Abstract:Herein, we reported a simple one-pot green method for preparation of fluorescent copper nanoclusters (Cu NCs) with a high quantum yield up to 8.6%, using glutathione as the stabilizing agent. The as-prepared Cu NCs had good water solubility, excellent stability and ultrafine size through the characterization of UV-vis absorption spectroscopy, fluorescence spectroscopy, transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), and Fourier transform infrared (FTIR) spectroscopy. Experimental results showed that the fluorescence of Cu NCs was linearly quenched by the Fe3+ concentrations in the range of 1-100 mu M, with the detection limit of 0.3 mu M. Abstract:Herein, we demonstrate a facile, straightforward and green method for synthesis of highly red-emitting fluorescent bimetallic Au/Ag nanoclusters (NCs) employing lipoic acid as both a reducing agent and a protecting agent. The as-prepared Au/Ag NCs exhibited bright red emission with a strong peak centered at 630 nm (quantum yield measured as 6.4%), and showed excellent stability and remarkable temperature-dependent fluorescent property. The fluorescence responses were reversible and linear to the environment temperature from 20 to 65 C with a resolution as high as 1.34 degrees C, which allows its potential application as a fluorescent nanothermometer. Moreover, Au/Ag NCs were also explored for selective and sensitive determination of Fe3+ based on the specific quenching of the fluorescence by Fe3+, and explored for fluorescence imaging Fe3+ in living HeLa cells thanks to their very low cytotoxicity. Abstract:A facile, one-pot synthetic method has been proposed to prepare water-soluble fluorescent copper nanoclusters (CuNCs) templated by nucleosides. The nucleoside-templated fluorescent CuNCs were further characterized by using various analytical techniques, such as transmission electron microscopy, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, and fluorescence spectroscopy. The role of various reactants such as ascorbic acid, nucleoside, and citrate buffer in the synthesis process of fluorescent CuNCs was explored. The results showed that nucleoside and ascorbic acid were very likey to respectively act as a stabilizer and a reductant to form nanoclusters, and citrate buffer acted as both pH regulator solution and a reducing agent. The fluorescence spectra of various nucleoside-templated CuNCs were finally combined with multivariate chemometrics analysis for discrimination of different nucleosides.

Abstract:Herein, an innovative and simple strategy for synthesizing high fluorescent Cu nanoclusters was successfully established while L-cysteine played a role as the stabilizer. Meaningfully, the current Cu nanoclusters together with a quantum yield of 14.3% were prepared in aqueous solution, indicating their extensive applications. Subsequently, the possible fluorescence mechanism was elucidated by fluorescence, UV-vis, HR-TEM, FTIR, XPS, and MS. Additionally, the CuNCs were employed for assaying Hg2+ on the basis of the interactions between Hg2+ and L-cysteine; thus facilitating the quenching of their fluorescence. The proposed analytical strategy permitted detections of Hg2+ in a linear range of 1.0 x 10(-7) mol L-1 x 10(-3) mol L-1, with a detection limit of 2.4 x 10(-8) mol L-1 at a signal-to-noise ratio of 3. Significantly, this CuNCs described here were further applied for coding and fluorescent staining, suggesting may broaden avenues toward diverse applications. (C) 2014 Elsevier B.V. Abstract:DNA strands have been used as templates to form different sized silver nanoclusters. Although DNA templated silver nanoclusters (NCs) probes which show outstanding fluorescence properties have been widely applied in chemical sensing and cellular imaging, synthesis of DNA template Au-Ag bimetallic nanoclusters remains a challenge. A facile one-step synthesis method was thus developed in this study to form Au-Ag NCs using C4-ATAT-C4 as template. C4-ATAT-C4 acted as a stabilizer for preventing aggregation of the Au-Ag NCs. The obtained Au-Ag NCs stabilized by C4-ATAT-C4 showed bright fluorescence and high stability. A series of experiments showed that temperature, citrate-citric acid buffer, sequence and concentration of DNA played an important role in the synthesis of fluorescent Au-Ag NCs. In addition, a ratiometric fluorescence probe was designed through combining the nucleotide sequence (C4-ATAT-C4) and hybridization sequence in the presence of the double-strand-chelating dye Super SYBR Green (SG). The combination of prepared Au-Ag NCs, Super SG, and perfectly matched DNA emitted fluorescence at 500 and 590 nm, respectively, when the composite was excited at 290 nm. A gastric cancer gene was also selectively detected by our developed ratiometric fluorescence probe.

