Abstract:In the structured illumination microscopy (SIM) family, interferometric implementations using traditional physical gratings or advanced liquid crystal on silicon spatial light modulators are popular but have the disadvantages of expensive and limited speed. A promising alternative of a digital micromirror device (DMD) has improved the situation for its lower cost and higher speed, furthermore, it has contributed to the proposal of a simple and compact projection DMD-SIM. However, the projection DMD-SIM method is accused of erroneously measuring the resolution because of the scattering characteristic of gold beads, despite the inferior experimental resolution that is because of the low fringe contrast and the resulting low signal-to-noise ratio (SNR). As an improvement, we use a high-brightness laser for illumination to achieve higher SNR, and the accompanying problems of speckle and inhomogeneity originating from the laser’s high coherence are solved by a high-frequency phase-randomization deformable mirror in conjunction with a square-core multimode fiber, which does not limit the image acquisition speed and with fewer energy losses compared to the existing methods. Also, in terms of the problem of low SNR, we apply a parallel-iteration Richardson–Lucy deconvolution algorithm for SIM reconstruction to further improve the imaging performance based on ensuring the resolution. Based on these advances, we image fluorescent nanoparticles and other non-biological and biological fluorescent samples, and the experimental results demonstrate the improved imaging performance of our modified system.
Wavelength-encoded Laser Particles for Massively Multiplexed Cell Tagging.

Abstract:Large-scale single-cell analyses have become increasingly important given the role of cellular heterogeneity in complex biological systems. However, no current techniques enable optical imaging of uniquely-tagged individual cells. Fluorescence-based approaches can only distinguish a handful of distinct cells or cell groups at a time because of spectral crosstalk between conventional fluorophores. Here we show a novel class of imaging probes emitting coherent laser light, called laser particles. Made of silica-coated semiconductor microcavities, these laser particles have single-mode emission over a broad range from 1170 to 1580 nm with sub-nm linewidths, enabling massive spectral multiplexing. We demonstrate the stability and biocompatibility of these probes in vitro and their utility for wavelength-multiplexed cell tagging and imaging. We demonstrate real-time tracking of thousands of individual cells in a 3D tumor model for several days showing different behavioral phenotypes. We expect laser particles will enable new approaches for single-cell analyses.

Abstract:A novel micro-fabrication technology using optical radiation pressure is presented. A simulation program has been written and simulation results have demonstrated that micro silica or diamond particles, trapped by focused laser beam, could be used as micro-fabrication tools for nanometer manufacturing. An experiment setup for laser trapping has been established and Silica particles, with diameter of 8 μm, has been successfully trapped in water and arbitrary-shaped diamond particles, whose size is about 3-8 μm, has been trapped and rotated as micro-fabrication tools. When trapped rigidity micro-particles tools was controlled by laser beam, the work stage was horizontally moved repeatedly so that a machining trace of 2nm on work piece has been attained.

Abstract:To study the spatio-temporal evolution of individual cells in an intact tissue or other specific microenvironment, organic fluorophores are widely used. Owing to the broad emission linewidth (full width at half maximum exceeding 50 nm) of the organic fluorophore molecules, only limited number of fluorophores with non-overlapping spectral signatures can be simultaneously observed on the spectral range of the spectrometer. Thus there is a need of optical probes of deterministic colors with non-overlapping narrow spectral signatures. In the recent past, a new class of biophotonic tool – called laser particles – has emerged, which is unique in terms of its narrow spectral linewidth arising from lasing. In the previous studies, however, the lasing wavelength of the individual laser particles has been random. In this presentation, we shall report the first step towards addressing this, by demonstrating fabrication of InGaAsP microdisk laser particles by active photoelectrochemical etching to lase in distinct deterministic narrow-band “colors”. The experimental pathway supported by a theoretical model to explain the results, would be presented. By careful choice of the material system coupled with the design of the disk geometry, we were able to demonstrate multiple unique “color bands” of single-mode lasing of all microdisks simultaneously emitting in the same narrow spectral band. Transferring these wavelength-tuned microdisks onto a PDMS substrate, we create free-standing precision laser particles. This technology could potentially open up a viable avenue to simultaneously identify and image over 20 unique biomarkers in the same spectral bandwidth as currently provided by 4 organic fluorophores.

