Abstract:BackgroundNear-infrared II (NIR-II, 900-1700 nm) fluorescence bioimaging with advantages of good biosafety, excellent spatial resolution, high sensitivity and contrast, has attracted great attentions in biomedical research fields. However, most nanoprobes used for NIR-II fluorescence imaging have poor tumor-targeting ability and therapeutic efficiency. To overcome these limitations, a novel NIR-II-emissive theranostic nanoplatform for imaging and treatment of cervical cancer was designed and prepared. The NIR-II-emissive dye IR-783 and chemotherapy drug doxorubicin (DOX) were encapsulated into liposomes, and the tumor-targeting peptide TMTP1 was conjugated to the surface of the liposomes to form IR-783-DOX-TMTP1 nanoparticles (NPs) via self-assembly methods.ResultsThe IR-783-DOX-TMTP1 NPs showed strong NIR-II emission, excellent biocompatibility, a long lifetime, and low toxicity. Further, high-definition NIR-II fluorescence microscopy images of ear blood vessels and intratumor blood vessels were obtained from IR-783-DOX-TMTP1 NPs-stained mice with high spatial resolution under 808 nm laser excitation. Moreover, IR-783-DOX-TMTP1 NPs showed strong tumor targeting ability and high efficiently chemotherapeutic character towards cervical tumors. ConclusionsThe novel targeting and NIR-II-emissive IR-783-DOX-TMTP1 NPs have potential in diagnosis and therapy for cervical cancer.

Abstract:A Pt prodrug polyphenol and gadolinium ion loaded cancer theranostics nanoplatform based on a mild acidic pH and thermal sensitive polymer was designed for photoacoustic (PA)/magnetic resonance (MR)/positron emission tomography (PET) multimodal imaging-guided chemo-photothermal combination therapy. The Pt drug release can be controlled by tumour-specific acidic pH and heat generated by external NIR irradiation. The nanoparticles were stable under normal physiological environments and released the drug under the acidic pH of tumours and NIR laser irradiation, which can reduce the side effects of the drug to normal organs. Moreover, the MR signal can be significantly enhanced (approximate to 3-fold increase in T-1 relaxivity) under the acidic tumour microenvironment, which is favorable for cancer diagnosis. The nanoparticles exhibited excellent tumour accumulation and led to complete tumour eradication with low power NIR laser irradiation. This promising approach provides a new avenue for imaging-guided combination therapy.

Abstract:A series of functional glycopolymer nanoparticles with 1,8‐naphthalimide motif was designed, synthesized and applied for tumor cell imaging. With the pH‐sensitive and aggregation‐induced emission (AIE) effect of the 1,8‐naphthalimide fluorescent probe, the presence of glucose‐based glycopolymers enhanced its water‐solubility and biocompatibility. Owing to the dual tumor‐targeting effects of the dense glucose part and the boronic ester modification, the obtained glycopolymers showed high affinity to tumor cells, with a much faster staining rate than normal cells, indicating a great potential for diagnosis and treatments of cancers.

ATernary Conjugated Polymer for NIR-II Fluorescence Imaging and Photothermal/Photodynamic Combination Therapy