Abstract:A trimethine cyanine dye (MTC) was developed for Cu2+ detection. MTC can exhibit different aggregation modes, such as monomers, dimers, H- and J-aggregates. Here, G-quadruplex (G2) with modification of an alkyne at the 5′ end could regulate J-aggregates of MTC to monomers. Upon addition of Cu2+, sodium ascorbate, and another sequence (G1) with azide modification at the 3′ end, a new sequence was ligated by click chemistry of azide-alkyne cycloaddition, accompanied by MTC monomers assembling into J-aggregates again. The colors of the solution turned pink to blue. This strategy shows high specificity and selectivity and can realize monitoring Cu2+ in tap water. This simple method uses only a Uv-vis instrument to finish Cu2+ detection. A cyanine dye transformation between J-aggregates and monomers was applied to monitor the Cu2+ level via Cu+-promoted azide-alkyne cycloaddition of two rich guanine sequences.

Abstract:Herein, the synthesis of Cu nanoclusters (CuNCs) templated with dopamine has been first successfully established by reducing CuSO4 in an aqueous medium, and this type of nanocluster emitted cyan high-fluorescence. Subsequently, the CuNCs were employed for assaying tetracycline (TC) on the basis of the quenching of the fluorescence intensity of CuNCs once TC was introduced. The proposed analytical strategy permitted the detection of TC in a linear range of 3.6 x 10(-6) to 1.0 x 10 (3) mol L (1) together with a detection limit of 9.2 x 10 (7) mol L-1 at a signal-to-noise ratio of 3. Abstract:Temperature measurement in biology and medical diagnostics, along with sensitive temperature probing in living cells, is of great importance; however, it still faces significant challenges. Metal nanoclusters (NCs) with attractive luminescent properties may be promising candidates to overcome such challenges. Here, a novel one-step synthetic method is presented to prepare highly fluorescent copper NCs (CuNCs) in ambient conditions by using glutathione (GSH) as both the reducing agent and the protective layer preventing the aggregation of the as-formed NCs. The resultant CuNCs, with an average diameter of 2.3 nm, contain 1–3 atoms and exhibit red fluorescence (λem = 610 nm) with high quantum yields (QYs, up to 5.0%). Interestingly, the fluorescence signal of the CuNCs is reversibly responsive to the environmental temperature in the range of 15–80 °C. Furthermore, as the CuNCs exhibit good biocompatibility, they can pervade the MC3T3-E1 cells and enable measurements over the physiological temperature range of 15–45 °C with the use of the confocal fluorescence imaging method. In view of the facile synthesis method and attractive fluorescence properties, the as-prepared CuNCs may be used as photoluminescence thermometers and biosensors.
Formation of copper nanoparticles on poly(thymine) through surface-initiated enzymatic polymerization and its application for DNA detection

Abstract:Poly(thymine) (polyT) and double-stranded DNA (dsDNA) can act as efficient templates for the formation of copper nanoparticles (CuNPs) at a low concentration of CuSO4, and the formed CuNPs emit excellent fluorescence. In this work, we demonstrated a new and facile strategy for the highly sensitive and selective detection of DNA on streptavidin-functionalized magnetic beads (SA-MB) using DNA-templated CuNPs as the fluorescent probe. Target DNA (tDNA) was hybridized with the capture DNA that was immobilized on the surface of SA-MB. Surface initiated enzymatic polymerization (SIEP) was employed as the signal amplification method to generate the polyT at the 3' end of tDNA for the formation of CuNPs. The incorporation of polyT by SIEP resulted in similar to 35.7 fold signal amplification compared to the dsDNA after hybridization without SIEP. A dose-response curve for detection of DNA was obtained, with a linear dynamic range of 0.1 nM to 10 nM. We showed that this method has a low pM limit of detection (LOD 98.2 pM) and it is also very sensitive to the mismatch type in a specific DNA sequence. In addition, it avoids rigorously controlled temperature, complex synthesis of the fluorescent probe and prelabeling of DNA strands and eliminates the use of sophisticated experimental techniques and equipment. Armed with these intriguing properties, the proposed system could provide an efficient tool for early diagnosis and risk assessment of malignancy.

Abstract:Novel luminescent silver nanoclusters (AgNCs) were synthesized utilizing DNA as templates by a simple, rapid and one-pot procedure. Luminescence studies indicated that these DNA-AgNCs exhibited strong emission with peak maximum at 624 nm. The fluorescence of the DNA-AgNCs was found to be quenched by Cu2+ enabling its detection with high sensitivity and selectivity.

