Abstract:In this paper, the method of HPLC-RF for determine streptomycin residue in royal jelly was studiedThe result showed that the pretreatment method could availably remove the interferences from impurities in. royal jelly, and the rate of recovery was 74.7%～95.3%, and the sensitivity was 5μg/kg which is much lower than that of limit the quantity of residue. This method has many excellences such as good stability, good veracity and low cost, and it is hopeful turn into an ideal method for determine the quality of royal jelly.

Abstract:In this study,a convenient liquid-liquid extraction method for sample preparation was introduced,which can enrich and purify chloramphenicol in royal jelly to a similar level compared to sample treated by solid phase extraction(SPE).This method showed no interference on the determination of chloramphenicol,and it greatly reduced the cost of sample preparation.

Abstract:A method has been developed to simultaneously determine chloramphenicol,sulfonamides,trimethoprims,fluoquinolones,lincosmides,nitroimidazoles and macrolides residues in royall jelly by hydrophilic interaction chromatography with tandem mass spectrometry.The experimental conditions such as extracting solvent,pH value,chromatographic column,mobile phase type and mass spectrometry condition were optimized.Under the optimal conditions,the calibration curves were linear in the range of 0.3-10 μg/kg for chloramphenicol,and 2-100 μg/kg for other target compounds.The limits of detection(S/N=3) were 0.1 μg/kg for chloramphenicol and 1 μg/kg for other residues.Recoveries at three different spiked levels ranged from 70% to 119% with relative standard deviations(n=3) of 4.3%-14.9%.The method is simple,accurate and is suitable for the analysis of 27 residues in royal jelly.

Abstract:Objective:To determine the pesticide multiresi-dues in vegetables effectively,a rapid and economical capi-llary gas chromatography method has been established for the determination of kelthane and pyrethriodes in vegetables.Methods:The pesticide residues in the samples were extracted with acetonitrile and then extracts concentrated with steam of nitrogen and cleaned up by silicon dioxide solid phase extraction column.Extracts were separated by HP-5(30 m×0.25 mm×0.25 μm) capillary column and detected by electroncapture detector.Results:The recoveries obtained from vege-tables were in the range of 67.8%~118.3%,with relative standard deviation of 3.2%~13.5%.Conclusion:This method is rapid,accurate and of less cost.It is suitable for the analysis of pesticide multi-residues in vegetables.

Abstract:The purpose of this paper using liquid-phase extraction as pre-treatment, five sulfonamide residues in royal jelly were determined by pre-column derivatization HPLC with fluorescence detector. The proposed method, with less reagent and cost, was simple, rapid and accurate. The detection limit was 1μg/kg, which could meet the need of the residue limit for sulfonamide in royal jelly of exportation.

Abstract:A method of high performance liquid chromatography coupled with mass spectrometry(HPLC-MS) for simultaneous determination of 23 pesticides in traditional Chinese medicine Paeoniae radix Alba was developed.P.radix was extracted with acetonitrile,and then purified using a preliminary sample treatment step based on primary-secondary amine(PSA) dispersive solid-phase extraction.Limits of quantification(LOQ) were between 0.75-12.17 μg/kg.Recovery between 70% and 120% were obtained for all pesticides at 0.01 and 0.1 mg/kg levels,except for polar pesticides such as thiamethoxam and imidacloprid.This method is accurate,reproducible and has good cleanup effect.It could be applied in pesticide residue analysis in P.Radix.

Abstract:A method for the determination and quantification of tau-fluvalinate,triadimenol and coumaphos in royal jelly was established by HPLC-MS-MS.Tau-fluvalinate,triadimenol and coumaphos residues were extracted by acetonitrile under alkaline condition,and clean-up of the extracts was carried out using HLB solid phase extraction(SPE) column.Quantification was conducted by HPLC-MS-MS coupled with multiple reaction-monitoring(MRM) system using electron spray ionization source in an external standard mode.The detection limits for coumaphos,triadimenol and tau-fluvalinate residues were 2,1 μg/kg and 1 μg/kg,respectively.The recovery rates of tau-fluvalinate,triadimenol and coumaphos residues at three spiked levels(5,10 μg/kg and 50 μg/kg) in different samples were in the range of 78.9%–102.1% with relative standard deviation of 2.1%–13.6%.The linear range of detection was 5–100 μg/kg(r > 0.9940).This method can be successfully applied to the qualitative and quantitative analysis of royal jelly samples.

