Abstract:Potassium antimonite was used to investigate the localisation of calcium in developing wheat anthers to examine the relationship between Ca 2+ and pollen development. During anther development, calcium precipitate formation increased in anther wall cells prior to microspore mother cell meiosis and appeared in microspores, suggesting the presence of a calcium influx from anther wall cells into the locule. Initially, the precipitates in microspore cytoplasm primarily accumulated in the mitochondria and destroyed their inner membranes (cisterns) to become small vacuoles, which expanded and fused, ultimately becoming a large vacuole during microspore vacuolisation. After microspore division and large vacuole decomposition, many calcium precipitates again accumulated in the small vacuoles, indicating that calcium from the large vacuole moved back into the cytoplasm of bicellular pollen.

Abstract:Calcium dynamics in the developing pollen of Chinese fir (Cunninghamia lanceolata) were investigated using an antimonate calcium precipitation method. After microspores were released from the tetrad, abundant calcium precipitates formed on the surface of microspores. A characteristic event during microspore development is the formation of a large vacuole which displaces the nucleus to the periphery and produces polarity, which initiates male gametophyte development. During the formation of the large vacuole, calcium precipitates moved into the large vacuole via mitochondria, suggesting that calcium abundance is related to vacuole biogenesis and formation. After microspore division, the large vacuole reorganised and calcium became concentrated in smaller vacuoles. The pollen wall of Chinese fir consists mostly of the intine, composed of polysaccharose, and the surface of the pollen grains is covered with a thin indefinite structure, the exine, which is composed of sporopollenin. The function of calcium during pollen development and the characteristics of the pollen wall of Chinese fir are discussed.

Abstract:The mechanism by which calcium regulates anther development remains unclear. This study investigated the relationship-between-calcium distribution and anther development in oil tea (Camellia oleifera Abel.) by using the potassium antimonite technique. Before the onset of microsporogenesis, abundant minute calcium precipitates appeared on the plasma membranes of microspore mother cells. Meanwhile, numerous precipitates accumulated in the tapetal cells. After meiosis, calcium precipitates appeared in young microspores. During microspore development, calcium precipitates mainly appeared in the small vacuoles of the cytoplasm. At the late microspore stage, a large vacuole formed, and the number of precipitates in the microspore decreased. The number of precipitates in the tapetal cells decreased as microsporogenesis proceeded. Then, calcium precipitates in the bicellular pollen cytoplasm again increased in number. During bicellular pollen development, the number of calcium precipitates decreased. As the pollen grains matured, only a few calcium precipitates were evident in the pollen cytoplasm. The results of this study, which show the-spatial and temporal features of calcium distribution during the anther development of C. oleifera, suggest that calcium distribution is related to anther development.

Abstract:Potassium antimonite was used to locate calcium in the anther of lettuce (Lactuca sativa L) during its development. At the early stage of anther development there were few calcium granules in microspore mother cells and the cells of anther wall. After meiosis of microspore mother cells, calcium granules first appeared in the tapetal cells in which some small secretive vacuoles containing many calcium granules were formed and secreted into locule. Then, the tapetal cells began to degenerate. At the late stage of microspore, tapetal cells completely degenerated and its protoplast masses moved into anther locule with many calcium granules. Few calcium granules were precipitated in the microspores just being released from tetrad, but some on the surface of exine. Then calcium granules appeared in the nucleus and cytoplasm of early microspores, as wall as in the exine. When microspores formed some small vacuoles containing some calcium granules, and then the small vacuoles fused to form a large vacuole, the calcium granules in the nucleus and cytoplasm evidently decreased, microspore developed to the late stage. The result suggested that calcium is related to the formation of large vacuole in microspores. The wall of microspore also is a main location of calcium granules during its developing. At early microspore some calcium granules began to accumulate in exine, which suggested calcium related with exine formation. At late stage of microspore, most of calcium granules were mainly deposited on the surface of exine. After the first mitosis of microspores, the large vacuole of bicellular pollen disappeared and calcium granules in the large vacuole went back to cytoplasm again. When bicellular pollen synthesized starches some calcium granules appeared on the surface of starches, which suggested calcium may regulate starch synthesis. With amount of starches increasing, calcium granules disappeared from pollen cytoplasm and only some of them located on the surface of pollen.

