Abstract:A new biomimetic surface named nano-micro binary polymer brushes is fabricated by large-area bench-top dip-pen nanodisplacement lithography technique. It is composed of gelatin-modified poly(glycidyl methacrylate) nanolines which are spaced by microstripes of poly(N-isopropylacrylamide). Cells are not only adhered and oriented well on the re-used surface, but also detachable from the surface with well-preserved extracellular matrix and aligned morphology.

Abstract:The intensive concern over the biosafety of nanomaterials demands the systematic study of the mechanisms underlying their biological effects. Many of the effects of nanomaterials can be attributed to their interactions with proteins and their impacts on protein function. On the other hand, nanomaterials show potential for a variety of biomedical applications, many of which also involve direct interactions with proteins. In this paper, we review some recent computational studies on this subject, especially those investigating the interactions of carbon and gold nanomaterials. Beside hydrophobic and pi-stacking interactions, the mode of interaction of carbon nanomaterials can also be regulated by their functional groups. The coatings of gold nanomaterials similarly adjust their mode of interaction, in addition to coordination interactions with the sulfur groups of cysteine residues and the imidazole groups of histidine residues. Nanomaterials can interact with multiple proteins and their impacts on protein activity are attributed to a wide spectrum of mechanisms. These findings on the mechanisms of nanomaterial-protein interactions can further guide the design and development of nanomaterials to realize their application in disease diagnosis and treatment.

Abstract:TiO2, Al2O3 and Au were used to modify the microstructured surfaces constructed by SiO2 microspheres, respectively. The protein adsorption capacity and cell response of the chemically modified surfaces were investigated. The results indicate that the protein adsorption is related to the chemical composition of the surface, the initial adhesion of the cells is closely related to the protein adsorption capacity, and the amount and orientation of protein adsorption both are considered as important factors affecting the cell proliferation.

Abstract:Regarding the importance of the biological effects of nanomaterials, there is still limited knowledge about the binding structure and stability of the protein corona on nanomaterials and the subsequent impacts. Here we designed a hard serum albumin protein corona (BSA) on CTAB-coated gold nanorods (AuNRs) and captured the structure of protein adsorption using synchrotron radiation X-ray absorption spectroscopy, microbeam X-ray fluorescent spectroscopy, and circular dichroism in combination with molecular dynamics simulations. The protein adsorption is attributed to at least 12 Au-S bonds and the stable corona reduced the cytotoxicity of CTAB/AuNRs. These combined strategies using physical, chemical, and biological approaches will improve our understanding of the protective effects of protein coronas against the toxicity of nanomaterials. These findings have shed light on a new strategy for studying interactions between proteins and nanomaterials, and this information will help further guide the rational design of nanomaterials for safe and effective biomedical applications.

Abstract:<p>Mesoporous silica nanoparticles (MSNs), one of the important porous materials, have garnered interest owing to their highly attractive physicochemical features and advantageous morphological attributes. They are of particular importance for use in diverse fields including, but not limited to, adsorption, catalysis, and medicine. Despite their intrinsic stable siliceous frameworks, excellent mechanical strength, and optimal morphological attributes, pristine MSNs suffer from poor drug loading efficiency, as well as compatibility and degradability issues for therapeutic, diagnostic, and tissue engineering purposes. Collectively, the desirable and beneficial properties of MSNs have been harnessed by modifying the surface of the siliceous frameworks through incorporating supramolecular assemblies and various metal species, and through incorporating supramolecular assemblies and various metal species and their conjugates. Substantial advancements of these innovative colloidal inorganic nanocontainers drive researchers in promoting them toward innovative applications like stimuli (light/ultrasound/magnetic)‐responsive delivery‐associated therapies with exceptional performance in vivo. Here, a brief overview of the fabrication of siliceous frameworks, along with discussions on the significant advances in engineering of MSNs, is provided. The scope of the advancement in terms of structural and physicochemical attributes and their effects on biomedical applications with a particular focus on recent studies is emphasized. Finally, interesting perspectives are recapitulated, along with the scope toward clinical translation.</p>

Abstract:Micro/nano structured surface has been shown to effectively enhance the biological performance of titanium alloy implants, exhibiting faster postoperative recovery. However, a common drawback of bio-functionalized surfaces featuring structured characteristics is their susceptibility to wear. This study proposed a novel micro/nano surface modification method using ultrasonic milling and anodization to enhance both biocompatibility and wear resistance of the implant surface. First, hierarchical structures are fabricated on the titanium surface by utilizing ultrasonic milling and anodization to generate micro-scale and nano-scale structures, respectively. Linear reciprocating tests, contact angle tests, and surface morphology observation were conducted to investigate the surface properties. Then, cell proliferative and adhesive abilities were tested on different modified surfaces by using mouse osteoblasts. Finally, nonosilver coatings were created on the structured surface by vapor deposition and the antibacterial performance was tested using staphylococcus aureus. The experiment results reveal a significant improvement in wear resistance, biological properties, and antibacterial capabilities of the processed surface, demonstrating the great application potential of the proposed method for implant surface modification.

