Abstract:Rh(III)-catalyzed C(sp(3)) - H bond aminocarbonylation of 8-methylquinolines and isocyanates has been realized under mild conditions. This approach is applicable to different aryl and alkyl isocyanates, leading to the synthesis of various alpha-quinolinyl amide compounds in moderate to excellent yields. A plausible mechanism for this transformation is proposed according to the experimental results obtained.

Abstract:Asymmetric expansion: A catalytic asymmetric ring-expansion reaction of the title compounds occurs in the presence of a Sc(OTf)(3) catalyst bearing an N,N'-dioxide-based ligand. Highly functionalized 2-quinolone derivatives containing a chiral C4-quaternary stereocenter were obtained in high yields and high levels of selectivity under mild reaction conditions (see scheme; Tf=trifluoromethanesulfonyl).

Abstract:A facile and novel approach to the synthesis of 2-phenylquinazolines was developed via a tandem reaction following sp(3) C-H functionalization. Twenty-five examples of 2-phenylquinazolines were obtained from easily available 2-aminobenzophenones and benzylic amines with good to excellent yields.

Abstract:An efficient method for the synthesis of substituted quinolines is described. By FeCl3-promoted C–C bond formation, 2,3-disubstituted and 2,3,4-trisubstituted quinolines were successfully synthesized from the reaction between imines and electron-deficient alkynes in good yields. The reaction can also be performed in a sequential three-component manner by using aromatic amines and aromatic aldehydes instead of the corresponding imines.

Abstract:An iron-catalyzed redox condensation of 2-nitrobenzyl alcohols, formic acid, and alcohols has been developed, which affords substituted quinolines with carbon dioxide and water as the only side products. With the use of formic acid as a redox moderator to fill the electron gap of the global redox condensation process, the reaction goes smoothly with a smaller amount of alcohol in comparison to previous reports (i.e. 1.2 equiv versus 3.3-4 equiv). The reaction goes equally well when 2-nitrobenzyl methyl ether was used instead of 2-nitrobenzyl alcohol under otherwise identical conditions, shedding a new light on the study of this quinoline synthetic method.

Abstract:Here we report a novel Rh-catalyzed C-H/C-H alkenylation of <i>N</i>-arylmethanimines with vinylene carbonate acting as a vinylene unit. Forty examples of C3,C4-nonsubstituted quinolines were achieved from commercially available starting materials. This identified process features an exceedingly simple system, a lower loading of catalyst, and the capacity for postfunctionalization with bioactive molecules.

Abstract:A wide range of isoquinoline ring-modified Quinap oxides with different steric and electronic variations have been constructed through a palladium-catalyzed imidoylative cyclization of arylethenyl isocyanides with 2-diphenylphosphinyl-1-naphthyl bromides. The Pd-catalysis plays dual roles in the formation of both axial C–C bond and isoquinoline ring. This de novo synthetic strategy features good functional group tolerance, high yields and easy scale-up, providing Quinap derivatives with substitution patterns that could not be obtained using coupling reactions. Chiral ligands 7 and 12 can be readily prepared by transformation of the resulting Quinap oxide to their BINOL esters, and have been proven to be superb chiral ligands for the copper-catalyzed enantioselective A3-coupling and alkynylation of quinoline reactions. In general, the enantioselectivies obtained using ligands 7 and 12 are excellent, and the ee values are higher than those using Quinap as ligand, even three times higher in some cases. Mechanism studies revealed that a monomeric copper(I) complex bearing a single chiral ligand was formed and served as the catalytically active species.

Abstract:C-N bond formation is crucial for the preparation of synthetic organic nitrogen-containing heterocyclic compounds. This work provides a facile and highly efficient strategy (ca. similar to 92 %) for the synthesis of quinazoline compounds, which involves tandem C-H/N-H bond functionalization using metal-free conditions without the need for an external oxidant. Meanwhile, the reaction system exhibits good functional group tolerance and a wide substrate scope. Furthermore, our corresponding isotope tracking experiments have shown that dimethyl sulfoxide acts as a carbon source. Finally, this work affords a new strategy toward the further design and synthesis of heteroaromatic rings.

Abstract:An electrochemical protocol for the synthesis of 2,3-disubstituted quinolines by reacting benzoxazinones with arylsulfonyl hydrazides was established under simple and mild conditions (room temperature and an undivided cell). The methodology does not require noble catalysts or external oxidants, providing a green and mild pathway for de novo synthesis of functional quinolines with excellent regioselectivity. Moreover, the method tolerated a variety of functional groups. Notably, the scaled-up experiment and follow-up procedures demonstrate the practicality of electrochemical synthesis.

Abstract:Multi-heteroatom heterocycle synthesis through direct C-H bond activation is methodologically appealing but synthetically challenging. An efficient double C-N bond formation sequence to prepare quinazolinones utilizing primary amides and oxadiazolones in a catalytic redox-neutral [CoCp*(CO)I-2]/AgSbF6 system, where oxadiazolone could function as an internal oxidant to maintain the catalytic cycle, is reported. Amide-directed C-H bond activation and oxadiazolone decarboxylation are key to the success of this traceless, atom- and step-economic, and cascade approach for the construction of the quinazolinone skeleton.

This method is enabled by a three-component cascade annulation of readily available aryl diazonium salts, nitriles, and alkynes. This reaction is catalyst- and additive-free. Various aryl diazonium salts, nitriles, and alkynes can participate in this transformation, and the yields are up to 83%.

Abstract:Without any metal catalyst, a simple and efficient method for the synthesis of E-2-styrylquinolines through sp3 C-H cross-dehydrogenative coupling of benzylamines with 2-methylquinolines mediated by NH4I under air is successfully developed. The oxidative olefination proceeded through deamination and sp3 C–H bond activation. A plausible mechanism is proposed for the construction of E-2-styrylquinolines.
