Abstract:Gold nanorods (GNRs) with suitable aspect ratio were synthesized with a template technique and then dispersed in a saturated sodium citrate solution by ultrasonication to form a GNR suspension. A GNR-modified electrode was fabricated using the GNR suspension. The oxidation of dopamine at the GNR/GC electrode exhibited surprisingly high electrocatalytic activity and adsorption-controlled characteristics. Square-wave voltammetry was used to detect dopamine. At the GNR/GC electrode, the linear concentration range of DA is from 1×10−8M to 1×10−7M and the detection limit (s/n=3) is as low as 5.5×10−9M. The current sensitivity is 3.280μA/μM, and 1000-fold ascorbic acid (AA) cannot interfere with the determination of DA. All these performances are greatly superior to those of the bare GC electrode.

Abstract:Chemical neurotransmission occurs at chemical synapses and endocrine glands, but up to now there was no means for direct monitoring of neurotransmitter exocytosis fluxes and their precise kinetics from inside an individual synapse. The fabrication of a novel finite conical nanoelectrode is reported perfectly suited in size and electrochemical properties for probing amperometrically inside what appears to be single synapses and monitoring individual vesicular exocytotic events in real time. This allowed obtaining direct and important physiological evidences which may yield important and new insights into the nature of synaptic communications.
Sensitive detection of dopamine using a platinum microelectrode modified by reduced graphene oxide and gold nanoparticles

Abstract:Platinum wire microelectrodes generally exhibited properties of low sensitivity but are easily contaminated where it comes to dopamine detection. In this paper, a platinum wire modified with gold nanoparticles (AuNPs) and reduced graphene oxide (rGO) was used to electrochemically detect dopamine in vivo. Electrochemical performances of microelectrodes (before and after surface modification) were investigated by impedance spectroscopy and cyclic voltammetry. The results showed that AuNPs/rGO composites significantly increase the electrode surface area and improve the electrocatalytic property of platinum wires. The modified electrodes exhibited excellent sensitivity for dopamine detection (LOD=16.57nM), as well as ascendant selectivity (simultaneous detection of ascorbic acid, dopamine, and uric acid), reproducibility (R.S.D.=3.98%), and stability (≫100 repetitive potential sweeps, 1-week lifespan). In vivo experiments showed that the proposed AuNPs/rGO electrode was also capable of detecting dopamine in complex physiological environments, particularly in the striatum of anesthetized rats. Hence, the proposed electrodes provided a potential application in in vivo multiplexing neurotransmitter detection in further research.

Abstract:Carbon fiber nanoelectrodes (tip diameter = ca. 100 nm) have been first used to monitor real-time dopamine release from single living vesicles of single rat pheochromocytoma (PC12) cells. The experiments show that active and inactive release sites exist on the surface of cells, and the spatial distributions have been differentiated even in the same active release zone. It is first demonstrated that multiple vesicles can sequentially release dopamine at the same site of the cell surface, which possibly plays the main role in the dopamine release from PC12 cells.

Abstract:To develop in vivo monitoring strategies of neurotransmitters involved in brain chemistry is a challenging work for progress in understanding the roles that biomolecules play in pathology and physiology. Here we report a new type of gold nanoparticle-sheathed glass capillary nanoelectrode (Au/GCNE) for sensing cerebral dopamine. First, a size-controlled needle-type quartz capillary was pulled with a laser puller. Then, the capillary tip exterior was chemically functionalized with colloidal gold nanoparticles by the seed-mediated growth protocol. Through insulating the above tip with cathodic electrophoretic paint followed by heating to tune the exposed area of gold-nanoparticle-film, the Au/GCNE with tip apex radius ranging from ~8.9 to ~500 nm can be prepared. Scanning electron microscopy (SEM) and steady-state voltammetry were utilized to characterize the effective radius of nanoelectrodes. The results showed that the tip apex radius of Au/GCNE was mainly affected by the pre-pulled capillary tip, the modified AuNPs and the cathodic electrophoretic paint. By taking advantage of the modified AuNPs and the enhanced electrochemical performance of the nanoelectrode, a wide dynamic linear range from 2.0×10(-8) M to 5.6×10(-6) M with a low detection limit of 1.0×10(-8) M (S/N=3), as well as good selectivity for dopamine, were first achieved with the Nafion-modified Au/GCNE. In addition, the designed glass substrates of Au/GCNE were mechanically stronger and their sharp tips aided in membrane penetration during implantation in the in vivo experiment. As a result, the Nafion-modified Au/GCNE was successfully applied for amperometrically monitoring dopamine in the striatum of anesthetic rats.

