Abstract:A method is developed that can rapidly produce blood vessel-like structures by bonding cell-laden electrospinning (ES) films layer by layer using fibrin glue within 90 min. This strategy allows control of cell type, cell orientation, and material composition in separate layers. Furthermore, ES films with thicker fibers (polylactic-co-glycolic acid, fiber diameter: ≈3.7 µm) are used as cell-seeding layers to facilitate the cell in-growth; those with thinner fibers (polylactic acid, fiber diameter: ≈1.8 µm) are used as outer reinforcing layers to improve the mechanical strength and reduce the liquid leakage of the scaffold. Cells grow, proliferate, and migrate well in the multilayered structure. This design aims at a new type of blood vessel substitute with flexible control of parameters and implementation of functions.
Fabrication of a tubular vascular scaffold with circumferential microchannels to induce oriented growth of smooth muscle cells

Abstract:BACKGROUND: In vascular tissue engineering, scaffolds that can induce smooth muscle cells align and orientate in circumferential direction, but not simple porous scaffold, would be welcome. OBJECTIVE: To observe the effect of circumferential microchannels on the in vitro induction of smooth muscle cells. METHODS: Electrospinning, melt spinning, and solvent adhesion techniques were combined and the as-prepared scaffolds were further modified by grafting collagen for improving their biocompatibility. Scanning electron microscope and fluorescence microscope were selected to characterize the alignment of smooth muscle cells on these biomimetic scaffolds. RESULTS AND CONCLUSION: Mixture of chloroform/alcohol with volume ratio at 5:95 was used to bond the elextro spun poly(lactic-co-glycolic acid) fibers and melt spun poly(ε-caprolactone-co-lactic acid) fibers successfully toward creating a biomimetic vascular scaffold. The scaffold surface was first introducing active carboxylate group by alkali hydrolyzing and then coupling with collagen by using carbodiimide. When the weaving angle in the melt spinning was suitable (30°), the pores of the scaffold could ensure the growth of smooth muscle cells on and in the scaffold homogenously. All the smooth muscle cells aligned along the microfibers and microchannels of the scaffolds, demonstrating that such novel type of scaffold had strong ability in inducing smooth muscle cells regenerating their microarchitecture in vivo. It is helpful in developing biomimetic tubular scaffold to induce blood vessel regeneration in vivo.

Abstract:The fabrication of heterogeneous microstructures, which exert precise control over the distribution of different cell types within biocompatible constructs, is important for many tissue engineering applications. Here, bioactive microfibers with tunable morphologies, structures, and components are generated and employed for creating different tissue constructs. Multibarrel capillary microfluidics with multiple laminar flows are used for continuously spinning these microfibers. With an immediate gelation reaction of the cell dispersed alginate solutions, the cell-laden alginate microfibers with the tunable morphologies and structures as the designed multiple laminar flows can be generated. The performances of the microfibers in cell culture are improved by incorporating bioactive polymers, such as extracellular matrix (ECM) or methacrylated gelatin (GelMA), into the alginate. It is demonstrated that a series of complex three-dimensional (3D) architectural cellular buildings, including biomimic vessels and scaffolds, can be created using these bioactive microfibers.

Abstract:The diameter of most blood vessels in cardiovascular and peripheral vascular system is less than 6 mm. Because the inner diameter of such vessels is small, a built-in stent often leads to thrombosis and other problems. It is an important goal to replace it directly with artificial vessels. This paper creatively proposed a preparation method of a small-diameter artificial vascular graft which can form a controllable microstructure on the inner wall and realize a multi-material composite. On the one hand, the inner wall of blood vessels containing direct writing structure is constructed by electrostatic direct writing and micro-imprinting technology to regulate cell behavior and promote endothelialization; on the other hand, the outer wall of blood vessels was prepared by electrospinning PCL to ensure the stability of mechanical properties of composite grafts. By optimizing the key parameters of the graft, a small-diameter artificial blood vessel with controllable microstructure on the inner wall is finally prepared. The corresponding performance characterization experimental results show that it has advantages in structure, mechanical properties, and promoting endothelialization.

