Abstract:Two-dimensional (2D) materials are of great significance to the materials community as a result of their high surface area and controllable surface properties. However, controlled preparation of biodegradable 2D structures with biological activity is difficult. In this work we demonstrate that by careful selection of building block structures and assembly conditions it is possible to use crystallization-driven self-assembly (CDSA) to assemble well-defined 2D nanostructures from poly(L-lactide) (PLLA)-based diblock glycopolymers. 1D glyco-cylinders and 2D diamond-shaped glyco-platelets are produced, where the underlying formation mechanism is revealed by dissipative particle dynamics simulations. Furthermore, we demonstrate that assembly of the polymers under mild degradation provides a straightforward route to hollow-cored platelets, a morphology that has previously proven laborious to access. The well-defined sizes and shapes of the glyco-platelets allow us to investigate macrophage activation efficiency and demonstrate clear size and shape effects, pointing toward potential applications in immunology.
Bisepoxide-Jeffamine Microgel Synthesis and Application Toward Heterogeneous Surface Morphology for Differential Neuronal/non-Neuronal Cell Responses in Vitro.

Abstract:Herein, a new non-vinylic type of cationic microgels (MG) was readily prepared from ethylene glycol diglycidyl ether and Jeffamine T-403 in water. The MG was responsive to both temperature and pH, and oxidatively stable as demonstrated by the hydrogen peroxide study. Using glass as a model substrate, its surface was easily imparted with a heterogeneous morphology by simply adsorbing MG dispersed in basic solution. Specifically, the morphology features patches made of a monolayer of connected yet individually recognizable MG. Through in vitro cell studies, we show that a mere change of the extent of surface coverage by such a patchy morphology can strike a balance in promoting adhesion and differentiation of neuron-like PC-12 cells and primary cortical neurons of chick embryo, without soliciting proliferative response from non-neuronal cells of NIH3T3 fibroblast and CTX astrocyte. This simple yet unconventional surface morphology created by MG could be leveraged in the future as an alternative strategy for neural interface engineering.

Abstract:A new biomimetic surface named nano-micro binary polymer brushes is fabricated by large-area bench-top dip-pen nanodisplacement lithography technique. It is composed of gelatin-modified poly(glycidyl methacrylate) nanolines which are spaced by microstripes of poly(N-isopropylacrylamide). Abstract:Hydrogels are of great significance in the field of bioactive delivery. This study designed the self-assembly of gallic acid-loaded nano cellulose crystals/chitosan (NC/CS) hydrogels via Nano bubbles (NBs). NC/CS NBs 4:2 hydrogels improved the mechanical properties compared to those without NBs. The hardness of NC/CS (4:2) NBs hydrogels was greatly enhanced by 1.15 +/- 0.05. The water-holding and swelling behavior can be tuned at different ratios. NC/CS NBs (4:2) showed the electrostatic interaction analyzed by FTIR, XRD, and XPS. SEM results displayed smoother and smaller pores along dense networks promoted by NBs. The antioxidant activity of hydrogels was increased by adding NBs (P < 0.05). In vitro and vivo release activity of gallic acid was higher in simulated intestinal fluid (SIF) at 4:2, depicting the controlled release mechanism. Thus, current work revealed that NBs and low concentrations of NC can be self-assembled with chitosan chains, producing a highly compact hydrogel structure.

Abstract:Engineered nanomaterials have been increasingly utilized in industry for various consumer products, environmental treatments, energy storage, and biomedical applications. Meanwhile, it has been established that certain nanomaterials can be toxic to biological cells from extensive experimental and theoretical studies. Despite that the exact molecular mechanisms of this nanomaterial toxicity are still not well understood, it is ubiquitous that their interactions with cell membranes, through either endocytosis or penetration (and thus potential lysis), act as the first step toward the inflammation or even the death of a cell. To facilitate the study of nanomaterial-membrane interactions, here we demonstrate a nanopore-based single-molecule approach that can be applied to monitor a specific nanomaterial-membrane interaction in real time. Combined with molecular dynamics and experimental approaches, we show how an ionic current can be used to detect membrane damage by a graphene nanosheet and illustrate the underlying molecular mechanism. More generally, we expect that measured transmembrane ionic currents (both DC and AC) can signify many particle-induced membrane modifications, such as hole formation, particle adsorption, and protein insertion.