Abstract:Calcium was localized in ovules of Plumbago zeylanica from 1 day before anthesis to 3 days after anthesis using potassium antimonate and transmission electron microscopy in pollinated and emasculated flowers. At 1 day before anthesis, embryo sacs (containing an egg cell, a central cell and zero to three accessory cells) appear mature and contain abundant calcium precipitates (ppts), in contrast to nucellar cells. At anthesis, the vacuoles of nucellar cells have enlarged, and micropylar cells, in particular, are heavily labeled with calcium ppts. As pollen tubes elongate through ovular tissues, ppts diminish in ovular cells and become concentrated in the pollen tube cell wall. After fertilization, the calcium ppts sharply diminish in fertilized ovules; in unfertilized ovules, calcium ppts remain abundant up to 3 days after anthesis (when unfertilized ovules are shed). The distribution of calcium in the ovule changes in apparent response to fertilization, suggesting that calcium content may be related to the attraction and receipt of the pollen tube. In contrast with conventionally-organized embryo sacs with synergids, Plumbago accumulates calcium in the egg cell.

Abstract:The stigma of Lycium barbarum is wet type and some material to be secreted to the surface of stigma to form a layer of tectum at anthesis.There were many vesicles in the tectum and numerous calcium precipitated grana were accumulated in the tectum vesicles.When pollen grains landed onto the stigma they began to hydrate and form many small vacuoles in which some calcium precipitated grana located,which made a swelling pressure in pollen.During pollen germinating in the tectum,the pollen tubes often traversed tectum vesicles and displayed many calcium precipitated grana on the tube wall,suggesting it to absorb calcium of the tectum vesicles.In the base of pollen tube some mitochondria with calcium precipitated grana swelled to form small vacuoles which fused to form a large vacuole.The pollen tube cytoplasm was pushed into the top of tube to make sure directional growth.This is first time to report that stigma tectum containing rich calcium in flowering plants.The calcium distribution in the stigma and pollen germination and tube growth were also discussed.

Abstract:Pollen is the male gametophyte of higher plants. Its major function is to deliver sperm cells to the ovule to ensure successful fertilization. During this process, many interactions occur among pollen tubes and pistil cells and tissues, and calcium ion (Ca2+) dynamics mediate these interactions among cells to ensure that pollen reaches the embryo sac. Although the precise functions of Ca2+ dynamics in the cells are unknown, we can speculate about its roles on the basis of its spatial and temporal characteristics during these interactions. The results of many studies indicate that calcium is a critical element that is strongly related to pollen germination and pollen tube growth.

Abstract:. Potassium antimonate was used to locate Ca 2+ in fertile and sterile anthers of a photoperiod-sensitive genic male-sterile rice ( Oryza sativa L. japonica ). During the development of fertile anthers, abundant calcium precipitates accumulated in the anther walls and on the surface of pollen grains and Ubish bodies at the late developmental stage of the microspore, but not in the cytoplasm of pollen grains. Following the accumulation of starch grains in pollen, calcium precipitates on pollen walls diminished and increased in parenchymatous cells of the connective tissue. In sterile anthers, calcium precipitates were abundant in the middle layer and endothecium, but not in the tapetum, as was found in fertile anthers. A special cell wall was observed between the tapetum and middle layer of sterile anthers that appeared to relate to distinctive calcium accumulation patterns and poor pollen wall formation in the loculi. The formation of different patterns of antimonate-induced calcium precipitates in the anthers of photoperiod-sensitive genic male-sterile rice indicates that anomalies in the distribution of calcium accumulation correlate with the failure of pollen development and pollen abortion.

Abstract:Calcium has an essential signaling, physiological, and regulatory role during sexual reproduction in flowering plants; elevation of calcium amounts is an accurate predictor of plant fertility. Calcium is present in three forms: (1) covalently bound calcium, (2) loosely bound calcium typically associated with fixed and mobile anions (ionic bonding); and (3) cytosolic free calcium-an important secondary messenger in cell signaling. Pollen often requires calcium for germination. Pollen tube elongation typically relies on external calcium stores in the pistil. Calcium establishes polarity of the pollen tube and forms a basis for pulsatory growth. Applying calcium on the tip may alter the axis; thus calcium may have a role in determining the directionality of tube elongation. In the ovary and ovule, an abundance of calcium signals receptivity, provides essential mineral nutrition, and guides the pollen tube in some plants. Calcium patterns in the embryo sac also correspond to synergid receptivity, reflecting programmed cell death in one synergid cell that triggers degeneration and prepares this cell to receive the pollen tube. Male gametes are released in the synergid, and fusion of the gametes requires calcium, according to in vitro fertilization studies. Fusion of plant gametes in vitro triggers calcium oscillations evident in both the zygote and primary endosperm during double fertilization that are similar to those in animals.

