Effect of bzr1 Gene Transformation on Characters of Fruit of Tomato

Abstract:Objective The transformation of BZR1, a BR signal transduction component to tomato and its effect on characters of fruit of tomato was investigated in present study. Method The bzr1 gene was introduced into tomato cultivar Zhongshusihao via tumefaciens-mediated transformation and confirmed by molecular test and genetic analysis. The content of main quality components and pigments, as well as ethylene evolution rate was analyzed in fruits of transgenic tomato line and non-transgenic line (control) at four typical developmental stages. The expression of BR biosynthetic and regulated genes, as well as ethylene biosynthetic, response, and receptor genes was also analyzed. Result Significantly higher contents of soluble solids, organic acid, soluble proteins, vitamin C, lycopene and total carotenold, as well as the ethylene evolution rate in fruit of turning stage were observed in transgenic tomato line compared with the control. The expression of BR regulated gene LeBR1 was higher in transgenic line than in the control at the stage of seedling and early three developmental stages of fruit. The expression of LeACS4 gene encoding ACC synthase, which catalyzes the rate-limiting step of ethylene biosynthesis, was higher in the fruit of transgenic line than that of control from the stage of mature green to turning. The expression of ethylene response gene ERF1 and receptor gene LeETR1 and LeETR3 in fruit of breaker stage was significantly higher in transgenic line than in the control. Conclusion It was concluded from the study that the transformation of bzr1gene to tomato increased the content of various nutrients in fruit of turning stage, which is the edible part of tomato, and therefore improved the nutrient quality of tomato.

Abstract:Rosaceae fruits are very important for fruit production in China and worldwide.Recently,significant advances in regeneration and genetic transformation of Rosaceae have been achieved and applied to various Rosaceae trees and fruits.However,the factors affecting the process of regeneration and transformation have not been reviewed in detail.This paper reviews recent progress in the regeneration pathway,factors affecting regeneration,genetically modified species and main traits,and the factors affecting Agrobacterium-mediated genetic transformation.The differences between species of Rosaceae fruits are emphasized,and the trends and areas for future studies are highlighted.
Proteomics as an approach to the understanding of the molecular physiology of fruit development and ripening

Abstract:Fruit ripening is a developmental complex process which occurs in higher plants and involves a number of stages displayed from immature to mature fruits that depend on the plant species and the environmental conditions. Nowadays, the importance of fruit ripening comes mainly from the link between this physiological process in plants and the economic repercussions as a result of one of the human activities, the agricultural industry. In most cases, fruit ripening is accompanied by colour changes due to different pigment content and increases in sugar levels, among others. Major physiological modifications that affect colour, texture, flavour, and aroma are under the control of both external （light and temperature） and internal （developmental gene regulation and hormonal control） factors. Differential proteomics of immature and mature fruits would be a useful tool to gain information on the molecular changes which occur during ripening, but also the investigation of fruits at different ripening stages will provide a dynamic picture of the whole transformation of fruits. Thus far different maturation profiles have been reported specific for each crop species. In this work, a thorough review of the proteomic database from fruit development and maturation of important crop species will be updated to understand the molecular physiology of fruits at ripening stages.

Abstract:Field investigation was conducted in Huarong County,Anxiang County,Datonghu District and Li County of Hunan Province from 2004 to 2005 to analyze the characteristic of yield constitution and the percentage of total seed-cotton yield by fruiting branch according to different types of growth and development of transgenic hybrid cotton.The different growth and development types were studied and classified into normal type,early-development and early-aging,early-development and early-maturing,late-development and late-maturing,two phase-growth.Six simulation models of percentage of total yield by fruiting branch corresponding to the different growth type were developed.Six simulation models were exponential-decrease,positive-skewness,negative-skewness,normal-distribution,double-peak and fluctuation-distribution,respectively.Test plots were conducted to study different growth and development types in the four counties and Hunan Agricultural University from 2006 to 2009.Models were validated by the data collected from test plots in different survey sites in different years.Models matched the different growth and development types well,and the simulated values agreed well with the observed values.The result showed that the model was accurate and applicable under different conditions,with the root mean square errors(RMSE) between simulated and observed values of the six simulation models varied from 0.3419 to 0.7092.

