Abstract:A general and transition-metal-free method for the construction of the carbazol-4-amine motif via a vinylogous Michael addition/cyclization/isomerization/elimination reaction of 3-nitroindoles with alkylidene malononitriles has been developed. This novel methodology allows the facile synthesis of a series of di- and trisubstituted carbazol-4-amine derivatives in moderate to good yields. A gram-scale experiment was successfully performed, highlighting the practicability of this method. Moreover, this strategy is also applicable to 3-nitrobenzothiophene, affording the corresponding dibenzo[ b, d]thiophen-1-amine derivatives in moderate yields.

Abstract:An unprecedented trifunctionalization of CC bonds in 2-(1-azidovinyl)-1,1 '-biphenyls has been successfully achieved using the NCS/AgNO2 system, leading to the preparation of 6-(dichloro(nitro)methyl)phenanthridines in moderate to good yields. In this process, the NCS/AgNO2 system serves as a NO2 radical initiator as well as a chloro group source. The present protocol is a rare example of the selective construction of densely functionalized phenanthridine derivatives in a one-pot manner.

Abstract:Herein, we report a general and practical nickel-catalyzed deaminative allenylation of amino acid derivatives with terminal alkynes. The well-designed, electron deficient, and sterically hindered amide-type NN2 pincer ligand was crucial to the success of this transformation, enabling the coupling to occur under mild conditions with high efficiency. The remarkable features of this chemistry are its good scalability, its broad substrate scope, functional group tolerance, and the efficient modification of peptides, drugs, and natural products.

Abstract:The transition-metal-catalyzed alkenylation strategy of propargylic alcohol derivatives provides an efficient protocol to access multifunctional products in a double-nucleophilic attack pattern. While limited relevant asymmetric examples have been reported via palladium catalysis, here we first demonstrate that a nonprecious Ni(0)-based chiral complex can efficiently promote the tandem substitution process between propargylic carbonates and N-trifluoroethyl ketimines via consecutive aza-vinylogous activations, finally accomplishing a (3 + 2) annulation reaction to afford products embedding a 4-methylene-3,4-dihydro-2H-pyrrole framework with high regio-, diastereo-, and enantiocontrol. Their assemblies with a few all-carbon-based vinylogous precursors are also successful, and enantioenriched adducts containing a 3-methylenecyclopentene scaffold are furnished effectively. The substitution patterns for both types of substrates are substantial, and an array of synthetic elaborations is conducted to deliver more versatile architectures with high application potential. In addition, density functional theory calculations and control experiments have been conducted to rationalize the catalytic pathways and regio- and enantioselectivity control.

Abstract:A convenient and efficient protocol for the synthesis of phenanthridine-6-carbonitriles has been successfully developed involving a copper(II)-catalyzed tandem annulation of vinyl azides and NaN3 in the presence of PhI(OAc)(2). A variety of functionalized phenanthridine-6-carbonitriles were synthesized in moderate to good yields under mild reaction conditions. Mechanistic study revealed that a radical pathway might be involved in this annulation reaction.

Abstract:A simple protocol to prepare the privileged 2-aminothiazoles promoted by ferrous sulfate heptahydrate via C-N bond formation from vinyl azides and commercially available potassium thiocyanate has been developed. A wide range of vinyl azides are tolerated to afford the expected polysubstituted 2-aminothiazoles in reasonably good yields. The use of the non-toxic substrates and catalyst renders the reaction more environmentally friendly than traditional approaches.

Abstract:Through vicinal difunctionalization of nitriles, a successful, innovative strategy for the synthesis of N-alkyl amidines is developed. This methodology capitalizes on commercially available nitriles, alcohols, and sulfonamides. The activation of alcohols by trifluoromethanesulfonic anhydride (Tf2O) plays a pivotal role throughout the entire reaction process. In comparison to existing amidine synthesis methods, this strategy showcases notable advantages, including the avoidance of toxic and hazardous azides, the exclusion of transition metal catalysts, and the use of mild reaction conditions. Importantly, this methodology provides an effective approach for late-stage modification of complex molecules and drugs, holding promise for the construction of molecular libraries in drug discovery.

Abstract:Azetidines, being four-membered N-heterocycles, possess significant potential in contemporary medicinal chemistry owing to their favorable pharmacokinetic properties. Regrettably, the incorporation of functionalized azetidines into pharmaceutical lead structures has been impeded by the absence of efficient synthetic methods for their synthesis. In this study, a Rh-catalyzed one-carbon ring expansion of aziridines with vinyl-N-triftosylhydrazones is presented, which facilitates the synthesis of high value-added 2-alkenyl azetidine products. This research represents the first example of ring expansion of aziridines enabled by vinyl carbenes. Additionally, a one-pot two-step protocol, initiated from cinnamaldehyde, was successfully achieved, offering a step-economical and facile approach for the synthesis of these compounds. The pivotal aspect of this successful transformation lies in the in-situ formation of an alkenyl aziridinium ylide intermediate. Experimental investigations, coupled with computational studies, suggest that a diradical pathway is involved in the reaction mechanism.

Abstract:Transformations enabling the synthesis of α-alkyl, α'-2-azinyl amines by addition of 2-heteroaryl-based nucleophiles to in situ-generated and non-activated alkyl-substituted iminium ions are extremely rare. Approaches involving classical 2-azinyl organometallics, such as the corresponding Grignard reagents, often fail to produce the desired products. Here, we report an operationally straightforward solution to this problem through the development of a multicomponent coupling process wherein a soft 2-azinyl indium nucleophile, generated in situ from the corresponding 2-iodo heteroarene and indium powder, adds to an iminium ion that is also formed directly in the reaction. This modular carbonyl azinylative amination (CAzA) displays a broad scope and only a metal reductant is needed to generate a reactive 2-azinyl nucleophile. Beyond the addition to iminium ions, the 2-azinyl addition to polyfluoromethyl ketones forms the corresponding tertiary alcohols. Together, the products of these reactions possess a high degree of functionality, are typically challenging to synthesize by other methods, and contain motifs recognized as privileged in the context of pharmaceuticals and agrochemicals.

Abstract:A sequential [3+2]/[2+1] annulation reaction of benzimidazole- and indole-derived acrylonitriles with vinylsulfonium salts has been developed for the first time. 37 examples of functionalized polycyclic molecules bearing three contiguous stereocenters were obtained as a only single diastereomer up to 98% yields. This method features simplicity, high efficiency and broad substrate scopes. Moreover, the synthetic utility was demonstrated with the gram-scale reactions and further transformations of the products.

Abstract:Despite their tremendous synthetic and pharmaceutical utility, primary azaaromatic amines remain elusive for access based on a generally applicable C-H functionalization strategy. An oxadiazolone-enabled approach is reported for convenient entry into N-unsubstituted 1-aminoisoquinolines through Co(III)-catalyzed redox-neutral, step-, atom-, and purification-economic C-H functionalization with alkynes. A 15N labeling experiment reveals the effectiveness of both oxadiazolone N atoms as directing sites. The installed primary amine can be harnessed as a synthetically useful handle for attachment of divergent appendages.