Abstract:In order to simultaneously achieve photothermal properties, photodynamic effects and second near-infrared (NIR-II 1000-1700 nm) fluorescence emission of conjugated polymers, herein a novel ternary conjugated polymer (BDP) was designed and synthesized by copolymerization of an electron-withdrawing monomer diketopyrrolopyrrole and two electron-donating monomers (benzodithiophene derivatives and 1,10-phenanthroline). Then water-soluble nanoparticles (BDP NPs) were further prepared by self-assembling with amphiphilic Pluronic F127 through nanoprecipitation method. The nanoparticles with a diameter of around 90 nm exhibited excellent stability in aqueous solution. The absorption and emission peaks of BDP NPs were determined to be 735 nm and 1039 nm, respectively. The mass extinction coefficient of the nanoparticles was as high as 4.67 mL/(mg center dot cm) the fluorescence quantum yield of BDP NPs was measured to be 0.62% by using IR-26 as a reference, and the photothermal conversion efficiency was 18.2%. Under 730 nm laser irradiation, the temperature of BDP NPs (150 mu g/mL) can reach 64 degrees C, which was high enough for photothermal therapy. Besides, the BDP NPs also produce reactive oxygen species with the same laser, which can be used for photodynamic therapy. Cell experiments confirmed that there was no obvious dark toxicity to both NIH3T3 cells and HeLa cells. While under NIR laser irradiation, strong phototoxicity toward cancer cells was observed, indicating the high therapeutic efficacy with the combination therapy. After intravenous injection of BDP NPs, the blood vessels in living mice can be observed which confirmed the high resolution of NIR-II imaging. Importantly, the BDP NPs can accumulate at the tumor sites at 12 h post-injection, indicating the possibility of NIR-II imaging guided combination therapy in vivo. Overall the obtained BDP NPs with high biocompatibility strong absorption in the first near-infrared (NIR-I, 650- 900 nm) range, intense emission in NIR-II region, and the ability to produce reactive oxygen species and hyperthermia were, excellent candidate for tumor diagnosis and phototherapy. In addition, this work also provides a promising method for the rational design of conjugated polymer-based high performance phototheranostic nanoplatforms. [GRAPHICS] .

Abstract:We report here the soft nanomaterial-based targeting polymersomes for near-infrared (NIR) fluorescence imaging to carry out in vivo tumor detection. Two polymersome-based NIR fluorescent probes were prepared through the self-assembly of amphiphilic block copolymers, poly(butadiene-b-ethylene oxide) (PEO-b-PBD). Each of them was encapsulated with distinct hydrophobic near-infrared dyes (DiD and DiR) and modified with different targeting ligands (anti-CEA antibody and anti-EGFR antibody), respectively. After simultaneous injection of these two probes into the tumor-bearing mice via tail vein, multispectral near-infrared fluorescence images were obtained. The results indicate that both probes are successfully directed to the tumor foci, where two distinguishable fluorescent signals were detected through the unmixed fluorescence images. By taking advantage of two targeting polymersome-based probes with distinct fluorescent features, the proposed multispectral near-infrared fluorescence imaging method can greatly improve the specificity and accuracy for in vivo tumor detection.
Ultrasmall Conjugated Polymer Nanoparticles with High Specificity for Targeted Cancer Cell Imaging

Abstract:Fluorescent and biocompatible organic nanoparticles have attracted great interest in cancer detection and imaging, but the nonspecific cellular uptake has limited the detection specificity and sensitivity. Herein, the authors report the ultrasmall conjugated polymer nanoparticles (CPNs) with bright far‐red/near‐infrared emission for targeted cancer imaging with high specificity. The sizes of the ultrasmall CPNs are around 6 nm (CPN6), while large CPNs show sizes around 30 nm (CPN30). Moreover, CPN6 exhibits largely improved fluorescence quantum yield (η) of 41% than CPN30 (25%). Benefiting from the ultrasmall size, bare CPN6 shows largely suppressed nonspecific cellular uptake as compared to CPN30, while cyclic arginine‐glycine‐aspartic acid (cRGD) functionalized CPN6 (cRGD‐CPN6) possesses excellent selectivity toward αvβ3 integrin overexpressed MDA‐MB‐231 cells over other cells in cell mixtures. The faster body clearance of CPN6 over CPN30 indicates its greater potentials as a noninvasive nanoprobe for in vivo and practical applications.
Nanoparticles destabilizing the cell membranes triggered by NIR light for cancer imaging and photo-immunotherapy

