Abstract:The anther development of Impatiens balsamina displays some special characters:(1)Anther wall consists of 6 layers of cells:one epidermis,two endotheciums,two middle layers and one tapedum.Tapetal cells are not clear during sporogenous stage and difficult to distinguish with sporogenous cells.The tapetal cells degenerate during the stage of microspore mother cell(MMC).(2)Some starch polysaccharides appear in the microspore mother cells which had formed a callose wall.After meiosis of MMCs,starches in early microspores disappear,and lipids were accumulated in early microspores.With microspore developing,late microspores formed several large vacuoles and lipids disappeared.After microspore division,comparing with the big vacuoles in vegetative cell disappearing,starches again appeared in the 2-cellular pollen.Nearly anthesis,mature pollen grains of I.balsaminaaccumulated many lipids and starches as storey materials.(3) During anther development of I.balsamina,some cells of anther wall degenerated and their cytoplasm transformed lipids for pollen absorbing,the tapetal cells degenerated and offered lipids for MMCs and microspores,and middle layer cells for bicellular pollen grains.

Abstract:The distribution of polysaccharides and lipids during the anther development of the Hymenocallis littoralis displayed some distinct features.A mass of lipids were accumulated in sporogenous cells.At microspore mother cell(MMC) stage,many lipids were still located in the MMCs which had formed a thick callose wall.At same time,lipids also increased in tapetal cells,but a few starches appeared in other cells of anther wall.After MMC meiosis,tetrad microspores were irregular and full of lipids.After released from tetrad,the irregular microspores began to form pollen wall.The lipids in the microspores decreased during the cell developing and forming many vacuoles,which made irregular microspores become round.In late microspore there were some polysaccharide materials appeared in the inner side of pollen wall.During the microspore stage tapetal cells degenerated,and the cell residue became many lipids.At early bicellular pollen stage,the vacuoles of the pollen disappeared and the lipid number increased evidently.The tapetal residue became massy lipid.The mature pollen of H.littoralis is bicellular and many lipids were accumulated in the pollen as a storage material.

Abstract:Polysaccharide and lipid in the anthers of Lycium barbarurn L. at different stages were examined with cytochemical techniques. At the stage of sporogenous cell, many starches have been storied in parenchyma around vascular bundle, epidermis and endothecium cells but no starches in sporogenous cells, tapetal and middle layer cells. At the stage of tetrad, there were many starches still in epidermis and endothecium, however tapetal cells began to accumulate lipid drops, suggesting that tapetal cells transformed polysaccharides into lipid. Tapetum degenerated at the late stage of microspore and the lipid drops moved into locule. During microspore development neither starches nor lipid drops were accumulated in the cell. After the division of microspore, some lipids drops appeared in 2-cellular pollen, and then some starches also appeared in the pollen. Two days before anthesis, there were many lipid drops and starches located in nearly mature pollen grains, suggesting that pollen of Lycium barbarurn L. has a function of transforming lipid into polysaccharide. The temporal and spatial features of polysaccharide and lipid material accumulated and distributed in anther during its development reflect the transformation of physiological function of the cells consisting of anther. This research will help us to understand the mechanism of anther development.

Abstract:Our goal was to determine the ultrastructure features and the dynamic changes in polysaccharides and neutral lipids in developing anthers of rose balsam (Impatiens balsamina), and to better understand the mechanisms controlling male reproductive development. Transmission electron microscope (TEM) techniques were used to study the ultrastructural characteristics of the anthers, and histochemical methods were used to determine levels of polysaccharides and lipids. The cytokinesis in the microsporocyte meiosis was simultaneous type, and microspore tetrads were mainly tetrahedral. The pollen exine began to form at the tetrad stage. The mature pollen grains were oval-shaped and bicellular. Accumulation of reserve substances began at the late microsporogenesis stage, and an abundance of starch grains and lipids were stored in pollen grains at anthesis. Polysaccharides and lipids changed in different stages and played an important role in anther development. Moreover, the calcium oxalate crystals may protect the pollen and suggest that calcium distribution is related to anther development.

Abstract:Bauhinia blakeana (Fabaceae) pollen is bicellular and contains only a vegetative and generative cell at anthesis. The pollen grain contents, including the generative cell, can be released in vitro following osmotic shock. In this study, when pollen grains were incubated in a 10% sucrose and 5%-15% polyethylene glycol 4500 (PEG 4500) medium, over 50% of the pollen grains germinated and formed a short pollen tube. However, the generative cell did not divide in in vitro pollen tubes. In vivo pollen tubes were induced to grow from the style using an in vivo-in vitro method. Sperm cells were subsequently isolated from broken pollen tubes.

Abstract:OBJECTIVE To reveal the feature of distribution and conversion of the nutritional ingredients during anther development of Houttuynia cordata.METHODS Semithin sections of anthers by phytohistochemistry were observed under light-microscope.RESULTS Some starches distributed in epidermis and endothecium.Theabnormality of plasmolysis appeared in tapetal cells at the period of microspore mother cell.Lipids accumulation was not abundant in mature pollen grains during the anther development.CONCLUSION Pollen grains are abortive because for deficiency of lipids in mature pollen grains.