Abstract:This article reports a novel laser-induced micro-fabrication method and its monitoring system for three-dimensional (3D) microstructures. The mechanism of the method is that a small zone of thermoplastic material melted by laser heating grows in liquid surrounding environment, solidifying into a convex microstructure, such as micro-dot or micro-pillar. A laser diode (808 nm) with maximum power output of 130 mW is used as power source, and a kind of paraffin mixed with stearic acid and paint serves as the thermoplastic material for 3D microstructure formation experiments. A light microscope system consisting of a charge-coupled device (CCD) and a computer is utilized to realize real-time observation of the micro-fabricating process. The distribution of local temperature rise on material surface created by laser irradiation is simulated. The effects of liquid environment on microstructure formation have been theoretically analyzed and experimentally studied. Experiments are further carried out to investigate the relationship between laser spot and fabricated microstructures. The results indicate that the widths of microdots or micro-pillars are mostly determined by the size of focal spot, and their heights increase with the enlargement of laser power density. With this method, a micro-dot array of Chinese characters meaning "China" has been successfully fabricated through computer programming. This method has the advantages of implementing direct, mask-less, real-time and inexpensive 3D microstructure fabrication. Abstract:Semiconductor-based laser particles (LPs) with exceptionally narrowband spectral emission have been used in biological systems for cell tagging purposes. Fabrication of these LPs typically requires highly specialized lithography and etching equipment, and is typically done in a cleanroom environment, hindering the broad adoption of this exciting new technology. Here, using only easily accessible laboratory equipment, we demonstrate a simple layer-by-layer fabrication strategy that overcomes this obstacle. We start from an indium phosphide (InP) substrate with multiple epitaxial indium gallium arsenide phosphide (InGaAsP) layers which are sequentially processed to yield LPs of various compositions and spectral properties. The LPs isolated from each layer are characterized, exhibiting excellent optical properties with lasing emission full width at half maximum as narrow as < 0.3 nm and typical thresholds of approximately 6 pJ upon excitation using a 3 ns pulse duration 1064 nm pump laser. The high quality of these particles renders them suitable for large-scale biological experiments including those requiring spectral multiplexing.

Abstract:In this paper, we present a focused femtosecond laser Bessel beam scanning technique for the rapid fabrication of large-area 3D complex microtube arrays. The femtosecond laser beam is converted into several Bessel beams by two-dimensional phase modulation using a spatial light modulator. By scanning the focused Bessel beam along a designed route, microtubes with variable size and flexible geometry are rapidly fabricated by two-photon polymerization. The fabrication time is reduced by two orders of magnitude in comparison with conventional point-to-point scanning. Moreover, we construct an effective microoperating system for single cell manipulation using microtube arrays, and demonstrate its use in the capture, transfer, and release of embryonic fibroblast mouse cells as well as human breast cancer cells. The new fabrication strategy provides a novel method for the rapid fabrication of functional devices using a flexibly tailored laser beam.

Abstract:We describe microfabrication of various materials by multiphoton absorption of femtosecond (120-150 fs) light pulses. The photo-modification at the focal point of tightly focused laser beam occurs inherently within a volume smaller than that defined by the diffraction limit. The achievable lateral resolution is compared with that obtainable by lithography which uses near-field optical microscopy. This technique can in principle lead to the realization of 3D optical memory and photonic crystals with arbitrary lattice in polymers and silica glass, 3D prototyping in polymerizable resins, and etching of 3D structures guided by the optically damaged pattern in silica. Another topic also discussed deals with laser micromanipulation using continuous wave laser operating at 1.06 /spl mu/m. Light-controlled revolution of liquid crystal droplet and volume-phase transition of gels are described.
High-resolution 3D Nanoprinting Based on Two-Step Absorption Via an Integrated Fiber-Coupled Laser Diode.

Abstract:Three-dimensional (3D) laser nanoprinting with high resolution and low cost is highly desirable for fabricating arbitrary 3D structures with fine feature size. In this work, we use a 405-nm integrated fiber-coupled continuous wave (cw) laser diode to establish an easy-to-build 3D nanoprinting system based on two-step absorption. Two-dimensional (2D) gratings with a sub-150-nm period and 3D woodpile nanostructures with a lateral period of 350 nm have been printed at a low speed. At a faster scan velocity of 1000 µm/s, 2D gratings with sub-200-nm resolution and sub-50-nm linewidth can still be fabricated with laser power less than 1 mW. The two-step absorption of the used benzil initiator enables us to use a second cw laser with 532-nm wavelength to enhance the polymerization with sub-100-nm feature size when starting with insufficient 405-nm laser power, which possess the potential to find applications in high-speed high-resolution parallel-writing and in situ manipulation.

Abstract:In this paper, we reviewed the fabrications of functional microcavity lasers in soft materials such as polymer and protein by femtosecond laser processing. High-quality (Q) microdisks with a laser dye (Rhodamine B, RhB) acting as gain medium were fabricated that produced whispering-gallery-mode (WGM) lasing output. We also obtained unidirectional lasing output with a low lasing threshold in a deformed spiral microcavity at room temperature. Photonic-molecule (PM) microlasers were prepared to investigate the interaction and coupling effects of different cavities, and it was found that the distance between the two disks plays an important role in the lasing behaviors. Single-mode lasing was realized from a stacked PM microlaser through Vernier effect. Furthermore we adopted the biocompatible materials, RhB-doped proteins as a host material and fabricated a three-dimensional (3D) WGM microlaser, which operated well both in air and aqueous environment. The sensing of the protein microlasers to Na2SO4 concentration was investigated. Our results of fabricating high-Q microlasers with different materials reveal the potential applications of femtosecond laser processing in the areas of integrated optoelectronic and ultrahigh sensitive bio-sensing devices.

