Abstract:Development of conjugated polymer-based water-soluble nanoparticles (NPs) for second near-infrared fluorescence (NIR-II FL, 1000–1700 nm) and NIR-II photoacoustic (PA) imaging-guided NIR-II photothermal therapy (PTT) is expected to gain favor in clinical anticancer applications. However, integrating these imaging and therapeutic functions into a single NIR-II absorption conjugated polymer BTC12 is a major challenge to accurately balance the nonradiative and radiative decay pathways. Herein, an NIR-II absorption conjugated polymer was designed and synthesized by coupling flexible nonconjugated segments into a polymer backbone to simultaneously achieve NIR-II FL, NIR-II PA imaging, and NIR-II PTT. The developed conjugated polymer-based water-soluble NPs (BTC12 NPs) showed satisfactory functionality with a promising photothermal conversion efficiency of 46.8%. An in vivo study demonstrated that BTC12 NPs can accurately acquire the tumor location and size via NIR-II FL/NIR-II PA imaging. Furthermore, BTC12 NPs showed marked antitumor efficacy by NIR-II PTT upon 1064 nm laser irradiation. This work provides a perspective for the design of multifunction conjugated polymers for NIR-II FL/NIR-II PA imaging-guided NIR-II PTT applications.

Abstract:The second near-infrared fluorescence imaging (NIR-II FI) guided NIR-II photothermal therapy (PTT) possesses advantages for cancer treatment because of its accurate diagnosis and enhanced therapeutic effects for cancer. However, developing a conjugated polymer with both excellent NIR-II fluorescence and NIR-II PTT properties encountered many hindrances, including strong nonradiative decay, low solubility in organic solution, and significant aggregation-caused quenching (ACQ) effect. Herein, a conjugated polymer TTQ-2TC with optimized absorption in NIR-II regions, good solubility in organic solvents, and bright NIR-II FI signals is obtained through introducing the alkyl-side-chain-modified bithiophene (2TC) as the electron-rich donor unit. The water-soluble nanoparticle (TTQ-2TC NPs) prepared from the conjugated polymer TTQ-2TC is used for precise NIR-II FI guided NIR-II PTT treatment. Abstract:Fluorescence imaging in the second near-infrared window (NIR-II) holds promise for real-time deep tissue imaging. In this work, we design a low-band gap conjugated polymers(pTB) with donor-acceptor (D-A) structures by Stille cross-coupling reaction, which can improve the quality of fluorescence imaging and effectiveness of photothermal therapy. In order to improve the water solubility and biocompatibility of pTB, we adopted nano-deposition technology to envelop the hydrophobic polymer p113 into amphiphilic copolymer (1,2-distearoyl-phosphatidylethanolamine-methyl-polyethyleneglycol conjugate (DSPE-mPEG)) shells for NIR-II water-soluble nanoparticles (p113-PEG). The structure, properties and morphology of the polymer were analyzed by 1H-NMR, UV-Vis, dynamic light scattering (DLS) and transmission electron microscopy (1EM). The results show that the conjugated polymer nanoparticles have not only a good absorption peak at 831 nm, but also a large Stokes shift of 206 nm. In addition, the average hydrodynamic radius of pTB-PEG NPs was around 69 nm and spherical morphology was observed from TEM. And the nanoparticles also possess excellent photostability and good biocompatibility in physiological environment such as phosphate buffer saline (PBS), Dulbecco's Modified Eagle Medium (DMEM) and fetal bovine serum (FBS), indicating the potential for further in vivo application. More importantly, we used MTT assay to analyze photothermal treatment toward human breast cancer (4T1) cells in vifro, and confocal laser scanning microscopy (CLSM) tests further indicated that this material has a good photothermal therapeutic effect. To study the NIR-II fluorescence characters, we firstly detected the maximum imaging depth of pTB-PEG NPs in vifro. With the help of long-wavelength emission (>1000 nm), the imaging depth is 6 mm, far exceeding the traditional near-infrared region (NIR-I) reagents. Under 808 nm laser irradiation, the high-resolution second near-infrared window (NIR-II) fluorescence imaging of healthy mouses' blood vascular system and tumor-bearing mice was effectively achieved. In general, conjugated polymer nanoparticles (pTB-PEG) have a good biocompatibility, excellent light stability and good optical properties. It is a promising NIR-II imaging probe with a wide range of the potential for clinical imaging and live tumor imaging.
ATernary Conjugated Polymer for NIR-II Fluorescence Imaging and Photothermal/Photodynamic Combination Therapy