Tailored Nano-Pillar Structures on Surfaces: Facile Formation and Multifunctional Properties

Abstract:Nano-pillar structured surfaces have prized for exceptional optoelectronic properties and biocompatibility but commonly complex, expensive, and inflexible in production. A systematic investigation of the formation, mechanisms, and properties of nanostructures induced by nano-capillaries remains unexplored. Through this study, the facile formation of nano-pillared surfaces induced by designable nano-capillaries on AAO template demonstrated significant improvements in antibacterial effect, spectral structural color and anti-reflection characteristics. Reverse molding of nano-pillar arrays with aspect ratios of up to 1.2 on polydimethylsiloxane (PDMS) films yielded a super-hydrophobic surface of 150° contact angle (CA). Through the utilization of computational fluid mechanics model, a direct correlation was demonstrated between the aspect ratio of nano-pillars and the viscosity of PDMS. The nano-pillared surfaces had remarkable antibacterial rates of 91 % for Escherichia coli and 89 % for Staphylococcus aureus. In addition, the structured surfaces exhibited color spanning from light blue to light red by tuning nano-pillars dimensions, incident light angle, and/or adding reflective metallic coating, which offered unique visualization effects for anti-counterfeiting. Moreover, the superior light-trapping and anti-reflection of the nano-pillared surface significantly increased the power conversion efficiency (PCE) 19 % from the base performance of the polysilicon solar cells.

Abstract:Precisely controlling the protein-nanomaterial interactions at selective sites is crucial in engineering biomolecule composite architectures with tailored nanostructures and functions for a variety of biomedical applications. This strategy, however, is only beginning to be explored. Here, we demonstrate the facet-specific assembly of proteins, such as albumin, immunoglobulin and protamine, on {100} facets of SrTiO 3 polyhedral nanocrystals, while none on {110} facets. Molecular dynamics simulations indicate the immobile surface hydration layer might play a barrier role to effectively prevent proteins adsorption on specific {110} facets. This work thus provides new insights into the fundamentally understanding of protein-nanomaterial interactions and open a novel, general and facile route to control the selective adsorption of various proteins on various nanocrystals.
'Nano on micro' hierarchical porous all carbon structures: an insight into interfacial interactions with bacteria

Abstract:Micron long carbon nanofibers (CNFs) were grown on porous carbon beads to give an active surface for rapid immobilization of guest molecules. The fabrication of nanostructures using a catalytic route involving chemical vapour deposition on a porous substrate was accomplished by the controlled synthesis of iron nanoclusters on the surface of porous carbon beads. The challenge of catalyst nanoparticle diffusion into the porous substrate was addressed by using iron coordinated ligand complexes and optimizing the loading percentage of metal salts onto beads. The effect of using tethered bottom up surface processed CNFs on the porous beads' morphologies was established using structural characterization. The protruding architecture of CNFs on the porous carbon surface was subjected to bacterial colonisation in order to determine the efficiency of cell conjugation onto hairy structures, particularly at a low concentration. The interfaces of immobilized bacteria on the textured surface were studied by varying the pH and external physical stimuli to check the biofilm formation. The strategy of fabricating all carbon porous beads, which had topologically controlled 'nano on micro' geometries, to give fast immobilization of guest molecules could be useful in the future for developing an active disinfectant surface.
Carbon Nanoscroll-Silk Crystallite Hybrid Structures with Controllable Hydration and Mechanical Properties

Abstract:Hybrid structures of nanomaterials (e.g. tubes, scrolls, threads, cages) and biomaterials (e.g. proteins) hold tremendous potential for applications as drug carriers, biosensors, tissue scaffolds, cancer therapeutic agents, etc. However, in many cases, the interacting forces at the nano-bio interfaces and their roles in controlling the structures and dynamics of nano-bio-hybrid systems are very complicated but poorly understood. In this study, we investigate the structure and mechanical behavior of a protein-based hybrid structure, i.e., a carbon nanoscroll (CNS)-silk crystallite with a hydration level controllable by an interlayer interaction in CNS. Our findings demonstrate that CNS with a reduced core size not only shields the crystallite from a weakening effect of water, but also markedly strengthens the crystallite. Besides water shielding, the enhanced strength arises from an enhanced interaction between the crystallite and CNS due to the enhanced interlayer interaction in CNS. In addition, the interfacial strength for pulling the crystallite out of the CNS-silk structure is found to be dependent on both the interlayer interaction energy in CNS as well as the sequence of protein at the CNS-silk interface. The present study is of significant value in designing drugs or protein delivery vehicles for biomedical applications, and serves as a general guide in designing novel devices based on rolled-up configurations of two-dimensional (2D) materials.
Enhancing the Biological Performance of Titanium Alloy through In Situ Modulation of the Surface Nanostructure: Near-Infrared-Responsive Antibacterial Function and Osteoinductivity