Abstract:In vivo monitoring neurochemicals with microelectrode is invasive and the damage to brain tissue may inevitably cause disturbance signals physiologically to the measurement.It is of great importance to reduce the electrode size and to decrease the damage.This study demonstrates a novel nanoelectrode preparation methodology for in vivo monitoring dopamine (DA) fluctuation in the living brain of rats with high dependability.The fabrication process of the gold nanoelectrode involving a few minutes consists of only two steps:1) growing gold nanoseeds on surface of tip of glassy capillary by ion sputtering;2) wet depositing a continuous conductive gold film composed of gold nanoparticles by dipping the capillary with gold nanoseeds into a freshly mixed chloroauric acid and hydroxylamine hydrochloride for one minute.The tip size of the well-prepared gold nanoelectrode was 300 ～400 nanometers.The gold nanoelectrode was able to detect DA and showed a good linearity with the concentration of DA ranging from 1.0 to 56.0 μmol· L-1 with a limit of detection as low as 0.14 μmol· L-1 (S/N=3).Benefiting from the excellent electrochemical performance,the gold nanoelectrode was successfully employed for catecholamine release in striatum of living rat brain.

Abstract:A single-walled carbon nanotube (SWNT) is covalently attached to the interior surface of a conical glass micropore electrode (GME) to create a novel amperometric dopamine sensor (SWNT/NH-GME). The SWNT/NH-GME combines the advantages of excellent transport properties of the cone-shaped micropore with the characteristics of a SWNT, exhibiting a dramatic electrocatalytic effect on the oxidation of dopamine (DA). Cyclic voltammetry and amperometric methods were employed to study the electrochemical behavior of the SWNT/NH-GME. The results showed that the SWNT/NH-GME sensor exhibited an excellent immunity from ascorbic acid interference and was able to measure DA concentrations with a detection limit of 4.2×10−7mol/L (S/N=3).
Spatial and Temporal Amperometric Monitoring Dopamine Release from Single PC12 Cells with Carbon Fiber Nanoelectrode

Abstract:Carbon fiber nanoelectrodes (200-300 nm) were firstly used to amperometricaly monitor the dopamine release from single PC12 cells with temporal resolution and especially more higher spatial resolution than those obtained by using microelectrodes. When the nanoelectrode was in a distance 1 mum above the PC12 cell, only one peak signal corresponding to a single vesicle exocytotic event was detected caused from stimulation with 1 mmol/L nicotine. The spatial difference of exocytosis was also detected by placing the electrode onto the different locations of the cell body, the results have demonstrated that the spatial distribution of dopamine in cells is not uniform and the time for stimulating secretion is very different. Nanoelectrodes electrochemical method can provide a powerful tool for observing the temporal and spatial characteristics of the secretion from single cells directly.