Abstract:Abstract Optical microcavities and microlasers were recently introduced as probes inside living cells and tissues. Their main advantages are spectrally narrow emission lines and high sensitivity to the environment. Despite numerous novel methods for optical imaging in strongly scattering biological tissues, imaging at single-cell resolution beyond the ballistic light transport regime remains very challenging. Here, we show that optical microcavity probes embedded inside cells enable three-dimensional localization and tracking of individual cells over extended time periods, as well as sensing of their environment, at depths well beyond the light transport length. This is achieved by utilizing unique spectral features of the whispering-gallery modes, which are unaffected by tissue scattering, absorption, and autofluorescence. In addition, microcavities can be functionalized for simultaneous sensing of various parameters, such as temperature or pH value, which extends their versatility beyond the capabilities of standard fluorescent labels.

Abstract:Laser microdissection technology is favored by biomedical researchers for its ability to rapidly and accurately isolate target cells and tissues. However, the precision cutting capabilities of existing laser microdissection systems are hindered by limitations in overall mechanical movement accuracy, resulting in suboptimal cutting quality. Additionally, the use of current laser microdissection systems for target acquisition may lead to tissue burns and reduced acquisition rates due to inherent flaws in the capture methods. To address these challenges and achieve precise and efficient separation and capture of cellular tissues, we integrated a digital micromirror device (DMD) into the existing system optics to modulate spatial light. This allows the system to not only implement the traditional point scanning cutting method but also utilize the projection cutting method.We have successfully cut various patterns on commonly used laser microdissection materials such as PET films and mouse tissues. Under projection cutting mode, we were able to achieve precise cutting of special shapes with a diameter of 7.5 micrometers in a single pass, which improved cutting precision and efficiency. Furthermore, we employed a negative pressure adsorption method to efficiently collect target substances. This approach not only resulted in a single-pass capture rate exceeding 90% for targets of different sizes but also enabled simultaneous capture of multiple targets, overcoming the limitations of traditional single-target capture and enhancing target capture efficiency, and avoiding potential tissue damage from lasers.In summary, the integration of the digital micromirror device into laser microdissection systems significantly enhances cutting precision and efficiency, overcoming limitations of traditional systems. This advancement demonstrates the accuracy and effectiveness of laser microdissection systems in isolating and capturing biological tissues, highlighting their potential in medical applications.
Femtosecond Mathieu Beams for Rapid Controllable Fabrication of Complex Microcages and Application in Trapping Microobjects

Abstract:Structured laser beam based microfabrication technology provides a rapid and flexible way to create some special microstructures. As an important member in the propagation of invariant structured optical fields, Mathieu beams (MBs) exhibit regular intensity distribution and diverse controllable parameters, which makes it extremely suitable for flexible fabrication of functional microstructures. In this study, MBs are generated by a phase-only spatial light modulator (SLM) and used for femtosecond laser two-photon polymerization (TPP) fabrication. Based on structured beams, a dynamic holographic processing method for controllable three-dimensional (3D) microcage fabrication has been presented. MBs with diverse intensity distributions are generated by controlling the phase factors imprinted on MBs with a SLM, including feature parity, ellipticity parameter q, and integer m. The focusing properties of MBs in a high numerical aperture laser microfabrication system are theoretically and experimentally investigated. On this basis, complex two-dimensional microstructures and functional 3D microcages are rapidly and flexibly fabricated by the controllable patterned focus, which enhances the fabrication speed by 2 orders of magnitude compared with conventional single-point TPP. The fabricated microcages act as a nontrivial tool for trapping and sorting microparticles with different sizes. Finally, culturing of budding yeasts is investigated with these microcages, which demonstrates its application as 3D cell culture scaffolds.
Light focusing and additive manufacturing through highly scattering media using upconversion nanoparticles

