Multiple Bio-Analytical Methods to Reveal Possible Molecular Mechanisms of Developmental Toxicity in Zebrafish Embryos/larvae Exposed to Tris(2-Butoxyethyl) Phosphate.

Abstract:The flame retardant tris(2-butoxyethyl) phosphate (TBEP) is a frequently detected contaminant in the environment, wildlife and human milk. The potentially toxic effects of TBEP and their underlying molecular mechanisms have not been elucidated. Here, zebrafish embryos were exposed to different concentrations of TBEP from 4 hours of post-fertilization (hpf) to 120 hpf, and effects on embryonic development and global protein expression patterns examined. Our results demonstrate that treatment with TBEP (0.8-100 mg/L) causes a concentration- and time-dependent decrease in embryonic survival and the hatching percentage. The median lethal concentration was 10.7 mg/L at 120 hpf. Furthermore, exposure to 150 or 800 mu g/L TBEP inhibited the degradation and utilization of vitellogenins and down-regulated the expression of proteins related to cation binding, and lipid transport, uptake and metabolism, accompanied by a decrease in heart rate and body length. Exposure to TBEP (150 or 800 mu g/L) also decreased the expression of proteins involved in cell proliferation and DNA repair, and led to an increased number of apoptotic cells in the tail region. Collectively, our results suggest that exposure to TBEP causes toxicity in the developing zebrafish by inhibiting the degradation and utilization of nutrients from the mother and inducing apoptosis. The Potential Endocrine Disruption of Pesticide Transformation Products (tps): the Blind Spot of Pesticide Risk Assessment.

Abstract:The ecological and health risk assessment of environmental pesticide residues have attracted ever-growing attention; however, their transformation products (TPs) have seldom been considered. Herein, we examined the endocrine-disrupting effects of 4 widely used pesticides as pyriproxyfen (Pyr), malathion (ML), benalaxyl (BX), and fenoxaprop-ethyl (FE), together with their 21 TPs through in vitro and in silico approaches, and found approximately 50% of the TPs exhibited stronger endocrine-disrupting effects than their corresponding parent compounds. Specifically, Pyr and 9 TPs (five TPs of Pyr, one of ML, one of BX, and two of FE) exhibited estrogen-disrupting effects, which were also confirmed by results of E-screen and pS2 expression assays, and molecular docking showed that certain hydroxylated TPs could well mimic the binding mode of estrogen with ERα. Meanwhile, two TPs of Pyr, ML and its TP demonstrated weak glucocorticoid antagonistic activities partially contributed by hydrogen bonds. We also discovered that in H295R cells, all the endocrine disruptors increased hormone secretion and the related gene expression levels. Conclusively, since an increasing number of pesticide TPs have been being detected in various environmental media, a more comprehensive understanding of the ecological risk of pesticide TPs is imperative for risk assessments more extensively and regulatory policy-making on pesticide restriction in the future.
Acute toxicity of dipterex and deltamethrin on laval Paramisgurnus dabryanus and Moina macrocopa

Abstract:The acute toxicity of dipterex and deltamethrin on 5 dph( days after hatching) laval Paramisgurnus dabryanus and Moina macrocopa was determined in static bioassay tests,when p H was 8. 2 ± 0. 1,water temperature was( 25 ± 2) ℃and dissolved oxygen was below 5 mg / L. The results showed that, the safe concentration of dipterex for 5 dph laval P. dabryanus and M. macrocopa were 0. 4 mg / L and 0. 0489 mg / L respectively and that the safe concentration of deltamethrin for 5 dph laval P. dabryanus and M. macrocopa were 0. 16 μg / L and 0. 0042 μg / L,respectively. The toxicities of these drugs was in order of Dipterex > Deltamethrin.