Abstract:A Au disk nanoelectrode down to 3 nm in radius was developed by a facile and reliable method and successfully applied for monitoring dopamine release from single living vesicles. A fine etched Au wire was coated with cathodic electrophoretic paint followed by polyimide, which retracted from the tip end during curing to expose the Au nanotip. By cyclic voltammetric scanning the above tip in 0.5 M KCl, the transformation of a core-shaped apex into a geometrically well-defined Au disk nanoelectrode with different dimensions can be controllably and reproducibly achieved. Scanning electron microscopy, transmission electron microscopy, and steady-state voltammetry were used to determine the size of nanoelectrodes. The results showed that the specific etching and insulation method not only avoids the use of toxic etching solution and the uncontrollable treatment to expose the tip but also makes possible the controllable and reproducible fabrication of Au disk nanoelectrode down to 3 nm in radius. The nanoelectrodes with well-demonstrated analytical performance were further applied for amperometrically monitoring dopamine release from single rat pheochromacytoma cells with high spatial resolution.

Abstract:We report a novel ultrasonic-aided fast and straightforward approach to fabricate Au microelectrodes by electroless deposition of nanostructured gold films on the rigid outer surface of pulled glass capillaries. Microelectrodes with tip diameters ranging from several hundred nanometers to several micrometers were fabricated within 20 min via three sequential ultrasonication steps. The ultrasonication technique has been validated to be a very effective route in engineering the morphology of Au film surfaces and improves the fabrication efficiency of Au microelectrodes. The nanostructured surfaces of the Au micro electrodes demonstrate excellent sensing activity and antifouling for dopamine oxidation. The microelectrodes were applied for measurement of catecholamines released from exocytosis events from single chromaffin cells and exhibited faster dynamic peak parameters, compared with carbon fiber microelectrodes. This report provides a generally accessible and complementary platform for analyzing catecholamines release events, which should be useful for new electrode designs and neurochemical sensing.

Abstract:Gold nanotadpoles were synthesized by a simple temperature-reducing seeding approach. A novel gold electrode modified with gold nanotadpoles through a sulphydryl-terminated self-assembled monolayer was then fabricated and exhibited strong promoting effect toward the electrochemical oxidation of dopamine (DA). The anodic peak currents obtained from the cyclic voltammetry (CV) were linearly dependent on DA concentration over the range of 3 x 10(-5)-1.8 x 10(-4) M with correlation coefficient of 0.999. The detection limit obtained was 0.1 microM. It was also successfully used for the selective detection of DA in the presence of ascorbic acid (AA). This modified electrode showed good selectivity and recovery.

Abstract:Neurotransmitters, the material bases for nerve signaling, play an important role in the physiological activities in the body. Accurate analysis of neurotransmitter concentration is of great significance for disease prevention and control. They are secreted in low concentrations; therefore highly sensitive detection methods are needed. In this paper, 3 spherical gold nanoparticles were prepared with diameters of 5 nm, 20 nm, and 50 nm. The gold nanoparticles were deposited on the surface of carbon fiber microelectrodes by a one-step potentiostatic process to obtain nanogold-modified carbon fiber microelectrodes (AuNPs/CFME). These electrodes were electrochemically characterized before and after modification. The electrochemical properties of dopamine, norepinephrine, ascorbic acid, and potassium ferricyanide were also studied on the microelectrodes before and after modification. The results demonstrated that AuNPs/CFME had an electrocatalytic influence upon catecholamines with positive charges with shielding effects against negatively charged substances. The smaller spherical gold nanoparticles had better catalytic shielding properties. Under the optimum conditions, the differential pulse voltammetry (DPV) peak current and the concentration of dopamine with high concentration of ascorbic acid displayed a linear range of 0.1 mu M to 10 mu M with a detection limit of 0.013 mu M based on a signal-to-noise ratio of 3. This work provides a simple and easy approach to selectively determine positively charged substances and shielding negatively charged compounds. The prepared gold nanoparticles were employed for the selective determination of dopamine in the presence of high concentrations of ascorbic acid in the mouse serum samples with excellent analytical performance. This work shows high selectivity for dopamine, which can be applied to its selective determination in the presence of high concentrations of ascorbic acid in biological systems.