Abstract:We used potassium antimonate to precipitate "exchangeable cellular Ca2+"-calcium that is sufficiently loosely bound to combine with antimonite, to investigate the feature of calcium distribution during anther development of Lycium barbarurn L. Before the stage of microspore mother cell, few calcium-induced precipitates were found in sporogenous cells and the somatic cells of anther wall. When microspore mother cell (MMC) preparing meiosis, calcium precipitates appeared in the cytoplasm of tapetal cells and callus wall surrounding MMC. After the meiosis of MMC,abundant calcium precipitates were accumulated in the cytoplasm of early microspores,and then in pollen wall, especially in the part of germ-pores. During the late microspore stage,a big vacuole formed and the nucleus was forced to move to peripheral region. Calcium precipitates decreased sharply and might dissolve in the large vacuole. After microspore mitosis, calcium precipitates appeared in the big vacuole of 2-cellular pollen,and then the vacuole disappeared. After that, the calcium precipitates again appeared in the cytoplasm of 2-cellular pollen, and the cytoplasm became densely and storage materials like starches accumulated inside the pollen grains. When pollen maturating,many small calcium precipitates distributed in its cytoplasm,especially in nucleus. The feature of calcium distribution in the anther of Lycium barbarurn L. means that it plays some biological roles during microspore development.

Abstract:Calcium distribution in fertile and sterile anthers of a thermosensitive male-sterile wheat genotype was investigated using an antimonate precipitation method. During fertile anther development, before meiosis of the microspore mother cells, calcium precipitates were apparent in tapetal cells of the anther wall. After meiosis, precipitates were detected in the early microspores and accumulated in the large vacuole of late microspores. After microspore division, following decomposition of the large vacuole, precipitates decreased in the bicellular pollen. The earliest abnormality in calcium precipitate distribution detected during sterile pollen development was the greater accumulation of precipitates in the cytoplasm and nucleus of late microspores. The sterile microspore can divide to form bicellular pollen, but the large vacuole of sterile bicellular pollen did not decompose and greater abundance of precipitates was retained in the large vacuole. Abnormal distribution of calcium precipitates in sterile pollen precedes structural changes, suggesting that abnormal calcium metabolism is associated with pollen abortion.

With microspore development many calcium precipitates accumulated in the germ pore, and then appeared in the small vacuoles of the cytoplasm. At a later microspore stage, a large vacuole formed and some precipitates were observed in the vacuole. The number of precipitates in tapetal cells decreased as microsporogenesis proceeded. After microgametogenesis began, calcium precipitates in the cytoplasm of pollen grains decreased. As pollen grains accumulated starch, a fewer were evident in the cytoplasm. Abstract:Potassium antimonate was used to locate Ca2+ in fertile and sterile anthers of Honglian-Yuetai cytoplasmic male-sterile rice(Oryza sativa L.). During the development of fertile and sterile anthers, abundant calcium precipitates accumulated on the outer surface of pollen walls at the binucleate stage while starch grains accumulated in pollen. There were no C2+a precipitates in the cytoplasm of fertile pollen grains. However, a great quantity of calcium precipitates accumulated in central vacuole of abortive pollen. There was not distribution of calcium precipitates in connective tissue of fertile anthers. Alternatively, in connective tissue of sterile anthers, abundant calcium precipitates located in vacuole and cytoplasm of vascular bundle sheath cells and its peripheral cells, vessels and sieve tubes, but few calcium precipitates in nuclei of parenchymatous cells. The changes of antimonate-induced calcium precipitates in the pollen and connective tissue of cytoplasmic male-sterile rice indicate that the anomalies in the distribution of calcium accumulation correlate with pollen abortion.
Calcium Function in Fertilization Process in Angiosperms

Abstract:The fertilization process in angiosperms in a broad sense begins since pollen grains land a stigma. The process continues as the pollen germinates, and pollen tube grows through pistil and finally reaches embryo sac in where fertilization occurs. In the process calcium plays important roles in controlling pollen tube growth during its journey toward embryo sac and promoting the fusion of both sperm and egg cells. It has been established in vitro experiment that tip-high intracellular calcium gradient is correlated with its growth and it is necessary for some extracellular calcium to keep the gradient. In trace of pollen tube in pistil, from stigma, transmitting tissue of style, micropyle of ovule to degenerated synergic of embryo sac, there is abundant calcium that quickly dissipates after pollen tube passed. It is the temporal and spatial characteristics of calcium in pollen tube and pistil that make pollen tube exactly reach embryo sac and insure fertilization. This review discusses recent advance in studies on calcium role in fertilization process of angiosperm, especially from calcium distribution in pistil to confirm its roles in fertilization of angiosperm.