Abstract:Cationic polymers have great potential for cancer therapy due to their unique interactions with cancer cells. However, their clinical application remains limited by their high toxicity. Here we show a cell membrane-targeting cationic polymer with antineoplastic activity (Pmt) and a second near-infrared (NIR-II) fluorescent biodegradable polymer with photosensitizer Bodipy units and reactive oxygen species (ROS) responsive thioketal bonds (PBodipy). Subsequently, these two polymers can self-assemble into antineoplastic nanoparticles (denoted mt-NPBodipy) which could further accumulate at the tumor and destroy cell membranes through electrostatic interactions, resulting in cell membrane destabilization. Meanwhile, the photosensitizer Bodipy produces ROS to induce damage to cell membranes, proteins, and DNAs to kill cancer cells concertedly, finally resulting in cell membrane lysis and cancer cell death. This work highlights the use of near-infrared light to spatially and temporarily control cationic polymers for photodynamic therapy, photo-immunotherapy, and NIR-II fluorescence for bio-imaging.
Hydrophilic Polymer-Brush Functional Fluorescent Nanoparticles to Suppress Nonspecific Interactions and Enhance Quantum Yield for Targeted Cell Imaging

Abstract:Hydrophilic poly(glyceryl monomethacrylate) (PGMMA) brush-functional nanoparticles were prepared via macromolecular chain-transfer-agent-adjusted reversible addition-fragmentation chain-transfer precipitation polymerization (RAFTPP). Hydroxyethyl anthrancene-9-carboxylate methacrylate (AnHEMA) was used as the comonomer to label the nanoparticles with fluorescence. Then, small molecular phenylboronic acid tags were modified on the nanoparticles for highly specific and sensitive sensing of glycans on living cells. The introduction of hydrophilic PGMMA brushes into nanoparticles can strongly improve their surface hydrophilicity, reduce their nonspecific interactions with biomolecules in aqueous medium, and give them strong fluorescence properties. Compared with fluorescent nanoparticles without grafted PGMMA brushes, grafted fluorescent nanoparticles showed an obviously enhanced fluorescence quantum yield (similar to 7%), longer lifetime (similar to 2 ns), and higher fluorescence stability due to the shielding effect of PGMMA. Using PGMMA-functional fluorescence nanoparticles as probes could effectively visualize and discriminate cancer cells from normal cells in a highly selective and nonviral manner. Fluorescence imaging of human hepatoma carcinoma cells (HepG-2) over normal hepatic cells (L-02) and of human cervical cancer cells (HeLa cells) over normal human cervical epithelial cells (HCerEpiC) using these nanoparticles was also demonstrated. This strategy demonstrates a flexible molecular brush approach for a highly efficient probe in advanced bioimaging applications.

Photodiagnosis and Photodynamic Therapy

Abstract:A novel aromatic-imide-based thermally activated delayed fluorescence material nano-micelle (TADF-NM) compound was prepared after being encapsulated with DSPE-mPEG2000 amphiphilic copolymers. TADF-NM had the preferential characteristics such as good biocompatibility, non-toxic and targeted ability, more importantly, TADF-NM efficiently isolated oxygen to enhance the fluorescence lifetime, and prevented quenching for using time-resolved fluorescence imaging (TRFI) in tumor cells. The fluorescence lifetime of TADF-NM was about 212 μs in PBS and 112 μs in tumor cells, which was the longest in fluorescence lifetime images based on TADF materials. These studies have shown that TADF-NM have excellent potential ability to overcome the interference of auto-fluorescence for being applied in confocal fluorescence tumor imaging.