Abstract:In this work, a highly sensitive and selective biomimetic electrochemical sensor for the amperometric detection of trace dopamine (DA) in human serums was achieved by gold nanoparticles (AuNPs) doped molecularly imprinted polymers (MIPs). Functionalized AuNPs (F-AuNPs), a novel functional monomer bearing aniline moieties on the surface of the AuNPs, were prepared via a direct synthesis method and then used to fabricate the conductive MIPs film on the modified electrode by electropolymerization method in the presence of DA and p-aminobenzenethiol (p-ATP). The obtained electrochemical sensor based on the conductive film of AuNPs doped MIPs (AuNPs@MIPs) could effectively minimize the interferences caused by ascorbic acid (AA) and uric acid (UA). The linear range for amperometric detection of DA was from 0.02 mu mol L-1 to 0.54 mu mol L-1 with the detection limit of 7.8 nmol L-1 (S/N=3). Abstract:We demonstrated potential features of gold nanoparticle bipyramid (AuNB) for an electrochemical biosensor. The facile synthesis method and controllable shape and size of the AuNB are achieved through the optimization of cetyltrimethylammonium chloride (CTAC) surfactant over citric acid ratio determining the control of typically spherical Au seed size and its transition into a penta-twinned crystal structure. We observe that the optimized ratio of CTAC and citric acid (CA) facilitates flocculation control in which Au seeds with size as tiny as ~14.8 nm could be attained and finally transformed into AuNB structures with an average length of ~55 nm with high reproducibility. To improve the electrochemical sensing performance of a screen-printed carbon electrode (SPCE), surface modification with AuNB via distinctive linking procedures effectively enhanced the electroactive surface area by 40%. Carried out for the detection of dopamine (DA), a neurotransmitter frequently linked to the risk of Parkinson's, Alzheimer's, and Huntington's diseases, the AuNB decorated-carbon electrode shows outstanding electrocatalytic activity that improves sensing performance, including high sensitivity, low detection limit, wide dynamic range, high selectivity against different analytes, such as ascorbic acid (AA), uric acid (UA) and urea, and excellent reproducibility.

Abstract:In vivo monitoring of dopamine (DA) in cerebral nervous system with no traumatic brain injury is still a challenging work for reliably evaluating the role that DA plays in physical and cognitive functionality. In this work, to reduce the traumatic injury on the brain tissue, we develop a glass-sealed Au nanoelectrode (GSNE) with nanoscale physical dimention for cerebral DA detection. First, an Au microwire was sealed into the pre-thinned silica capillary. Then, the obtained Au/capillary assembly was pulled into two tips followed by polishing away the excess glass to expose the Au nanotip. The morphology of GSNE was characterized by scanning electron microscopy and transmission electron microscopy. The results showed that this nanoelectrode can be quickly fabricated with high reproducibility. By further electrodepositing cluster-like gold nanostructure on GSNE and modifying Nafion, amperometric detection of DA with high sensitivity and good selectivity were achieved on the Nafion-modified Au/GSNE. Moreover, the Nafion-modified Au/GSNE possessed advantages of easy tissue insertion and minimum tissue disturbance. Thus, the developed electrochemical nanosensor was successfully used for amperometrically monitoring DA level in the striatum of anesthetic rats.
Dopamine Measurement Using Engineered CNT–CQD–Polymer Coatings on Pt Microelectrodes