Abstract:The poor clinical performance of titanium and its alloy implants is mainly attributed to their lack of antibacterial ability and poor osseointegration. The key and challenge lie in how to enhance their osteoinductivity while imparting antibacterial capability. In this study, a titanium oxide metasurface with light-responsive behavior was constructed on the surface of titanium alloy using an alkaline-acid bidirectional hydrothermal method. The effects of the acid type, acid concentration, hydrothermal time, hydrothermal temperature, and subsequent heat treatments on the optical behavior of the metasurface were systematically investigated with a focus on exploring the influence of the metasurface and photodynamic reaction on the osteogenic activity of osteoblasts. Results show that the type of acid and heat treatment significantly affect the light absorption of the titanium alloy surface, with HCl and post-heat-treatment favoring redshift in the light absorption. Under 808 nm near-infrared (NIR) irradiation for 10 min, in vitro antibacterial experiments demonstrate that the antibacterial rate of the metasurface titanium alloy against Staphylococcus aureus and Escherichia coli were 96.87% and 99.27%, respectively. In vitro cell experiments demonstrate that the nanostructure facilitates cell adhesion, proliferation, differentiation, and expression of osteogenic-related genes. Surprisingly, the nanostructure promoted the expression of relevant osteogenic genes of MC3T3-E1 under 808 nm NIR irradiation. This study provides a method for the surface modification of titanium alloy implants.

Abstract:Immune rejection, poor biocompatibility and cytotoxicity have seriously stalled the widespread application of biometallic materials. To overcome these problems, biometallic materials with fast and sufficient osseointegration, antibacterial properties and long-term stability have attracted the attention of researchers worldwide. Surface modification is currently used as a general strategy to develop material coatings that will overcome these challenging requirements and achieve the successful performance of implants. In this study, we proposed a substrate surface-modification strategy based on biofilm CsgA proteins that promote rapid cell attachment, proliferation, and stabilization of the cytoskeleton. CsgA-based nano-coating is easy to fabricate and has superior performance, which is expected to expand the application of medical implants.

Abstract:We demonstrate that hierarchical nanostructured metal sub-microtubes with evenly distributed nanoscale pores on sidewalls can be synthesized though electrodeposition of metals on electrospun fiber templates and subsequent wet etching. Due to the hierarchical nanostructure and uniform "hot spots" on sidewalls, these porous sub-microtubes exhibit higher surface-enhanced Raman scattering (SERS) activities than both smooth metal sub-microtubes and nanoporous thin films. The synthetic process is simple, inexpensive, and effective, and therefore is a suitable methodology for large-scale production of reliable and reproducible SERS substrates.

Abstract:The topography at the micro/nanoscale level for biomaterial surfaces has been thought to play vital roles in their interactions with cells. However, discovering the interdisciplinary mechanisms underlying how cells respond to micro-nanostructured topography features still remains a challenge. In this work, ∼37 μm 3D printing used titanium microspheres and their further hierarchical micro-nanostructured spheres through hydrothermal treatment were adopted to construct typical model surface topographies to study the preosteoblastic cell responses (adhesion, proliferation, and differentiation). We here demonstrated that not only the hierarchical micro-nanostructured surface topography but also their distribution density played critical role on cell cytocompatibility. The microstructured topography feature surface with middle-density distributed titanium microspheres showed significantly enhanced cell responses, which might be attributed to the better cellular interaction due to the cell aggregates. However, the hierarchical micro-nanostructured topography surface, regardless of the distribution density of titanium microspheres, improved the cell-surface interactions because of the enhanced initial protein adsorption, thereby reducing the cell aggregates and consequently their responses. This work, therefore, provides new insights into the fundamental understanding of cell-material interactions and will have a profound impact on further designing micro-nanostructured topography surfaces to control cell responses.
Enhanced initial protein adsorption on an engineered nanostructured cubic zirconia

