Abstract:The 15-day intact adult male assay was used to evaluate effects of isoflurane on the testes and sexual hormones. Forty adult male Sprague-Dawley rats were divided into five groups exposed to air containing 0, 50, 300, 1800 or 10,800 ppm isoflurane. After the treatments, serum was collected for the hormones assay. The right testis was to be used for daily sperm production. The left testis was processed for histopathology and electron microscopy observation. Daily sperm productions were significantly decreased at doses of 300, 1800 and 10,800 ppm. Impaired seminiferous tubules were noted at doses of 300, 1800 and 10,800 ppm. Ultrastructural changes included nucleus agglutination of spermatocytes, big lipid drops and autophagosome in cytoplasm. The serum follicle-stimulating hormone and testosterone concentrations reduced significantly at doses of 1800 and 10,800 ppm. Isoflurane induced impairments of seminiferous tubules and spermatogenesis. The testicular damages caused by isoflurane can be related to the imbalances in the sexual hormones.

Abstract:OBJECTIVE:To study the effects of inhaled anesthetics on human sperm motility in vitro.METHODS:Sperm samples were obtained from 20 healthy men by masturbation and prepared by the swim-up technique. The effects of isoflurane and sevoflurane at the clinical concentration (1.4%-5.6%) and high concentration (5.6%-84%) on human sperm motility in vitro were observed at 25 degrees C by the computer-assisted sperm analysis (CASA).RESULTS:The sperm vitality and motility were significantly increased on 0.5-4 h exposure to isoflurane at the clinical concentration and decreased gradually at high concentration (42%-84%). The effect of isoflurane on human sperm motility and vitality at the clinical concentration was reversible when the anesthetic withdrawn. Sevoflurane had no effects on human sperm motility and vitality at either the clinical or high concentration.CONCLUSION:Isoflurane has a reversible increasing effect at the clinical concentration and a significant decreasing effect at the high concentration on the motility and vitality of human sperm, while sevoflurane does not affect human sperm motility and vitality at either concentration.
The effects of isoflurane on human sperm motility in asthenospermia in vitro

Abstract:To investigate the effects of isoflurane on human sperm motility in asthenospermia in vitro.After 10 asthenospermic and 10 normospermic samples were collected,each was divided into 12 0.1mL aliquots,then the 12 aliquots were allocated to three equal groups.After 0.1mL semen was added into the culture capsules,the four aliquots of each group were exposed to 0L(as control),5L,10L and 20L isoflurane(not contact directly with semen and the corresponding vol% of isoflurane is 0,1.4,2.8,and 5.6 respectively),and then the culture capsules were sealed immediately.The test was performed at 25℃ and the motility and viability of sperm were analysed by computer-assisted sperm analysis at 0.5h,1h and 2h respectively.The results showed that ① Compared with control,1.4%～5.6% isoflurane obviously increased the motility and viability of asthenosperm at 0.5h and 1h,effected no significant difference at 2h,and normospermia treated with 1.4%～5.6% isoflurane showed an significant increase in the motility and viability of sperm at 0.5h,1h and 2h as compared with control;② 5.6% isoflurane obviously increased the motility and viability of asthenosperm at 0.5h and 1h,effected no significant difference at 2h,and normospermia treated with 5.6% isoflurane had a significant increase the motility and viability of sperm at 0.5h,1h and 2h as compared with 1.4% or 2.8% isoflurane;③ 1.4%～5.6% isoflurane markedly decreased the motility and viability of asthenosperm at 1h and 2h as compared with previous time,and sperm parameters in normospermia showed no significant difference between three time;④ The increasing amplitude of motility and viability in asthenospermia treated with 5.6% isoflurane for 0.5h were higher than those in normospermia.Isoflurane not only increased sperm motility in asthenospermia and normospermia,but also the increasing amplitude of motility and viability in asthenospermia were higher than those in normospermia.
[Effects of Gestational Isoflurane Exposure on Postnatal Memory and Learning in Rats].