Abstract:Light-based additive manufacturing holds great potential in the field of bioprinting due to its exceptional spatial resolution, enabling the reconstruction of intricate tissue structures. However, printing through biological tissues is severely limited due to the strong optical scattering within the tissues. The propagation of light is scrambled to form random speckle patterns, making it impossible to print features at the diffraction-limited size with conventional printing approaches. The poor tissue penetration depth of ultra-violet or blue light, which is commonly used to trigger photopolymerization, further limits the fabrication of high cell-density tissue constructs. Recently, several strategies based on wavefront shaping have been developed to manipulate the light and refocus it inside scattering media to a diffraction-limited spot. In this study, we present a high-resolution additive manufacturing technique using upconversion nanoparticles and a wavefront shaping method that does not require measurement from an invasive detector, i.e., it is a non-invasive technique. Upconversion nanoparticles convert near-infrared light to ultraviolet and visible light. The ultraviolet light serves as a light source for photopolymerization and the visible light as a guide star for digital light shaping. The incident light pattern is manipulated using the feedback information of the guide star to focus light through the tissue. In this way, we experimentally demonstrate that near-infrared light can be non-invasively focused through a strongly scattering medium. By exploiting the optical memory effect, we further demonstrate micro-meter resolution additive manufacturing through highly scattering media such as a 300-{\mu}m-thick chicken breast. This study provides a proof of concept of high-resolution additive manufacturing through turbid media with potential application in tissue engineering.

Abstract:Due to the stimulated emission amplification, lasers with excellent characteristics, including the high energy density, ultra-narrow spectral linewidth, and high directionality, are extremely favorable for sensing, detection, and imaging. Bringing these merits into the micro/nano scale, micro/nano lasers with miniaturized device sizes further enable outstanding spatial and temporal confinement, greatly boosting the light-matter interaction and bridging the size mismatch between light and biomolecules. Thanks to these advantages, micro/nano lasers have drawn widespread attention and opened new opportunities for a variety of biomedical and biochemical applications. In this paper, we review recent developments in biomolecular sensing and cellular analysis based on micro/nano lasers. We first describe the fundamental building blocks of micro/nano lasers, with discussions on gain material considerations, cavity structures, and pumping. We then review recent applications using micro/nano lasers as biosensors and bioprobes, including biomolecule (mainly proteins and DNAs) sensing, wavelength-multiplexed cell labeling/tracking/probing, and high-resolution cellular/tissue bioimaging. Finally, an outlook of the challenges and potential developments of micro/nano lasers for biological sensing and clinical applications is provided.

Abstract:The miniaturization of lasers has opened up a wide variety of new applications,including on-chip optical communications,laser displays,medical imaging,and sensing.A representative example is the cell laser that emerged in 2011,in which a single biological cell expressing green fluorescent protein generated laser emission under optical pumping with nanojoule pulse inside a Fabry-Pérot microcavity.1 The ongoing miniaturization of lasers enables their incorporation into live cells,lead-ing to the achievement of intracellular microlasers.2 This significant progress has equipped individual cells with a distinctive coherent light source characterized by high brightness and sub-nanometer linewidth,paving the way for high-performance applications in microlaser-based cell sensing,tagging,and imaging.3,4 For example,a remarkable achievement in massively multiplexed cell tagging was demonstrated by Seok-Hyun Yun and his coworkers in 2019,through the utilization of wavelength-encoded laser particles formed by semiconductor microdisks.5
Fully-automatic laser welding and micro-sculpting with universal in situ inline coherent imaging

Abstract:Though new affordable high power laser technologies make possible many processing applications in science and industry, depth control remains a serious technical challenge. Here we show that inline coherent imaging, with line rates up to 312 kHz and microsecond-duration capture times, is capable of directly measuring laser penetration depth in a process as violent as kW-class keyhole welding. We exploit ICI's high speed, high dynamic range and robustness to interference from other optical sources to achieve fully automatic, adaptive control of laser welding as well as ablation, achieving micron-scale sculpting in vastly different heterogeneous biological materials.

Abstract:Micronanofabrication technologies developed so far pursue faithful conversion from digital models to matter structures. This is vital for microdevices in optics, mechanics, and electronics, where device shape and size matter. However, biotissues grow under rich environmental factor interactions, as demands novel manufacturing approaches for biomimetic and biological fabrication. Here, a concept of dynamic laser prototyping is reported, which is based on a new finding of a multilayered three-dimensional (3D) wrinkling phenomenon. The 3D wrinkling started with formation of a photocrosslinked hydrogel sheet by femtosecond laser direct writing. It was followed by spontaneous self-bending of the sheet, caused by a purposely designed sheet-substrate stress mismatch. The flower blooming process has been successfully mimicked, indicating broader usages of the technology in biotissue-growth-related manufacturing.

Abstract:Three-dimensional microstructures with aspect ratios larger than 20 and a solidified wall with a thickness down to 2.0 mu m have been fabricated by the one-photon absorption photopolymerization technique. Lateral and longitudinal spatial resolutions in a single-line scanning process of as high as 1.0 mu m and 0.3 mu m, respectively, are reproducible. An experimental system consisting of a common picosecond pulsed laser and an optical microscope makes laser microfabrication easy to realize, implying a broad application potential.
