Abstract:Herein, we demonstrate a facile, straightforward and green method for synthesis of highly red-emitting fluorescent bimetallic Au/Ag nanoclusters (NCs) employing lipoic acid as both a reducing agent and a protecting agent. The as-prepared Au/Ag NCs exhibited bright red emission with a strong peak centered at 630 nm (quantum yield measured as 6.4%), and showed excellent stability and remarkable temperature-dependent fluorescent property. The fluorescence responses were reversible and linear to the environment temperature from 20 to 65 C with a resolution as high as 1.34 degrees C, which allows its potential application as a fluorescent nanothermometer. Moreover, Au/Ag NCs were also explored for selective and sensitive determination of Fe3+ based on the specific quenching of the fluorescence by Fe3+, and explored for fluorescence imaging Fe3+ in living HeLa cells thanks to their very low cytotoxicity. Abstract:Techniques for detecting glucose are developing at a breathtaking speed because diabetes mellitus can cause many serious complications, such as blindness, high blood pressure heart disease and kidney failure. Herein, water soluble NaYF 4 :Eu 3+ @Ag core-shell nanocrystals for glucose detection with lower detection limit have been successfully developed, using NaYF 4 :Eu 3+ cores as the energy donors and Ag shells as the efficient quenchers through energy transfer. After immobilization of glucose oxidase (GOx) on the surface of NaYF 4 :Eu 3+ @Ag core-shell nanocrystals, the Ag shells can be decomposed in the presence of glucose, accompanied by down-shifting luminescence recovery. The limit of detection of NaYF 4 :Eu 3+ @Ag was 0.12 μmol L −1 . Therefore, the NaYF 4 :Eu 3+ @Ag can be easily extended to the detection of a variety of H 2 O 2 -involved analytes.

Abstract:A fluorescent-colorimetric dual-signal platform, N, S co-doped carbon dots functionalized silver nanoparticles (NS-CDs-AgNPs), was designed in situ by reducing AgNO3 in the presence of N, S co-doped carbon dots (NS-CDs) under the assistance of microwave irradiation for glucose determination. With the formation of silver nanoparticles (AgNPs), the intrinsic fluorescence of NS-CDs was quenched, showing the fluorescence state was off. Whereas the fluorescence of NS-CDs can be switched on when a trace amount of H2O2 was added. Based on this novel phenomenon, the peroxidase-like activity of NS-CDs-AgNPs by using 3,3',5,5'-tetramethylbenzidine (TMB) chromogen and H2O2 as substrates was evaluated. The Km values of the prepared probe for H2O2 and TMB were 0.84 mM and 0.01 mM with the Vm of 6.65×10-8 M·S-1 and 3.01×10-8 M·S-1, respectively. The results showed that NS-CDs-AgNPs had good peroxidase-like activity and strong affinity to TMB and H2O2. It confirmed that there is a redox interaction between AgNPs and H2O2, and H2O2 can oxidize Ag to produce Ag+, which is the main reason that the fluorescence of NS-CDs-AgNPs can be activated by H2O2. The hydroxyl radical (·OH) was formed in the process of reaction, which can further oxidize TMB for color reaction. Meanwhile, glucose can be oxidized to produce H2O2 in the presence of glucose oxidase (GOx). Based on the phenomenon, a fluorimetric and colorimetric dual-mode sensor for glucose detection was established. Satisfactory results were obtained with the linear range of 0.1-80 μM for fluorimetric mode and 0.5-5 μM for colorimetric mode, respectively. Additionally, the LOD was below 0.32 μM and 0.21 μM, respectively. The method was successfully applied to determine the glucose in human serum with satisfactory recovery and RSD.

Abstract:The accurate detection of glucose at cellular level remains a big challenge. In this study, a signal amplification strategy mediated by silver nanocube (AgNC), glucose oxidase (GOx), and silver ion fluorescence probe (denoted as AgNC-GOx/Ag+-FP) is proposed for amplified intracellular glucose detection. The AgNC is oxidized into Ag+ by H2O2 generated from GOx-catalyzed glucose oxidation reaction, and Ag+ remarkably enhances the red fluorescence of Ag+-FP. Our results show that AgNC-GOx/Ag+-FP is highly sensitive and specific to glucose and H2O2. Afterward, the feasibility of using AgNC-GOx/Ag+-FP to detect intracellular glucose is verified in five different cell lines. In summary, a sensitive and specific fluorescence amplification strategy has been developed for intracellular glucose detection.