Abstract:A maternal fetal rat model was developed to study the effects of gestational isoflurane exposure on postnatal memory and learning and investigate the potential mechanisms. Pregnant rats at gestational day 14 were exposed to 1.3% isoflurane for 4h. Spatial learning and memory of the offspring were examined using the Morris Water Maze. The expression levels of C/EBP homologous transcription factor protein (CHOP) and caspase-12 in the hippocampus of the pups were determined by immunohistochemistry and western blot analysis. Simultaneously, the ultrastructure changes of synapse in the hippocampal CA1 and dentate gyrus region were also observed by transmission electron microscopy (TEM). Prenatal exposure to isoflurane impaired postnatal spatial memory and learning in the offspring rats as shown by the longer escape latency and the fewer times of original platform crossing in the Morris Water Maze test. The number of CHOP and caspase-12 positive neurons significantly increased by 138% and 147% respectively in the hippocampus of isoflurane-exposed pups, as well as the levels of CHOP and caspase-12 protein. Furthermore, TEM studies showed changes of synaptic ultrastructure in isoflurane-exposed hippocampus characterized by the decreased synapse number, the widened synaptic cleft and the thinned postsynaptic densities. These results demonstrate that gestational exposure to a clinically relevant concentration of isoflurane could cause neuron apoptosis, changes of synaptic structure, and postnatal spatial memory and learning impairments in offspring. Our study further showed that the up-regulation of CHOP and caspase-12 may contribute to isoflurane-induced neuron apoptosis.
Effects of diazepam and midazolam on contraction of rat uterine smooth muscles

Abstract:AIM To compare effects of diazepam and midazolam on the isolated pregnant and nonpregnant rat uterine smooth muscles.METHODS Uterine strips from the pregnant and nonpregnant rats isolated and prepared.After contractions became regular,strips were exposed to cumulative concentrations(0.01-300 μmol·L~(-1)) of diazepam and midazolam.Contractile amplitude and frequency of the isolated uterine smooth muscles were recorded by MedLab Biological Signal Collection System.RESULTS Both drugs inhibited contractile amplitude in a concentration-dependent manner on myometrium from non-pregnant and pregnant rats.On myometrium of non-pregnant rats,the IC_(50) of diazepam and midazolam was 280.6 and 1.1 μmol·L~(-1),respectively.On myometrium of pregnant rats,the IC_(50) of diazepam and midazolam was 52.2 and 28.2 μmol·L~(-1),respectively.Diazepam had slight inhibitory effect on the contractile frequency,while midazolam completely depressed the contractile activity at the highest concentration.CONCLUSION The inhibitory effect of midazolam is stronger than that of diazepam,especially on the myometrium from nonpregnant rats,providing experimental basis for clinical use of midazolam on parturition.
The Effect of Sevoflurane on the Cognitive Function of Rats and Its Association with the Inhibition of Synaptic Transmission.

Abstract:To observe the effects of different concentrations of sevoflurane on synaptotagmin 1 (Syt1) expression, synaptic long term depression (LTD), and paired pulse depression (PPD) in the rat hippocampus as well as to investigate the association between these effects and the cognitive function of rats. A total of 24 male Sprague-Dawley (SD) rats were selected and randomly divided into 3 groups: the control group (group A), which inhaled air; group B, which inhaled 0.65 minimum alveolar concentration (MAC) sevoflurane for 2 h; and group C, which inhaled 1.30 MAC sevoflurane for 2 h. The subsequent experiments were performed after one day. (1) Y maze tests were performed, and the expression of Syt1 in hippocampal tissues was detected using western blot. (2) The changes in LTD and PPD in rat hippocampal slices were examined using electrophysiological techniques. Compared to the control group, the cognitive function was decreased and Syt1 expression in the hippocampus was significantly decreased in rats in the 1.30 MAC sevoflurane inhalation group. After 60 min of low frequency stimulation, the amplitudes of population spike (PS) potentials in rat hippocampal slices were significantly decreased. After induction of PPD, the P2/P1 ratio was significantly increased. No indicators in the 0.65 MAC sevoflurane inhalation group showed any significant changes. Inhalation of high concentrations of sevoflurane significantly reduced Syt1 protein levels in the rat hippocampus, significantly inhibited the release of presynaptic neurotransmitters, and reduced the efficiency of synaptic transmission, thus causing memory impairment.