Abstract:Preparing complex non-spherical assemblies of elongated nanoparticles and exploring their topological conformations is a challenge due to liquid crystals' mobility and elastic distortion. Here, we fabricated a variety of non-spherical liquid crystal assemblies of chitin nanocrystals (ChNCs) in a coagulation bath containing sodium triphosphate (STP) by drop impact assembly method, and the forming mechanism and internal topology were systematically investigated. The collection height, ChNCs concentration, and STP concentration have significant influence on the shape and size of the assembled structures. Long-range ordered structures and long-lived topological textures of the ChNCs liquid crystal can be obtained since a molecular interaction of hydrogen bonding and electrostatic attractions between ChNCs and STP occur during the impact assembly. Rheological and kinetic analysis suggested the shear thinning behavior of the ChNCs liquid crystals and the rapid gelation phenomenon of ChNCs induced by STP. Morphology results showed that the rod-like ChNCs in the non-spherical assemblies were orderly and closely arranged with periodic repetition and layered structure. The non-spherical assemblies of ChNCs liquid crystals can be used as carriers of carbon nanotubes, magnetic Fe<sub>3</sub>O<sub>4</sub> nanoparticles, synthesized polymers, and anticancer drugs for functional composite applications. The drop impact assembly method of ChNCs liquid crystal structure is highly controllable on the composition, morphology, and function, which shows promising applications in energy, environmental-friendly, and bioactive materials.

Abstract:Using nanostructured substrates for enhancing cell adhesion is an important method. However, the physical mechanism of cell adhesion on nanostructured substrate is not well understood yet, particularly the evolution process of membrane during cell adhesion. Here, we propose an analytical model to study quantitatively the evolution of membrane during adhesion process on the nanostructured substrate. The effects of substrate surface topography, receptor-ligand binding, substrate property and membrane property were investigated. We found a small nanopillar radius, low nanopillar density, large binding rate, large receptor density, low non-specific energy, or large stretching modulus can result in a large adhesion rate and a large adhesion depth. Besides, the evolution of membrane on truncated nanocone arrays was also studied by the model. The theoretical results are in good agreement with the experimental observations, which implied that our studies can provide a useful guide to control cell adhesion process and cell motion on nanostructured substrates.

Abstract:A new and easy method of stimuli-triggered growth and removal of a bioreducible nanoshell on nanoparticles is reported. The results show that pH or temperature could induce the aggregation of disulfide-contained branched polymers at the surface of nanoparticles; subsequently, the aggregated polymers could undergo intermolecular disulfide exchange to cross-link the aggregated polymers, forming a bioreducible polymer shell around nanoparticles. When these nanoparticles with a polymer shell are treated with glutathione (GSH) or d,l-dithiothreitol (DTT), the polymer shell could be easily removed from the nanoparticles. The potential application of this method is demonstrated by easily growing and removing a bioreducible shell from liposomes, and improvement of in vivo gene transfection activity of liposomes with a bioreducible PEG shell.
Investigation of MC3T3-E1 cell behavior on the surface of GRGDS-coupled chitosan.

Abstract:The GRGDS (Gly-Arg-Gly-Asp-Ser) peptide has intermediate affinity to alpha(v)beta(3) and alpha(IIb)beta(3), which are the integrins most reported to be involved in bone function. In this study, biomimetic chitosan films modified with GRGDS peptide were prepared and were used as a substrate for the in vitro culture of MC3T3-E1 cells in order to investigate the effect of GRGDS modification on MC3T3-E1 cell behavior. The results of electron spectroscopy for chemical analysis (ESCA), attenuated total reflection-Fourier transform infrared spectra (ATR-FTIR), and amino acid analysis (AAA) demonstrated that the chitosan films were successfully modified with GRGDS peptides and that the surface density of the immobilized GRGDS was on the order of 10(-9) mol/cm(2). The immobilization of the GRGDS sequence on chitosan as well as the peptide concentration play a significant role in MC3T3-E1 cell behavior. MC3T3-E1 cell attachment, proliferation, migration, differentiation, and mineralization were remarkably greater on GRGDS-coupled chitosan than on unmodified chitosan. Besides, the degree of acceleration of these biological processes was found to be dependent on peptide density. Competitive inhibition of MC3T3-E1 cell attachment using soluble GRGDS peptides indicated that the interaction of MC3T3-E1 cells with the surface of the materials was ligand-specific. Cytoskeleton organization in the fully spread MC3T3-E1 cells was highly obvious on GRGDS-coupled chitosan when compared to the lack of actin fibers noted in the round MC3T3-E1 cells on unmodified chitosan. These results suggest that MC3T3-E1 cell function can be modulated. in a peptide density-dependent manner, by the immolization of GRGDS peptide on chitosan used for scaffold-based bone tissue engineering.

