Abstract:Objective To establish a solid phase extraction(SPE)-high performance liquid chromatography(HPLC)method for determination of melamine in foods. Methods Melamine in sample was extracted with extractive agent. After centrifugation, the supernatant fluid was purified by Waters Oasis MCX cartridge,and detected with HPLC method. Chromatographic column of TIANHE C18(250 mm×4.6 mm,5 μm) was used at 35 ℃ and mobile phase was water(containing 0.01 mol/L 1-hexanesulfonic acid sodium salt and 0.01 mol/L citric acid)∶ acetonitrile = 94∶6 (V∶V) at a flow rate of 1.0 ml/min. Injection volume was 20 μl. Results In the range of 0.80 to 80.0 mg/L, the regression equation was y=84.64 x-6.127 1 with the correlation coefficient of 0.999 98. The detection limit of melamine in foods was 0.5 mg/kg. The average rates of recovery were 88.0%-96.0% and the relative standard deviation was less than 5%. Conclusion The method established in the present paper is applicable to determination of melamine in foods with good precision,accuracy and sensitivity.

Abstract:Mercury (Hg) is a toxic element that exists in various forms, each with its characteristics and impacts on the environment. Methylmercury (MeHg) bioaccumulates in the food chain and identifying mercury species is crucial to assessing risks to ecology and health. Therefore, accurate, sensitive, and broad-range analytical methods are essential for the determination of methylmercury. This study optimized and validated a procedure for Hg speciation analysis in rice, fish, and soil using different extractants, followed by liquid chromatography inductively-coupled plasma mass spectrometry (LC ICP-MS) using certified references. Microwave-assisted extraction (MAE) parameters were optimized using 25 % KOH, resulting in improved sensitivity, accuracy and processing time. The validated method achieved a limit of quantitation (LOQ) of 12 µg kg−1 and showed acceptable percentage recoveries (75.3–98.1 %) in all matrices, enabling precise determination of MeHg (%RSDr; 3.76–5.78, %RSDR; 2.34–6.75) and Hg in a large number of samples within a short time frame. The validated method is particularly well-suited for rice, followed by fish and then soil. Therefore, the method utilizing microwave-assisted extraction and LC ICP-MS ensures precise methylmercury determination in rice, fish, and soil, crucial for evaluating environmental and health risks associated with mercury bioaccumulation.

Abstract:We described the hyphenation of photo-induced chemical vapor generation with high performance liquid chromatography–atomic fluorescence spectrometry (HPLC–AFS) for the quantification of inorganic mercury, methylmercury (MeHg) and ethylmercury (EtHg). In the developed procedure, formic acid in mobile phase was used for the photodecomposition of organomercury compounds and reduction of Hg2 + to mercury vapor under microwave/ultraviolet (MW/UV) irradiation. We optimized the proposed method studying the influence of several operating parameters, including the type of organic acid and its concentration, MW power, composition of HPLC mobile phase and catalytic action of TiO2 nanoparticles. Under the optimized conditions, the limits of detection were 0.15, 0.15 and 0.35 μg L− 1 for inorganic mercury, MeHg and EtHg, respectively. The developed method was validated by determination of the main analytical figures of merit and applied to the analysis of three certified reference materials. The online interfacing of liquid chromatography with photochemical-vapor generation–atomic fluorescence for mercury determination is simple, environmentally friendly, and represents an attractive alternative to the conventional tetrahydroborate (THB) system.

