Abstract:Sulfur dioxide (SO,) is a dangerous environmental pollutant. Excessive intake of it may cause some respiratory diseases and even lung cancer. The development of effective methods for detection of SO2 is of great importance for the environment and physiology. Herein, we have designed and synthesized a novel two-photon (TP) and deep-red emission ratiometric fluorescent probe (CP) for detection of SO2. Notably, the novel probe CP exhibited ultrafast response to SO2 in less than 5 s and displayed a great emission shift (195 nm) and a large emission signal ratio variation (enhancement from 0.1347 to 100.14). In addition, the unique probe was successfully employed for imaging SO2 not only in the mitochondria of living cells but also in brain tissues and zebrafishes.

Abstract:As a reactive sulfur species, sulfur dioxide (SO2) and its derivatives play crucial role in various physiological processes, which can maintain redox homeostasis at normal levels and lead to the occurrence of many diseases at abnormal levels. So, the development of a suitable fluorescent probe is a crucial step in advancing our understanding of the role of SO2 derivatives in living organisms. Herein, we developed a near-infrared fluorescent probe (SP) based on the ICT mechanism to monitor SO2 derivatives in living organisms in a ratiometric manner. The probe SP exhibited excellent selectivity, good sensitivity, fast response rate (within 50 s), and low detection limit (1.79 µM). In addition, the cell experiment results suggested that the SP has been successfully employed for the real-time monitoring of endogenous and exogenous SO2 derivatives with negligible cytotoxicity. Moreover, SP was effective in detecting SO2 derivatives in mice.

Abstract:Sulfur dioxide (SO2) is one of the most important reactive sulfur species widely found in living organisms. SO2 is recognized as an important regulatory factor that can modulate the cardiovascular system, with functions such as relaxing blood vessels, lowering blood pressure and acting as an anti-oxidation. Compared to fluorescence turn-on type probes, a ratiometric fluorescent probe can avoid the background fluorescence of the probe and the interference from other environmental factors. Herein, a new ratiometric-type fluorescent probe with emission in the near infrared area was developed. Upon interaction with SO2, the emission at 750 nm decreased and a new fluorescent peak was observed in the visible region simultaneously. The novel probe showed excellent selectivity toward SO2 among other species. In addition, the new probe exhibited little toxicity to HeLa cells and was used for imaging SO2 in solution and the cell environment successfully with a fluorescence change in both the green and red channels.

Abstract:SO2 has been recently identified as an essential gas messenger followed by NO, CO and H2S. However, abnormal concentrations of SO2 in our bodies can cause many diseases. Thus, the real-time monitoring of SO2 to well define the generation, physiological and pathological functions of SO2 is urgently needed. In this work, we developed a novel SO2 fluorescent probe on the basis of the conjugation of semi-cyanine and coumarin derivate dyes with superior features, such as near-infrared (NIR) and two-photon dual-mode monitoring, a large Stokes shift (175 nm), ultrafast response towards SO2 (within 10 s), high selectivity and photostability. Furthermore, this probe could sense SO2 by dual colorimetric and fluorescence means. In biological imaging, the probe was able to trace exogenous and endogenous SO2 in living cells, mitochondria, E. coli, zebrafish and mice under an NIR and two-photon dual-mode. These results demonstrated that the probe has strong potential for studying the physiological and pathological functions of SO2in vitro and in vivo.

However, it has been documented that excessive intake of SO2 could cause serious health problems. Therefore, it is very necessary to develop a novel SO2 probe with excellent performance. Herein, a novel fluorescent probe (Ph-CN) based on the ICT-ESIPT mechanism for monitoring SO2 with the C[double bond, length as m-dash]N double bond was developed. Furthermore, Ph-CN exhibits various splendid features, such as near-infrared (NIR) and ratiometric detection, real-time detection (<5 s), good selectivity and photostability, large Stokes shift (230 nm) and large emission shift (200 nm). Most importantly, the probe Ph-CN can realize the detection of SO2 in HeLa cells and in vivo.