Abstract:Objective This study was designed to study the effect of Diltiazem on patency rate of arteriovenous anastomosis in rat and how it works.Methods 24 SD rats were divided into control group and experimental group,12 rats in each group.Experimental group rats were gavaged with Diltiazem after vascular anastomosis.Control group rats were gavaged with water.By comparing the patency rate and the thickness of artery to make sure whether Diltiazem will affect the patency rate.;By comparing the clotting time,prothrombin time,artial thromboplastin time,and serum thromboxane B2 levels to explore the pathway of diltiazem.Results The patency rate was 75％ in the experimental group and 25％ in control group.Compared with the control group,experimental group venous blood vessels in the film segment was significantly thicker,clotting time was prolonged,TXB2 levels in blood was decreased,the differences were statistically significant(P ＜ 0.05).There were no significant difference in prothrombin time and partial thromboplastin between two groups (P ＞ 0.05).Conclusion Diltiazem can inhibit the secretion of TXB2,antagonize the effct of antiplatelet,and increase the patency rate of vascular anastomosis in rats.

Abstract:AIMS:To evaluate effects of diltiazem on platelet hyper-reactivity in situations associated with endothelial injury and their possible relationship to cytosolic calcium concentration.METHODS:Blood samples were collected at seven time points from 35 patients undergoing percutaneous transluminal coronary angioplasty (PTCA) who received combined diltiazem and aspirin/ticlopidine therapy or aspirin/ticlopidine therapy alone. Platelet expression of glycoprotein IIb/IIIa and P-selectin, production of thromboxane B(2), and cytosolic calcium concentration were measured, respectively, by whole blood flow cytometry, radioimmunoassay, and fluorospectrophotometry. The effects of diltiazem of different concentrations on expression of glycoprotein IIb/IIIa and P-selectin were also studied in vitro in blood samples from patients with chronic stable angina.RESULTS:Of the two treatments, aspirin/ticlopidine therapy did not prevent an acute increase of expression of glycoprotein IIb/IIIa and P-selectin and plasma thromboxane B(2) five minutes and 10 minutes after first inflation and 10 minutes after PTCA, whereas combined diltiazem and aspirin/ticlopidine therapy had a significant inhibitory effect. In the group receiving aspirin/ticlopidine therapy, there was a short term increase of platelet [Ca(2+)](i) immediately after PTCA which was significantly reduced by diltiazem treatment. Expression of glycoprotein IIb/IIIa and P-selectin was significantly inhibited in vitro by diltiazem in the concentration of 200 ng/ml or higher, but not 50 ng/ml.CONCLUSIONS:Combined diltiazem and aspirin/ticlopidine therapy significantly inhibited platelet activation that continued in the presence of conventional aspirin/ticlopidine treatment. Antiplatelet effects of diltiazem were probably a consequence of reduction of platelet [Ca(2+)](i) and may only be achieved in higher than therapeutic concentrations.

Abstract:In recent years,a large number of pre-clinical cell and animal studies have shown that isoflurane as the representative of a variety of clinical commonly used inhaled anesthetics may induce neurotoxicity,including neuroapoptosis,neurodegeneration,and long-term neurocognitive and behav? ioral deficits. Mitochondrial dysfunction and oxidative stress play an important role in the side effect. This paper reviews the currently relatively clear mechanisms by which isoflurane causes postoperative cognitive dysfunction and prevention methods. Specific mechanisms of these effects include inhibition of the hydrolysis of brain-derived neurotrophic factor precursor in hippocampal neurons,promotion of neuronal apoptosis, overactivation of 1,4,5-inositol trisphosphate receptor, intracellular calcium imbalance,acetylcholine inhibition,inhibition of long-term potentiation. At the same time,current preventive studies are introduced,such as dexmedetomidine and xenon.
Influence of Diltiazem on the Behavior of Zolpidem-Treated Mice in the Elevated-Plus Maze Test