Abstract:In order to simultaneously achieve photothermal properties, photodynamic effects and second near-infrared (NIR-II 1000-1700 nm) fluorescence emission of conjugated polymers, herein a novel ternary conjugated polymer (BDP) was designed and synthesized by copolymerization of an electron-withdrawing monomer diketopyrrolopyrrole and two electron-donating monomers (benzodithiophene derivatives and 1,10-phenanthroline). Then water-soluble nanoparticles (BDP NPs) were further prepared by self-assembling with amphiphilic Pluronic F127 through nanoprecipitation method. The nanoparticles with a diameter of around 90 nm exhibited excellent stability in aqueous solution. The absorption and emission peaks of BDP NPs were determined to be 735 nm and 1039 nm, respectively. The mass extinction coefficient of the nanoparticles was as high as 4.67 mL/(mg center dot cm) the fluorescence quantum yield of BDP NPs was measured to be 0.62% by using IR-26 as a reference, and the photothermal conversion efficiency was 18.2%. Under 730 nm laser irradiation, the temperature of BDP NPs (150 mu g/mL) can reach 64 degrees C, which was high enough for photothermal therapy. Besides, the BDP NPs also produce reactive oxygen species with the same laser, which can be used for photodynamic therapy. Cell experiments confirmed that there was no obvious dark toxicity to both NIH3T3 cells and HeLa cells. While under NIR laser irradiation, strong phototoxicity toward cancer cells was observed, indicating the high therapeutic efficacy with the combination therapy. After intravenous injection of BDP NPs, the blood vessels in living mice can be observed which confirmed the high resolution of NIR-II imaging. Importantly, the BDP NPs can accumulate at the tumor sites at 12 h post-injection, indicating the possibility of NIR-II imaging guided combination therapy in vivo. Overall the obtained BDP NPs with high biocompatibility strong absorption in the first near-infrared (NIR-I, 650- 900 nm) range, intense emission in NIR-II region, and the ability to produce reactive oxygen species and hyperthermia were, excellent candidate for tumor diagnosis and phototherapy. In addition, this work also provides a promising method for the rational design of conjugated polymer-based high performance phototheranostic nanoplatforms. [GRAPHICS] .
NIR-Absorbing Water-Soluble Conjugated Polymer Dots for Photoacoustic Imaging-Guided Photothermal/Photodynamic Synergetic Cancer Therapy

Abstract:“Theranostics” become increasingly significant in current personalized precision medicine. Herein, we developed a new NIR-absorbing photo-theranostic agent based on water-soluble diketopyrrolopyrrole (DPP) conjugated polymer (WSCP) dots. The WSCPs can be easily self-assembled into WSCP dots under ultrasonication only, instead of any other nano-technology. Compared to the monomers of WSCPs, WSCP dots have no fluorescence emission but produce photoacoustic (PA) signal detected upon laser irradiation due to the reduced energy loss from excited state. PA imaging in vivo indicated that WSCP dots can accumulate at tumor site within 4 h post-injection. More importantly, WSCP dots not only generate heat with a photothermal conversion efficiency of ∼54%, but also produce reactive oxygen species (ROS, QY ∼13%). Furthermore, in vitro and in vivo experiments confirmed effective inhibition of tumor growth by WSCP dots via synergetic photothermal/photodynamic therapy. All results indicate a great potential of WSCP dots as highly efficient theranostic agents in PA imaging-guided synergetic cancer treatment.

Abstract:Fluorescence imaging in the second near-infrared (NIR-II) window has emerged as a promising imaging method for cancer diagnosis because of its superior properties such as deep penetration depth and high signal to background ratio. The integration of NIR-II imaging and NIR-II photothermal therapy (PTT) has shown great potential in the field of biomedical research and preclinical practice. Semiconducting polymers have several featured advantages, such as easily tunable optical properties, high photostability, and reliable biocompatibility. However, most semiconducting polymer nanoparticles (SPNs) formed using nanoprecipitation method have high accumulation in reticulum endoplasmic system (RES), and their maximum absorption is located in the first near-infrared (NIR-I) window. Therefore, it is required to develop new NIR-II emissive SPNs with the absorption peaks over 1000 nm, which can be triggered by NIR-II light for deep tissue PTT. Herein, we develop an amphiphilic semiconducting polymer (ASP) with the maximum absorption located in the NIR-II window. The amphiphilic feature of ASP drives it to self-assemble into nanoparticles in aqueous media, which show excellent optical properties, low toxicity, and proper size. ASP exhibits not only bright NIR-II fluorescence but also prominent photothermal performance under 1064 nm laser irradiation. Impressively, ASP shows superior accumulation ability in the tumor compared with that in liver or spleen. Taken together, the overall properties of ASP make it a promising NIR-II phototheranostic agent for cancer diagnosis and treatment.