Abstract:Nowadays, many studies focus on the preparation of small-diameter vascular scaffolds, but some issues still existed that need to be urgently solved, but there are still problems such as slow endothelialization and failure to keep the vessels open for a long time that need to be solved. In this paper, a composite process of preparing triple-layer vascular scaffolds with microchannel structure in the inner layer is proposed to guide cell growth and accelerate the endothelialization process. The PCL pattern was printed as the intermediate layer of the vascular scaffold by 3D printing method, and the width of the microchannel in the inner layer of the scaffold was controlled by the printing spacing. The inner and outer layers were prepared by electrospinning. Heparin is often used as an anticoagulant in vascular tissue engineering to improve the problem of thrombosis and blockage of vascular scaffold after transplantation into the body. Here, the inner layer carries heparin immobilized with silk fibroin by coaxial electrospinning, so that heparin can be released slowly. The addition of silk fibroin also improved the hydrophilicity of PCL electrospinning film by water contact angle test. The mechanical properties of vascular scaffolds with double intermediate layers (DIL) are better than those with single intermediate layers (SIL) and also better than pure electrospinning scaffolds. In vitro cell experiments showed that cells grow directionally on microchannel structures of inner layer and randomly on electrospinning film without microchannel structure. The results showed that the microchannels were more conducive to cell growth and proliferation at a diameter of 0.25 mm. The composite electrospinning films with silk fibroin and heparin facilitated cell adhesion and proliferation compared to the pure PCL films. All these results indicate that the vascular scaffolds have potential applications in clinic for vascular tissue regeneration.

Abstract:Currently, large, thick, and complex tissue vascularization is one of the research focuses of tissue engineering. Numerous studies have proven that microvascular systems can be developed by cultivating endothelial cells in a hydrogel/scaffold structure. As the sources of adult endothelial cells are very limited and very easily degraded, it is better to induce stem cells into endothelial cells. In this article, a grid poly(d,l-lactic- co-glycolic acid) structure with defined internal channels was fabricated using a low-temperature deposition manufacturing technique under computer direction. In a fibrinogen mixture, an aqueous adipose-derived stem cell fibrinogen mixture was incorporated into the internal walls of the poly(d,l-lactic- co-glycolic acid) scaffold and stabilized with thrombin solution. After several days of in vitro culture, the adipose-derived stem cells immobilized in the fibrin hydrogel were induced into endothelial-like cells with endothelial growth factor and basic fibroblast growth factor. Morphological and biological properties of the composite cell/fibrin–poly(d,l-lactic- co-glycolic acid) construct were characterized.

Abstract:Nature has created many perfect helical microstructures, including DNA, collagen fibrils, and helical blood vessels, to achieve unique physiological functions. While previous studies have developed a number of microfabrication strategies, the preparation of complex helical structures and cell-laden helical structures for biomimetic applications remains challenging. In this study, a one-step microfluidics-based methodology is presented for preparing complex helical hydrogel microfibers and cell-laden helical hydrogel microfibers. Several types of complex helical structures, including multilayer helical microfibers and superhelical hollow microfibers with helical channels, are prepared by simply tuning the flow rates or modifying the geometry of microfluidic device. With the decent perfusability, the hollow microfibers may simulate the structural characteristics of helical blood vessels and create swirling blood flow in a blood-vessel-on-chip setup. Such hydrogel-based helical microstructures may potentially be used in areas such as blood vessel tissue engineering, organ-on-chips, drug screening, and biological actuators.

Abstract:Modelling organ-blood barriers through the inclusion of microvessel networks within tissue models could lead to more physiologically accurate results, especially since organ-blood barriers are crucial to the normal function, drug transport, and disease states of vascularized organs. Microvessel networks are difficult to form, since they push the practical limit of most fabrication methods, and it is difficult to coax vascular cells to self-assemble into structures larger than capillaries. Here we present a method for rapidly forming networks of microvessel-like structures using sacrificial, alginate structures. Specifically, we encapsulated endothelial cells within short alginate threads, then embedded them in collagen gel. Following enzymatic degradation of the alginate, the collagen gel contained a network of hollow channels seeded with cells, all surrounding a perfusable central channel. This method uses a 3D printed coaxial extruder and syringe pumps to generate short threads in a way that is repeatable and easily transferrable to other labs. The cell-laden, sacrificial alginate threads can be frozen after fabrication and thawed before embedding without significant loss of cell viability. The ability to freeze the threads enables future scale up and ease of use. Within millifluidic devices that restrict access to media, the threads enhance cell survival under static conditions. These results indicate the potential for use of this method in a range of tissue engineering applications.