Abstract:In most biophysiological processes, sulfur dioxide (SO 2 ) is an important intracellular signaling molecule that plays an important role. The change of SO 2 in cells are closely related to various diseases such as neurological disorders and lung cancer, so it is necessary to develop fluorescent probes with the ability to accurately detect SO 2 during physiological processes. In this work, we designed and synthesized a multifunctional fluorescent probe TIS. TIS has excellent properties such as near-infrared emission, large stokes shift, excellent SO 2 detection capabilities, low detection limit, high specificity and visualization of color change before and after reaction. Simultaneously, TIS has low cytotoxicity, good biocompatibility, clear cell imaging capability and mitochondrial targeting ability. In addition, the ability of TIS to be applied to different material surfaces for latent fingerprint fluorescence imaging was also explored. TIS provides an excellent method for the accurate detection of SO 2 derivatives and shows great potential applications in near-infrared cellular imaging and latent fingerprint fluorescence imaging.

Abstract:A novel mitochondria-targeted ratiometric near-infrared fluorescence probe NDMBT for Sulfur dioxide (SO2) derivatives was constructed based on the SO2 derivatives-triggered Michael addition reaction. It displayed ultrafast response time (within 10s), large hypsochromic shift (260nm), high photostability, excellent selectivity and high sensitivity in aqueous media with a detection limit of 43nM. More importantly, it was successfully applied to imaging of the enzymatically generated SO2 derivatives in mitochondria of live cells.

Abstract:Sulfur dioxide derivatives (HSO3− and SO32−) play an important role in food preservative, antibacterial, antioxidant and other aspects, so it is urgent for us to develop more efficient detection methods to broaden their application in biochemical research and related disease diagnosis. Fluorescent probes are of particular interest because of their simplicity and high temporal and spatial resolution. Herein, we constructed a new near-infrared (NIR) fluorescence probe, CQC, composed of coumarin fluorophore and quinoline fluorophore, for detecting SO2 derivatives. The near-infrared emission probe CQC with a large Stokes shift (260 nm) not only kept the distance between the two emission peaks large enough (165 nm), but also had a particularly high energy transfer efficiency (99.5%), and was particularly sensitive to the detection of HSO3−/SO32− (LOD: 0.1 μM). The powerful probe CQC succeeded in real-time visualizing endogenous HSO3−/SO32− in living cells.

Abstract:A ratiometric fluorescent probe (L-HF3) based on an efficient FRET platform constructed by 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) and 3-hydroxyflavone (HF) fluorophores for detection of sulfur dioxide derivatives (HSO3−/SO32−) was developed. The probe featured very large (pseudo) Stokes shifts, good selectivity and high sensitivity, high FRET efficiency and high resolution of the emission bands. The probe was also mitochondria-targeted and could detect exogenous and endogenous bisulfite in living cells.

Abstract:A promising near-infrared emissive and mitochondria-targeted fluorescence probe (SNB) for the ratiometric detection of sulfur dioxide derivatives with a novel reaction mechanism was developed on the basis of FRET and the ICT platform. Probe SNB showed favorable fluorescence properties, including an active NIR emission signal (671 nm), ultra-large Stokes shift (251 nm) and an ultra-broad emission band gap (193 nm). Furthermore, SNB was applied to detect sulfur dioxide derivatives with a low detection limit (17 nM), excellent sensitivity and exceptional selectivity. Of greater significance, SNB also successfully implemented the real-time and ratiometric surveillance for exogenous or endogenous sulfur dioxide derivatives in living HeLa, L-O2 and HepG2 cells. Interestingly, SNB possessed a subcellular organelle targeting ability with a preferred co-localization coefficient of 0.91 for mitochondria, which revealed its promising potential in the detection of subcellular sulfur dioxide derivatives.

Abstract:Inflammation is a complex physiological response involving various cellular and molecular events. Sulfur dioxide (SO2), which is usually in the form of HSO3- and SO32- under physiological conditions, plays a crucial role in the regulation of inflammation and diseases. Frequency upconversion luminescence (FUCL) can realize the unique anti-Stokes process of long-wavelength excitation to short-wavelength emission; thus, it is a highly promising optical method for in vivo imaging due to its deep tissue penetration, low photo-damage, etc. Therefore, we developed a near-infrared FUCL probe NIRX-1 for the detection of HSO3-. NIRX-1 had a fast response (80 s), a low detection limit (0.43 μM), and a high selectivity towards HSO3-. In addition, NIRX-1 had a deep light penetration ability due to the near-infrared excitation at 808 nm, and was able to detection of HSO3- in living cells and mice. Lastly, NIRX-1 was employed in the imaging of HSO3- in an inflammation mouse model through FUCL imaging techniques. All these features make NIRX-1 a good candidate for the investigate of SO2-associated physiological and pathological processes.