Abstract:The normal development of anthers and the formation of functional pollen are the prerequisites for successful pollination and fertilization. In this study, we observed dynamic changes in inflorescence and anther development in the chinquapin (Castanea henryi) using stereomicroscopy, light microscopy, and transmission electron microscopy. We found that cytokinesis during meiosis in microsporocytes was of the simultaneous type, and that the tetrads were mainly tetrahedral. Mature pollen grains contained two cells with three germ pores. The anther wall was of the basic type and composed of epidermis, endothecium, middle layers, and tapetum. Mature anthers had no middle layer and tapetum. The tapetum was of the glandular type. At the early microspore stage, a large number of starch granules appeared in the endothecium, which was deformed at the late microspore stage. Lipid droplets appeared in tapetum during the early microspore stage, and a few lipid droplets were still found during tapetum degeneration. The mature pollen accumulated a large amount of starch and lipids. These findings demonstrated that the anther wall provides nutrients and protection for pollen development. There is relatively stable correspondence between the external morphological characteristics of male flowers and internal structure of anther development.

Abstract:Polysaccharides was detected using the periodic-acid-Schiff's (PAS) technique and lipid detected using Sudan black during the anther development of Allium cepa L. Before the meiosis of microspore mother cells there were a few lipid drops in endothecium cells and little polysaccharides in tapetal cells which did not differentiate completely in young anthers. At the stage of tetrad there were still few polysaccharides and lipid material in young anther, and only cell wall of anther wall and callose wall of tetrads displayed the feature of polysaccharids. The size of tapetal cells began to increase at this stage. During microspore development the tapetal cells reached its maximal size, and many lipid drops were accumulated in the cells. However, few lipid drops and starches appeared in microspores. At early stage of 2-celled pollen, the vegetative cell of 2-celled pollen began to accumulate starches. Tapetal cells degenerated at this stage and its lipid drops concentrated to form lipid block. Then the starches in 2-celled pollen disappeared with pollen development, and many lipid drops were accumulated in vegetative cell of nearly mature pollen.

Abstract:The Periodic Acid-Schiff (PAS) reaction was used to test for the presence of starch, and Sudan black was applied to detect neutral lipids (lipid drops) during anther development in Ipomoea cairica. Starch was observed in sporogenous and anther wall cells. Prior to meiosis in microspore mother cells (MMCs), starch was not evident, although small lipid drops appeared. A substantial amount of starch accumulated in the epidermal and endothecium cells, but starch and lipid drops were not identified in the tapetum. At the tetrad stage, starch appeared in microspores, and the amount of starch remained constant in anther cells. Early in the microspore stage, the tapetum degenerated, starch remained in the microspores, and lipid drops were completely lost. A large and conspicuous vacuole was formed in the microspores and lipid drops reformed in the cells at the late microspore stage. Following microspore division, the vegetative cell of bicellular pollen grains was filled with starch and lipid drops, and the inner middle layer degenerated. Finally, at anthesis, lipid drops and starch constituted the stored nutritive material in the mature pollen.

Abstract:With pollen development, a big vacuole appeared in the pollen and the starch grains in endothecium and tapetum disappeared. Then the big vacuole decomposed and some starch grains appeared in pollen. Tapetal cells degenerated with pollen development. Some lipid material accumulated in tapetal cells and the lipid material in pollen grains also increased. The most store material in mature pollen is lipid and small part of starches. The process of nutrition material accumulated in pollen with special time and part is also a developmental feature.

Abstract:Dendrobium catenatum is a widely cultivated Chinese orchid herb rich in abundant secondary metabolites, such as terpenes. However, terpene distribution and characterization of terpene biosynthesis-related genes remain unknown in D. catenatum. In this study, metabolic profiling was performed to analyze terpene distribution in the root, stem, leaf, and flower of D. catenatum. A total of 74 terpene compounds were identified and classified. Clustering analysis revealed that terpene compounds exhibited a tissue-specific accumulation, including monoterpenes in the flowers, sesquiterpenes in the stems, and triterpenes in the roots. Transcriptome analysis revealed that the ‘terpenoid backbone biosynthesis’ pathway was only significantly enriched in root vs. flower. The expression of terpene biosynthesis-related genes was spatiotemporal in the flowers. Prenylsynthase-terpene synthases (PS-TPSs) are the largest and core enzymes for generating terpene diversity. By systematic sequence analysis of six species, 318 PS-TPSs were classified into 10 groups and 51 DcaPS-TPSs were found in eight of them. Eighteen DcaPS-TPSs were regulated by circadian rhythm under drought stress. Most of the DcaPS-TPSs were influenced by cold stress and fungi infection. The cis-element of the majority of the DcaPS-TPS promoters was related to abiotic stress and plant development. Methyl jasmonate levels were significantly associated with DcaTPSs expression and terpene biosynthesis. These results provide insight into further functional investigation of DcaPS-TPSs and the regulation of terpene biosynthesis in Dendrobium.