Abstract:There are some problems in the current land requisition, which cause rapid decrease of farmland, and the guarantee problem of the farmers' benefits. This paper sets up a model to analyze the land price and the distributing of the welfare and the lucre in the process of farmland conversion. Then the writer analyzes China land requisition system by the model, and gives some suggestion to innovate the current land requisition system.

Abstract:The factors influencing the quantity of cultivated land are many and c omplex,so a satisfactory model is hard to make by adopting the common regression method. Taking Sichuan Province as an example, this paper conducts a quantitati ve analysis of the factors which influence the quantity of cultivated land and c oncludes that there are four main factors influencing the quantity of cultivated land in Sichuan Province, namely, economic development, influences of policies, natural disasters, and advances of agricultural science and technology. The pap er also point out that policies alone can not solve the problem of cultivated la nd protection. Rather,it's necessary to take measures combining economy, policy , science and technology to achieve the goal.

Abstract:Simulation modeling is a particularly important tool for estimating the impacts of climate change on agricultural productivity,so as to attract much attentions and applications.The development process of modeling of climate change impacts on agricultural productivity was given,including the modeling of climate change impacts on agricultural productivity by changing climate parameters artificlally,linking to climate scenarios,and integrating with other models for compositive simulations.Basing on the analysis on the key issues of making choice between empirical and mechanistic models,field and region scale,determined and probabilistic climate scenarios,the indeterminacy and problems existed and prospects were given.
Development of Multi-Cultivar Models for Predicting the Soluble Solid Content and Firmness of European Pear (pyrus Communis L.) Using Portable Vis–nir Spectroscopy

Abstract:The feasibility of a multi-cultivar model's development for the on-site determination of the internal qualities of European pear (Pyrus communis L.) cv. 'Abate', 'Cascade', 'Conference', 'Red Cornice' and Wujiuxiang' was investigated. In total, 240 pear samples, 48 for each cultivar, were collected during fruit post-ripening period under retail stage conditions to explore the performance of a technique for developing soluble solid content and firmness models with partial least squares (PLS) and multiple linear regression (MLR) methods A commercially available portable visible and near-infrared spectrometer was used to collect the absorbance spectra (500-1010 nm) of pears. A hybrid method, orthogonal signal correction (OSC) combined with other spectral pre-process methods, such as standardization, second-order derivation and Savitzkye-Golay smoothing, was employed to filter out the variations in signals caused by different pear cultivars. Compared with the routine PLS and MLR models, the performances of the OSC PLS and OSC MLR models resulted in higher ratios of prediction to deviation. A new external sample set obtained at the retail stage was used to assess the performances of the best multi-cultivar models of soluble solid content and firmness, which resulted in greater coefficients of determination for prediction of 0.87 and 0.66, respectively, and lower root mean square errors of prediction of 0.45% and 8.11 N, respectively. The results highlighted the potential of portable Vis-NIR instruments for the on site testing of internal quality indices of European pears. The development of a multi-cultivar model for fruit is necessary and significant, in particular for improving the performance of the firmness model.

Abstract:New model industrialization is not just industrial information,but the advancement of national economy structure.China is a large agricultural country,the mission of Chinese industrialization is to achieve modernization of industrialization in agriculture sector.At present,because the inverse structure between the agricultural small department and large amount of farmers,leading to agriculture small-scale production do not adapt to the modern big market,the land high investment do not adjust to the agricultural low productivity,the farmer low income do not fit the national economy high growth.These issues become very sharp.It is fundamental contradiction in the development of Chinese modernization.So we believe that the meaning of new model industrialization with Chinese characteristic should be a fusion of the industry and agriculture,return profit to agriculture,and put industrial capital into agriculture,improve mechanical technology,reform agriculture to the industry mode of production.It means to deepen internal division of labor in agricultural industry,develop agriculture industrialization,improve the agricultural productivity,and push forward national economy being modernized.

