Abstract:A divergent synthesis of amides and nitriles from vinyl azides and p-quinone methides (p-QMs) was developed. The p-QMs could be activated by BF3-Et2O and then nucleophilically attacked by vinyl azides, leading to divergent rearrangement toward amides and nitriles.

Abstract:A palladium-catalyzed highly regioselective C-H allylation/annulation reaction of N-sulfonyl amides with secondary or tertiary allylic alcohols has been developed to construct 3,4-dihydroisoquinolones bearing a synthetically valuable vinyl substituent. This cascade cyclization approach of allylic alcohols involving C-H allylation has not been reported previously. The commercially available allylic alcohol substrates, the only by-product of water, and the used terminal oxidant of O2 provide environmentally benign advantages.

Abstract:A method for the synthesis of amide-containing molecules was developed using vinyl azides as an enamine-type nucleophile towards carbon electrophiles, such as imines, aldehydes, and carbocations that were generated from alcohols in the presence of BF3OEt2. After nucleophilic attack of the vinyl azide, a substituent of the resulting iminodiazonium ion intermediate migrates to form a nitrilium ion, which is hydrolyzed to afford the corresponding amide.

Abstract:Atom- and step-economy in IBX assisted diversity-oriented synthesis is achieved with a versatile AQ auxiliary α-amino acid analogs offering rapid access to polycyclic spiro-quinolines featuring a quaternary stereocenter in 20%–91% yields under mild conditions via 7,8-dearomatization of quinolines. Free of a preinstalled activation group is highlight of this intramolecular oxidation spiroannulation tandem reaction. This type of N-heterospirocycles, traditionally difficult to access, may open the door to a potentially interest scaffold for synthetic and medicinal chemistry.

Abstract:Utilizing simple precursors for the exquisite construction of molecules with diversity and complexity is urgent. Herein, a novel A3-X type reaction for functionalized 3-haloquinolines has been developed by PhI (OAc)2-mediated four-component tandem addition/cyclization/oxidation/aromatization process with simple aromatic amine, aldehyde, alkyne and halide salt. Broad functional group tolerance, very mild condition, easy for further transformation illustrate the concision, high efficiency and practicability of this A3-X type reaction.

Abstract:The para-quinone methides (p-QMs) are activated by Lewis acid and then attacked by diazo compounds. The following rearrangement leads to nitrogen gas extrusion and C-C double bond formation to constitute a metathesis reaction process. Therefore, the diazoester is transformed into tetrasubstituted alkenes, whereas the diazo-oxindole delivers the quinolinone products. Furthermore, the 13C-labeling experiments were also conducted to elucidate a possible mechanism.

Abstract:The first iridium-catalyzed enantioselective coupling reaction of vinyl azides and vinyl benzoxazinones is presented. Vinyl azides underwent a tandem allylic alkylation/interrupted Schmidt rearrangement process to produce enantioenriched 3,4-dihydroquinolin-2(1 H )-imines, a new class of N-heterocycles. In the presence of CH 3 CO 2 H, a conventional asymmetric allylic substitution occurred to provide access to nonracemic allylic amides. The synthetic transformations of the product were carried out to construct chiral amines, amides and N-hetero polycycles.

Abstract:Highly functionalized quinolines and pyridines could be synthesized by BF3⋅OEt2-mediated reactions of vinyl azides with N-aryl and N-alkenyl aldimines, respectively. The reaction mechanism could be characterized as formal [4+2]-annulation, including unprecedented enamine-type nucleophilic attack of vinyl azides to aldimines and subsequent nucleophilic cyclization onto the resulting iminodiazonium ion moieties.

Abstract:Here we report a novel Rh-catalyzed C-H/C-H alkenylation of <i>N</i>-arylmethanimines with vinylene carbonate acting as a vinylene unit. Forty examples of C3,C4-nonsubstituted quinolines were achieved from commercially available starting materials. This identified process features an exceedingly simple system, a lower loading of catalyst, and the capacity for postfunctionalization with bioactive molecules.

Abstract:We have successfully developed a novel synthetic route for the synthesis of naphthyridinones. This was achieved through a reaction between formyl-phenylpropiolamides and arylamines, utilizing an intramolecular Povarov reaction. Notably, this reaction exhibited remarkable tolerance towards a wide range of substituents, resulting in the formation of diverse products using simple reaction conditions. Additionally, we discovered that the same substrates can undergo intramolecular carbonyl-alkyne metathesis reactions in the absence of anilines. This versatile transformation enabled the synthesis of quinolinones and chromen-2-ones with high yields.

Abstract:The synthesis of a biologically relevant 2-amino- N 3-alkylamido 4-quinazolinone has been accomplished in four steps from commercially available materials using design principles from both modular and divergent synthesis. N 3-alkylation of 2-chloro-4(3 H )-quinazolinone using methyl bromoacetate, followed by C2-amination produced a suitable scaffold for introducing molecular diversity. Optimization of alkylation conditions afforded full regioselectivity, enabling exclusive access to the N -alkylated isomer. Subsequent C2-amination using piperidine, pyrrolidine, or diethylamine, followed by amide bond formation using variously substituted phenethylamines, generated fifteen unique 4-quinazolinones bearing C2-amino and N 3-alkylamido substituents. These efforts highlight the reciprocal influence of C2 and N 3 substitution on functionalization at either position, establish an effective synthetic pathway toward 2, N 3-disubstituted 4-quinazolinones, and enable preliminary bioactivity studies while providing an experiential learning opportunity for undergraduate student researchers.

Abstract:Mn-III-catalyzed functionalization of vinyl azides via phosphorylation was developed to synthesize various beta-keto phosphine oxides in high yields. The transformation is operated in mild conditions and tolerant of a range of functional groups. Abstract:Here, a catalyst-controlled chemodivergent 1,6-addition of p-quinone methides with alkynes was developed, affording diverse alkynyl- and vinyl-substituted diarymethanes. In this transformation, copper catalyzed the direct 1,6-addition of p-QMs with alkynes, while iron promoted the three components reaction of p-QMs, alkynes, and halogens from iron salts or added HX acid. The salient features of this transformation include completely controllable chemoselectivity, mild conditions, inexpensive catalysts, and good substrates scopes.

Abstract:Mn(III)-mediated formal [3+3]-annulation has been developed using readily available vinyl azides and cyclopropanols with a wide range of substituents. Vinyl azides were successfully applied as a three-atom unit including one nitrogen to prepare pyridines and δ-lactams by the reactions with monocyclic cyclopropanols as well as to construct 2-azabicyclo[3.3.1] and 2-azabicyclo[4.3.1] frameworks with bicyclic cyclopropanols, bicyclo[3.1.0]hexan-1-ols, and bicyclo[4.1.0]heptan-1-ols. These reactions were initiated by a radical addition of β-carbonyl radicals, generated by the one-electron oxidation of cyclopropanols with Mn(III), to vinyl azides to give iminyl radicals, which cyclized with the intramolecular carbonyl groups. In addition, application of the present methodology to a synthesis of the quaternary indole alkaloid, melinonine-E, was accomplished.