Abstract:The pollen developmental characteristics in plants with pollinia remain unknown. In this study, Dendrobium officinale (Orchidaceae) pollinium development was investigated on the basis of a histochemical examination of polysaccharides and lipids. The D. officinale anther wall is composed of one layer of epidermal cells, two layers of endothecial cells, one layer of middle layer cells, and one layer of tapetal cells. The microspore mother cells, which have no obvious callose wall structure before meiosis, undergo simultaneous microsporogenesis. After meiosis, four microspores in a tetrad do not separate and subsequently develop as pollen tetrads, which ulti-mately form two pollinia. During microspore development, sporopollenin covers the pollinium surface and forms the exine of the pollinium, but the pollen tetrads inside the pollinium lack the exine structure. A pollen germinal pore was not detected in the pollinium exine. An in vitro analysis of pollen germination revealed that pollen grains at the surface of pollinia covered with an exine layer do not germinate. Furthermore, pollen tubes are mainly produced by the pollen grains inside the pollinium.

Abstract:The regulation of nutrient transportation and transformation in developing anthers is very complex. We analyzed the distribution and features of polysaccharides and lipids in the developing anthers of tomatoes using histochemical methods. Some starches appeared in the connective somatic tissue of anthers during the sporogenous cell stage. Before meiosis of the microspore mother cell,a thick polysaccharide callose wall was formed,accompanied by a reduction in the connective tissue starches. During the tetrad stage after meiosis,the polysaccharide material in the anther did not change. At the early microspore stage,the starches in the connective cells again increased,and polysaccharide material appeared in the partial intine of pollen. At the late microspore stage,a large vacuole formed that did not contain lipids or starches,and only the pollen wall contained red polysaccharides. At this stage,the connective somatic cell starch amounts decreased,and the tapetal cells changed shape and degenerated. After microspore division,abundant lipids appeared in the bicellular pollen,and starches accumulated following pollen development. As the anthers matured,many lipids and some starches accumulated in the epidermal cells. Nutrient metabolism within the tomato pollen characteristically accumulated lipids first and then starches,while the mature pollen accumulated starches and lipids simultaneously. This characteristic pattern of nutrient metabolism in tomato pollen shows species specificity among plants.

Abstract:We analyzed anther development in Amomum villosum Lour. (Zingiberaceae) using the periodic acid-Schiff's technique and Sudan black staining to test for the presence of starch and lipids, respectively. Our analyses showed that microspore mother cells of A. villosum lack typical callose walls, and numerous lipid granules appear in the cells early in development. Some starch granules are present in anther wall cells, but not in tapetal cells. After meiosis, numerous lipid granules remain unchanged in the microspores. During microspore development, some small starch granules first appear in the central cell region, and then the starch granules increase in size. After microspore division, the bicellular pollen grains become filled with starch and lipids, and remain in this state until the pollen grains reach maturity. At anthesis, the anther wall of A. villosum consists of several layers of endothecium cells with an evidently thickened radial wall, and some layers of parenchyma cells containing numerous starch granules.
Transcriptome and metabolome analysis of the developmental changes in Cynanchum thesioides anther

Abstract:Cynanchum thesioides, a xerophytic species utilized both as a medicinal herb and a food source, plays a significant role in arid and desert ecosystem management. Its inflorescence is an umbellate cyme, each carrying nearly a thousand flowers; however, its fruiting rate remains remarkably low. The normal development of the anther is a necessary prerequisite for plants to produce seeds. However, our understanding of the anther development process in Cynanchum thesioides remains limited. To better understand the pollen development process in Cynanchum thesioides, the stages of pollen development were determined through paraffin sectioning, and observations were made on the distribution characteristics of polysaccharides and lipid droplets in the pollen development of Cynanchum thesioides using Periodic Acid-Schiff stain (PAS) and 0.5% Sudan Black B tissue staining. Concurrently, the gene expression patterns and metabolite profiles were delineated across various developmental stages of Cynanchum thesioides anthers (T1: microspore stage, T2: tetrad stage, T3: mononuclear stage, and T4: maturation stage). The findings revealed that Cynanchum thesioides pollen is in an aggregate form. Polysaccharides gradually accumulate during maturation and lipid droplets form a surrounding membrane, thereby preventing pollen dispersion. Furthermore, transcriptomic and metabolomic analyses across distinct developmental phases uncovered a plethora of differentially expressed genes and metabolites associated with the flavonoid biosynthesis pathway. Flavonoid levels exhibited dynamic changes concurrent with anther development, aligning with the gene regulatory patterns of the corresponding biosynthetic pathways. The study identified 63 differentially accumulated flavonoid compounds and 21 differentially expressed genes associated with flavonoid biosynthesis. Weighted gene co-expression network analysis revealed six MYB and ten bHLH transcription factors as key candidates involved in flavonoid biosynthesis, with CtbHLH (Cluster-6587.1050) and CtMYB (Cluster-6587.31743) specifically regulating structural genes within the pathway. These findings underscore the pivotal role of flavonoid biosynthesis in anther development of Cynanchum thesioides. In conclusion, this research offers a comprehensive insight into the anther development process in Cynanchum thesioides.
