The Role of Soluble Fas, Soluble Fas Ligand and Cytokines in the Pathogenesis of Type 1 Diabetes Mellitus

Abstract:Objective To investigate the role of the concentration of soluble Fas (sFas), soluble Fas ligand (sFasL) and cytokines in the pathogenesis of type 1 diabetes mellitus (DM). Methods The concentrations of sFas, sFasL, IFN-γ and IL-1 in sera isolated from 32 patients with type 1 DM and 20 healthy individuals were determined by the BAS-ELISA. Results The concentrations of sFas, interferon-γ (IFN-γ) and interleukin-1 (IL-1) in sera from 32 patients with type 1 DM were (1 546±685, 1 074±451 and 1 406±721)ng/L respectively, which were significantly higher than those of healthy individuals. Although the serum level of sFasL of type 1 DM patients was (211±73)ng/L which was lower than that of the healthy control but the difference was not significant. High concentrations of both sFas and IFN-γ were found in the sera of 12 patients with type 1 DM, while low concentrations of both sFas and IFN-γ were found in the sera of 9 patients with type 1 DM. Conclusion The serum levels of sFas, IFN-γ and IL-1 of the patients with type 1 DM are significantly higher than those of healthy individuals. There is positive corralation between serum sFas and IFN-γ (r=0.79, P<0.05). Analysing the concentrations of the sFas, IFN-γ and IL-1 in the sera from patients with type 1 diabetes mellitus may be helpful for evaluating the lesion of β cells of the patients with type 1 DM and useful for diagnosis and treatment.
