Abstract:Herein,an organocatalytic enantioselective desymmetrizing hydroalkoxylationof 1,4- and 1,8-diynes is disclosed, which represents a unique chiralBronsted acid-catalyzed desymmetrization of diynes. This protocolprovides a facile and practical access to enantioenriched 1,3-diaminopropanolderivatives and & gamma;-butyrolactones with a wide substrate scopeand generally high enantioselectivities. Besides, the backbones ofthese products constitute the structural core of numerous bioactivemolecules, and they can serve as valuable precursors for the expeditiousassembly of versatile N- and O-heterocycles. Abstract:The integration of organocatalysis and enzyme catalysis in one‐pot cascade processes allows for the efficient construction of complex molecular architectures with high levels of stereocontrol. However, challenges related to reaction compatibility between both processes are often a limitation for the development of efficient synthetic routes. In this study, we describe the combination of an enzymatic aerobic oxidation followed by the squaramide‐mediated asymmetric formation of C‐P and C‐C bonds to access important building blocks such as chiral α‐hydroxyphosphonates and β‐nitro alcohols in good yields and enantiomeric ratios. This sequential process is conducted in a one‐pot fashion within a biphasic system and represents a pioneering example of a chemoenzymatic cascade involving aerobic biooxidation and an organocatalytic step operating under hydrogen‐bond activation mode and under mild reaction conditions.

Abstract:A mild and efficient enantioselective one-pot synthesis of β-sulfonyl ketones and trisubstituted tetrahydrothiophenes via introducing 1,4-dithiane-2,5-diol to the simple kinetic resolution of β-sulfonyl ketones has been described herein. The one-pot reaction sequence including kinetic resolution and cascade sulfa-Michael/Aldol reaction proceeded successively to afford corresponding sulfonyl ketones and tetrahydrothiophenes with high enantioselectivities (85%–98% ee and 84%–95% ee).

Abstract:(R)-Phenylglycinol is an important chiral building block for pharmaceutical and fine chemical industry, and its efficient synthesis from cheap and commercially available starting materials is challenging and highly desirable. Herein, a new three-step one-pot cascade system employing epoxide hydrolase, glycerol dehydrogenase, and omega-transaminase was designed for the asymmetric synthesis of (R)-phenylglycinol from racemic styrene oxide. A cofactor self-sufficient system employing AlaDH/L-Ala was utilized for the regeneration of expensive cofactor NAD(+) and removal of by-product pyruvate. Furthermore, in situ product removal by cation resin adsorption was used to drive the thermodynamic equilibrium of the cascade reaction to the direction of product generation. Finally, optically pure (R)-phenylglycinol was successfully produced from racemic styrene oxide with high yield (81.9 %) and excellent enantioselectivity (99 % ee).

Abstract:An efficient one-pot, two-step process was disclosed for production of chiral α,β-epoxy ketones from benzaldehydes and acetophenones catalyzed by imidazolium-modified poly(l-leucine). Two effective reaction systems with complementary high enantioselectivities (up to 98% ee) or satisfactory yields (up to 89%) have been developed. Importantly, the poly(amino acid) catalyst can be easily recovered and recycled for ten times without losing its catalytic efficiency in terms of both enantioselectivity and yield.

Abstract:A double-enzyme-catalyzed strategy for the synthesis of enantiocomplementary vicinal fluoro alcohols through a one-pot, three-step process including lipase-catalyzed hydrolysis, spontaneous decarboxylative fluorination, and subsequent ketoreductase-catalyzed reduction was developed. With this approach, beta-ketonic esters were converted to the corresponding vicinal fluoro alcohols with high isolated yields (up to 92%) and stereoselectivities (up to 99%). This new cascade process addresses some issues in comparison with traditional methods such as environmentally hazardous reaction conditions and low stereoselectivity outcome.

Abstract:An Rh 2(OAc)4/Zr-3-I-binol co-catalyzed enantioselective three-component cascade reaction of diazo compounds, ethyl 4-hydroxybutenoate and furfurals has been designed to build complex fused O-heterocycles in a one pot fashion. The reaction was conducted in CH2Cl2 or CHCl3 at ambient temperature with 3 A molecular sieves as an additive. The multi- component process was completed within 3 h to give multi-component products and the following Diels-Alder (DA) cycliza- tion product was observed in the next several hours. It was found that the DA process was slow but could be completed after 7~12 d by standing the crude reaction mixture after evaporating solvent. Following a one-pot epoxidation of the product further increases the molecular complexity to give fused epoxidation products bearing multiple chiral carbon centers. Espe- cially to be noted, all of the products are very easy to be purified by simple recrystallization in mixture solvent of ethyl ace- tate (EA)/petroleum ether (PE, 60~90 ℃) and do not need any column chromatography method. Using the developed method, hydroepoxyisochromene derivatives with up to eight contiguous chiral carbon centers have been readily prepared from simple starting materials with high diastereoselectivity (up to 99/1 dr) and enantioselectivity (up to 99% ee). The abso- lute stereochemistry of product was determined by the single crystal X-ray analysis as a chair conformation with absolute configurations. The possible mechanisms were theoretically investigated by DFT calculation and proposed as an oxonium ylide-trapping three-component reaction subsequently followed with a spontaneous intramolecular Diels-Alder process and an epoxidation at the end. The developed three-component cascade reaction provides a rapid access to the construction of chiral fused polycyclic O-heterocycles with molecular complexity. Using cinchona alkaloid-derived bifunctional thiourea as the catalyst, chiral β-hydroxy-α-amino acid derivatives can be obtained in excellent yields and enantioselectivities (up to 95% yield and 92% ee) after acidic hydrolysis. This protocol provides a straightforward method to access multiple substituted β-hydroxy-α-amino acid derivatives with high enantiomeric purity.

Abstract:An efficient kinetic resolution of racemic oxaziridines has been realized via catalytic asymmetric α-hydroxylation of available β-keto esters. In the presence of a chiral bifunctional guanidine catalyst, a variety of optically active oxaziridines and chiral α-hydroxy β-keto esters were generated with excellent results (ee's of up to 99% and 97% and yields of up to 44% and 54%, respectively).

Abstract:Chiral benzo five-membered heterocyclic spirocyclopropanes are an important class of parent core structures with pharmacological activity. A novel organocatalytic one-pot cascade ether oxidation iminium-ion activation strategy for the asymmetric spirocyclopropylation of benzofuran-2-ones and indolin-2-ones from allyl tert-butyl ethers/ pent-2,4-dienyl ethyl ethers with excellent enantioselectivity (ee% up to > 99) and diastereoselectivity(dr.% up to 91:9) has been developed. This process involves the successful dynamic kinetic resolution of racemic 3-bromobenzofuran-2-ones or 3-bromoindolin-2-ones. Its synthetic application will provide a new aminocatalytic cascade tool for the efficient synthesis of complex molecules.