Abstract:This work demonstrated the feasibility of mercury speciation analysis by anion exchange chromatographic separation with inductively coupled plasma mass spectrometry detection. For the first time, by complexing with the mobile phase containing 3-mercapto-1-propanesulfonate into negatively charged complexes, fast separation of inorganic mercury (Hg2+), monomethylmercury (MeHg), ethylmercury (EtHg) and phenylmercury (PhHg) was achieved within 5 min on a 12.5-mm strong anion exchange column. The detection limits for Hg2+, MeHg, EtHg and PhHg were 0.008, 0.024, 0.029 and 0.034 mu g L-1, respectively. The relative standard deviations of peak height and peak area (5.0 mu g L-1 for each Hg species) were all below 3%. The determined contents of Hg2+, MeHg and total Hg in a certified reference material of fish tissue by the proposed method were in good accordance with the certified values with satisfactory recoveries. The relative errors for determining MeHg and total mercury were -2.4% and -1.2%, respectively, with an acceptable range for spike recoveries of 94-101%. Mercury speciation in 11 fish samples were then analyzed after the pretreated procedure. Abstract:Adsorbed by sulfhydryl cotton in acidic solution,methylmercury in drinking water was determined by gas chromatograph with ECD.The solid phase extraction was performed by using a multi-channel paralleling device and influence of pH value in water samples on recovery was observed.It was found that good linearity of method obtained in range from 0.050 mg/L to 1.00 mg/L,detection limits 0.03 ng/L at sample volume 1.0 L,RSD of standard and parallel samples <5.0%,spiked recoveries >80%.Some suggestions were made for accurate data by using QA/QC of laboratory including blank through entire procedure along with samples,solution blank,parallel samples,and spiked recoveries.

Abstract:A simple, rapid and efficient sample preconcentration method of ultrasonic assisted cloud point extraction ( CPE ) coupled with atomic fluorescence spectrometry ( AFS ) was developed for the determination of trace mercury in water samples. In this method, Thio-Michler's ketone was used as a chelating agent and Triton X-114 was used as a surfactant to enrich trace mercury in the surfactant phase. Before AFS analysis, tributyl phosphate was added as an antifoam. Under the optimal extraction conditions, such as pH, the amount of chelating agent and surfactant, the detection limit of the method is 0.003 μg/L, the relative standard deviation is 4.2–8.6% and standard recovery rate is 85–106%. The results show that ultrasonic assisted CPE-AFS can be applied to the determination of trace mercury in clean water samples, such as surface water and tap water, with lower detection limit, good accuracy and precision and can meet the requirements of higher sensitivity for sample analysis.

Abstract:Mercury is a major pollutant in food crops. In this study, we synthesized an anti-mercury monoclonal antibody (mAb; IC50 was 0.606 ng mL-1) with high sensitivity and specificity and different immunogens and coating antigens and developed an immuno-chromatographic assay (ICA) for the detection of mercury in rice. The ICA strip had a visible detection limit of 20 ng g-1 and a cut-off value of 500 ng g-1 in rice. The performance of the ICA strip was consistent with that of ICP-MS and ic-ELISA. The recoveries of mercury in rice ranged from 94.5% to 113.7% with ic-ELISA and from 93.6% to 116.45% with ICP-MS. Qualitative analysis by ICA can be obtained with the naked eye. The ICA strip is an effective and practical method for the rapid and high-throughput determination of mercury in rice.

Abstract:A method for the determination of methylmercury(MeHg) in organism was developed using a direct Hg analyzer(DMA-80).The analysis of MeHg in biota samples was performed by separating of MeHg from sample matrix with hydrobromic acid,extraction of MeHg with toluene and back-extraction of MeHg from organic solvent to cysteine aqueous solution.MeHg in cysteine aqueous solution was determined by the mercury analyzer.The relative error between the determined and the certified values of standard reference materials(GBW 10029,SRM 2976) was less than 4%,and the relative standard deviation of the determined values was less than 2%.The detection limit of MeHg was 0.64 ng·g-1.The total extraction efficiency was between 95% and 99%.This method can yield accurate and reliable result,and allows routine analysis of both total mercury and MeHg in organisms using the same instrument.