Abstract:The extensive use of carbamate pesticides has led to a range of environmental and health problems, such as surface and groundwater contamination, and endocrine disorders in organisms. In this study, we focused on examining the effects of toxic exposure to the carbamate pesticide methomyl on the hatching, morphology, immunity and developmental gene expression levels in zebrafish embryos. Four concentrations of methomyl (0, 2, 20, and 200 μg/L) were administered to zebrafish embryos for a period of 96 h. The study found that exposure to methomyl accelerated the hatching process of zebrafish embryos, with the strongest effect recorded at the concentration of 2 μg/L. Methomyl exposure also trigged significantly reductions in heart rate and caused abnormalities in larvae morphology, and it also stimulated the synthesis and release of several inflammatory factors such as IL-1β, IL-6, TNF-α and INF-α, lowered the IgM contents, ultimately enhancing inflammatory response and interfering with immune function. All of these showed the significant effects on exposure time, concentration and their interaction (Time × Concentration). Furthermore, the body length of zebrafish exposed to methomyl for 96 h was significantly shorter, particularly at higher concentrations (200 μg/L). Methomyl also affected the expression levels of genes associated with development (down-regulated igf1, bmp2b, vasa, dazl and piwi genes), demonstrating strong developmental toxicity and disruption of the endocrine system, with the most observed at the concentration of 200 μg/L and 96 h exposure to methomyl. The results of this study provide valuable reference information on the potential damage of methomyl concentrations in the environment on fish embryo development, while also supplementing present research on the immunotoxicity of methomyl.

Abstract:Plasticizer, especially di-2-ethyl hexyl phthalate ( DEHP ) , is widely used in various types of plastic product. With the news of adding plasticizer in food continually exposed, the harm of plasticizer to human health has attracted public attention gradually. The in vi-vo accumulation of plasticizer and its metabolites can lead to the multiple organs damage. The reproductive organs accumulation of plas-ticizer can contribute to significant reproductive system damage and its accumulation in liver can lead to the oxidative stress and cause the liver cell apoptosis. In addition, plasticizer can be used as an adjuvant to affect the immune system. This review systematically elab-orates the reproductive toxicity, liver toxicity and immune toxicity of DEHP, and the relevant mechanisms.
The Gene Expression Profile of Monochamus Alternatus in Response to Deltamethrin Exposure

Abstract:To identify additional genes that are potentially associated with deltamethrin toxicology, differentially expressed gene (DGE) libraries of Monochamus alternatus after short-term exposure to a sub-lethal concentration of deltamethrin were prepared. The transcripts were sequenced using the Illumina sequencing platform. After cleaning and quality checks, the total numbers of clean reads were 7.066 and 7.139 million in the control and deltamethrin-exposed libraries, respectively. We found that 4886 and 2083 unique genes were significantly up- and down-regulated, respectively. The gene ontology enrichment analysis identified that 4032, 3379 and 8323 DEGs were involved in cellular components, molecular functions and biological processes, respectively, and showed that the genes were distributed among more than 50 categories. The Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis indicated that the top 20 enriched pathways included the glutathione metabolism pathway and the antigen processing and presentation pathway. There were 42 DEGs enriched in the antigen processing and presentation pathway, including those encoding the 70-kDa heat shock protein, protein disulfide isomerase and calreticulin, all of which were up-regulated. There were 61 DEGs enriched in the glutathione metabolism pathway, and these included the up-regulated DEGs encoding glutamate–cysteine ligase, glutathione synthase and glutathione reductase and the down-regulated DEGs encoding glutathione S-transferase, glutathione peroxidase and ornithine decarboxylase. This study is the first step toward achieving an understanding of the profile of the secondary targets of deltamethrin in M. alternatus, and the results may provide insights to further explore the functions of the target genes in detoxification and resistance to deltamethrin.
Integration of metabolomics and transcriptomics reveals the toxicological mechanism of deltamethrin exposure in Chinese mitten crab Eriocheir sinensis