Abstract:The present study was undertaken to investigate the effect of diltiazem, a L-type calcium channel blocker (CCB), on the behavior of zolpidem-treated mice in the elevated plus-maze (EPM). Atypical benzodiazepine zolpidem significantly increased the percentage of open arm entries without influencing the total entries and total distance and average speed at the dose of 5 mg/kg (p.o.). Co-administration of zolpidem (2 mg/kg, p.o.) and diltiazem (5, 10 and 20 mg/kg, p.o.) significantly increased both the time spent and arm entries in the open arms without influencing the total entries and spontaneous activity notwithstanding that zolpidem at dose up to 2 mg/kg (p.o.) and diltiazem at dose up to 20 mg/kg (p.o.) did not show any effects on mice behavior in EPM. Zolpidem also attenuated the anxiogenic effect of 1-(3-Chlorophenyl)piperazine (mCPP, 0.7 mg/kg, i.p.) and 5-hydroxytryptophan (5-HTP, 30 mg/kg, i.p.). Even though the zolpidem at 1 mg/kg and diltiazem at 5 mg/kg were ineffective on mCPP-induced anxiety, respectively, the co-administration of zolpidem (1 mg/kg, i.p.) and diltiazem (5 mg/kg, p.o.) showed inhibitory effect on mCPP-induced anxiety in mice. These results suggested that diltiazem, a L-type CCB may augment the anxiolytic-like effect of zolpidem and also indicated that calcium channel modulation maybe involved in the anxiolytic-like properties of zolpidem.
Dual effects of isoflurane on proliferation, differentiation, and survival in human neuroprogenitor cells

Abstract:Background: Previous studies have demonstrated that isoflurane can provide both neuroprotection and neurotoxicity in various tissue culture models and in rodent developing brains. The cellular and molecular mechanisms mediating these dual effects are not clear, but the exposure level and duration of isoflurane appear to be determinant factors. Methods: Using the ReNcell CX (Millipore, Billerica, MA) human neural progenitor cell line, the authors investigated the impact of prolonged exposure to varying isoflurane concentrations on cell survival and neurogenesis. In addition, the authors assessed the impact of short isoflurane preconditioning on elevation of cytosolic Ca concentration and cytotoxic effects mediated by prolonged isoflurane exposures and the contribution of inositol-1,4,5-trisphosphate or ryanodine receptor activation to these processes. Results: Short exposures to low isoflurane concentrations promote proliferation and differentiation of ReNcell CX cells, with no cell damage. However, prolonged exposures to high isoflurane concentrations induced significant ReNcell CX cell damage and inhibited cell proliferation. These prolonged exposures suppressed neuronal cell fate and promoted glial cell fate. Preconditioning of ReNcell CX cultures with short exposures to low concentrations of isoflurane ameliorated the effects of prolonged exposures to isoflurane. Pretreatment of ReNcell cultures with inositol-1,4,5-trisphosphate or ryanodine receptor antagonists mostly prevented isoflurane-mediated effects on survival, proliferation, and differentiation. Finally, isoflurane-preconditioned cultures showed significantly less isoflurane-evoked changes in calcium concentration. Conclusion: The commonly used general anesthetic isoflurane exerts dual effects on neuronal stem cell survival, proliferation, and differentiation, which may be attributed to differential regulation of calcium release through activation of endoplasmic reticulum localized inositol-1,4,5-trisphosphate and/or ryanodine receptors.
Postconditioning of Sevoflurane and Propofol is Associated with Mitochondrial Permeability Transition Pore

Abstract:Background: Sevoflurane and propofol are effective cardioprotective anaesthetic agents, though the cardioprotection of propofol has not been shown in humans. Their roles and underlying mechanisms in anesthetic postconditioning are unclear. Mitochondrial permeability transition pore (MPTP) opening is a major cause of ischemia-reperfusion injury. Here we investigated sevoflurane-and propofol-induced postconditioning and their relationship with MPTP. Methods: Isolated perfused rat hearts were exposed to 40 min of ischemia followed by 1 h of reperfusion. During the first 15 min of reperfusion, hearts were treated with either control buffer (CTRL group) or buffer containing 20 μmol/L atractyloside (ATR group), 3% (v/v) sevoflurane (SPC group), 50 μmol/L propofol (PPC group), or the combination of atractyloside with respective anesthetics (SPC+ATR and PPC+ATR groups). Infarct size was determined by dividing the total necrotic area of the left ventricle by the total left ventricular slice area (percent necrotic area). Results: Hearts treated with sevoflurane or propofol showed significantly better recovery of coronary flow, end-diastolic pressures, left ventricular developed pressure and derivatives compared with controls. Sevoflurane resulted in more protective alteration of hemodynamics at most time point of reperfusion than propofol. These improvements were paralleled with the reduction of lactate dehydrogenase release and the decrease of infarct size (SPC vs CTRL: (17.48±2.70)% vs (48.47±6.03)%, P<0.05; PPC vs CTRL: (35.60±2.10)% vs (48.47±6.03)%, P<0.05). SPC group had less infarct size than PPC group (SPC vs PPC: (17.48±2.70)% vs (35.60±2.10)%, P<0.05). Atractyloside coadministration attenuated or completely blocked the cardioprotective effect of postconditioning of sevoflurane and propofol. Conclusion: Postconditioning of sevoflurane and propofol has cardioprotective effect against ischemia-reperfusion injury of heart, which is associated with inhibition of MPTP opening. Compared to propofol, sevoflurane provides superior protection of functional recovery and infarct size.