Abstract:Quinolones (QNs) is one kind of antibiotics, and in recent years its residue exceeding limits has been found frequently in animal-origin exportation food such as bee products. In this study, liquid-liquid extraction was used to pre-treat royal jelly sample, and then five QNs residues in royal jelly sample were determined by high-performance liquid chromatography coupled with fluorescence detector. The results showed that the detection limits of ofloxacin, enrofloxacin, ciprofloxacin, norfloxacin and danofloxacin are 5 μg/kg, 2.5 μg/kg, 2.5 μg/kg, 2.5 μg/kg and 1 μg/kg, respectively, which meet the detection demand of allowable limits of QNs in royal jelly. The proposed method is simple, rapid, accurate and low-reagent consumption and cost.

Abstract:The sample was diluted with pH 4.0 of Na2EDTA-Mcllvaine buffer solution,and pesticides and antibiotics in the sample were extracted with a mixture of acetic acid-acetonitrile (1 + 19),and the extract was stratified by salting-out agents.The supernatant was cleaned up with purification pipe filled with magnetic C18 and PSA.Luna C18 column was used as stationary phase,and a mixture of 0.1％ (φ) formic acid solution and acetonitrile in various ratios as mobile phase in gradient elution.ESI+ and MRM were adopted in MS.Linear relationships between values of peak area and mass concentration of 30 compounds were found in definite ranges,with detection limits (3S/N) in the range of 0.3-4.1 μg · kg 1.Test for recovery was made by standard addition method,giving results in the range of 71.1％-116％,with RSDs (n=5) ranged from 0.80％ to 21％.

Abstract:A liquid chromatographic method with mass spectrometric detection was developed for simultaneous determination of lincomycin and macrolide antibiotic residues in royal jelly.The samples were extracted by acetonitrile -ammonia(97+3)and concentrated to dryness,then dissolved in pH=8.0 phosphoric acid and was back-extracted by ethyl acetate.The use of the classical liquid-liquid extraction for lincomycin and macrolide antibiotic residues in royal jelly saves great cost of detection.The sample pre-treatment method is simple and rapid,speeding up the detection process.The detection limit is 0.2～0.9μg/kg.

Abstract:A method was developed for the determination of four insecticide residues in honey and royal jelly by gas chromatography-negative chemical ionization mass spectrometry (GC-NCI/MS). The honey and royal jelly samples were treated with different preparation methods as the result of the different components. The honey sample was extracted with ethyl acetate and cleaned up with primary second amine, and the royal jelly sample was extracted with acetonitrile-water (1:1, v/v), and cleaned up with a C18 solid-phase extraction column. Finally, the extracts of the honey and royal jelly were analyzed by GC-NCI/MS in selected ion monitoring (SIM) mode separately. External standard calibration method was used for quantification. The linearities of calibration curves of the four insecticides were good with the correlation coefficients greater than 0.99 in the range of 50-500 microg/L. The limits of the detection (LODs) of the four insecticides were in the range of 0.12- 5.0 microg/kg, and the limits of the quantification (LOQs) were in the range of 0.40-16.5 microg/kg. The recoveries of the four insecticides spiked in honey and royal jelly at three spiked levels (10, 15 and 20 microg/kg) were in the range of 78.2 -110.0%, and the relative standard deviations (RSDs) were all below 14%. The sensitivity and selectivity of this method were good with no interfering peaks. The proposed method is simple quick and effective to analyze the four insecticide residues in honey and royal jelly.

