Abstract:Difunctionalization of alkenes represent a convenient way to complex molecular structure. Herein, visible light-induced intramolecular carbodi(tri)fluoromethylthiolation of alkenes was reported. CC and C-S were formed in the procedure. Various fluoroalkylthiol substituted 2,3-dihydrobenzofurans (DHB) was prepared. Aryldiazonium salts derived from o-allyloxy anilines were used as precursors of aryl radicals. The following cyclization occurred, and the resulting alkyl radicals were trapped by shelf-stable reagent PhSO2SRf (Rf= SCF2H, SCF3). The direct difluoromethylthiolation on arenes was not observed. A variety of atoms and functional groups were tolerated.

Abstract:A convenient approach for the synthesis of 3-difluoroalkyl phthalides has been developed from phthalaldehydic acids and difluoroenoxysilanes by using relatively inexpensive p-toluenesulfonic acid monohydrate (PTSA) as a catalyst. A series of 3-difluoroalkyl phthalides and cyclic difluoroalkyl ethers were obtained in up to 99% yield. The products obtained could be readily converted into difluoroalkyl phthalide derivatives by simple modifications.

Abstract:A hexafluoroisopropanol (HFIP)-catalyzed difluoroalkylation of propargylic alcohols with difluoroenoxysilanes to access structurally diverse tetrasubstituted difluoroalkyl allenes has been developed. This convenient procedure enables the rapid construction of highly functionalized multisubstituted fluorinated allenes in a mild and straightforward way. Furthermore, the synthetic potential of this methodology has been demonstrated by the facile synthesis of various structurally interesting fluorine-containing molecules such as gem-difluorosubstituted dihydropyran, tetrasubstituted CF2H-allene, and multisubstituted fluorinated cyclopentanone derivatives.

Abstract:Hexafluoroisopropanol (HFIP)-catalyzed direct dehydroxydifluoro-alkylation of benzylic and allylic alcohols with difluoroenoxysilanes is developed. This procedure enables the synthesis of a broad range of alpha,alpha-difluoroketones, a class of highly valuable intermediates and building blocks in medicinal and organic chemistry. Here, we have demonstrated for the first time that HFIP could act as a powerful catalyst for fluorinated carbon-carbon bond formation. The application of this protocol in late-stage dehydroxydifluoroalkylation of potentially bioactive drugs and natural products has also been carried out.

Abstract:A novel efficient HFIP-catalyzed synthesis of structurally diverse 2,2-difluoro-3-hydroxy-1,4-diketone derivatives from readily available glyoxal monohydrates and difluoroenoxysilanes is described. This convenient protocol is induced by the distinctive fluorine effect of the reactants and the fluoroalcohol catalyst, which represents the first application of fluoroalcohol catalysis in a Mukaiyama aldol reaction.

Abstract:A mild and general procedure for the difluoromethylation of organochalcogen compounds has been accomplished via sulfoxide and selenoxide-directed [3,3]-sigmatropic rearrangement/Haller-Bauer reaction with difluoroenoxysilanes. The reactions showed good functional group compatibility, wide substrate applicability, and facile scalability. The synthetic utility is highlighted by the iterative use of this protocol to construct multi-CF2H-containing chalcogenides and the late-stage modification of pharmaceuticals.

Abstract:Herein, we report the first example of Rh-catalyzed C(sp 2 )–H difluoroallylation of α,β-unsaturated amides with 3-bromo-3,3-difluoropropene under mild conditions. The features of this protocol for difluoroallylation are unprecedented vinylic substrates, ethanol as a green solvent, good functional group and air compatibility, and scale-up synthesis with low catalyst loading.

Abstract:In contrast with unactivated alkenes, the corresponding hydrotrifluoromethylation of styrene has remained challenging due to the strong propensity of styrene for oligomerization and polymerization. On the basis of our newly developed trifluoromethylation reagent, TFSP, herein we present a general method for the hydrotrifluoromethylation of styrene under photoredox catalysis. The substrate scope was further extended to unactivated alkenes, acrylates, acrylamides, and vinyl-heteroatom-substituted alkenes. The tunability of this method was showcased via the relevant deprotonative trifluoromethylation and trifluoromethyltrifluoroethoxylation reactions.