Abstract:AIM:Aquaporins (AQPs) are water-channels that play important roles in brain water homeostasis and cerebral edema induced by brain injury. This study aimed to investigate the relationship between AQP4, bone morphogenetic protein 4 (BMP4)/Smad1/5/8 signaling pathway and isoflurane post-conditiong, which has effects on brain edema in rats with cerebral ischemia/reperfusion (I/R) injury. METHODS:Cerebral I/R injury was induced in rats by using the middle cerebral artery occlusion (MCAO) model for 90min, followed by 24h of reperfusion. Isoflurane post-conditioning (ISO) group received 90min ischemia and underwent 1.5% isoflurane post-conditioning for 60min after initiating reperfusion. Neurobehavior, brain water content, thionine staining and 2, 3, 5-triphenyl tetrazolium chloride staining were evaluated to measure levels of brain edema and damage. Expressions of AQP4, BMP4, Smad1/5/8 and phosphorylated Smad1/5/8 were detected by using Western blot, quantitative real-time polymerase chain reaction (qRT-PCR), and immunofluorescence (IF) staining. RESULTS:Compared with the Sham group, neurological behavior score, brain infarct volume and water content of MCAO model rats increased with reperfusion injury. However, in the ISO group, cell edema and damage of brain was significantly ameliorated (P<0.01). qRT-PCR showed less AQP4 mRNA expression in the hippocampal tissue of the ISO group than in the I/R group (P<0.01). Western blot and immunofluorescence results showed similar changes in protein levels of both groups. Related protein expressions showed expressions of BMP4 and Smad1/5/8 increased in the ISO group (P<0.01), whereas total Smad1/5/8 expression didn't change in all groups. When BMP4 inhibitor (LDN193189) was injected, expression levels of AQP4 increased and neuronal density decreased (P<0.05). By contrast, expression levels of BMP4 did not change significantly after pre-injection of AQP4 inhibitor (TGN020) (P>0.05), but neuronal density increased (P<0.05). CONCLUSION:Isoflurane post-conditioning may inhibit occurrence of brain edema and reduce cerebral I/R injury through down-regulating expression of AQP4, This process may be related to the activation of BMP4/Smad1/5/8 signaling pathway.

Abstract:BACKGROUND: Isoflurane has been demonstrated to limit myocardial ischemic injury. This effect is hypothesized to be mediated in part via effects on mitochondria. We investigated the hypothesis that isoflurane maintains mitochondrial respiratory chain functionality, in turn limiting mitochondrial damage and mitochondrial membrane disintegration during myocardial ischemic injury. METHODS: Mice (9-12 weeks of age) received isoflurane (1.0 minimum alveolar concentration) 36 hours before a 30-minute coronary artery occlusion that was followed by 24 hours of reperfusion. Cardiac mitochondria were isolated at a time point corresponding to 4 hours of reperfusion. 2,3,5-Triphenyltetrazoliumchloride staining was used to determine myocardial infarct size. Mitochondrial respiratory chain functionality was investigated using blue native polyacrylamide gel electrophoresis, as well as specific biochemical assays. Mitochondrial lipid peroxidation was quantified via the formation of malondialdehyde; mitochondrial membrane integrity was assessed by Ca2+-induced swelling. Protein identification was achieved via liquid chromatography mass spectrometry/mass spectrometry. RESULTS: Thirty-one mice were studied. Mice receiving isoflurane displayed a reduced myocardial infarct size (P = 0.0011 versus ischemia/reperfusion [I/R]), accompanied by a preserved activity of respiratory complex III (P = 0.0008 versus I/R). Isoflurane stabilized mitochondrial supercomplexes consisting of oligomers from complex III/IV (P = 0.0086 versus I/R). Alleviation of mitochondrial damage after isoflurane treatment was further demonstrated as decreased malondialdehyde formation (P = 0.0019 versus I/R) as well as a diminished susceptibility to Ca2+-induced swelling (P = 0.0010 versus I/R). CONCLUSIONS: Our findings support the hypothesis that isoflurane protects the heart from ischemic injury by maintaining the in vivo functionality of the mitochondrial respiratory chain. These effects may result in part from the preservation of mitochondrial supramolecular organization and minimized oxidative damage, circumventing the loss of mitochondrial membrane integrity.
The effect of mitochondrial calcium uniporter opener spermine on diazoxide against focal cerebral ischemia--reperfusion injury in rats