Abstract:NIR-II imaging-guided phototherapy is an attractive, yet challenging, tumor treatment strategy. By monitoring the accumulation of phototherapy reagents at the tumor site through imaging and determining the appropriate therapy window, the therapeutic effect could be significantly improved. Probes with NIR-II (1000-1700 nm) fluorescence emission and a large Stokes shift hold great promise for fluorescence imaging with deep penetration, minimized self-quenching, and high spatiotemporal resolution. However, due to the lack of a suitable molecular framework, the design of a simple small-molecule dye with a large Stokes shift and NIR-II fluorescence emission has rarely been reported. Herein, we prepare an asymmetric D-π-A type NIR-II fluorescence probe (TBy). The probe is incapsulated in an amphiphilic polymer and modified with a fibronectin targeting peptide CREKA, which could recognize the fibrin-fibronectin complex overexpressed in multiple malignant tumors. The nanoparticles thus constructed (TByC-NPs) have maximum fluorescence emission at 1037 nm with a large Stokes shift of 426 nm, which is the largest Stokes shift among organic NIR-II fluorescent dyes reported in the literature. The TByC-NPs exhibit a good NIR-II imaging performance, active tumor targeting, and good photothermal and photodynamic capabilities. In vitro and in vivo studies verify that the TByC nanoplatform shows outstanding biocompatibility for NIR-II imaging-guided phototherapy and provides an excellent antitumor effect.
Fabrication of conjugated polymers with aggregation-induced near-infrared-II emission for efficient phototheranostics

Abstract:Conjugated polymers (CPs), which emit in the second near-infrared window (NIR-II, 1000-1700 nm), are used as biomaterials for NIR-II fluorescence imaging because of their adjustable photophysical properties and high optical stability. However, the fluorescence signal of conventional CPs is quenched in an aggregated state due to strong π-π stacking, which results in the closure of the radiation attenuation pathway. To solve this problem, the aggregation-induced emission effect is considered a reasonable strategy for enhancing the aggregative fluorescence of IR-II emitters. We herein report NIR-II conjugated polymers with typical AIE characteristics (αAIE > 3) by changing the side chain structure of receptor units and the conjugation degree of donors. Conjugated polymer nanoparticles (PoBVT NPs) exhibit outstanding performance in NIR-II fluorescence imaging (QY = 1.94%) and highly effective photothermal therapy (η = 45%). In vivo studies have shown that the location of tumors can be accurately obtained by NIR-II FL/NIR-II PA imaging, and there is a significant anti-tumor effect after laser irradiation. This work offers prospects for the design of multifunctional conjugated polymers for NIR-II FL/PA imaging to guide NIR-II PTT applications.

Abstract:Our exploiting versatile multimodal theranostic agent aims to integrate the complementary superiorities of photoacoustic imaging (PAI), second near-infrared (NIR-II, 1000-1700) fluorescence and T1-weighted magnetic resonance imaging (MRI) with an ultimate objective of perfecting cancer diagnosis, thus improving cancer therapy efficacy. Herein, we engineered and prepared a water-soluble gadolinium-chelated conjugated polymer-based theranostic nanomedicine (PFTQ-PEG-Gd NPs) for in vivo tri-mode PA/MR/NIR-II imaging-guided tumor photothermal therapy (PTT).Methods: We firstly constructed a semiconducting polymer composed of low-bandgap donor-acceptor (D-A) which afforded the strong NIR absorption for PAI/PTT and long fluorescence emission to NIR-II region for in vivo imaging. Then, the remaining carboxyl groups of the polymeric NPs could effectively chelate with Gd3+ ions for MRI. The in vitro characteristics of the PFTQ-PEG-Gd NPs were studied and the in vivo multimode imaging as well as anti-tumor efficacy of the NPs was evaluated using 4T1 tumor-bearing mice.Results: The obtained theranostic agent showed excellent chemical and optical stability as well as low biotoxicity. After 24 h of systemic administration using PQTF-PEG-Gd NPs, the tumor sites of living mice exhibited obvious enhancement in PA, NIR-II fluorescence and positive MR signal intensities. Better still, a conspicuous tumor growth restraint was detected under NIR light irradiation after administration of PQTF-PEG-Gd NPs, indicating the efficient photothermal potency of the nano-agent.Conclusion: we triumphantly designed and synthesized a novel and omnipotent semiconducting polymer nanoparticles-based theranostic platform for PAI, NIR-II fluorescence imaging as well as positive MRI-guided tumor PTT in living mice. We expect that such a novel organic nano-platform manifests a great promise for high spatial resolution and deep penetration cancer theranostics.