Abstract:The advantages of fluorescence bioimaging in the second near-infrared (NIR II, 1000-1700 nm) window are well known; however, current NIR II fluorescent probes for in vivo tumor imaging still have many shortcomings, such as low fluorescence efficiency, unstable performance under in vivo environments, and inefficient enrichment at tumor sites. In this study, Ag2Te quantum dots (QDs) that emit light at a wavelength of 1300 nm are assembled with poly(lactic-co-glycolic acid) and further encapsulated within cancer cell membranes to overcome the shortcomings mentioned above. The as-prepared approximate to 100 nm biomimetic nanobioprobes exhibit ultrabright (approximate to 60 times greater than that of free Ag2Te QDs) and highly stable (approximate to 97% maintenance after laser radiation for 1 h) fluorescence in the NIR II window. By combining the active homotypic tumor targeting capability derived from the source cell membrane with the passive enhanced permeation and retention effect, improved accumulation at tumor sites ((31 +/- 2)% injection dose per gram of tumor) and a high tumor-to-normal tissue ratio (13.3 +/- 0.7) are achieved. In summary, a new biomimetic NIR II fluorescent nanobioprobe with ultrabright and stable fluorescence, homotypic targeting and good biocompatibility for enhanced in vivo tumor imaging is developed in this study.
Cancer Cell Membrane-Biomimetic Nanoprobes with Two-Photon Excitation and Near-Infrared Emission for Intravital Tumor Fluorescence Imaging

Abstract:Biomimetic fluorescent nanoprobes capable of emitting near-infrared (NIR) fluorescence (λmax ≈ 720 nm) upon excitation of 800 nm light were developed. The key conjugated polymer enabled two-photon absorption and Förster resonance energy transfer (FRET) processes within the nanoprobes, which imparted the nanoprobes with ideal NIR-incoming-NIR-outgoing fluorescence features. The cancer cell membrane (CM) coating endowed these nanoprobes with perfect biocompatibility and highly specific targeting ability to homologous tumors. It was believed that CM encapsulation provided an additional protecting layer for the photoactive components residing in the core of nanoprobes for retaining their intrinsic fluorescing ability in the physiological milieu. The long-term structural integrity, excellent photostability (fluorescence decrease <10% upon 30 min illumination of 800 nm pulse laser), high NIR fluorescence quantum yield (∼20%), and long in vivo circulation time of the target nanoprobes were also confirmed. The ability of these feature-packed nanoprobes for circumventing the challenges of absorption and light scattering caused by cellular structures and tissues was definitely confirmed via in vivo and in vitro experiments. The superior performances of these nanoprobes in terms of fluorescence signaling as well as targeting specificity were verified in intravital fluorescence imaging on tumor-bearing model mice. Specifically, these nanoprobes unequivocally enabled high-resolution visualization of the fine heterogeneous architectures of intravital tumor tissue, which proclaims the great potential of this type of probe for high-contrast fluorescence detection of thick biological samples in practical applications.

Abstract:Fluorescence imaging in the second near-infrared window (NIR-II) holds promise for real-time deep tissue imaging. In this work, we design a low-band gap conjugated polymers(pTB) with donor-acceptor (D-A) structures by Stille cross-coupling reaction, which can improve the quality of fluorescence imaging and effectiveness of photothermal therapy. In order to improve the water solubility and biocompatibility of pTB, we adopted nano-deposition technology to envelop the hydrophobic polymer p113 into amphiphilic copolymer (1,2-distearoyl-phosphatidylethanolamine-methyl-polyethyleneglycol conjugate (DSPE-mPEG)) shells for NIR-II water-soluble nanoparticles (p113-PEG). The structure, properties and morphology of the polymer were analyzed by 1H-NMR, UV-Vis, dynamic light scattering (DLS) and transmission electron microscopy (1EM). The results show that the conjugated polymer nanoparticles have not only a good absorption peak at 831 nm, but also a large Stokes shift of 206 nm. In addition, the average hydrodynamic radius of pTB-PEG NPs was around 69 nm and spherical morphology was observed from TEM. And the nanoparticles also possess excellent photostability and good biocompatibility in physiological environment such as phosphate buffer saline (PBS), Dulbecco's Modified Eagle Medium (DMEM) and fetal bovine serum (FBS), indicating the potential for further in vivo application. More importantly, we used MTT assay to analyze photothermal treatment toward human breast cancer (4T1) cells in vifro, and confocal laser scanning microscopy (CLSM) tests further indicated that this material has a good photothermal therapeutic effect. To study the NIR-II fluorescence characters, we firstly detected the maximum imaging depth of pTB-PEG NPs in vifro. With the help of long-wavelength emission (>1000 nm), the imaging depth is 6 mm, far exceeding the traditional near-infrared region (NIR-I) reagents. Under 808 nm laser irradiation, the high-resolution second near-infrared window (NIR-II) fluorescence imaging of healthy mouses' blood vascular system and tumor-bearing mice was effectively achieved. In general, conjugated polymer nanoparticles (pTB-PEG) have a good biocompatibility, excellent light stability and good optical properties. It is a promising NIR-II imaging probe with a wide range of the potential for clinical imaging and live tumor imaging.
Delicately Designed Cancer Cell Membrane-Camouflaged Nanoparticles for Targeted 19 F MR/PA/FL Imaging-Guided Photothermal Therapy