Abstract:Cell cultures for tissue engineering are traditionally prepared on two-dimensional or three-dimensional scaffolds with simple pores; however, this limits mass transportation, which is necessary for cell viability and function. In this paper, an innovative method is proposed for fabricating porous scaffolds with designed complex micro-architectures. Channels devised by computer-aided design were used to simulate features of blood vessels in native rat liver. Rapid prototyping and microreplication were used to produce a negative polydimethylsiloxane mold, and then a planar porous scaffold with predefined microchannel parameters was obtained by freeze-drying a silk fibroin/gelatin solution of an optimized concentration. After seeding with rat primary hepatocytes, the planar scaffold was rolled up to build spatial channels. By reconstructing the three-dimensional channel model in the scaffold in the form of micro-computed topography data and observing the cross-sections of the scroll, we confirmed that the bent channels were still interconnected, with restricted deviations. A comparison of the primary hepatocyte culture in the scaffolds with and without the devised channels proved that our design influenced cell organization and improved cell survival and proliferation. This method can be used for the construction of complex tissues for implantation and for culturing cells in vitro for biological tests and observations.

Abstract:Layer-by-layer (LbL) assembly is widely applied as a coating technique for the nanoscale control of architecture and related properties. However, its translational applications are limited by the time-consuming and laborious nature of the process. Inspired by the blood-clotting process, herein, we develop a shear-flow-driven LbL (SF-LbL) self-assembly approach that accelerates the adsorption rate of macromolecules by mechanically configuring the polymer chain via a coil-stretch transition, which effectively simplifies and speeds the diffusion-controlled assembly process. The structural characteristics and surface homogeneity of the SF-LbL films are improved, and diverse three-dimensional structures can be achieved. Functional SF-LbL-assembled surfaces for corneal modification are successfully fabricated, and the surface of wounded rat corneas and skin can be directly decorated in situ with SF-LbL nanofilms due to the advantages of this approach. Furthermore, in situ SF-LbL self-assembly has promise as a simple approach for the wound dressing for interventional therapeutics in the clinic, as illustrated by the successful in situ fabrication of drug-free layers consisting of chitosan and heparin on the dorsal skin of diabetic mice to rescue defective wound healing. This bioinspired self-assembly approach is expected to provide a robust and versatile platform with which to explore the surface engineering of nanofilms in science, engineering, and medicine.
Rapid Assembling Organ Prototypes with Controllable Cell-Laden Multi-Scale Sheets

Abstract:A native organ has heterogeneous structures, strength, and cell components. It is a big challenge to fabricate organ prototypes with controllable shapes, strength, and cells. Herein, a hybrid method is developed to fabricate organ prototypes with controlled cell deposition by integrating extrusion-based 3D printing, electrospinning, and 3D bioprinting. Multi-scale sheets were first fabricated by 3D printing and electrospinning; then, all the sheets were assembled into organ prototypes by sol–gel reaction during bioprinting. With this method, macroscale structures fabricated by 3D printing ensure the customized structures and provide mechanical support, nanoscale structures fabricated by electrospinning offer a favorable environment for cell growth, and different types of cells with controllable densities are deposited in accurate locations by bioprinting. The results show that L929 mouse fibroblasts encapsulated in the structures exhibited over 90% survival within 10 days and maintained a high proliferation rate. Furthermore, the cells grew in spherical shapes first and then migrated to the nanoscale fibers showing stretched morphology. Additionally, a branched vascular structure was successfully fabricated using the presented method. Compared with other methods, this strategy offers an easy way to simultaneously realize the shape control, nanofibrous structures, and cell accurate deposition, which will have potential applications in tissue engineering.

Abstract:Engineering three-dimensional (3D) scaffolds with in vivo like architecture and function has shown great potential for tissue regeneration. Here we developed a facile microfluidic-based strategy for the continuous fabrication of cell-laden microfibers with hierarchically organized architecture. We show that photolithographically fabricated microfluidic devices offer a simple and reliable way to create anatomically inspired complex structures. Furthermore, the use of photo-cross-linkable methacrylated alginate allows modulation of both the mechanical properties and biological activity of the hydrogels for targeted applications. Via this approach, multilayered hollow microfibers were continuously fabricated, which can be easily assembled in situ, using 3D printing, into a larger, tissue-like construct. Importantly, this biomimetic approach promoted the development of phenotypical functions of the target tissue. As a model to engineer a complex tissue construct, osteon-like fiber was biomimetically engineered, and enhanced vasculogenic and osteogenic expression were observed in the encapsulated human umbilical cord vein endothelial cells and osteoblast-like MG63 cells respectively within the osteon fibers. The capability of this approach to create functional building blocks will be advantageous for bottom-up regeneration of complex, large tissue defects and, more broadly, will benefit a variety of applications in tissue engineering and biomedical research.
3D Bioprinting of Vessel-like Structures with Multilevel Fluidic Channels