Abstract:Development of versatile phototheranostics is highly desirable for cancer theranostics. Herein, a novel organic conjugated polymer (named DPP-TT) with excellent optical properties was designed and prepared. Based on single-component DPP-TT, single DPP-TT NPs as versatile phototheranostics were developed by a simple nanoprecipitation method. The obtained NPs exhibited good water solubility, excellent biocompatibility, outstanding photostability, strong NIR-I light absorption, and appropriate NIR-II fluorescence emission. Importantly, such NPs presented high photothermal conversion efficiency. From the investigations performed in vitro and in vivo, it was observed that DPP-TT NPs show remarkable performance for cancer theranostics, benefiting from single 808 nm laser-induced tri-modal (NIR-II fluorescence/photoacoustic/thermal) imaging-guided photothermal therapy.

Abstract:<p>Development of near-infrared-II (NIR-II) light responsive nano-agents with high photothermal stability, high photothermal conversion efficiency (PCE), and excellent biocompatibility for photoacoustic (PA) imaging-guided photothermal therapy (PTT) is of tremendous significance. In spite of the superiority of organic semiconducting polymer nanoparticles (OSPNs) in PA imaging-guided PTT, the limited absorption in the first NIR (NIR-I) window and metastable nanostructure of OSPNs resulting from commonly used preparation methods based on nanoprecipitation or reprecipitation compromise their <em>in vivo</em> phototheranostic performance. Herein we design and synthesize a novel NIR-II absorbing organic semiconducting polymer amphiphile (OSPA) to enhance the structural stability of OSPNs. With prominent optical properties, low toxicity, and a suitable size, OSPA not only efficiently labels and kills cancer cells under NIR-II irradiation but also accumulates at the tumor of living mice upon intravenous injection, allowing efficient NIR-II light-triggered phototheranostics toward tumor. The developed OSPA has promising potential for fabricating multifunctional nanoplatforms to enable multimodal theranostics.</p>

Abstract:Optical imaging in the second near-infrared (NIR-II) windows reduces much more autofluorescence and photon scattering from biological tissues and allows further tissue penetration depth and superior spatial resolution in living bodies. Herein, a fused-ring 2,2' - ( (2Z,2'Z)-( (12,13-bis(2-ethylhexyl)-3,9-diundecyl-12,13-dihydro-[1,2,5] thiadiazolo [3,4-e]thieno [2,'' 3 '' :4',5'] thieno-[2',3' :4,5] pyrrolo [ 3,2-g] thieno [2',3' :4,5] thieno [3,2-b] indole-2,10-diyl) bis(methanylylidene))bis (5,6-difluoro-3-oxo-2,3-dihydro-1H-indene-2,1-diylidene))dimalononitrile (TPBT) molecule was explored as a multifunctional tumor theranostic reagent for photothermal/photodynamic therapy guided by NIR-II imaging. The TPBT molecule has an electron-deficient core with a ladder-type multi-fused ring and shows a narrow band gap that can enhance the near-infrared absorption. The J-aggregative TPBT NPs were formed by nanoprecipitation with great bathochromic shift in absorption and emission spectra, which endows them with ideal fluorescence imaging ability in the NIR-II region. Moreover, TPBT NPs present both higher photothermal conversion efficiency (similar to 36.5%) and effective ROS generation ability, making them excellent candidate for cancer photothermal/photodynamic therapy. Moreover, the biocompatible TPBT NPs can effectively passively target tumor sites due to their enhanced permeability and retention effect for more precision treatment. Thus, TPBT NPs as a multifunctional phototheranostic agent in the NIR-II region present promising potential in clinical cancer NIR-II imaging-guided phototherapy.