Abstract:This study investigated the toxicological mechanism of deltamethrin on Chinese mitten crab Eriocheir sinensis juveniles in fresh water. We first conducted an acute toxicity test, followed by laboratory methods to detect changes in immune-related indices in terms of antioxidant enzyme markers, lipid metabolism-related genes, and autophagy-related and apoptosis genes. The acute toxicity (96-h LC50) of deltamethrin to E. sinensis was 7.195 μg/L. After 48 h of exposure, serum showed elevated immune-related indices (P < 0.05) for alkaline phosphatase (AKP), acid phosphatase (ACP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), complement components C3 and C4, and the key pro-inflammatory cytokines interleukin-6, interleukin-1β, and tumor necrosis factor alpha (TNF-α). In hepatopancreas at 48 h, indicators related to the antioxidant system, namely superoxide dismutase (SOD) and glutathione (GSH), were significantly elevated, whereas nitric oxide and total antioxidant capacity (T-AOC) were decreased (P < 0.05). In contrast, lipid metabolism indices for triglyceride (TG), total cholesterol (TC), and malondialdehyde (MDA) were increased (P < 0.05). Transcriptomics and metabolomics revealed that exposure to deltamethrin disrupted the lipid metabolic process in the hepatopancreas mainly by altering fatty acid synthesis, amino acid metabolism, immune signaling, and autophagy activation, while the exposure increased the content of phospholipids and cholesterol but decreased the levels of amino acids and palmitoleic acid. Quantitative genetics revealed significantly aberrantly expressed (P < 0.05) lipid metabolism-related genes, including acc1, fasn, scd1, and pnpla2, all key genes involved in lipid accumulation. Deltamethrin exposure also significantly altered (P < 0.05) gene expression levels for Toll-like receptor (tlr), myeloid differentiation factor 88 (myd88), crustin1, anti-lipopolysaccharide factor isoform 3 (alf3), tumor necrosis factor alpha (tnf-α), and NF-κB transcription factor relish. Furthermore, deltamethrin activated the toll-like receptor/major myeloid differentiation response gene 88/nuclear factor kappa-light-chain-enhancer of activated B cells (TLR/MyD88/NF-kB) signaling pathway, which activates a nonspecific immune response in E. sinensis. Additionally, carnitine palmitoyltransferase 1 A (cpt1a), cytochrome c (cyt-c), adenosine 5'-monophosphate (amp)-activated protein kinase (ampk), the autophagosomal protein microtubule-associated protein 1 light chain 3c (lc3c), and the autophagy-related proteins beclin1, atg5, atg12 were significantly induced (P < 0.05) in the adenosine monophosphate-activated protein kinase/rapamycin (AMPK/mTOR) signaling pathway. These changes resulted in excess free radicals, causing oxidative stress in the mitochondrial membrane, promoting mitochondrial autophagy. The results confirm that deltamethrin exposure can induce hepatopancreatic injury by promoting mitochondrial autophagy, activating an immune response, and inhibiting lipid metabolism. Overall, this study provides multi-level information to reveal the toxic effects of deltamethrin on E. sinensis.

Abstract:The increase of diseases can be attributable to the turbulence of the immune system function, and this trend could be at least partially bottom of new and modified patterns of exposure to chemicals in environment. Among chemicals of concern, pesticides can be included. The present review describes the definition of immunotoxicity, the immunotoxicity test methods, the relation of pesticide and immune response and the immunotoxicity of pesticides in aquatic species, terrestrial species, mammals and human. The importance of studying the immunotoxicity of chiral pesticides is also introduced and prospected.