Abstract:In this study, 3D hydrogel-based vascular structures with multilevel fluidic channels (macro-channel for mechanical stimulation and microchannel for nutrient delivery and chemical stimulation) were fabricated by extrusion-based three-dimensional (3D) bioprinting, which could be integrated into organ-on-chip devices that would better simulate the microenvironment of blood vessels. In this approach, partially cross-linked hollow alginate filaments loading fibroblasts and smooth muscle cells were extruded through a coaxial nozzle and then printed along a rotated rod template, and endothelial cells were seeded into the inner wall. Because of the fusion of adjacent hollow filaments, two-level fluidic channels, including a macro-channel in the middle formed from the cylindrical template and a microchannel around the wall resulted from the hollow filaments were formed. By this method, different shapes of vessellike structures of millimeter diameter were printed. The structures printed using 4% alginate exhibited ultimate strength of 0.184 MPa, and L929 mouse fibroblasts encapsulated in the structures showed over 90% survival within 1 week. As a proof of concept, an envisioned load system of both mechanical and chemical stimulation was demonstrated. In addition, a vascular circulation flow system, a cerebral artery surgery simulator, and a cell coculture model were fabricated to demonstrate potential tissue engineering applications of these printed structures.

Abstract:The spatial cellular alignment and multi-layer structure are vitally important for the physiological functions of natural blood vessels. However, the two features are difficult to be constructed in one scaffold simultaneously, especially in the small-diameter vascular scaffold. Here we report a general strategy to construct a gelatin-based biomimetic three-layer vascular scaffold with spatial alignment features mimicking the natural structure of blood vessels. By using a sequential electrospinning strategy combined with folding and rolling manipulation, a three-layer vascular scaffold with inner and middle layers spatially perpendicular to each other was obtained. The special features of this scaffold could fully mimic the natural multi-layer structures of blood vessels and also possess great potential for spatial arrangement guidance of corresponding cells in blood vessels.

Abstract:Bioprinting of vascular tissues holds great potential in tissue engineering and regenerative medicine. However, challenges remain in fabricating biocompatible and versatile scaffolds for the rapid engineering of vascular tissues and vascularized organs. Here, we report novel bioink-enabled microfluidic printing of tunable hollow microfibers for the rapid formation of blood vessels. By compositing biomaterials including sodium alginate, gelatin methacrylate (GelMA), and glycidyl-methacrylate silk fibroin (SilkMA), we prepared a novel composite bioink with excellent printability and biocompatibility. This composite bioink can be printed into hollow microfibers with tunable dimensions using a microfluidic co-axial printing. After seeding human umbilical vein endothelial cells (HUVEC) into the hollow chambers via a microfluidic prefusion device, these cells can adhere to, grow, proliferate, and then cover the internal surface of the printed hollow scaffolds to form vessel-like tissue structures within three days. By combining the unique composite bioink, microfluidic printing of vascular scaffolds, and microfluidic cell seeding and culturing, our strategy can fabricate vascular-like tissue structures with high viability and tunable dimension within three days. The presented method may engineer in vitro vasculatures for the broad applications in basic research and translational medicine including in vitro disease models, tissue microcirculation, and tissue transplantation.

Abstract:In this work, a novel strategy was developed to fabricate prevascularized cell-layer blood vessels in thick tissues and small-diameter blood vessel substitutes using three-dimensional (3D) bioprinting technology. These thick vascularized tissues were comprised of cells, a decellularized extracellular matrix (dECM), and a vasculature of multilevel sizes and multibranch architectures. Pluronic F127 (PF 127) was used as a sacrificial material for the formation of the vasculature through a multi-nozzle 3D bioprinting system. After printing, Pluronic F127 was removed to obtain multilevel hollow channels for the attachment of human umbilical vein endothelial cells (HUVECs). To reconstruct functional small-diameter blood vessel substitutes, a supporting scaffold (SE1700) with a double-layer circular structure was first bioprinted. Human aortic vascular smooth muscle cells (HA-VSMCs), HUVECs, and human dermal fibroblasts–neonatal (HDF-n) were separately used to form the media, intima, and adventitia through perfusion into the corresponding location of the supporting scaffold. In particular, the dECM was used as the matrix of the small-diameter blood vessel substitutes. After culture in vitro for 48 h, fluorescent images revealed that cells maintained their viability and that the samples maintained structural integrity. In addition, we analyzed the mechanical properties of the printed scaffold and found that its elastic modulus approximated that of the natural aorta. These findings demonstrate the feasibility of fabricating different kinds of vessels to imitate the structure and function of the human vascular system using 3D bioprinting technology.