Abstract:By introducing non‐conjugated fragments and AIE motifs through a dual‐enhancement strategy and alternating donor‐acceptor copolymerization with BBT electron‐deficient units, conjugated polymers with the AIE effect are developed with both high‐quality fluorescence imaging and high photothermal conversion efficiencies, enabling NIR‐II (1000‐1700 nm) fluorescence imaging‐guided NIR‐II photothermolysis for use in oncology therapy. The development of conjugated polymer–based water‐soluble nanoparticles for near‐infrared‐II (NIR‐II) fluorescence (FL; 1000–1700 nm)‐guided photothermal therapy holds promise in advancing cancer treatment. However, excessive nonradiative decay leads to almost complete quenching of conjugated polymers' fluorescence. Therefore, a critical challenge is to suppress nonradiative decay while maintaining high‐quality fluorescence imaging and excellent photothermal conversion efficiency. In this study, a series of NIR‐II‐conjugated polymers with aggregation‐induced emission (AIE) effects are designed and synthesized using the Stille coupling reaction. The dual enhancement strategy of modulating the AIE units and introducing non‐conjugated backbone into the polymer backbone resulted in BCT1 with a high αAIE value of 3.27. BCT1 nanoparticles exhibit excellent NIR‐II fluorescence, a high photothermal conversion efficiency of 70.51%, and a tenfold enhancement in fluorescence compared with BT1. Both in vitro and in vivo experiments validated their good biocompatibility and outstanding performance in NIR‐II fluorescence imaging for accurately determining the location of tumors. This study provides a novel strategy and method for designing and developing multifunctional conjugated polymers for NIR‐II fluorescence imaging–guided photothermal therapy.

Abstract:Photodynamic therapy (PDT) and photothermal therapy (PTT) are two powerful tools to tackle cancer in the biomedical field due to their super tumor ablation efficiency and minimal damage to normal tissue. However, it is still a tricky challenge to combine these two treatment modalities into a single agent for single wavelength laser triggered efficient tumor therapy. Herein, water-soluble nanoparticles consisting of diketopyrrolopyrrole-based conjugated polymer and amphipathic Pluronic F127 were intelligently devised and fabricated, which possessed outstanding optical properties, superb biocompatibility, excellent photostability. Surprisingly, the nanoparticles exhibited moderate singlet oxygen yield (2.34%) and photothermal conversion efficiency (41.5%) under an 808 nm laser illumination, which was in favor of achieving high-performance PDT/PTT combination therapy. Moreover, the evident antitumor effect of the nanoparticles was confirmed by in vitro and in vivo experiments. This work provides a promising approach to rationally design near infrared absorbing organic polymer nanoplatforms for combination cancer therapy.

Abstract:Photodynamic therapy (PDT) has become a promising treatment approach against cancer due to low side effects and high therapeutic efficacy. However, the limitations of photosensitizers such as poor water solubility and lack of targeting ability hindered the clinical application of PDT. Here, we synthesized a phosphorescent conjugated polymer brush (PPF-Ir-g-(POEGMA-b-PGal)) in which a small photosensitizer iridium(III) complex was covalently attached to the conjugated backbone. A further hydrophilic polymer (POEGMA) and glycopolymer polygalactose (PGal), which has a specific binding ability with Hep G2 tumors, were grafted from the conjugated backbone via atom transfer radical polymerization (ATRP) and click reaction. The brush structure rendered PPF-Ir-g-(POEGMA-b-PGal) exhibited excellent water solubility. Meanwhile, PPF-Ir-g-(POEGMA-b-PGal) showed efficient properties of producing singlet oxygen. The photodynamic effect of the PPF-Ir-g-(POEGMA-b-PGal) photosensitizer was evaluated in Hep G2 cells via the MTT assay and flow cytometry and the results indicated that this photosensitizer can efficiently cause the death of cancer cells. Additionally, an antitumor study of PPF-Ir-g-(POEGMA-b-PGal) was conducted in vivo with Hep G2 tumor bearing nude mice, and the results show that the xenograft tumors were significantly inhibited. In summary, PPF-Ir-g-(POEGMA-b-PGal) exhibited fine water solubility and high PDT efficiency both in vitro and in vivo. Our study may further encourage the applications of conjugated polymer brush based photosensitizers for PDT in tumor treatment.

