Acceleration of Hen Egg White Lysozyme Amyloid Fibrillation by Single- or Few-Layer Molybdenum Disulfide Nanosheets

Abstract:Since molybdenum disulfide has attracted considerable attention as a result of its promising applications in vivo, it is of great importance to investigate the potential effects of MoS2 on the biomolecules. Here, we paid attention to explore the relationship between MoS2 nanosheets and the protein amyloid fibrillation. Hen egg white lysozyme was used as a model protein. By using atomic force microscopy and thioflavin T fluorescence spectroscopy, it was found that two kinds of MoS2 nanosheets led to an enhancement at the fibrillation stage by decreasing the lag time. Compared with few-layer MoS2 nanosheets, the role of single-layer MoS2 nanosheets in accelerating lysozyme fibrillation was much significant. In addition, according to the decreased intrinsic fluorescence signals and the increased beta-sheet content of lysozyme in circular dichroism, we speculated that a mechanism might be due to strong noncovalent interactions and protein conformational changes, which might induce a local high concentration of lysozyme on the nanosheet surface and then favor the nucleation. The work may lay a good foundation for the research of interactions between MoS2 and proteins. Furthermore, it can enrich our understanding of the possibility of inducing diseases, especially amyloidosis, by nanomaterials as an environmental factor.