Abstract:Pyrethroids are a class of highly effective, broad-spectrum, less toxic, biodegradable synthetic pesticides. However, despite the extremely wide application of pyrethroids, there are many problems, such as insecticide resistance, lethal/sub-lethal toxicity to mammals, aquatic organisms or other beneficial organisms. The objectives of this review were to cover the main structures, synthesis, steroisomers, mechanisms of action, anti-mosquito activities, resistance, photodegradation and toxicities of pyrethroids. That was to provide a reference for synthesizing or screening novel pyrethroids with low insecticide resistance and low toxicity to beneficial organisms, evaluating the environmental pollution of pyrethroids and its metabolites. Besides, pyrethroids are mainly used for the control of vectors such as insects, and the non-target organisms are mammals, aquatic organisms etc. While maintaining the insecticidal activity is important, its toxic effects on non-target organisms should be also considered. Pyrethroid resistance is present not only in insect mosquitoes but also in environmental microorganisms, which results in anti-pyrethroids resistance (APR) strains. Besides, photodegradation product dibenzofurans is harmful to mammals and environment. Additionally, pyrethroid metabolites may have higher hormonal interference than the parents. Particularly, delivery of pyrethroids in nanoform can reduce the discharge of more toxic substances (such as organic solvents, etc.) to the environment.

Abstract:Biochemical synthetic pyrethroids, deltamethrin are presently used insecticides for the control of mosquito vector-borne diseases in worldwide. Mosquito re-emergence with diseases becoming a serious problem due to development of insecticide resistance. The comprehensive knowledge on the underlying mechanisms of resistance against deltamethrin is required for implementation of an efficient vector control programme. The assessment of the biological fitness of a mosquito strain exposed to insecticide pressure is extremely vital because it provides information on the development of resistance. In the present study, the adult stage of malaria vector, Anopheles stephensi , was designated for the study of deltamethrin resistance (F40 generations). The non-blood-fed, laboratory-reared females to sub-lethal doses of deltamethrin (0.004%, 0.005%, 0.007%, or 0.01%) exposed to every generation for up to F40. The adult mosquito susceptibility was performed by WHO standard method for evaluation. After 24 h, mortality was recorded in both treated and control groups. Therefore, the biological fitness characteristics such as feeding, fecundity, hatchability, egg retention, immature duration, adult emergence, and adult life span were studied to assess the exposed deltamethrin under selection pressure as compared to the unexposed (control) population. The laboratory selection of An. stephensi exposed deltamethrin over the generations were diminished its biological fitness. Information on biological fitness including reproductive potential of mosquito strain under selection pressure against deltamethrin is incredibly necessary because it would facilitate in resistance management. Baseline information gives in this experiment will guide for future studies on the susceptibilities of wild malaria mosquito populations in India.

[Progress in Developmental Toxicity of Methylmercury].

Abstract:Mercury is a common pollutant in the environment and it can be turned into methylmercury (MeHg) under the metabolism of microbe. MeHg is a confirmed teratogen in human. It can impair the germ cells and influence the growth and development of the offspring. MeHg can easily pass through the placental barrier, accumulate in the fetus and change the behavior and perceptibility of the second generation related to the function of central nervous system. Recently, the investigations on the mechanism about developmental toxicity of MeHg have progressed much, so we make a review in this field as follows.

Abstract:Imidacloprid,a typical neonicotinoid insecticide,has the characteristics of broad-spectrum insect control and low toxicity to mammals,and therefore is widely used.Now imidacloprid exists in both environment and human body,and its potential toxic effects,particularly genetic toxicity,have raised great concern.China is the largest country producing and exporting imidacloprid,and its annual output accounts for about two-thirds of the global total production.However,few data report the human exposure to imidacloprid in China.Given the widespread presence of imidacloprid and its potential genetic toxicity,future research should focus on its genetic damage to human being and relevant mechanism.
Toxicological effects and defense mechanisms induced by beta-cypermethrin in Drosophila melanogaster