Abstract:A new method was established to determine the content of total mercury in chemical raw material with sulfamethoxazole as research object.Mercury was determined by cold vapor atomic absorption spectrometry with pretreatment of sample by microwave digestion.The detection wavelength was 253.7nm.There was a good linear relationship between the absorbance and the concentration of mercury in the range of 0—18ng·mL-1,and the correlation coefficient was 0.9996.The average additional standard recovery was 112.7%,and the detection limit was 0.02μg·g-1.This method has good specific,high sensitive with accurate and rapid,and can be applied to turning out a universal method for the determination of total mercury in chemical raw material,and can provide basis for drug quality control,simultaneously.

Abstract:Methylmercury is one of the most virulent pollutants and does a lot of harm to human health through bioconcentration,biomagnifications and transfer by food chain,although it is usually in the trace levels in the environment.With more attention being attached to the environmental safety and human healthy,it became necessary to develop high sensitive,high accurate,fast,simplified and economic detection technology for methylmercury.This article introduced systematically some determination methods for methylmercury in environmental samples and discussed the advantages and shortcomings of different detection methods in case of pretreatment,separation procedure and final detector.On that base,we took an outlook for the determination method in the future.GC in cooperation with ICP-MS or ID-ICP-MS would be widely used because of high sensitive and accurate.Biosensor would be a new technology,which is more useful to detect methylmercury in polluted water and sediment,and reducing its detection limit is the key task for this method.

Abstract:A method for the determination and confirmation of methylene blue (MB) in aquatic products was developed. Residues of MB were extracted from homogenized tissues with acetonitrile/sodium acetate buffer solution, and simply cleaned up with dichloromethane liquid/liquid extraction. After concentration and dissolution, the sample solutions were cleaned up by the neutral alumina and weak cation-exchange solid phase extraction (SPE) cartridge, prior to LC-MS/MS analysis. MB was determined at 1.0-20 mu g/kg in eel, toasted eel and shrimp, with a limit of quantification of 0.5 mu g/kg. Recovery for MB was between 73.0% and 108.3%. This method is fast, exact and sensitive. It can be applied to determine MB in aquatic products.

Abstract:Arsenic speciation in paddy rice is of considerable interest due to its impact on the food safety and human health. In this study, a simple methodology was developed to simultaneously extract and analyze As species in rice from China. Arsenic species, including arsenite (As(III)), arsenate (As(V)), dimethylarsinic acid (DMA), and monomethylarsonic acid (MMA), were extracted by methanol-water (50:50, v/v) containing 0.02 mol L−1 nitric acid with a microwave-assisted procedure, and then determined by high performance liquid chromatography–hydride generation-atomic fluorescence spectrometry (HPLC–HG-AFS). The results showed that the method has good efficiency (>90%) for rice, indicating that there were no significant losses or transformations of arsenic during sample treatment and analysis. The limits of quantification (LOQ) of the method were 8.0, 20, 12, and 12 ng g−1 for As(III), As(V), DMA, and MMA, respectively. When this method was applied to the analysis of rice, As(III) had the highest concentration, followed by DMA, As(V), and MMA. The estimated weekly intake of inorganic As from rice by Chinese people accounted for 11.83% of the provisional tolerable weekly intake. The As speciation results in this study suggest that the risk associated with As in rice to human health may be negligible.

Abstract:To explore the possibility of using chemical extraction methods to predict phytoavailability/bioaccumulation of soil-bound MeHg, MeHg extractions by three widely-used extractants (CaCl2, DTPA, and (NH4)2S2O3) were compared with MeHg accumulation in rice grains. Despite of variations in characteristics of different soils, MeHg extracted by (NH4)2S2O3 (highly affinitive to MeHg) correlated well with grain MeHg levels. Thus (NH4)2S2O3 extraction, solubilizing not only weakly-bound and but also strongly-bound MeHg, may provide a measure of ‘phytoavailable MeHg pool’ for rice plants. Besides, a better prediction of grain MeHg levels was obtained when growing condition of rice plants was also considered. However, MeHg extracted by CaCl2 or DTPA, possibly quantifying ‘exchangeable MeHg pool’ or ‘weakly-complexed MeHg pool’ in soils, may not indicate phytoavailable MeHg or predict grain MeHg levels. Our results provided the possibility of predicting MeHg phytoavailability/bioaccumulation by (NH4)2S2O3 extraction, which could be useful in screening soils for rice cultivation in contaminated areas.