Abstract:Motivated by experimentally observed biocompatibility enhancement of nanoengineered cubic zirconia ZrO2 coatings to mesenchymal stromal cells, we have carried out computational analysis of the initial immobilization of one of known structural fragment of the adhesive protein (fibronectin) on the corresponding surface. We constructed an atomistic model of the zirconia nano-hillock of 3-fold symmetry based on AFM and TEM images. First-principle quantum-mechanical calculations show a substantial variation of electrostatic potential at the hillock due to the presence of surface features such as edges and vertexes. Using an implemented Monte Carlo simulated annealing method we found the orientation of the immobilized protein on the zirconia surface (both flat and nanostructured) and contribution of the each amino acid residue from the protein sequence to the adsorption energy. Accounting for the variation of the dielectric permittivity at the protein-implant interface we use a model distance-dependent dielectric function to describe the inter-atom electrostatic (Coulomb) interactions in the adsorption potential. We find that the initial immobilization of the rigid protein fragment on the nanostructured pyramidal ZrO2 surface is achieved with magnitude of adsorption energy larger than the one for the protein on the smooth (flat) surface. The strong attractive electrostatic interactions are a major factor in the enhanced adsorption at nanostructured surface that is absent in the case of adsorption on the flat uncharged surface. We show that the best electrostatic and steric fit of the protein to the inorganic surface corresponds to a minimum of the adsorption energy determined by the non-covalent interactions.

Abstract:Surface chemistry is essential for the biomedical applications of functional nanomaterials. Here, a supramolecular container-based surface engineering approach is designed to impart excellent water dispersibility and precisely control the orientation of surface targeting ligands of the nanoparticles. An acyclic cucurbituril (aCB) molecular container is used as a chemical bridge to incorporate nanoparticles and targeting ligands via a bilateral host-guest complexation, enabling the bioactive moieties of targeting ligands to be fully exposed and faced outward to facilitate biological targeting. The enhanced biological targeting effect as well as targeted imaging performance of aCB-engineered nanoparticles are demonstrated in vitro and in vivo. Molecular dynamic simulations illustrate a tight binding of targeting ligand to the relevant receptor with the assistance of the aCB molecular container for the enhanced targeting efficiency, representing an attractive extension of supramolecular chemistry-based technology for nanoparticle surface engineering and supramolecularly regulated biological targeting.
Biological and Mechanical Effects of Micro-Nanostructured Titanium Surface on an Osteoblastic Cell Line In vitro and Osteointegration In vivo

Abstract:Hybrid micro-nanostructure implant surface was produced on titanium (Ti) surface by acid etching and anodic oxidation to improve the biological and mechanical properties. The biological properties of the micro-nanostructure were investigated by simulated body fluid (SBF) soaking test and MC3T3-E1 cell co-culture experiment. The cell proliferation, spreading, and bone sialoprotein (BSP) gene expression were examined by MTT, SEM, and reverse transcription-polymerase chain reaction (RT-PCR), respectively. In addition, the mechanical properties were evaluated by instrumented nanoindentation test and friction-wear test. Furthermore, the effect of the micro-nanostructure surface on implant osteointegration was examined by in vivo experiment. The results showed that the formation of bone-like apatite was accelerated on the micro-nanostructured Ti surface after immersion in simulated body fluid, and the proliferation, spreading, and BSP gene expression of the MC3T3-E1 cells were also upregulated on the modified surface. The micro-nanostructured Ti surface displayed decreased friction coefficient, stiffness value, and Young's modulus which were much closer to those of the cortical bone, compared to the polished Ti surface. This suggested much better mechanical match to the surrounding bone tissue of the micro-nanostructured Ti surface. Furthermore, the in vivo animal experiment showed that after implantation in the rat femora, the micro-nanostructure surface displayed higher bonding strength between bone tissues and implant; hematoxylin and eosin (H&E) staining suggested that much compact osteoid tissue was observed at the interface of Micro-nano-Ti-bone than polished Ti-bone interface after implantation. Based on these results mentioned above, it was concluded that the improved biological and mechanical properties of the micro-nanostructure endowed Ti surface with good biocompatibility and better osteointegration, implying the enlarged application of the micro-nanostructure surface Ti implants in future.
Biological self-protection inspired engineering of nanomaterials to construct a robust bio-nano system for environmental applications

Abstract:Nanomaterials can empower microbial-based chemical production or pollutant removal, e.g., nano zero-valent iron (nZVI) as an electron source to enhance microbial reducing pollutants. Constructing bio-nano interfaces is critical for bio-nano system operation, but low interfacial compatibility due to nanotoxicity challenges the system performance. Inspired by microorganisms' resistance to nanotoxicity by secreting extracellular polymeric substances (EPS), which can act as electron shuttling media, we design a highly compatible bio-nano interface by modifying nZVI with EPS, markedly improving the performance of a bio-nano system consisting of nZVI and bacteria. EPS modification reduced membrane damage and oxidative stress induced by nZVI. Moreover, EPS alleviated nZVI agglomeration and probably reduced bacterial rejection of nZVI by wrapping camouflage, contributing to the bio-nano interface formation, thereby facilitating nZVI to provide electrons for bacterial reducing pollutant via membrane-anchoring cytochrome c. This work provides a strategy for designing a highly biocompatible interface to construct robust and efficient bio-nano systems for environmental implication.