Abstract:Apple trees are grown worldwide, and consuming fresh apple fruit is associated with many health benefits. China produces about half of the world's apple supply. However, apple growing in China differs sharply from that in western countries in terms of the prevalent diseases and corresponding management strategies. For instance, family-owned small-scale orchards dominate China's apple industry, and manual bagging of fruit has been a long-standing practice for controlling fruit diseases. In recent years, rural labor shortages have been increasingly challenging the traditional production system, and China's apple industry is experiencing a rapid transition to much larger-scale enterprises featuring high-density orchards with advanced automation and mechanization. Associated with this transition are new challenges and grower demands that are changing the face of apple disease management. This Feature Article summarizes the ongoing transformation of China's apple industry in the context of sustainable disease management.

Abstract:As a type of agricultural robots, fruit-picking robot will have great application potential. Many studies on fruit picking robot working in fields or in greenhouse have been performed abroad in recent years. The paper introduced the design rules of picking manipulator mechanism from aspects of type-synthesis and dimension-synthesis. At the same time, The performance indices including operational space, measure of manipulability, obstacles avoidance, redundant space and posture diversity was analyzed in details along with the algorithm of each performance index, Which provides the important theoretical reference for design and performances evaluation of picking manipulator mechanism. Abstract:Abstract Background There are many studies on business model innovation and evolution in academia, but there are few studies on the specific process of business model evolution and relevant elements of business model innovation under a specific background. It is not clear how to realize the development mechanism of digital agriculture, especially in the context of intelligent agriculture. Combined with the existing literature, this paper believes that the evolution of business model begins with external and internal driving forces, and is divided into different stages according to the changes of the constituent elements of the existing business model. On this basis, the evolution mechanism of smart agriculture business model and its impact on customer emotion regulation are discussed. Research Objects and Methods Using the method of case study, this paper analyzes the market psychology in the development of “Jingdong farm” intelligent agricultural business model. The study found that its development has experienced three stages: preliminary exploration, brand construction and farm construction. This paper makes a psychological analysis from four aspects: value proposition, value creation, value creation and value creation. The value transfer and value acquisition in each stage are carried out in turn. In order to verify the impact of the algorithm on customer emotion, this study uses the self-efficacy scale and the abbreviated teacher self-efficacy scale compiled by Tschannen, Moran and Hoy. The scale has 12 items, including three dimensions: teaching strategy efficiency, student incentive efficiency and class management efficiency. Using Likert's 9-point evaluation method, it gives 1-9 points respectively from “completely unable” to “fully able”. The ol coefficients of each dimension of the original scale were 0.86, 0.81 and 0.86 respectively. In this study, the only coefficients of the three dimensions are 0.84, 0.83 and 0.81 respectively. Results The results show that: first, the value proposition of smart agriculture has changed from bilateral to multilateral, aiming to integrate multilateral resources and provide consumers with better products and services; Second, take “Circulation” as the center and expand to the whole industry chain management mode centered on “production”, “Circulation” and “sales”; Third, intelligent agriculture breaks through the original value boundary and realizes value appreciation by improving agricultural production efficiency and consumers' ability to meet their own value. Conclusion Most of the existing studies solve the production problems of agricultural industry from the perspective of digital technology, and pay less attention to the evolution mechanism of agricultural industry business model. Any technological innovation will inevitably lead to the change of its business model. For example, the birth of Internet technology has changed the traditional business model. The evolution of business model is not a one-time process, but a gradual process of constantly adjusting its own business logic. Therefore, combined with the business model theory, this paper discusses the mechanism of the evolution from traditional agriculture to intelligent agriculture. In view of this, this paper attempts to take Jingdong farm as a typical case, take its development process as a clue, answer the dynamic evolution mechanism of smart agriculture, and then put forward the overall framework of the transformation and upgrading from traditional agriculture to smart agriculture. Abstract:Cucumber is one of the most widely grown vegetables in China and an indispensable fresh fruit in the diet. With the development of society, the demand of people for cucumber quality is higher and higher. Therefore, cultivating high-quality cucumber varieties is one of the main goals of cucumber breeding. With the rapid development of biotechnology such as molecular marker, cucumber quality control network is becoming clear. In this review, we describe the formation mechanism of cucumber fruit quality from three aspects: (1) the commercial quality of cucumber fruit, (2) nutritional quality formation, and (3) flavor quality of cucumber fruit. In addition, the determinants of cucumber fruit quality were summarized from two aspects of genetic regulation and cultivation methods in order to provide ideas for cucumber researchers and cultivators to improve fruit quality.