Abstract:An automated method for determination of methylmercury in sediments was developed using headspace coupled to gas chromatography with atomic fluorescence spectrometry detection and aqueous phase ethylation. The method avoids the problems associated with purge and trap techniques. The volume of ethylating agent was optimized and headspace only or headspace trap modes were evaluated. The results showed that the latter mode provided better performance for methylmercury determination than the former. An organic extraction of methylmercury from sediment samples was used involving the use of copper(II) sulfate, potassium bromide, and sulfuric acid. The method was verified by analysis of sediment reference materials (IAEA-405; IAEA-433) and the results were in good agreement with certified values. Due to the low detection limit, the method is appropriate for analysis of contaminated and uncontaminated sediment samples at ultratrace levels.

Abstract:Driven by the significant need for characterization of the chemical speciation of arsenic in food, this work developed a method for rapid determination of four common arsenic species, namely, arsenite, arsenate, monomethyl arsenic acid, and dimethyl arsenic acid, in vegetables using microwave-assisted extraction, followed by detection with HPLC hyphenated to inductively coupled plasma-mass spectrometry. Initial screening results showed that microwave-assisted extraction using 1% HNO3 exhibited the highest overall efficiencies for all arsenic species without causing significant degradation of the organic ones. With the aid of response surface methodology, the optimum conditions established for extraction of arsenic species from vegetables were: 500 mg of freeze-dried vegetable sample, extracted by closed vessel microwave-assisted extraction using 10 mL of 2% (v/v) HNO3 at 90 °C for 17 min. Application of the method in the analysis of 24 market vegetable samples indicates that the extraction efficiencies for total arsenic species were in the range of 91.4-106%. Arsenite and arsenate were found to be the predominant arsenic species in the vegetables, which suggests that vegetable consumption could be an important route of inorganic arsenic exposure for the population with a heavy vegetable diet in arsenic polluted regions. Abstract:In this work, a direct solid sampling device based on modified graphite furnace electrothermal vaporization (GF-ETV) with inductively coupled plasma mass spectrometry (ICP-MS) was established for the simultaneous detection of trace selenium and cadmium in rice samples. A bypass gas was first designed in GF-ETV to improve the device’s analytical sensitivity and precision. The ashing and vaporization conditions, the flow rates of the Ar carrier and the bypass gases of ICP-MS were all investigated. Under the optimized conditions, the limits of detection (LODs) for Se and Cd were 0.5 μg kg−1 and 0.16 μg kg−1, respectively; the relative standard deviations (RSDs) of repeated measurements were within 8% (n = 6). The recoveries of Cd and Se in rice samples were in the range of 89−112% compared with the microwave digestion ICP-MS method, indicating good accuracy and precision for the simultaneous detection of Se and Cd in rice matrix. The whole analysis time is <3 min without the sample digestion process, fulfilling the fast detection of Se and Cd in rice samples to protect food safety.
Source Apportionment of Speciated Mercury in Chinese Rice Grain Using a High-Resolution Model