Abstract:An analytical method was developed for the simultaneous determination of 23 pesticide residues in various commercial honeys. Meanwhile, the characteristic ions and fragmentation mechanism of three pesticides in the process of electron ionization mass spectrometry (EI/MS) were evaluated. After the optimization of different parameters such as the extraction solvent, pesticides were extracted from honey with ethyl acetate in an ultrasonic bath, cleaned up on a Florisil column by an elution of mixture of hexane and ethyl acetate (7: 3,v/v), and analyzed by gas chromatography-electron ionization mass spectrometry (GC-EUMS) in the selected ion monitoring mode (SIM) with PCB103 as internal standard. Recovery studies were performed at 50, 100 and 200 mu g/kg fortification levels for each pesticide, and the recoveries ranged from 82% to 120% with relative standard deviations between 1.8% and 11.0% for different pesticides. The limit of detection was less than 10.0 mu g/kg for all the pesticides. The developed method was linear in the range of 10-500 mu g/kg, with correlation coefficients larger than 0.995. Finally, the developed analytical method has been successfully applied to the determination of pesticide residues in several honey samples.
Optimization of Nitromidazoles Residues in Royal Jelly by Liquid Chromatography Tandem Mass Spectrometry

Abstract:A optimization method was developed for determination of 3 nitromidazoles,metronidazole,ronidazole and dimetridazole in royal jelly using Agilent 1200 liquid chromatography and API 3200 tandem mass spectrometry. Royal jelly was extracted by 2%perchloric acid and then counter extraction with acetic ether and then degreasing with hexane.The determination of purified samples were achieved by liquid chromatography tandem mass spectrometry.The procedure was validated by spiking 3 nitromidazoles(metronidazole,ronidazole and dimetridazole ) at 3 levels(0.5,5, 20μg·kg-1 ):linearity(r0.9990 ),accuracy(recovery=90.5%-127%),precision(intra-day CV≤11.4%,interday CV≤14.6%) and sensitivity(metronidazole and dimetridazole LOQ=0.1μg/kg,ronidazole LOQ=0.2μg/kg ) were met the requirement of method for determination of nitromidazoles residues.The results showed that this method was a highly productive procedure for optimization of the nitromidazoles residues in royal jelly.

Abstract:The application of capillary electrophoresis (CE) to the separation and determination of the active ingredient, 10-hydroxy-2-decenoic acid, in royal jelly with direct on-column UV detection at 214 nm is described. Using a cathodic injection and anodic detection scheme, 10-hydroxy-2-decenoic acid (10-HDA) was separated and detected in less than 10 min in a fused silica capillary column with a phosphate buffer at pH 7.3 with an applied voltage of 20 KV followed by direct UV detection. The use of cetyltrimethylammonium bromide (CTAB) as electroosmotic flow modifier allows the rapid separation of 10-HDA from other constituents in royal jelly by reversing the direction of electroosmotic flow. The influence of organic solvents in the electrolyte on separation selectivity is also discussed.

Abstract:An analytical method based on sugaring-out assisted liquid-liquid extraction (SULLE) and HPLC-fluorescence detection has been developed for the determination of bisphenol A (BPA) and bisphenol B (BPB) in royal jelly. Experimental parameters of SULLE including the extraction solvent, amount of phase separation agent, initial concentration of acetonitrile (ACN), and sample size were investigated. The optimal SULLE procedure involves 0.3 g sample extracted by 2 mL ACN-water mixture (50%, v/v), followed by phase separation with the addition of 0.4 g glucose. The obtained extract was injected into the reversed-phase HPLC system without clean-up step. The chromatography separation of BPA and BPB from royal jelly matrix was achieved in the mobile phase composed of 40% ACN and 60% water with isocratic elution at 1.0 mL/min. Limits of quantification for BPA and BPB were 40 μg/kg and 45 μg/kg, respectively, and limits of detection for BPA and BPB were 16 μg/kg and 18 μg/kg, respectively. Average recoveries of BPA and BPB at three spiked levels were in the range of 88.32~93.59% and 95.14~97.48%, respectively. Precision expressed as relative standard deviations (RSDs) in the interday and intraday analysis were all lower than 5%. The developed method was applied to the analysis of ten commercial and eight raw royal jelly samples. Results indicated that none of BPA and BPB were detected in these 18 samples. The proposed method is simple, fast, and sensitive, and could be used for the monitoring of BPA and BPB contamination in royal jelly.

