Induction of immune tolerance by injection of guinea pig vascular endothelial cells intraperitoneally in xenotransplantation heart (guinea pig-rat) model

Abstract:Objective By injecting guinea pig specific vascular endothelial cells into the peritoneal cavity of rats, the feasibility of immune tolerance induction in guinea pig to rat heart transplantation model was investigated.Methods Guinea pig endothelial cells were injected intraperitoneally 14 days before heart transplantation and survival time of donor hearts was observed. All specimens of heart grafts were inspected by HE staining and immunohistological staining for IgM and C 3. The experimental rats were divided into the following groups: Group A served as experimental control group, Group B received 2×10 6 guinea pig specific endothelial cells. Group C was treated with cyclophosphamide (CY) 40 mg/kg. Group D was treated with CY 40 mg/kg in combination with 2.5×10 6 guinea pig specific endothelial cells.Results Intraperitoneal injection of guinea pig specific endothelial cells could prolong the survival time of transplanted heart significantly. In the group B, mean graft survival time was (35.12±5.24) ?min,but (14.75±7.22) min in the group A (P0.001) and (42.34±5.43) min in the group C (P0.05). Group D [(50.33± 6.21) min] showed a longer survival time as compared with group C (P0.05). The typical features of hyperacute rejection were observed by pathological studies in the specimens of all groups.Conclusions Intraperitoneal injection of guinea pig specific vascular endothelial cells has the effect of prolonging survival time of donor hearts and inducing immune tolerance.Combined use of guinea pig specific vascular endothelial cells with CY can strengthen induction of immune tolerance.