Abstract:In the past few years, fluorescent gold nanoclusters (AuNCs) have gained much attention in many areas of physics, chemistry, materials science, and biosciences due to their unique physical, electrical, and optical properties. Herein, we reported for the first time the synthesis of water soluble, monodispersed AuNCs by using methionine both as a reductant and a stabilizer. The synthetic process is green and simple, and the resulting AuNCs capped by methionine (Met-AuNCs) would be biocompatible with bioorganisms. UV-visible absorption, photoluminescence, X-ray photoelectron spectroscopy (XPS), and Fourier transform infrared (FTIR) were carried out to demonstrate the chemical composition and optical properties of the as-prepared Met-AuNCs. The Met-AuNCs possess many attractive features including intense yellow fluorescence (emission maximum at 530nm), a long fluorescence lifetime (181ns and 1651ns), high colloidal stability (pH-, temperature-, salt- and time-stability), and a large Stoke's shift (110nm), holding great promise as late-model analytical tools for life science and environmental studies. Moreover, the as-synthesized Met-AuNCs can serve as an efficient fluorescent probe for selective detection of Cu(2+) by fluorescence quenching. The limit of detection for Cu(2+) was determined to be 7.9nM and linear response over the Cu(2+) concentrations range from 50nM to 8μM. Furthermore, the new-constructed probe allows simple and rapid detection of the concentrations of Cu(2+) in soil, with results demonstrating its great feasibility for the determination of copper in real samples.

Abstract:Because of bioaccumulation of food chain and disability of biodegradation, concentration of toxic mercury ions (Hg2+) in the environment dramatically varies from picomolar to micromolar, indicating the importance of well-performed Hg2+ analytical methods. Herein, reticular DNA is constructed by introducing thymine (T)-Hg2+-T nodes in poly(T) DNA, and copper nanoclusters (CuNCs) with aggregate morphology are prepared using this reticular DNA as a template. Intriguingly, the prepared CuNCs exhibit enhanced fluorescence. Meanwhile, the reticular DNA reveals evident resistance to enzyme digestion, further clarifying the fluorescence enhancement of CuNCs. Relying on the dual function of DNA manipulation, a high signal-to-noise ratio biosensor is designed. This analytical approach can quantify Hg2+ in a very wide range (50 pM to 500 mu M) with an ultralow detection limit (16 pM). Besides, depending on the specific interaction between Hg2+ and reduced L-glutathione (GSH), this biosensor is able to evaluate the inhibition of GSH toward Hg2+. In addition, pollution of Hg2+ in three lakes is tested using this method, and the obtained results are in accord with those from inductively coupled plasma mass spectrometry. In general, this work provides an alternative way to regulate the properties of DNA-templated nanomaterials and indicates the applicability of this way by fabricating an advanced biosensor.

Abstract:A facile strategy for designing a glutathione-capped Mn-doped CdS/ZnS/CdS core/shell/shell QD-based fluorescent nanosensor has been developed for rapid, selective, and ultrasensitive detection of Cu2+. In this nanosensor, glutathione provided the binding and recognition sites for Cu2+, accumulating Cu2+ on the QD surface. The energy transfer pathway from the host to the Mn dopant was sensitive to the interferences from non-radiative recombination pathways caused by Cu2+, resulting in photoluminescence quenching. The nanosensor responded to Cu2+ within one minute and exhibited good selectivity to Cu2+ over other metal ions and good linear correlation over the concentration range of 1 nM to 100 nM, with a detection limit as low as 0.74 nM. Moreover, this nanosensor avoids the self-quenching problem and autofluorescence in biosystems due to the substantial Stokes shift and long lifetime, and thus can be used to monitor Cu2+ in living cells.

Abstract:Due to the excellent characteristics, fluorescent copper nanoclusters (Cu NCs) have aroused great interest in recent years. Herein, the simple prepared, environmentally friendly fluorescent Cu NCs were synthesized by using trypsin as the stabilizer and applied for the determination of tetracycline. Uniformly dispersed Try-Cu NCs were obtained with average size of 3.5 ± 0.3 nm and some excellent merits of good water solubility, UV light stability and salt stability. Emission peaks around 460.0 nm were visibly quenched by tetracycline based on static quenching mechanism and inner filter effect (IFE). Two excellent linear relationships were observed between ln(F0/F) and tetracycline concentrations in the range of 1-100 μM and 100-300 μM with limit of detection (LOD) of 0.084 μM. Meanwhile, this nanoprobe exhibited an apparent selectivity for tetracycline detection. Moreover, Try-Cu NCs were successfully employed to determine tetracycline in serum and milk samples after facile pretreatment with satisfactory recovery rates and credible standard deviation. The results suggested that this as-prepared Try-Cu NCs had excellent application prospects in the future.