Abstract:Potassium antimonate was used to locate loosely bound calcium in the stigma and style of tobacco. The tobacco stigma is wet and covered by a thick layer of glycoprotein exudate at anthesis. The exudate contains abundant vesicles, which are densely labeled with calcium precipitates. When pollen grains arrive at the stigma, become hydrated, and as the pollen swells, Ca 2+ precipitates accumulate at the aperture. Calcium precipitates that accumulate in pollen cytoplasm are initially concentrated within small vacuoles, but as germination proceeds these appear to fuse, forming prominent, densely labeled vesicles that preferentially accumulate near the proximal region of the growing tube. Although the stigma has abundant particles, few calcium precipitates are observed in the transmitting tissue from anthesis to 11 h after pollination. However, at 22 h after pollination, accumulation of calcium increases distally from the stigmatic interface with the transmitting tissue through the length of the style to the ovary. An examination of flowering plants with differing floral biology will be needed to understand the role of loosely bound calcium accumulation and its relationship to tissue-level changes in calcium uptake, maintenance of other calcium pools, including [Ca 2+ ] cyt , and in pollen and style maturation during the progamic phase.
The Primary Observation of Calcium Distribution During the Anther Development of Tobacco

Abstract:Potassium antimonite was used to locate calcium in the fertile and sterile anthers of a genic male sterile Chinese cabbage (Brassica campestris L. ssp. chinensis Makino) to probe the relation between Ca(2+) and fertility and sterility of anthers of the cabbage. During fertile anther development, calcium granules increase in number in anther wall cells after meiosis, and then appeared also in locule, suggesting a calcium influx into locule from anther wall cells (Plate I-4). Then the number of calcium granules in microspore cytoplasm also increased at early stage (Plate II-1), accumulated mainly on the membrane of small vacuoles which were fusing to form big ones to make a polarity in the cell and to prepare asymmetric division of microspore (Plate II-3,4). After microspore division and the big vacuole decomposition, many calcium granules accumulated again on the membrane of the vacuoles (Plate III-1,2), displaying calcium regulates vacuole formation and decomposition during pollen development. In sterile anthers, abnormal distribution of calcium granules first appeared in callus wall of microspore mother cell (Plate IV-1). However, only a few calcium granules appeared in early microspores, which then could not form small vacuoles and finally a big vacuole (Plate IV-2,3). The aborting microspores degenerate by cytoplasm shrinking (Plate IV-5,6). The difference pattern of distribution of calcium granules between the fertile and sterile anthers indicates that anomalies in the distribution of calcium accumulation are correlated with the failure of pollen development and pollen abortion.
Effect of photoperiod on calcium distribution in photoperiod-sensitive cytoplasmic male-sterile wheat during anther development

Abstract:Potassium antimontate was used to locate Ca 2+ in fertile and sterile anthers of photoperiod-sensitive cytoplasmic male-sterile wheat (Triticum aestivum L.).The quantity and distribution of calcium precipitates change in microspore and pollen during microsporogenesis and pollen development. The calcium precipitates gradually accumulated on the surface of tetraspore or pollen until pollen mature. They were abundant on the surface of the trinucleate pollen, but not in the cytoplasm. Calcium was transported in anther wall and intervening connective tissue via symplast and apoplast system, meeting the need of microspore and pollen development. Abortive pollens were observed in the anthers developed under long daylight exposure. Abundant calcium precipitates were distributed along the edge of the degradative pollen plasma and in the locular matrix, but little in the anther wall and intervening connective tissue of the early abortive pollen. Calcium precipitates accumulated on the surface of pollen was less than fertile pollen, but abundant calcium appear in the epiderms, endotheciums, middle layers of anther wall and cells of connective tissue. The results indicate that abnormalities in the distribution of calcium correlate with the failure of pollen development in photoperiod-sensitive cytoplasmic male-sterile wheat.

Abstract:Potassium antimonite was used to deposit calcium in the stigma and style of lettuce (Lactuca sativa L.) before and after pollination. The stigma of lettuce is two splits. Abundant calcium granules are displayed in the wall of papillae on the receptive surface of stigma before and after pollination, which may facilitate pollen germination. However, a few calcium granules in the wall of epidermis cell on no-receptive surface. Calcium distribution in style presents a gradient in transmitting tissue and parenchyma cells from the top to the base of the style before pollination. After pollination, calcium in transmitting tissue distinctly increased and its gradient distribution became more evident. Pollen tubes grow in the intercellular gaps of transmitting tissue. When pollen tubes grew into transmitting tissue, calcium granules in parenchyma around transmitting tissue decreased, suggesting a calcium movement was controlled by pollen tubes. The calcium gradient distribution also appeared in the trachea of vascular bundle of style. In general, calcium in style displays a feature of time-special distribution: transmitting tissue doesn't need much more calcium that is only stored in the parenchyma before pollination. However, calcium in parenchyma cells may be transported to transmitting tissue and make the latter contain more calcium to form an evident calcium gradient and meet the requirement of pollen tubes directionally growing after pollination. This is the second sample of calcium gradient existing in style, which was found by using potassium antimonite method.