Abstract:Background: Recent research has indicated that mitochondrial adenosine triphosphate-sensitive potassium channels play an important role in cerebral protection, which involves in attenuating the calcium of mitochondria. However, the effect of diazoxide on cerebral ischemia-reperfusion and the role of spermine, the agonist of mitochondrial calcium uniporter (MCU), remain unknown. Objective: We investigated the effect of MCU opener spermine on diazoxide against focal cerebral ischemia-reperfusion injury in rats. Methods: Adult male Wistar rats were randomly divided into 5 groups: the Sham group, the I/R group, the Dzx + I/R group, the Dzx + Sper + I/R group, and the Sper + I/R group. Rats were exposed to 2-hour ischemia and 24-hour reperfusion. Diazoxide were administrated 30 minutes before ischemia, and spermine were given 10 minutes before reperfusion. Rats in the Sham group did not experience the process of ischemia-reperfusion. After 24-hour reperfusion, rats were given neurological performance tests, overdosed with general anesthesia, and then their brains were excised for infarct volume, pathological changes, and biochemical evaluation and analysis. Results: Rats in the Dzx + I/R group displayed improved neurological deficits and decreased infarct volume and oxidative stress (evidenced by decreased nitric oxide and malondialdehyde but increased antioxidant enzymes [eg, glutathione peroxide and superoxide dismutase]) caused by ischemia-reperfusion. The beneficial effects of diazoxide were significantly attenuated by spermine treatment. Rats in the Sper + I/R group displayed worse neurological deficits, larger infarct volume and more oxidative stress, and less antioxidant enzymes than those in the Dzx + I/R. Conclusions: Our results suggested that diazoxide, which improved neurological deficits and decreased infarct volume and oxidative stress against ischemia-reperfusion injury, is mediated by spermine.
Persistent mitoK ATP Activation Is Involved in the Isoflurane-induced Cytotoxicity

Abstract:Isoflurane exposure induces apoptosis in cultured cells and in the developing brain, while the underlying mechanism remains largely unclarified. This study was designed to determine whether the disruption of mitoK ATP -mediated ATP balance was involved in the cytotoxicity of isoflurane. Human neuroglioma cells U251 and 7-day-old mice were treated with isoflurane. A specific mitoK ATP antagonist 5-HD was used, and the cellular ATP levels, NAD + /NADH ratios, and mitochondrial transmembrane potential (ΔΨ m ) were measured. Our data showed that the blockage of mitoK ATP by 5-HD mitigated the isoflurane-induced ΔΨ m disruption, reactive oxygen species (ROS) accumulation, and apoptosis in U251 cells. Moreover, we found that the toxic effect of isoflurane was not observed in the first 2-h exposure; instead, the cellular ATP levels and NAD + /NADH ratios were markedly increased. The reduction of ATP levels and NAD + /NADH ratios was only detected after this initial phase. This dynamical effect of isoflurane was blocked by 5-HD. In contrast, a ROS scavenger NAC sustained the isoflurane-induced ATP elevation. Similar results were observed in animal studies. And again, 5-HD attenuated isoflurane-induced cognitive disorders in the Intellicage test, a system that assesses place learning behavior in a social environment. Our study uncovered a potential mechanism underlying isoflurane’s toxicity with a therapeutic future.
Effect of Opioid Analgesics on Human Sperm Motility in Vitro