Abstract:Nucleotides, nucleosides and nucleobases play a greater role in the physiological activity of organisms which are highly present in royal jelly (RJ). The objective of the present study is to develop a HPLC method to simultaneous determine nucleotides, nucleosides and nucleobases in RJ and access them in fresh and commercial RJ samples. The LOD and LOQ were 12.2-99.6 mu g/L and 40.8-289.4 mu g/L, respectively with nearly 100.9% recoveries. Except uric acid, all other compounds were found in RJ samples. Significant difference in the average content of compounds in fresh (2682.93 mg/kg) and commercial samples (3152.78 mg/kg) were observed. AMP, adenosine and adenine were found predominant in all the samples. Significant higher levels of ATP, ADP and AMP was seen in fresh RJ samples, and IMP, uridine, guanosine, and thymidine was seen in commercial RJ samples. The investigated compounds can be used as indexes for assessment RJ freshness and quality. (C) 2014 Elsevier Ltd. All rights reserved.

Abstract:An analytical method for determination for tebufenozide,methoxyfenozide and Fuxian residue in cabbage,pakchoi and tomato by high performance liquid chromatography(HPLC) was developed.Samples were extracted by blending with acidic methanol/water.The extract was purified first by n-hexane to remove some fatty impurities,then extracted with dichloromethane to discard water-soluble impurities,and finally cleaned by Florisil column chromatography.The HPLC conditions : Agilent HC-C18,methanol-water=75:25(V/V) as mobile phase,and UV detection at 230nm.The average fortified recoveries for tebufenozide,methoxyfenozide and Fuxian in cabbage,pakchoi and tomato samples were 76.9%～105%.Relative standard deviation was less than 10%,and the limit of detection of three pesticides in cabbage,pakchoi and tomato samples were 0.05mg/kg,respectively.

Abstract:A simple, rapid, and reliable liquid-liquid extraction coupled to GC-MS method was developed and validated for the quantification of 22 phthalate esters (PAEs) in honey and royal jelly. Instrument parameters for GC-MS were tested to obtain the satisfactory separation between 22 PAEs with high sensitivity. The extraction procedure was optimized in order to achieve the best recovery. The following criteria were used to validate the developed method: linearity, LOD, lower LOQ, precision, accuracy, matrix effect and carry-over. Correlation coefficients were >0.999 by applying the linear regression model based on the least-squares method with a weighting factor (1/x). The intra- and interday precision were within 12.7% in terms of RSD, and the accuracy was within -11.8% in terms of relative error. The mean extraction recoveries ranged between 80.1 and 110.9% for honey and royal jelly. No significant matrix effect and carry-over for PAEs were observed for the analysis of honey and royal jelly samples. A total of 20 real samples were analyzed for a mini-survey using the developed method. Seven PAEs in honey samples and five PAEs in royal jelly samples were found, indicating potential contamination with several PAEs.

Abstract:A high performance liquid chromatographic (HPLC) method for simultaneous determination of 12 pesticides in vegetable and fruit samples was developed and evaluated. The analytical procedure involved in ultrasonic extraction with ethyl acetate, purification using a Florisil SPE column (6 mL, 1 000 mg) and n-hexane-dichloromethane (1: 1, v/v). The eluate was blown to dryness under a stream of nitrogen and the residue was dissolved in 0. 10 mL of methanol, followed by separation and quantitative analysis by using reversed-phase HPLC with gradient elution for the baseline separation of the 12 pesticides and programmed ultraviolet wavelength shift detection in 32 min. The detection limits of the 12 pesticides ranged from 0. 14 to 2. 65 ng. The average recoveries ranged from 62. 2% to 118. 2% with the relative standard deviation (RSD) range of 0. 56% -11. 8% for the spiked fruit samples, and from 52. 1% to 124. 6% with RSD range of 0. 89% - 18. 4% for the spiked vegetables. The method was applied to determining multiresidual pesticides in vegetables and fruit for a total of 40 samples. It is concluded that this method is rapid, accurate, sensitive and reproducible for determining trace pesticides in vegetable and fruit samples.
