Abstract:The radical aryl migration reactions provide a synthetic methodology capable of constructing interesting building blocks that are not readily accessible via traditional processes. However, efficient methods to perform radical 1,4-aryl migration in stereoselective manner are fully challenging, and particularly functionalized starting materials and harsh reaction conditions are usually required, much less asymmetric catalytic versions. Here, we reported the breakthrough in diastereo- and enantioselective catalytic aerobic oxidative radical addition/hydroxylation/1,4-aryl migration processes from bisaryl-substituted ethenes and indanone derivatives. We disclosed that the combination of CuI/O-2 oxidative system and chiral Lewis acid-involved asymmetric catalysis synergistically enabled the direct generation of optically enriched 3-arylated indanone derivatives under mild reaction conditions. The aerobic oxidative addition provided a straightforward and in situ generation of diaryl alcohol and distal radical for 1,4-aryl migration. An easily available chiral magnesium complex catalyst could relay chiral information in radical addition and 1,4-aryl migration with high diastereo- and enantioselectivity.

Abstract:Radical difunctionalization of aliphatic alkynes provides direct access to valuable multi-substituted alkenes, but achieving a high level of chemo- and stereo-control remains a formidable challenge. Herein we report a novel photoredox neutral alkyne difunctionalization through functional group migration followed by radical-polar crossover and energy transfer-enabled stereoconvergent isomerization of alkenes. In this sequence, a hydroxyalkyl and an aryl group are incorporated concomitantly into an alkyne, leading to diversely functionalized E-allyl alcohols. The scope of alkynes is noteworthy, and the reaction tolerates aliphatic alkynes containing hydrogen donating C-H bonds which are prone to intramolecular hydrogen atom transfer. The protocol features broad functional group compatibility, high product diversity and exclusive chemo- and stereoselectivity, thus providing a practical strategy for the elusive radical difunctionalization of unactivated alkynes.

Abstract:A visible‐light induced radical 1,2‐aryl migration of α,α‐diarylallyl alcohols was developed under mild and metal‐free conditions. Commercially available CHCl3 and CH2Cl2 were used as tri‐ and dichloromethyl radical precursors. Structurally diverse β‐polochloromethylated ketones were obtained in good to moderate yields via neophyl‐type rearrangement. Besides, the polychloromethyl group can be easily transformed into other useful functional groups.

Abstract:The direct alkylation of the α-position of aldehydes is an effective method for accessing a wide range of structurally diverse aldehydes, yet tert-alkylation has proven to be a challenging task. In this study, we present a novel radical-mediated tert-alkylation approach targeting the α-position of aldehydes, enabling the synthesis of complex aliphatic aldehydes. The transformation is initiated by the interaction between an in situ generated enamine intermediate and α-bromo sulfone, forming an electron donor-acceptor (EDA) complex, followed by consecutive 1,4- and 1,3-functional group migrations. This protocol operates under metal-free and mild photochemical conditions, delivering a broad scope of products and providing new mechanistic insights into radical rearrangement reactions.

Abstract:Abstract Currently, the selective activation of C(sp 3 )–F bonds and C–C bonds constitute one of the most widely used procedures for the synthesis of high-value products that range from pharmaceuticals to agrochemical applications. While numerous examples of these two methods have been reported in their respective fields, the processes which merge the activation of both single C(sp 3 )-F bonds and C–C bonds in one step still remain elusive. Here, we demonstrate the controllable defluoroalkylation–distal functionalization of trifluoromethylarenes with unactivated alkenes via distal heteroaryl migration. This is proposed to proceed via tandem C(sp 3 )–F and C–C bond cleavage using visible-light photoredox catalysis combined with Lewis acid activation. This strategy provides facile and flexible access to multiply functionalized α , α -difluorobenzylic ketones in useful yields (up to 88%) under mild conditions. The products can be further transformed into other valuable compounds, demonstrating the method’s utility.

Abstract:Heteroarenes are important units in organic chemistry and are ubiquitous in natural products, pharmaceuticals, and numerous artificial molecules. Despite great efforts devoted to accessing heteroarenes, the development of new methods to efficiently produce heteroarenes remains a long-term interest. Abstract:Herein we report a "carbonylative migration" strategy for the acylation‐esterification type double functionalization of unactivated alkenes using alkyloxalkyl chlorides and CO as the reagents. The transformation is proceeded by the alkoxycarbonyl radical addition to unactivated alkenes, followed by the insertion of carbon monoxide to induce intramolecular migration of heteroaryl groups, which is different from the traditional reaction modes. The reaction conditions were mild and well tolerated with varieties of functional groups. A variety of 1,4‐dicarbonyl compounds with different ester groups were produced easily which has high potential applications in biology, medicine, and other fields.

The migration is triggered by various fluoroalkyl radicals, leading to valuable multifunctionalized ketones. The comparisons of migratory aptitude for O-/S-containing heteroaryls are comprehensively investigated. The origin of the chemoselective migration could be partially attributed to the discrepancy in the energy level of the LUMO of each heteroaryl group.