Abstract:Photoredox-catalyzed hydrodifluoromethylation of alkenes has become an effective method to introduce difluoromethyl group into organic molecules. As the reported methods involve either photocatalysts or superstoichiometric amounts of additives, we herein describe a simple alternative without using photocatalyst or additive for the hydrodifluoromethylation of alkenes, through photoactivation of difluoromethyltriphenylphosphonium iodide salt. Mechanistic studies shed light on how the transformation takes place.

Abstract:(DHQD)(2)PHAL-catalyzed asymmetric allylic trifluoromethylation of Morita-Baylis-Hillman adducts using a Rupert-Prakash reagent is reported. This transformation provided the S(N)2' trifluoromethylated products with good yields and excellent enantioselectivities at room temperature. It was also found that the reaction could be accelerated using acetonitrile as cosolvent.

Abstract:The synthesis of medicinally interesting aryldifluoromethylated compounds has drawn significant research attention in recent years. Herein, we report an unprecedented iron-mediated process for the selective defluorination of trifluoromethylarenes to achieve the 1,2-difluoroalkylthiolation of alkenes. Preliminary mechanistic studies revealed that thiolate anion, trifluoromethylarene, and iron cation could form an electron donor-acceptor (EDA) complex, which induced selective defluorination and then difunctionalization of alkenes to obtain aryldifluoromethylated products. The generated aryldifluoromethylated compounds make it difficult to form an EDA complex again, thus avoiding excessive defluorination. This conversion has concise and ambient reaction conditions and provides an alternative solution for obtaining difluorobenzylic intermediates.

Abstract:Atom transfer radical addition and Heck-type reaction between alkenes and ethyl iododifluoroacetate by using organophosphine compounds as catalysts have been developed. The reaction proceeds under mild reaction conditions with excellent functional group tolerance and high chemo- and regioselectivities. Mechanistic study indicates that the reaction might be initiated via noncovalent interactions between an organophosphine catalyst and a carbon–iodine bond.

Abstract:Although defluorinative functionalization of trifluoromethylarenes has been studied by several research groups, different kinds of catalysts are requested accordingly based on specific substrate families to obtain corresponding RCF2Ar compounds. Herein, we report a general photocatalytic approach for selective hydrodefluorination (HDF) and defluoroalkylation (DFA) of ArCF3. Using electronically-different arenethiolates as photosensitizer, 88 gem-difluoromethyl compounds were synthesized from a broad range of electronically-variable trifluoromethylarenes, including natural products and bioactive molecules. Our mechanistic studies uncovered photo-induced Electron Donor-Acceptor (EDA) complexation followed by the silane-assisted C-F bond activation.

Abstract:Trifluoromethylthiolation/oxidation of styrenes with easiy-handling AgSCF3 used as the trifluoromethylthiolating reagent has been described, which furnished a series of α-trifluoromethylthio acetophenons. A variety of substituted styrene could be bifunctionalized via radical process in a mixed solvent of dimethyl sulfoxide (DMSO) and CHCl3, with potassium persulfate used as the oxidant under mild conditions. A simple and efficent method for the facile construction of various α-trifluoromethylthio acetophenons is thus demonstrated.

Abstract:In this study, we explored the reactivity of meta‐trifluoromethanesulfonate (OTf) substituted diaryliodonium salts under alkaline conditions. Two distinct pathways with direct arylation of pyrrole derivatives and Diels‐Alder cycloaddition via aryne intermediates, have been found upon the relative alkalinity strength of potassium carbonates or hydroxides. This approach offers a straightforward and efficient route to synthesize heterocyclic aromatics under mild reaction conditions with tolerance of various functional groups.