Abstract:In this work, an efficient and sensitive fluorometric sensor was developed to detect silver ions (Ag+). It is based on the cytosine–Ag+–cytosine (C–Ag+–C) structure via a dual-signal amplification strategy using glucose oxidase (GOx) and the hybridization chain reaction (HCR). A silver-coated glass slide (SCGS) acts as an ideal material for separation. Cytosine rich (C-rich) capture DNA (C-DNA) assembled themselves on the SCGS via Ag–S bonds and hybridized with signal DNA (S-DNA) to trigger the HCR. With specific base-pairing, the S-DNA and HCR products bind on the SCGS. Then, the GOx–biotin–streptavidin (SA) complexes bind to the HCR products through SA–biotin interactions. Owing to the formation of a particular C–Ag+–C structure between two neighboring C-rich C-DNA on the SCGS, the C-DNA/S-DNA/HP1-GOx/HP2-GOx complex gradually moved away from the SCGS as the concentration of Ag+ increased and the combined GOx fell into the buffer. H2O2 could be generated during the oxidation of glucose, catalyzed by GOx in the buffer. Afterward, H2O2 could oxidize the substrate (3-(p-hydroxyphenyl)-propanoic acid) when Horseradish peroxidase was present, giving rise to blue fluorescence. The proposed strategy reached a limit of detection (LOD) of 1.8 pmol L−1 with a linear detection range of 5 to 1000 pmol L−1 for Ag+. Moreover, this assay has been commendably used for the detection of Ag+ in actual samples with fairly good results.

Abstract:In this work, biomolecule-stabilized Au nanoclusters were demonstrated as a novel fluorescence probe for sensitive and selective detection of glucose. The fluorescence of Au nanoclusters was found to be quenched effectively by the enzymatically generated hydrogen peroxide (H2O2). By virtue of the specific response, the present assay allowed for the selective determination of glucose in the range of 1.0×10−5M to 0.5×10−3M with a detection limit of 5.0×10−6M. The absorption spectroscopy, X-ray photoelectron spectroscopy (XPS) and fluorescence decay studies were then performed to discuss the quenching mechanism. In addition, we demonstrated the application of the present approach in real serum samples, which suggested its great potential for diagnostic purposes.

Abstract:Herein, we report the design of a single-excitation/double-emission ratiometric fluorescence nanosensor for the determination of glucose. The sensing system combines glucose oxidation catalyzed by glucose oxidase, Fenton chemistry, Fe3+-sensitive fluorescent gold nanoclusters (AuNCs), and Fe3+-inert fluorescent graphene quantum dots (GQDs). We used orange-fluorescent AuNCs co-modified with bovine serum albumin and 3-mercaptopropionic acid as the indicator probe, and GQDs with the same excitation wavelength as the BSA/MPA-AuNCs, but with different emission wavelength, as the reference probe. The fluorescence intensity-ratio between 420 nm and 575 nm (F420/F575) was used to quantitatively determine glucose with a low detection limit of 0.18 μM, and the nanosensor was successfully used to detect glucose in human serum. This ratiometric fluorescence sensing system, based on AuNCs and GQDs, ensures sensitive and convenient determination of glucose, and has broad application prospects for biomedical-analysis applications.

Abstract:We present a new glucose oxidase (GOx)-mediated strategy for detecting glucose based on carbon nanodots supported on silver nanoparticles (C-dots/AgNPs) as nanocomplexes. The strategy involves three processes: quenching of C-dots' fluorescence by AgNPs, production of H2O2 from GOx-catalyzed oxidation of glucose, and H2O2-induced etching of AgNPs. In the C-dots/AgNPs complex, AgNPs act as a "nanoquencher" to decrease C-dots fluorescence by surface plasmon-enhanced energy transfer (SPEET) from C-dots (donor) to AgNPs (acceptor). The H2O2 formed by GOx-catalyzed oxidation of glucose etches the AgNPs to silver ions, thus freeing the C-dots from the AgNPs surfaces and restoring the C-dots' fluorescence. Therefore, the increase in fluorescence depends directly on the concentration of H2O2, which, in turn, depends on the concentration of glucose. The strategy allows the quantitative analysis of glucose with a detection limit of 1.39 μM. The method based on C-dots/AgNPs offers the following advantages: simplicity of design and facile preparation of nanomaterials, as well as low experimental cost, because chemical modification and separation procedures are not needed.

Abstract:In this paper, a novel fluorescent detection method for glucose and lactic acid was developed based on fluorescent iron nanoclusters (Fe NCs). The Fe NCs prepared using hemin as the main raw material exhibited excellent water solubility, bright red fluorescence, and super sensitive response to hydrogen peroxide (H2O2). This paper demonstrates that Fe NCs exhibit excellent peroxide-like activity, catalyzing H2O2 to produce hydroxyl radicals (•OH) that can quench the red fluorescence of Fe NCs. In this paper, a new type of glucose sensor was established by combining Fe NCs with glucose oxidase (GluOx). With the increase in glucose content, the fluorescence of Fe NCs decreases correspondingly, and the glucose content can be detected in the scope of 0–200 μmol·L−1 (μM). Similarly, the lactic acid sensor can also be established by combining Fe NCs with lactate oxidase (LacOx). With the increase in lactic acid concentration, the fluorescence of Fe NCs decreases correspondingly, and the lactic acid content can be detected in the range of 0–100 μM. Furthermore, Fe NCs were used in the preparation of gel test strip, which can be used to detect H2O2, glucose and lactic acid successfully by the changes of fluorescent intensity.