Abstract:This study aims to develop a microelectrode array-based neural probe that can record dopamine activity with high stability and sensitivity. To mimic the high stability of the gold standard method (carbon fiber electrodes), the microfabricated platinum microelectrode is coated with carbon-based nanomaterials. Carboxyl-functionalized multi-walled carbon nanotubes (COOH-MWCNTs) and carbon quantum dots (CQDs) were selected for this purpose, while a conductive polymer like poly (3-4-ethylene dioxythiophene) (PEDOT) or polypyrrole (PPy) serves as a stable interface between the platinum of the electrode and the carbon-based nanomaterials through a co-electrodeposition process. Based on our comparison between different conducting polymers and the addition of CQD, the CNT–CQD–PPy modified microelectrode outperforms its counterparts: CNT–CQD–PEDOT, CNT–PPy, CNT–PEDOT, and bare Pt microelectrode. The CNT–CQD–PPy modified microelectrode has a higher conductivity, stability, and sensitivity while achieving a remarkable limit of detection (LOD) of 35.20 ± 0.77 nM. Using fast-scan cyclic voltammetry (FSCV), these modified electrodes successfully measured dopamine's redox peaks while exhibiting consistent and reliable responses over extensive use. This electrode modification not only paves the way for real-time, precise dopamine sensing using microfabricated electrodes but also offers a novel electrochemical sensor for in vivo studies of neural network dynamics and neurological disorders.

Abstract:Dopamine (DA), a catecholamine neurotransmitter , is crucial in brain signal transmission . Monitoring cytoplasmic DA levels can reflect changes in metabolic factors and provide valuable information for researching the mechanisms involved in neurodegenerative diseases. However, the in-situ detection of intracellular DA is constrained by its low contents in small-sized single cells. In this work, we report that noble metal (Au, Pt)-modified carbon fiber micro-nanoelectrodes are capable of real-time detection of DA in single cells with excellent sensitivity, selectivity, and anti-contamination capabilities. Notably, noble metals can be modified on the electrode surface through electrochemical deposition to enhance the conductivity of the electrode and the oxidation current of DA by 50 %. The nanosensors can work stably and continuously in rat adrenal pheochromocytoma cells (PC12) to monitor changes in DA levels upon K + stimulation. The functionalized carbon fibers based nanosensors will provide excellent prospects for DA analysis in the brains of living animals.

Abstract:In this research, a manganese dioxide (MnO2) nanowires/chitosan-modified gold (Au) electrode was successfully fabricated for amperometric determination of dopamine (DA) at a very low working potential (0.00V vs. SCE). The electrochemical properties of the modified Au electrode were characterized by cyclic voltammetry and electrochemical impedance spectroscopy. The detection scheme was proposed to involve two major reactions: the chemical oxidation of DA to o-dopaminoquinone by MnO2 nanowires, and then the electro-reduction of o-dopaminoquinone to DA at a low working potential (0.00V vs. SCE), resulting in a distinct reduction response current. The modified Au electrode displays good amperometric response to DA in a wide concentration range 0.10μM to 1.10mM. The fabrication of MnO2 nanowires/chitosan-modified Au electrode is simple and highly reproducible (RSD=3.8%, n=5). It exhibits excellent analytical performance in terms of low working potential (0.00V vs. SCE), long shelf-life (90% of its initial response after 1-month storage) and high repeatability (RSD=2.2%, n=5). Finally, it was applied to determine DA in real samples directly with high accuracy (99%) and good recovery (99.7–103%), confirming its promising application in routine DA analysis.

Abstract:One big challenge of fabricating nanosensors for spatially resolved electrochemical detection of neurochemicals, such as dopamine (DA), is the difficulty to assembly nanometer-scale patternable and integrated sensors. In this work we develop a novel approach to precisely manufacture nano-Au-electrode (NAE) using lithographic fabrication technique, and characterize the NAE for DA detection. A negative photoresist, SU-8, is used as a substrate and protection layer for the 127-nm Au active sensing layer. The cross surface morphology and thickness of the Au layer are imaged by scanning electron microscopy and an interference microscopy. This NAE could be precisely controlled, repeatedly fabricated and conveniently renewed for several times. The electrochemical sensitivity and selectivity of the NAE towards DA detection are significantly higher than those of a standard Au thin-film electrode. This work demonstrates that the NAE could be used as an attractive means for electrochemically sensing and recording DA. (c) 2012 Elsevier B.V. All rights reserved.