Abstract:Sulfur dioxide (SO 2 ), as a known important sulfur oxide in RSS, has an essential role in achieving the homeostasis of organisms. The development of a highly sensitive and selective fluorescent probe for monitoring SO 2 is of great interest in understanding its pathophysiology. Herein, we developed a new near-infrared FRET-based fluorescent probe QD , in which quinolinium fluorophore was selected as the donor and the dicyanoisophorone fluorophore was selected as the acceptor. Probe QD had dual colorimetric and ratiometric fluorescence, larger Stokes shift (277 nm) and longer emission wavelength (677 nm). The fluorescence spectrum showed that QD was highly selective and sensitive with excellent linear response concentration range of 2.5 to 80 μM. Importantly, probe QD allowed detection of SO 2 derivatives and bioimaging of lysosome-targeted in live cells.

Abstract:SO2 and its derivatives (bisulfite/sulfite) play crucial roles in several physiological processes. Therefore, development of reliable analytical methods for monitoring SO2 and its derivatives in biological systems is very significant. In this paper, a FRET-based two-photon fluorescent turn-on probe, A-HCy, was proposed for specific detection of SO2 derivatives through the bisulfite/sulfite-promoted Michael addition reaction. In this FRET system, an acedan (2-acetyl-6-dialkylaminonaphthalene) moiety was selected as a two-photon donor and a hemicyanine derivative served as both the quencher and the recognition unit for bisulfite/sulfite. A-HCy exhibited excellent selectivity and rapid response to HSO3− with a detection limit of 0.24 μM. More importantly, probe A-HCy was first successfully applied in two-photon fluorescence imaging of biological SO2 derivatives in living cells and tissues, suggesting its great potential for practical application in biological systems.

Abstract:Sulfur dioxide (SO2), a possible endogenous gaseous signaling molecule, has been found to be correlated with various diseases in living organisms. Thus, the development of effective methods for detection of SO2 are of great physiological and pathological significance. In this work, we report a ratiometric NIR fluorescent probe Cou-Rh for monitoring bisulfite on basis of Michael addition reaction. This probe is prepared from coumarin by acid-catalyzed cyclization reaction. The addition of bisulfite results in a significant decrease of fluorescence intensity at 695 nm and enhancement at 550 nm. The probe also displays high selectivity over various biologically related analytes with nanomolar limit of detection. What's more, fluorescence imaging further confirms that the probe is capable of detecting bisulfite in living cells and zebrafish larvae. The results demonstrate that probe Cou-Rh can be employed for a useful scaffold to study the physiological function of sulfur dioxide.

Abstract:The development of rapid, convenient, and sensitive methods for selective detection SO2 derivatives is of considerable significance in biomedical research and has become a topical objective. Herein four probes (DS1, DS2, DB1 and DB2) based on pi-conjugated indolium/benzoindolium systems were designed for detection bisulfite/sulfite. The results indicated that probes DB1 and DB2 showed rapid response, high selectivity and sensitivity for bisulfite/sulfite anions with significant ratiometric fluorescent signal changes. Moreover, DB1 and DB2 were successfully applied for bioimaging SO2 derivatives in living cells under one-and two-photon confocal laser scanning microscopy. Abstract:A novel chromenylium-based fluorescent probe was exploited for sulphur dioxide (SO₂) detecting. The probe displayed a remarkable fluorescence turn-on response towards SO₂ based on the nucleophilic addition reaction to the carbon-carbon double bond with 105 nm Stock shift. The probe was successfully applied for the quantification of SO₂.The linear detection range was from 0-160 μM with the detection limit as low as 99.27 nM. It also exhibited high selectivity for SO₂ than other reactive species and amino acids. Furthermore, cell staining experiments indicated that the probe was cell membrane permeable and could be used for high-performance imaging of SO₂ in living cells. The superior properties of the probe made it highly promising for use in chemical and biological applications.