Abstract:AbstractWe report an unprecedented, efficient nickel‐catalysed radical relay for the remote cross‐electrophile coupling of β‐bromo‐α‐benzylamino acid esters with aryl bromides via 1,4‐aryl migration/arylation cascades. β‐Bromo‐α‐benzylamino acid esters are considered as unique molecular scaffolds allowing for aryl migration reactions, which are conceptually novel variants for the radical Truce–Smiles rearrangement. This reaction enables the formation of two new C(sp3)−C(sp2) bonds using a bench‐stable Ni/bipyridine/Zn system featuring a broad substrate scope and excellent diastereoselectivity, which provides an effective platform for the remote aryl group migration and arylation of amino acid esters via redox‐neutral C(sp3)−C(sp2) bond cleavage. Mechanistically, this cascade reaction is accomplished by combining two powerful catalytic cycles consisting of a cross‐electrophile coupling and radical 1,4‐aryl migration through the generation of C(sp3)‐centred radical intermediates from the homolysis of C(sp3)−Br bonds and the switching of the transient alkyl radical into a robust α‐aminoalkyl radical.

Abstract:Functional group translocation is undoubtedly a pivotal synthetic transformation in organic chemistry. Numerous types of reactions involving radical 1,2-aryl or 1,4-aryl migration via electron transfer mechanism have been extensively investigated. Nevertheless, energy-transfer enabled 1,4-arylation remains unknown. Herein we disclose that an unprecedented di-π-ethane rearrangement featuring 1,4-aryl migration facilitated by energy transfer catalysis under visible light conditions. The newly developed mild protocol exhibits tolerance towards diverse functional groups and enables the migration of a multitude of aromatic rings, encompassing both electron-withdrawing and electron-rich functional groups. The open-shell strategy has also found successful application in the modification of several drugs. Large-scale experiments, continuous-flow experiment, and versatile manipulation of products have demonstrated the robustness and potential utility of this synthetic method. Preliminary mechanistic studies have supported the involvement of radical species in this di-π-ethane rearrangement and have also provided evidence for the energy transfer mechanism.

Abstract:Herein, a palladium-catalyzed 1,3-alkynyl migration of allylic alcohol for the synthesis of β-alkynyl ketone was described. This intramolecular rearrangement reaction demonstrated an enhanced reactivity compared to the traditional intermolecular alkynylation by circumventing the dimerization of alkynes, exhibiting a specific selectivity toward β-alkynyl elimination. Moreover, this reaction featured wide substrate scope, good functional group tolerance, and 100% atom economy.

Abstract:Comprehensive Summary α‐Fluoroalkyl (Rf) alcohols are privileged motifs in drugs and pharmaceutically active compounds. As such, it is highly desirable to develop efficient methods for assembling these scaffolds. Herein, a visible‐light‐induced cascade radical cyclization of alkenyl fluoroalkyl ketones is developed, producing a variety of decorate α‐Rf cycloalkanols in promising yields with up to >20:1 dr selectivity. A radical chain mechanism involving an intramolecular radical addition to the α‐Rf carbonyl group and a subsequent intermolecular hydrogen atom transfer (HAT) has been proposed. Density functional theory (DFT) calculations indicate that a fluorine effect contributes to the radical addition to carbonyls by lowering the π* (C=O) orbital energy of CORf and energy barrier of the HAT between alkoxy radicals and THF, which may be valuable for controllable transformations of fluorinated molecules. Abstract:AbstractA radical-mediated hetaryl functionalization of nonactivated alkenes through distal ipso-migration of O- or S-containing hetaryls was developed. Furyl, benzofuryl, thienyl, and benzothienyl groups showed satisfactory migratory abilities. A variety of heteroatom-centered radicals, including azido, trifluoromethylsulfanyl, and silyl radicals readily trigger the migration cascade, and a new C–heteroatom and C–C bond are concomitantly constructed in the reaction. This method provides an efficient approach to the synthesis of high-valued complex O- or S-hetaryl compounds.

Abstract:Radical-mediated difunctionalization of alkenes is a powerful tactic for olefin utilization. However, the transformation of unactivated alkenes still remains a formidable challenge. Now a conceptually new docking-migration strategy is presented for the difunctionalization of alkenes with photo-redox catalysis. Both activated and unactivated alkenes are suitable substrates. A vast array of functional groups are compatible with the mild reaction conditions. Heteroaryl and difluoromethyl are concomitantly incorporated into alkenes, leading to synthetically valuable products that are readily converted into a variety of fluorine-containing molecules. The protocol provides a kinetic control of stereoselectivity for cycloalkenes to generate the unusual cis-products, and offers an efficient approach for the late-stage functionalization of complex natural products and drug molecules. A portfolio of dual-function reagents are prepared for the elusive radical difunctionalization of alkenes.