Abstract:Our exploration of multimodal nanoprobes aims to combine photoacoustic (PA) imaging, 19F magnetic resonance (MR), and fluorescence (FL) imaging, which offers complementary advantages such as high spatial resolution, unlimited penetration, and high sensitivity to enable more refined images for accurate tumor diagnoses. In this research, perfluorocarbons (PFCs) and indocyanine green (ICG) are encapsulated by poly(lactic-co-glycolic acid) (PLGA) for intravital 19F MR/FL/PA tri-modal imaging-guided photothermal therapy. Then, it is coated with an A549 cancer cell membrane (AM) to fabricate versatile theranostic nanoprobes (). After systemic administration, FLI reveals time-dependent tumor homing of NPs with high sensitivity, 19F MRI provides tumor localization of NPs without background signal interference, and PAI illustrates the detailed distribution of NPs inside the tumor with high spatial resolution. What is more, accumulated in the tumor area exhibit a prominent photothermal effect (48.4 °C) under near infrared (NIR) laser irradiation and realize an enhanced antitumor response in vivo. Abstract:Abstract Purely organic room-temperature phosphorescence has attracted attention for bioimaging but can be quenched in aqueous systems. Here we report a water-soluble ultralong organic room-temperature phosphorescent supramolecular polymer by combining cucurbit[n]uril (CB[7], CB[8]) and hyaluronic acid (HA) as a tumor-targeting ligand conjugated to a 4-(4-bromophenyl)pyridin-1-ium bromide (BrBP) phosphor. The result shows that CB[7] mediated pseudorotaxane polymer CB[7]/HA–BrBP changes from small spherical aggregates to a linear array, whereas complexation with CB[8] results in biaxial pseudorotaxane polymer CB[8]/HA–BrBP which transforms to relatively large aggregates. Owing to the more stable 1:2 inclusion complex between CB[8] and BrBP and the multiple hydrogen bonds, this supramolecular polymer has ultralong purely organic RTP lifetime in water up to 4.33 ms with a quantum yield of 7.58%. Benefiting from the targeting property of HA, this supramolecular polymer is successfully applied for cancer cell targeted phosphorescence imaging of mitochondria.

Abstract:B. Liu and co-workers report on page 3093 a new strategy to realize bight farred/near-infrared (FR/NIR) fluorescent conjugated polymer (CP) nanoparticles (NPs) through polymer design and matrix encapsulation. The obtained NPs have 27% quantum yield in aqueous media with improved thermal-/photo-stabilities and larger Stokes shift as compared to commercially available quantum dots (Qdot 655) and organic dyes (Alexa Fluor 555 and Rhodamine 6G). In vivo studies of the NPs on a hepatoma H22 tumorbearing mouse model reveal that they could serve as an efficient and safe FR/NIR fluorescent probe for targeted in vivo fluorescence imaging and cancer in a high contrast and specific manner.