Abstract:Shear stress is an important mechanical stimulus that plays a critical role in modulating cell functions. In this study, we investigated the regulating effects of shear stress on the internalization of cell membrane proteins in a microfluidic chip. A hairpin-type DNA probe was developed and indiscriminately anchored to the cell surface, acting as an indicator for the membrane proteins. When cells were exposed to shear stress generated from fluid cell medium containing external proteins, strong fluorescence was emanated from intra-cellular regions. With intensive investigation, results revealed that shear stress could enhance the specific cell endocytosis pathway and promote membrane protein internalization. This process was indicated by the enhanced intracellular fluorescence, generated from the internalized and mitochondria accumulated DNA probes. This study not only uncovered new cellular mechanotransduction mechanisms but also provided a versatile method that enabled in situ and dynamic indication of cell responses to mechanical stimuli.
Generation of a Hydrophobic Protrusion on Nanoparticles to Improve the Membrane‐Anchoring Ability and Cellular Internalization

Abstract:Controlling the nanoparticle-cell membrane interaction to achieve easy and fast membrane anchoring and cellular internalization is of great importance in a variety of biomedical applications. Here we report a simple and versatile strategy to maneuver the nanoparticle-cell membrane interaction by creating a tunable hydrophobic protrusion on Janus particles through swelling-induced symmetry breaking. When the Janus particle contacts cell membrane, the protrusion will induce membrane wrapping, leading the particles to docking to the membrane, followed by drawing the whole particles into the cell. The efficiencies of both membrane anchoring and cellular internalization can be promoted by optimizing the size of the protrusion. In vitro, the Janus particles can quickly anchor to the cell membrane in 1 h and be internalized within 24 h, regardless of the types of cells involved. In vivo, the Janus particles can effectively anchor to the brain and skin tissues to provide a high retention in these tissues after intracerebroventricular, intrahippocampal, or subcutaneous injection. This strategy involving the creation of a hydrophobic protrusion on Janus particles to tune the cell-membrane interaction holds great potential in nanoparticle-based biomedical applications.

Abstract:Anchorage-dependent contact-inhibited growth usually refers to on-surface cell proliferation inhibited by the proximity of other cells. This phenomenon, prominent in nature, has yet to be achieved with polymeric micelles. Here, we report the control living supra-macromolecular self-assembly of elongated micelles with a liquid crystalline core onto a hydrophobic substrate via the synergetic interactions between the substrate and aggregates dispersed in solution. In this system, seed formation is a transient phenomenon induced by the adsorption and rearrangement of the core-swollen aggregates. The seeds then trigger the growth of elongated micelles onto the substrate in a living controllable manner until the contact with the substrate is disrupted. Brownian dynamic simulations show that this unique behavior is due to the fusion of the aggregates onto both ends of the anchored seeds. Abstract:Cells respond to the mechanical signals from their surroundings and integrate physiochemical signals to initiate intricate mechanochemical processes. While many studies indicate that topological features of biomaterials impact cellular behaviors profoundly, little research has focused on the nuclear response to a mechanical force generated by a topological surface. Here, we fabricated a polymeric micropillar array with an appropriate dimension to induce a severe self-deformation of cell nuclei and investigated how the nuclear shape changed over time. Intriguingly, the nuclei of mesenchymal stem cells (MSCs) on the poly(lactide-co-glycolide) (PLGA) micropillars exhibited a significant initial deformation followed by a partial recovery, which led to an "overshoot" phenomenon. The treatment of cytochalasin D suppressed the recovery of nuclei, which indicated the involvement of actin cytoskeleton in regulating the recovery at the second stage of nuclear deformation. Additionally, we found that MSCs exhibited different overshoot extents from their differentiated lineage, osteoblasts. These findings enrich the understanding of the role of the cell nucleus in mechanotransduction. As the first quantitative report on nonmonotonic kinetic process of self-deformation of a cell organelle on biomaterials with unique topological surfaces, this study sheds new insight into cell-biomaterial interactions.
Surface-chemistry effect on cellular response of luminescent plasmonic silver nanoparticles.

Abstract:Cellular response of inorganic nanoparticles (NPs) is strongly dependent on their surface chemistry. By taking advantage of robust single-particle fluorescence and giant Raman enhancements of unique polycrystalline silver NPs (AgNPs), we quantitatively investigated effects of two well-known surface chemistries, passive PEGylation and active c-RGD peptide conjugation, on in vitro behaviors of AgNPs at high temporal and spatial resolution as well as chemical level using fluorescence and Raman microscopy. The results show that specific c-RGD peptide-alpha(v)beta(3) integrin interactions not only induced endosome formation more rapidly, enhanced constrained diffusion, but also minimized nonspecific chemical interactions between the NPs and intracellular biomolecules than passive PEGylation chemistry; as a result, surface enhanced Raman scattering (SERS) signals of c-RGD peptides were well resolved inside endosomes in the live cells, while Raman signals of PEGylated AgNPs remained unresolvable due to interference of surrounding biomolecules, opening up an opportunity to investigate specific ligand receptor interactions in real time at the chemical level.

We have observed a substantial impact of carbon nanotube on the arrangement of the cholesterol domain.