Abstract:Rice grain consumption is a primary pathway of human mercury exposure. To trace the source of rice grain mercury in China, we developed a rice paddy mercury transport and transformation model with a grid resolution of 1 km x 1 km by using the unit cell mass conservation method. The simulated total mercury (THg) and methylmercury (MeHg) concentrations in Chinese rice grain ranged from 0.08 to 243.6 and 0.03 to 238.6 mu g/ kg, respectively, in 2017. Approximately, 81.3% of the national average rice grain THg concentration was due to atmospheric mercury deposition. However, soil heterogeneity, especially the variation in soil mercury, led to the wide rice grain THg distribution across grids. Approximately, 64.8% of the national average rice grain MeHg concentration was due to soil mercury. In situ methylation was the main pathway via which the rice grain MeHg concentration was increased. The coupled impact of high mercury input and methylation potential led to extremely high rice grain MeHg in partial grids among Guizhou province and junctions with surrounding provinces. The spatial variation in soil organic matter significantly impacted the methylation potential among grids, especially in Northeast China. Based on the high-resolution rice grain THg concentration, we identified 0.72% of grids as heavily polluted THg grids (rice grain THg > 20 mu g/kg). These grids mainly corresponded to areas in which the human activities of nonferrous metal smelting, cement clinker production, and mercury and other metal mining were conducted. Thus, we recommended measures that are targeted at the control of heavy pollution of rice grain by THg according to the pollution sources. In addition, we observed a wide spatial variation range of MeHg to THg ratios not only in China but also in other regions of the world, which highlights the potential risk of rice intake.

Abstract:Human exposure to inorganic arsenic (iAs) via rice consumption is of increasing concern. In the present study, microwave-assisted dispersive liquid-liquid micro-extraction (MADLLME) and electrothermal atomic absorption spectrometry (ETAAS) were developed for the speciation of iAs in rice samples. After microwave-assisted digestion, the As(III) ion reacted with diethyldithiophosphoric acid (DDTP) to form an As-DDTP complex and was extracted at the same time. Some parameters affecting digestion, complex formation, and extraction were studied and optimised. Under the optimised conditions, a detection limit of 0.2 µg kg(-1) with a correlation coefficient of 0.9901 were obtained with a calibration curve in the range of 0.5-200 µg kg(-1). Total iAs was determined after reduction of As(V) to As(III) with sodium thiosulfate and potassium iodide, and As(V) was calculated by difference. The proposed extraction procedure was successfully applied for the determination of iAs ions in certified reference materials (NIST CRM 1568a and NMIJ CRM 7503a) and 10 rice samples produced in Iran and other Asian countries.
Headspace Solid-Phase Microextraction Coupled to Miniaturized Microplasma Optical Emission Spectrometry for Detection of Mercury and Lead.

Abstract:Despite increased interest in microplasma atomic spectrometry over the past several years, its applications to field analysis of real samples still remain limited due to its low sensitivity. In this work, headspace solid-phase microextraction (HS-SPME) using PDMS/DVB fiber was used as a sample introduction method to improve the sensitivity of a miniature (6.0 cm length × 5.0 cm width × 2.8 cm height) low power (<10 W) microplasma point discharge optical emission spectrometer (PD-OES) for the determination of Hg and Pb after their derivatization with sodium tetraethylborate (NaBEt4). Owing to its advantages of combining sampling, separation, and preconcentration of analytes from sample matrices into a single step, HS-SPME not only simplifies sample pretreatment and the experimental setup but also eliminates interferences from matrices and water vapor while significantly improving analytical performance. In comparison to photochemical vapor generation PD-OES, limits of detection for Hg and Pb were enhanced by at least 100-fold, providing great potential for field analysis of these and other trace elements in real samples. Under optimal conditions, the limits of detection for Hg and Pb were 0.001 and 0.003 μg L-1, respectively, with relative standard deviations (RSDs) better than 2.1% and 4.5% at a concentration of 1 μg L-1. The accuracy of the HS-SPME-PD-OES system was validated by analysis of three Certified Reference Materials, including DORM-4 (Hg, 412 ± 36 μg kg-1; Pb, 404 ± 62 μg kg-1), TORT-3 (Hg, 292 ± 22 μg kg-1; Pb, 225 ± 18 μg kg-1) and SRM1568b (Hg, 5.53 ± 0.58 μg kg-1), providing analytical results in excellent agreement with certified values at the 95% confidence level. This method was also successfully used for the analysis of several real rice and water samples with satisfactory results (90-130% recoveries of added spikes).
