MRI study of a nude mouse hepatic metastasis model with human colon carcinoma

Abstract:Objective To produce a nude mouse hepatic metastasis model with human colon carcinoma, and to investigate methods and manifestations of routine MR imaging(such as T1WI and T2WI) and diffusion weighted imaging (DWI). Methods SW480, a human colon cancer cell line, was implanted subcutaneously to the costal regions of 2 nude mice by injection (1 × 107/mL, 0.5 mL). The mice beared cancer were anesthetized,the tumors were resected and divided into small pieces of 1 mm3. Thirty-six athymic mice (half female, half male) were anesthetized, and their abdomen were entered through an upper midline incision. The tumor pieces were injected into their livers. These mice were scanned to acquire TSE-T1WI, T2WI and EPI-DWI. Results The successful rate of inoculation was 100% (36/36), and the maximum diameter of tumors in liver of mice could be 0.7~2 cm after 2~6 weeks. All tumors were detected clearly on T1WI, T2WI images, and shown as iso-intensity on T1WI and hyper-intensity on T2WI. Comparing with T2WI, images of DWI had conspicuous geometry deformation (4 mice), moderate to slight geometry deformation (14 mice) which did not affect the detection of tumors in mice livers, and no geometry deformation (18 mice). Conclusions The nude mouse hepatic metastasis model with SW480 colon cancer cell line, with ideal implantation and tumor growth rate, are convenient for MRI observation. MR images can well reflect the pathologic changes. The nude mouse model is suitable for molecular imaging research of human colon cancer.