Abstract:Objective To assess the effects of fentanyl (F),alfentanil (A),sufentaril (S),butorphanol (B),dezocine (D) and pentazocine (P) on human sperm motility in vitro.Methods Twenty samples of semen with normal sperm motility were collected.The semen was mixed with HTF liquid culture medium and sperm concentration was adjusted to (40-80) × 106/ml.Then semen suspension 50 μl was added into the culture dishes.The semen suspensions were randomly divided into 3 groups:group Ⅰ control-semen suspension 50 μl + 0.9％ NaCl 50 μl.GroupⅡ single opioid-semen suspension 50 μl + 50 μl of 4 concentrations of F,A,S,B,D and P (10-3,10-4,10-5,10-7 mol/L).Group Ⅲ combined opioid-naloxone-semen suspension 50 μl + 25 μl of 3concentrations of naloxone (2 × 10-8,1 × 10-7,2 × 10-7 mol/L) + 25 μl of 4 concentrations of F,A,S,B and D.After being incubated for 1 h the sperm motility was assessed by sperm analyzer and the amplitude of change in sperm motility was calculated.Median inhibition concentration (IC50) and complete inhibition concentration ( IC100 ) of the 6 opioids were calculated from concentration-response curve.Results ( 1 ) The shape of concentration-response curve for F,A and S was straight line.The maximal decrease in sperm motility was (56 ± 5)％(F),(58±7)％ (A) and(79±:6)％ (S).(2) The shape of concentration-response curve for B,D and P was sigmoid.Low concentrations of B,D and P did not affect sperm motility.Median and high concentrations of B and D could inhibit sperm motility.The maximal inhibition was 100％.High concentration of P increased sperm motility.The maximal increase was (19 ± 6)％.(3) Naloxone shifted the concentration-response curve for F,A,S,B and D to the right,and decreased the maximal inhibitory effect of F,A and S dose-dependently but did not affect the maximal inhibitory effect of B and D.(4) IC50 was in the order of A≈ F ＞ D ＞ S≈- B.IC100 was D ＞ B.Conclusion F,A and S inhibit sperm motility by acting on opioid receptor on human sperms while B and D act through opioid or nonopioid pathways.P can increase sperm motility.

Abstract:Objective To study the effects of lidocaine on sevoflurane postconditioning-induced cardioprotection.Methods Ischemic status was kept for 40 rain in isolated perfused rat hearts followed by 1 h of reperfusion.Sevoflurane(3%) was administered at the beginning of reperfusion for 15 rain with or without lidocaine (20 μg/ml) perfusion.The direct mitochondrial permeability transition pore (MPTP) inhibitor Cyclosporin A (CsA,0.2 μmol/L) was co-administered in the presence or absence of lidocaine.LVDP,LVEDP,+dp/dtmax were recorded and infarct size was measured with TTC staining.Results Sevoflurane postconditioning significantly improved the recovery of ischernic myocardial function and decreased the infarct size of rat hearts (P＜0.05),which was abolished by lidocaine perfusion.The inhibition of lidocaine on sevoflurane posteonditioning effect was reversed by CsA.Conclusion Sevoflurane postconditioning effectively protects myocardium against ischemia/reperfusion injury,and higher concentration of lidocaine inhibits this protective effect by opening MPTP.
Effects of Isoflurane on Synthesis of Phosphatidylcholine in Adult Rat Lungs

Abstract:Objective To investigate whether inhaled different concentrations of isoflurane(Iso)may affect the synthesis of phosphatidylcholine(PC)in mature lungs.Methods Seventy-eight adult male Wistar rats were randomly divided into four groups.Furthermore,for each of group A to D,3 subgroups(n=6)were established according to the duration of the gas administration.High performance liquid chromatography(HPLC)was employed to deter mine the PC concentration in bronchoalveolar lavage fluid(BALF).The mRNA expression and protein of cytidine triphosphate:phosphocholine cytidylyltransferase(CCT)were measured by reverse transcription polymerase chain reaction(RT-PCR)and Western blot.Results The level of mRNA and protein of CCT in lung homogenates were not significantly different among all groups.Compared with group E and 4 h subgroup,in group B2 and C2,both the PC concentration in BALF and the expression of microsomal CCT in pulmonary tissue reached the nadir(P0.05)and recovered at 2 hours after the cessation.During 8 h exposure,in group B2 and C2,the PC concentration and the expression of microsomal CCT were lower than in group D2(P0.05),while both parameters in group C2 were also lower than in group A2(P0.05).Conclusion Isoflurane(1.5%,2.0%)inhalation for a longer duration(8 hours)may reversibly impair the de novo phospholipids synthesis in rat lung,of which the mechanism may be associated with the decreased activity of CCT.