Abstract:The fluorescence of nanoparticles has attracted much attention in recent research, but in many cases the underlying mechanisms are difficult to evaluate due to the polydispersity of nanoparticles and their unknown structures, in particular the surface structures. Recent breakthroughs in the syntheses and structure determinations of well-defined gold nanoclusters provide opportunities to conduct in-depth investigations. Devising well-defined nanocluster sensors based on fluorescence change is of particular interest not only for scientific studies but also for practical applications. Herein, the potential of the glutathionate (SG)-capped Au25 nanocluster as a silver ion sensor is evaluated. The Ag+ detection limit of approximately 200 nM, based on the fluorescence enhancement and good linear fluorescence response in the silver ion concentration range from 20 nM to 11 mu M, in combination with the good selectivity among 20 types of metal cations, makes Au25(SG)18 a good candidate for fluorescent sensors for silver ions. Further experiments reveal three important factors responsible for the unique fluorescence enhancement caused by silver ions: 1) the oxidation state change of Au25(SG)18; 2) the interaction of neutral silver species (Ag0, reduced by Au25(SG)18-) with Au25(SG)18; and 3) the interaction of Ag+ with Au25(SG)18. Experiments demonstrate the very different chemistry of hydrophobic Au25(SC2H4Ph)18 and hydrophilic Au25(SG)18 in the reaction with silver ions. This work indicates another potential application of gold nanoclusters, offers new strategies for nanocluster-based chemical sensing, and reveals a new way to influence nanocluster chemistry for potential applications.

Abstract:A fiber-optic surface plasmon resonance (SPR) biosensor based on a silver-coated hollow fiber (HF) structure for glucose detection is presented. The sensor surface was immobilized with 4-mercaptophenylboronic acid (PMBA) acting as a glucose recognition monolayer. Then, gold nanoparticles (AuNPs) modified with 2-aminoethanethiol (2-AET) and PMBA were introduced onto the sensor surface after glucose was captured to enhance the wavelength shift of the SPR phenomenon excited by the light transmitted in the wall of the HF sensor. Instead of the conventional one-step sensitization pretreatment commonly used in the deposition process of silver films for fiber-optic SPR sensors, a sensitization-activation two-step activation method was adopted in the fabrication of the proposed sensor. Experiments for glucose detection were performed on the fabricated sensors in the concentration range of 1 nM-1 mM. Results showed that the sensor fabricated by the two-step activation method has a much larger shift of resonance wavelength than the sensor fabricated using the one-step sensitization method. The resonance wavelength shift was found to be linear to the logarithm of the concentration in the range of 1 nM-1 mM. The sensor achieved a limit of detection (LOD) of as low as 1 nM, which is at least an order of magnitude lower than that of other fiber-optic sensors for glucose detection reported previously. The presented HF glucose sensor has the potential for biosensing applications and provides a large reference value in the study of optical fiber SPR sensors for biosensing.

Abstract:We developed a simple, non-enzymatic approach for the colorimetric detection of glucose based on a gold nanoparticles (Au NPs) assisted silver mirror reaction (AuSMR). The linear range of the concentration of glucose is from 0.04 mM to 1 mM, and the lowest concentration that can be distinguished by the naked eye is 10 nM. This approach has been successfully used for detecting glucose in serum.

Abstract:A non-enzymatic copper-doped zinc oxide glucose sensor, which is covered with silver nanowires (AgNWs) and modified with Nafion, can be employed to measure glucose with a concentration of 2–14 mM. In this study, X-ray photoelectron spectroscopy (XPS) was used to analyze the film of the working electrodes, and the AgNWs on the electrode surface were analyzed by a field-emission scanning electron microscope (FE-SEM). Also, the AgNW/CZO structure was investigated through the use of an atomic force microscope (AFM) and an ultraviolet/visible (UV/Vis) spectrophotometer. Finally, we used electrochemical impedance spectroscopy (EIS) and the voltage–time (V–T) measurement system to measure the sensing characteristics of the thin film. The potentiometric non-enzymatic glucose sensor has the average sensitivity of 10.31 mV/mM, and the linearity is 0.997. Also, the sensor has the response time with 15 s, and the interference experiment proved that it has good anti-interference and excellent repeatability (relative standard deviation (RSD) = 3%). Besides, the sensor still maintains a certain degree of average sensitivity at high temperatures, and it has the limit of detection (LOD) with 0.272 mM.