Abstract:The synthesis of water-soluble symmetric molecules with donor-acceptor-donor (D-A-D) structure is reported. The compound is connected by π bridge with 2-bromofluorene external polyethylene glycol 2000 as the shielding unit, and donor component and pyrrolopyrrole (DPP) as the acceptor unit. The D-A-D double donor fluorescent molecule P2-DPP is obtained by coupling reaction. The absorption peak and emission peak of the fluorescent molecule P2-DPP are 600 and 1020 nm, respectively. It has potential excellent imaging characteristics. It does not need to use nanoparticles formed by the DSPE-MPEG amphiphilic block to form micelles. The quantum yield reaches 0.6% and the penetration depth can reach 10 mm. The chemical is capable of achieving liver and renal metabolism. It has a good application prospect in the photothermal therapy of mouse tumors and realizes the integration of biological diagnosis and treatment.
Dual-modal Imaging-Guided Agent Based on NIR-II Aggregation-Induced Emission Luminogens with Balanced Phototheranostic Performance

Abstract:Phototherapy has garnered considerable interest for its potential to revolutionize conventional cancer treatment. Organic materials with near-infrared II (NIR-II, 1000-1700 nm) fluorescence and photothermal effects are key for precise tumor diagnosis and treatment, yet optimizing their output for higher resolution and reduced photodamage remains a challenge. Herein, a multifunctional NIR-II photosensitizer (LSC) has been developed using the aggregation-induced emission (AIE) technology. The utilization of Thieno[3,2-b]thiophene as an electron-rich and bulky donor/acceptor bridge has allowed for the elongation of conjugation length and distortion of the AIE main chain. This strategic modification effectively enhances the electron push-pull effect, endowing the LSC with a Stokes shift of over 400 nm and AIE characteristics. We have successfully built-up stable nanoparticles called FA-LSC NPs using a nano-precipitation method. These nanoparticles exhibit high NIR-II fluorescent brightness (ε × QY = 1064 M-1 cm-1) and photothermal conversion efficiency (41%). Furthermore, the biocompatible FA-LSC NPs demonstrate effective tumor accumulation and exceptional photothermal therapeutic efficacy both in vitro and in vivo. These nanoparticles were applied to fluorescence-photothermal dual-mode imaging-guided photothermal ablation in a Hela tumor xenograft mouse model, resulting in favorable photothermal clearance outcomes.

Abstract:Spatiotemporally manipulating in situ immobilization of theranostic agents within cancer cells to improve their bioavailability is highly significant yet challenging in tumor diagnosis and treatment. Herein, as a proof-of concept, we for the first time report a tumor-targetable near-infrared (NIR) probe DACF with photoaffinity crosslinking characteristics for enhanced tumor imaging and therapeutic applications. This probe possesses great tumor-targeting capability, intensive NIR/photoacoustic (PA) signals, and predominant photothermal effect, allowing for sensitive imaging and effective photothermal therapy (PTT) of tumors. Most notably, upon 405 nm laser illumination, DACF could be covalently immobilized within tumor cells through photocrosslinking reaction between photolabile diazirine group and surrounding biomolecules resulting in enhanced tumor accumulation and prolonged retention simultaneously, which significantly facilitates the imaging and PTT efficacy of tumors in vivo. We therefore believe that our current approach would provide a new insight in achieving precise cancer theranostics.

Abstract:Photothermal therapy is a promising phototherapeutic modality that has been widely studied in cancer therapy. However, because of the influence of heat shock protein (HSP), the therapeutic efficacy of photothermal therapy (PTT) is significantly suppressed. To improve the therapeutic efficacy, different tumor-specific therapeutic modalities have been chosen to combine with PTT. However, most of them rely on endogenous stimuli to trigger combination therapy, which may suffer from the issue of incomplete activation. Herein, we develop a PTT/thermodynamic combination therapeutic nanosystem whose therapeutic process is controlled by an external stimulus, near-infrared (NIR) light. The nanosystem (ADPPTN) is composed of a second NIR (NIR-II) fluorescent semiconducting polymer (SP) (DPPT) as the core, and a carboxyl group-decorated amphiphilic copolymer (PSMA-PEG) as the shell with an azo-containing compound (AIPH) loaded via electrostatic interaction. Under 808 nm laser irradiation, DPPT can generate heat to conduct PTT, while the elevated temperature may further trigger the release of AIPH radicals, conducting thermodynamic therapy (TDT). In addition, the NIR-II fluorescence signal emitted from DPPT can light the tumor. Compared with the nanoparticles without AIPH (DPPTN), ADPPTN has better anticancer efficacy under laser irradiation both in vitro and in vivo. Thus, our study provides an NIR-II fluorescence imaging-guided PTT/TDT combination therapeutic nanosystem for efficient cancer theranostics.
