained when the two reactions are carried out sequentially with their respective substrates2,7. However, such reactions are difficult to develop because chemical and enzymatic catalysts generally operate in different media at different temperatures and can deactivate each other1–9. Owing to these constraints, the vast majority of cooperative chemoenzymatic processes that have been reported over the past 30 years can be divided into just two categories: chemoenzymatic dynamic kinetic resolutions of racemic alcohols and amines, and enzymatic reactions requiring the simultaneous regeneration of a cofactor2,4,5. New approaches to the development of chemoenzymatic reactions are needed to enable valuable chemical transformations beyond this scope. Here we report a class of cooperative chemoenzymatic reaction that combines photocatalysts that isomerize alkenes with ene-reductases that reduce carbon–carbon double bonds to generate valuable enantioenriched products. This method enables the stereoconvergent reduction of E/Z mixtures of alkenes or reduction of the unreactive stereoisomers of alkenes in yields and enantiomeric excesses that match those obtained from the reduction of the pure, more reactive isomers. The system affords a range of enantioenriched precursors to biologically active compounds. More generally, these results show that the compatibility between photocatalysts and enzymes enables chemoenzymatic processes beyond cofactor regeneration and provides a general strategy for converting stereoselective enzymatic reactions into stereoconvergent ones.

Abstract:Enzymes are increasingly explored for use in asymmetric synthesis1–3, but their applications are generally limited by the reactions available to naturally occurring enzymes. Recently, interest in photocatalysis4 has spurred the discovery of novel reactivity from known enzymes5. However, so far photoinduced enzymatic catalysis6 has not been used for the cross-coupling of two molecules. For example, the intermolecular coupling of alkenes with α-halo carbonyl compounds through a visible-light-induced radical hydroalkylation, which could provide access to important γ-chiral carbonyl compounds, has not yet been achieved by enzymes. The major challenges are the inherent poor photoreactivity of enzymes and the difficulty in achieving stereochemical control of the remote prochiral radical intermediate7. Here we report a visible-light-induced intermolecular radical hydroalkylation of terminal alkenes that does not occur naturally, catalysed by an ‘ene’ reductase using readily available α-halo carbonyl compounds as reactants. This method provides an efficient approach to the synthesis of various carbonyl compounds bearing a γ-stereocentre with excellent yields and enantioselectivities (up to 99 per cent yield with 99 per cent enantiomeric excess), which otherwise are difficult to access using chemocatalysis. Mechanistic studies suggest that the formation of the complex of the substrates (α-halo carbonyl compounds) and the ‘ene’ reductase triggers the enantioselective photoinduced radical reaction. Our work further expands the reactivity repertoire of biocatalytic, synthetically useful asymmetric transformations by the merger of photocatalysis and enzyme catalysis. A transformation in which an ‘ene’ reductase catalyses the visible-light-induced intermolecular radical hydroalkylation of alkenes gives carbonyl compounds with a remote stereocentre in high yield and enantioselectivity.

Abstract:A series of cinchona-derived N-oxide asymmetric phase-transfer catalysts were synthesized and applied in the enantioselective photo-organocatalytic alpha-hydroxylation of beta-keto esters and beta-keto amides (23 examples) using molecular oxygen in excellent yields (up to 98%) and high enantioselectivities (up to 83% ee).