Rapid Fabrication of Cell-Laden Microfibers for Construction of Aligned Biomimetic Tissue

Abstract:Bottom-up engineering of tissue constructs is being rapidly developed and broadly applied in biomanufacturing. As one type of building block, cell-laden microfibers are promising for reconstruction of oriented structures and functions of linear tissues, such as skeletal muscles, myocardia, and spinal cord tissues. Herein, we propose wet-spinning method with agitating collection, wherein alginate-based material is extruded into an agitated CaCl2 bath with a magnetic rotor acting as the microfiber collector. By applying this method, we achieve rapid fabrication and oriented collection of hydrogel microfibers with diameters ranging from 100 to 400 μm. In addition, we encapsulate myoblasts in the hydrogel to form cell-laden microfibers, which show a high viability (more than 94%) during in vitro culture. Moreover, the method allows to fabricate of cell-laden core-sheath microfibers and hollow microfibers. We also fabricate 3D constructs using various methods of microfiber assembly like weaving and braiding. The assembling results suggest that the proposed method is a promising technology for bottom-up engineering of aligned biomimetic tissue constructs.

Abstract:Aligned electrospun fibers used for the fabrication of tubular scaffolds possess the ability to regulate cellular alignment and relevant functional expression, with applications in tissue engineering. Despite significant progress in the fabrication of small-diameter vascular grafts (SDVGs) over the past decade, several challenges remain; one of the most problematic of these is the fabrication of aligned nanofibers for multilayer SDVGs. Furthermore, delamination between each layer is difficult to avoid during the fabrication of multilayer structures. This study introduces a new fabrication method for minute delamination four-layer tubular scaffolds (FLTSs) that consist of an interior layer with highly longitudinal aligned nanofibers, two middle layers composed of electrospun sloped and circumferentially aligned fibers, and an exterior layer comprising random fibers. These FLTSs are used to simulate the structures and functions of native blood vessels. Here, thermoplastic polyurethane (TPU)/polycaprolactone (PCL)/polyethylene glycol (PEG) were electrospun to fabricate FLTSs or tubular scaffolds with completely random fibers layer (RLTSs). The surface wettability of the TPU/PCL/PEG tubular scaffold was tested by water contact angle analysis. In particular, compared with RLTSs, FLTSs showed excellent mechanical properties, with higher circumferential and longitudinal tensile properties. Furthermore, the high viability of the human umbilical vein endothelial cells (HUVECs) on the FLTSs indicated the biocompatibility of the tubular scaffolds comparing to RLTSs. The aligned and random composite structure of the FLTSs are conducive to promoting the growth of HUVECs, and the cell adhesion and proliferation on these scaffolds was found to be superior to that on RLTSs. These results demonstrate that the fabricated FLTSs have the potential for application in vascular tissue regeneration and clinical arterial replacements.

Abstract:It is a huge challenge to engineer vascular networks in vital organ tissue engineering. Although the incorporation of artificial microfluidic network into thick tissue-engineered constructs has shown great promise, most of the existing microfluidic strategies are limited to generate rectangle cross-sectional channels rather than circular vessels in soft hydrogels. Here we present a facile approach to fabricate branched microfluidic network with circular cross-sections in gelatin hydrogels by combining micromolding and enzymatically-crosslinking mechanism. Partially crosslinked hydrogel slides with predefined semi-circular channels were molded, assembled and in situ fully crosslinked to form a seamless and circular microfluidic network. The bonding strength of the resultant gelatin hydrogels was investigated. The morphology and the dimension of the resultant circular channels were characterized using scanning electron microscopy (SEM) and micro-computerized tomography (μCT). Computational fluid dynamic simulation shows that the fabrication error had little effect on the distribution of flow field but affected the maximum velocity in comparison with designed models. The microfluidic gelatin hydrogel facilitates the attachment and spreading of human umbilical endothelial cells (HUVECs) to form a uniform endothelialized layer around the circular channel surface, which successfully exhibited barrier functions. The presented method might provide a simple way to fabricate circular microfluidic networks in biologically-relevant hydrogels to advance various applications of in vitro tissue models, organ-on-a-chip systems and tissue engineering.
