Association Between Serum IL-18 Level and Cytokine in Type 1 Diabetes Mellitus in Children

Abstract:Objective To investigate the serum IL-18 level in type 1 diabetes mellitus(T1DM) in children and the positive rate of islet cell antibody(ICA) and glutamic acid decarboxylase antibody(GADA),and explore the association of autoantibody status and serum cytokine levels in children patients with T1DM.Methods The positive rates of ICA and GADA in 108 T1DM patients and 110 controls and serum levels of interleukin(IL)-18,IL-12,IL-10,IFN-gamma(IFN-γ),IL-4 in 48 newly diagnosed T1DM patients and 40 controls were detected by enzyme linked immunosorbent assay(ELISA).Results The serum IL-18 levels in newly diagnosed T1DM patients were significantly higher than in age-and sex-matched controls(P0.05),while there was no significant difference between T1DM patients and controls on the serum levels of IFN-γ,IL-4,IL-10,and IL-12.Among 108 T1DM patients,the positive rates of ICA and GADA were 50.9% and 49.1% respectively,both significantly higher than in controls with P value less than 0.05.And among TIDM patients whose courses were within or over one year,the positive rates of GADA were 54.9% and 37.8% respectively,without statistical significance(P0.05).There was positive correlation between IL-18 and autoantibody,and also the amount of the autoantibody,but not between IFN-γ,IL-4 and IL-10 and autoantibody.Conclusions The serum IL-18 level of newly diagnosed T1DM increases significantly,and positively correlated with the amount of autoantibody.So the serum IL-18 level might be a useful predictor for T1DM.
