Abstract:Asymmetric synthesis of 3-hydroxyl-2-alkanones is achieved via one-pot, tandem organocatalytic aminoxylation of aldehydes and chemoselective CH2N2-induced homologation. Accelerating effect of water is observed in α-aminoxylation. MgCl2, a Lewis acid additive, improves the chemoselectivity of the diazomethane homologation to 6:1 in favor of ketone.

Abstract:A one-pot approach has been developed for the synthesis of α-ketothioamide derivatives from sulfur ylides, nitrosobenzenes, and thioacetic acid. This protocol is carried out under mild reaction conditions in generally moderate to excellent yields without any precious catalysts, affording the derivatives with structural diversity. Additionally, a possible mechanism for this chemical transformation is proposed.

Abstract:A strategy for the synthesis of alpha-alkylated ketones via one-pot, sequential, catalytic hydration of alkynes and alpha-alkylation with alcohols was proposed and successfully accomplished. In the presence of chloro[1,3-bis(2,6-diisopropylphenyl) imidazol-2-ylidene] gold(I) ([(IPr) AuCl])/silver triflate (AgOTf) and water, a series of alkynes are first hydrated to give the corresponding methyl ketones, which in turn are alpha-alkylated by adding pentamethylcyclopentadienyliridium-(III) chloride ([Cp*IrCl2](2))/KOtBu and alcohols to the reaction mixture. Notably, this reaction is highly attractive because of easily available starting materials, high atom efficiency, good to excellent yields, and minimal consumption of chemicals and energy. The present research will also facilitate the development of one-pot, sequential, catalytic reactions based on "hydrogen autotransfer" (or "hydrogen-borrowing") processes.

Abstract:Three-component reaction between aryl glyoxals, aryl amines, and guanidine afforded N , N -di ( α -amino aryl ketone) derivatives. This method include two C–N bonds formation in a one-pot and single synthetic step in neutral and simple reaction conditions. All reactions were conducted in EtOH as solvent at reflux conditions without using any catalyst, and the stable products were obtained in high yields. The structures of all products were proved by 1 H and 13 C NMR, IR spectral and elemental analysis data.

Abstract:A three-step, one-pot tandem reaction including radical nucleophilic alkylation/cyclization/aromatization was developed using 0.3 equivalents of silver(I) acetate (AgOAc) as the catalyst and 2 equivalents of ammonium persulfate as the oxidant. This strategy is highly efficient for the assembly of pentacyclic complex carbazoles from aryl-fused bromobenzoquinones and indol-3-ylpropanoic acid acids in 52-72% overall yields (three steps). This new approach provides a significant improvement over the previously reported methods and would greatly facilitate analog library construction of pentacyclic complex carbazoles and benefit further biological evaluation of these compounds.

Abstract:A cascade Wolff rearrangement and double N‐acylation reaction has been developed for the synthesis of fused pyrimidinediones in one‐pot under air. These reactions were performed with α‐diazoketones and 2‐aminopyridines or 3‐aminopyrazoles under catalyst‐ and additive‐free conditions. The protocol avoided traditional purification procedures, such as organic solvent extraction and column chromatography. The synthesis of biologically active molecules and gram‐scale experiment demonstrated the potential applications of this reaction.

Abstract:Synthesis of α-heterosubstituted ketones was achieved through sulfur mediated difunctionalization of internal alkynes in one-pot. The reaction design involves: phenyl substituted internal alkyne attacking the triflic anhydride activated diphenyl sulfoxide to give a sulfonium vinyl triflate intermediate, hydrolysis to give the α-sulfonium ketone, and then substitution with various nucleophiles. This method provides a unified route to access α-amino ketones, α-acyloxy ketones, α-thio ketones, α-halo ketones, α-hydroxy ketones, and related heterocyclic structures, in a rapid fashion.

Abstract:A novel and green approach for the efficient synthesis of β-amido ketones using VB1–Al2O3 as a heterogeneous catalyst for the first time from an aldehyde, enolizable ketones, and an amide in EtOH is described. The present methodology has several advantages from the economical and environmental points of view, such as simple procedure, low catalyst loading, high atom economy, and good yields.

Abstract:In this paper, a novel cascade reaction of caesium carbonate-promoted Michael addition and lactonization for the one-pot synthesis of 3-alkyl-3- N -substituted aminobenzofuran-2(3 H )-one derivatives has been established based on the screening of the alkaline reagents and optimization of reaction conditions, in which the N -substituted ( ortho -hydroxy)aryl glycine esters were used as the Michael donors to react with different α , β-unsaturated carbonyl compounds. In the case of using the asymmetric starting material, the epimers could be successfully separated by conventional chromatography. In addition, plausible mechanisms were suggested and the absolute configuration of the epimer was analysed. All the chemical structures of unreported benzofuran-2(3 H )-one derivatives were characterized by 1 H nuclear magnetic resonance (NMR), 13 C NMR, IR and high-resolution mass spectrometry (HRMS).

Abstract:We have developed a palladium‐catalyzed cross‐coupling reaction of isocyanides with α‐diazoacetates to form active ketenimines and following a 1,4‐diazabicyclo[2.2.2]octane (DABCO) catalyzed aza‐Mannich type reaction with various 2‐mercaptoimidazoles for the synthesis of 1,3‐bis(β‐aminoacrylate)‐substituted 2‐mercaptoimidazole derivatives with structural diversity. In addition, the use of 1H‐benzo[d]imidazol‐2‐ol as the reaction partner also allowed the formation of 1,3‐bis(β‐aminoacrylate)‐substituted 2‐benzimidazolinone derivatives with moderate yields (34‐52%). In the aza‐Mannich reaction, the regioselective N‐attack rather than S‐attack or O‐attack at the electrophilic central carbon of ketenimines was observed. This tandem sequence is efficient since two C=C and two C‐N bonds are consecutively created in one‐pot.

Abstract:The chemoselective [4+2] annulation/aromatization reactions between benzofuran‐derived azadienes and N‐Ts cyanamides are developed, affording a convenient method for synthesizing benzofuro[3,2‐d]pyrimidin‐2‐amines under mild conditions. Herein, N‐Ts cyanamides selectively participated in reactions absolutely via carbodiimide anion intermediates and the corresponding cyanamide anion intermediates derived products were not observed. The proposed chemoselective stepwise reaction mechanism was well supported by DFT calculations.

Abstract:2-Aryl-3-alkoxycarbonyl chromones were effectively constructed from aryl aldehydes and 3-(2-(methoxymethoxy)phenyl)propiolates via a cascade Lewis acid catalyzed phenol ether deprotection/aldehyde olefination/intramolecular oxa-Michael addition reaction, and a sequential oxidation. Abstract:A novel one-pot synthesis of ,-unsaturated methyl ketone/nitro compounds from benzylic amines through an oxidation-aldol/Henry reaction is reported. The reaction proceeded well by using MCPBA as oxidant and CuCl(2)2H2O as catalyst. A variety of functionalized ,-unsaturated methyl ketone/nitro compounds were assembled in moderate yields by application of this catalytic one-pot reaction.

Abstract:An efficient copper-catalyzed strategy for the synthesis of α-ketoamides via cross-coupling of methyl ketones and pyridin-2-amines is described. This transformation has provided a simple process for the formation of C−N and C=O bonds to prepare α-ketoamides, which are important substrates and intermediates for the preparation of fine chemicals. The reaction mechanism is investigated, which suggests that the reaction proceeds via a radical pathway.