Abstract:Optical imaging in the second near-infrared (NIR-II) windows reduces much more autofluorescence and photon scattering from biological tissues and allows further tissue penetration depth and superior spatial resolution in living bodies. Herein, a fused-ring 2,2' - ( (2Z,2'Z)-( (12,13-bis(2-ethylhexyl)-3,9-diundecyl-12,13-dihydro-[1,2,5] thiadiazolo [3,4-e]thieno [2,'' 3 '' :4',5'] thieno-[2',3' :4,5] pyrrolo [ 3,2-g] thieno [2',3' :4,5] thieno [3,2-b] indole-2,10-diyl) bis(methanylylidene))bis (5,6-difluoro-3-oxo-2,3-dihydro-1H-indene-2,1-diylidene))dimalononitrile (TPBT) molecule was explored as a multifunctional tumor theranostic reagent for photothermal/photodynamic therapy guided by NIR-II imaging. The TPBT molecule has an electron-deficient core with a ladder-type multi-fused ring and shows a narrow band gap that can enhance the near-infrared absorption. The J-aggregative TPBT NPs were formed by nanoprecipitation with great bathochromic shift in absorption and emission spectra, which endows them with ideal fluorescence imaging ability in the NIR-II region. Moreover, TPBT NPs present both higher photothermal conversion efficiency (similar to 36.5%) and effective ROS generation ability, making them excellent candidate for cancer photothermal/photodynamic therapy. Moreover, the biocompatible TPBT NPs can effectively passively target tumor sites due to their enhanced permeability and retention effect for more precision treatment. Thus, TPBT NPs as a multifunctional phototheranostic agent in the NIR-II region present promising potential in clinical cancer NIR-II imaging-guided phototherapy.

Abstract:Simultaneous targeted cancer imaging, therapy and real-time therapeutic monitoring can prevent over-or under-treatment. This work describes the design of a multifunctional nanomicelle for recognition and precise near-infrared (NIR) cancer therapy. The nanomicelle encapsulates a new pH-activatable fluorescent probe and a robust NIR photosensitizer, R16FP, and is functionalized with a newly screened cancer-specific aptamer for targeting viable cancer cells. The fluorescent probe can light up the lysosomes for real-time imaging. Upon NIR irradiation, R16FP-mediated generation of reactive oxygen species causes lysosomal destruction and subsequently trigger lysosomal cell death. Meanwhile the fluorescent probe can reflect the cellular status and in situ visualize the treatment process. This protocol can provide molecular information for precise therapy and therapeutic monitoring.

Abstract:A multifunctional theranostic platform based on conjugated polymer nanoparticles (CPNs) with tumor targeting, fluorescence detection, photodynamic therapy (PDT), and photothermal therapy (PTT) is developed for effective cancer imaging and therapy. Two conjugated polymers, poly[9,9-bis(2-(2-(2-methoxyethoxy)ethoxy)-ethyl)fluorenyldivinylene]-alt-4,7-(2,1,3-benzothiadiazole) with bright red emission and photosensitizing ability and poly[(4,4,9,9-tetrakis(4-(octyloxy)phenyl)-4,9-dihydro-s-indacenol-dithiophene-2,7-diyl)-alt-co-4,9-bis(thiophen-2-yl)-6,7-bis(4-(hexyloxy)phenyl)-thiadiazolo-quinoxaline] with strong near-infrared absorption and excellent photothermal conversion ability are co-loaded into one single CPN via encapsulation approach using lipid-polyethylene glycol as the matrix. The obtained co-loaded CPNs show sizes of around 30 nm with a high singlet oxygen quantum yield of 60.4% and an effective photothermal conversion efficiency of 47.6%. The CPN surface is further decorated with anti-HER2 affibody, which bestows the resultant anti-HER2-CPNs superior selectivity toward tumor cells with HER2 overexpression both in vitro and in vivo. Under light irradiation, the PDT and PTT show synergistic therapeutic efficacy, which provides new opportunities for the development of multifunctional biocompatible organic materials in cancer therapy.