Abstract:Widespread use of the pyrethroid insecticide beta-cypermethrin (beta-CYP) has led to adverse effects on nontarget populations within agroecosystems. Despite the efficacy of beta-CYP in pest control, its toxicological and defense mechanisms remain incompletely understood. In the present study, we explored the toxicological effects, antioxidant mechanisms and immune response against beta-CYP using Drosophila melanogaster, a well-established model organism for the study of insect biology, to represent the broader class of nontarget organisms. We exposed Drosophila larvae to 0.667 μg/mL beta-CYP and revealed that delayed development and caused intestinal epithelial damage in larvae. To gain insights into the molecular underpinnings of these effects, RNA sequencing analysis and quantitative polymerase chain reaction validation were performed. These analyses revealed that the messenger RNA levels of glutathione S-transferase were increased, third instar larvae exhibited an increase in reactive oxygen species content and a corresponding increase in antioxidant enzyme activity in response to beta-CYP exposure, indicating an upregulated response to oxidative stress. Beta-CYP also activated Hippo pathway to resist apoptosis and promote cell proliferation. Moreover, beta-CYP induced melanization and Toll immune pathways involved in immune response in Drosophila larvae, specifically the Toll pathway gene Drs. This activation suggests that Drosophila increases antioxidant defenses and promotes mitosis in damaged tissues as compensatory mechanisms to mitigate the cytotoxic effects of beta-CYP. These findings provide new insight into the mechanisms of beta-CYP-induced toxicity and the defense mechanisms in insects; they may also inform strategies for the sustainable use of insecticides and the development of mitigation measures to protect nontarget species in agroecosystems.
Mixture toxicity of thiophanate-methyl and fenvalerate to embryonic zebrafish (Danio rerio) and its underlying mechanism

Abstract:Though pesticide mixtures can reflect the real-life situation in the water ecosystem, the quantification of their toxicity is still not fully understood. Combined effects of thiophanate-methyl (THM) and fenvalerate (FEN) on embryonic zebrafish (Danio rerio) and underlying mechanism were conducted in this study. Results showed that the 96-h LC50 values of THM to D. rerio at different growth periods ranged from 12.1 to 26.1 mg L−1, which were lower in comparison with those of FVR ranging from 0.025 to 2.8 mg L−1. Mixture of THM and FVR exhibited a synergetic response to zebrafish embryos. Activities of Cu/Zn-SOD, POD, caspase 3 and caspase 9 were significantly different in most of single and mixture administrations compared with the control group. In addition, five genes (P53, Cu/Zn-sod, crh, ERα and IL-8) associated with oxidative stress, cellular apoptosis, immune system and endocrine system showed greater variations of expressions when administrated to pesticide mixtures compared with single chemicals. Our experimental results exhibited that mixtures of thiophanate-methyl and fenvalerate produced higher toxicity towards aqueous vertebrates than when determined singly. Collectively, upcoming environmental risk assessments established according to single administrations might not be enough to protect the water ecosystem.

Abstract:The acute immobilization toxicity effects expressed as pI50(-lg EC50) of sixteen triazole pesticides on D.magna in 48 hours were studied.2D-QSAR models were carried out with some quantitative chemical parameters calculated by G03W program with 6-31G* basis,including the energy of the highest occupied molecular orbital(EHOMO) and energies of its two neighborhood orbits(EHOMO-1 and EHOMO-2),the energy of the lowest unoccupied molecular orbit(ELUMO) and energies of its two neighborhood orbits(ELUMO+1 and ELUMO+2),atomic charges(Q),electrostatic potencial charges(EQ),dipole moment(μ),the first-order polarizability(α),the first-order hyperpolarizability(αH),lipid/water partition coefficient(Clg P),heat of formation(EHF),hydration energy(EHE),molar refractivity(Rm) etc.These models would provide the theoretical basis for predicting the pI50 of the compounds with a similar structure.Additionally,the predicting models could initially explaine the mechanism of toxicity to D.magna,and scientifically design triazole pesticides with high efficiency and low toxicity from perspective of molecular structure.

Abstract:In the past,chiral pesticides were explored as racemic mixtures in environmental fate and ecotoxicities.However,as a result of the enantioselective interactions with chiral molecules,the enantiomers of chiral pesticides may have different potential toxicities to humans and animals.The present review introduced recent findings on enantioselective endocrine disruption,neurotoxicity,immunotoxicity,reproductive and developmental toxicity of organophosphorus,organochlorines,pyrethroids and phenylpyrazole pesticides,as well as the underlying mechanisms.Furthermore,the developmental trends of future investigations were discussed.
