Abstract:A rhodium‐catalyzed intramolecular [3+2] cycloaddition of 1‐allene‐vinylcyclopropanes (1‐allene‐VCPs) to construct cyclopentane‐embedded bicyclic structures with quaternary stereocenters at the bridgehead position has been developed with good to excellent yields. In addition, this transformation could also be conducted on a large scale with reduced catalyst loading, which could be useful for further derivatizations and applications.

Abstract:Vinylcyclopropane (VCP) has been well applied as a five-carbon component, rather than a three-carbon component, in transition-metal catalyzed cycloadditions. Here we demonstrate a Rh(l)-catalyzed [3 + 2] reaction of trans-VCP-enes, where VCP acts as a three-carbon synthon to furnish five-membered carbocycles. This novel cycloaddition is efficient in generating bicyclic cyclopentanes in good yields from simple and easily prepared substrates. When cis-VCP-ene is used as the substrate, VCP acts as a five-carbon unit to give a [5 + 2] cycloadduct. Rationalization of the [3 + 2] and [5 + 2] cycloadditions of VCP-enes has been proposed.

Abstract:Through the combined use of computational (density functional theory) and experimental studies, a new [Rh(CO)(2)Cl](2) catalyzed two-component [5+2+1] cycloaddition of ene-vinylcyclopropanes and CO for the synthesis of fused bicyclic cyclooctenones has been designed and experimentally realized. The key point behind this design is to turn a disfavored reductive elimination of (sp(3))C-Rh-C(sp(3)) to an easier migratory reductive elimination of (sp(3))C-Rh-CO-C(sp(3)) by the introduction of CO. These reactions proceed in good yields for substrates with various tether types and substituents, providing a flexible, convenient, efficient, and stereocontrolled method for the construction of bicyclic cyclooctenones.

Abstract:Previously reported was that cis-ene-vinylcyclopropanes (cis-ene-VCPs) underwent Rh-catalyzed [5+2] reaction to give 5,7-fused bicyclic products, where vinylcyclopropane (VCP) acts as five-carbon synthon. Unfortunately, this reaction had very limited scope. Replacing the 2 pi component of cis-ene-VCPs to allene moiety, the corresponding cis-allene-VCPs did not undergo the expected normal [5+2] cycloaddition to give 5,7-fused bicyclic products. Instead, the challenging bicyclo[ 4.3.1]decane skeleton was obtained via an unprecedented bridged [5+2] cycloaddition. DFT calculations were applied to understand why this bridged [5+2] reaction is favored over the anticipated but not realized normal [5+2] reaction.

Abstract:Transition metal-catalyzed [4+2+1] reactions of dienes (or diene derivatives such as vinylallenes), alkynes/alkenes, and CO (or carbenes) are expected to be the most straightforward approach to synthesize challenging seven-membered ring compounds, but so far only limited successes have been realized. Here, an unexpected three-component [4+2+1] reaction between two vinylallenes and CO was discovered to give highly functionalized tropone derivatives under mild conditions, where one vinylallene acts as a C4 synthon, the other vinylallene as a C2 synthon, and CO as a C1 synthon. It was proposed that this reaction occurred via oxidative cyclization of the diene part of one vinylallene molecule, followed by insertion of the terminal alkene part of the allene moiety in another vinylallene, into the Rh-C bond of five-membered rhodacycle. Then, CO insertion and reductive elimination gave the [4+2+1] cycloadduct. Further experimental exploration of why ene/yne-vinylallenes and CO gave monocyclic tropone derivatives instead of 6/7-bicyclic ring products were reported here.

The possible competing [4+1], [4+2], and [2+2+1] cycloadditions were disfavored, making the present reaction an efficient way to access functionalized 5/7 rings.

Abstract:A formal [3+3+1] reaction through gold(I)‐catalyzed enyne cycloisomerization of enyne‐methylenecyclopropanes and rhodium(I)‐catalyzed [6+1] reaction of vinylspiropentanes and CO has been developed to synthesize aza‐6/7 bicyclic compounds, hexahydrocyclohepta[c]pyridinones. DFT calculations reveal that four steps are involved in the [6+1] cycloaddition of vinylspiropentanes and CO, including oxidative addition of cyclopropane to Rh, β−C elimination, CO insertion and reductive elimination.

Abstract:Described herein is an unprecedented example of generating the endocyclic donor–donor vinyl carbene achieved by the dirhodium(II)-catalyzed enynal cycloisomerization. The endocyclic vinyl carbene intermediate could be trapped by a diversity of alkenes, including 1,1-disubstituted alkenes, dienes, and α-methyl styrenes, which could undergo [2 + 1], [4 + 3] cycloaddition and an unexpected formal allylation, respectively, giving spiro and fused polycyclic heterocycles. In these cascade transformations, up to three rings and three stereocenters were constructed in one step with good selectivities. The enantioselective versions were also achieved with up to 84% ee. Mechanistic studies revealed that a concerted intramolecular [2 + 3] cycloaddition was the key step for the generation of vinyl Rh(II) carbene. Detailed studies were conducted to elucidate the intriguing mechanism of the formal allylation.

Abstract:A palladium-catalyzed ring-opening cyclization of (E) & (Z)-ene-vinylidenecyclopropanes has been developed via an intramolecular [3+2] cycloaddition process in the presence of sterically bulky biaryl phosphine ligand, stereoselectively affording fused cis- & trans-bicyclo[4.3.0] skeleton products in good yields with a broad substrate scope and good functional tolerance. Plausible reaction mechanism was proposed on the basis of previous work and the DFT calculations.

Abstract:The first rhodium-catalyzed intermolecular [3+2] cycloaddition reaction of vinyl aziridines and allenes for the synthesis of enantioenriched functionalized pyrrolidines was realized. [3+2] cycloaddition with the proximal C=C bond of N-allenamides gave 3-methylene-pyrrolidines in high regio- and diastereoselectivity, whereas, 2-methylene-pyrrolidines were obtained as the major products by the cycloadditions of vinyl aziridines with the distal C=C bond of allenes. Use of readily available starting materials, a broad substrate scope, high selectivity, mild reaction conditions, as well as versatile functionalization of the cycloadducts make this approach very practical and attractive.
