Abstract:β-Cypermethrin (β-CYP), one of most important pyrethroids, is widely used to control insects, and has been detected in organisms, including human. Pyrethroids have been shown to pose neurotoxicity, hepatotoxicity, endocrine disruption and reproductive risks in mammals. However, research in immunotoxicity of pyrethroids, especially their metabolites, is limited. A common metabolite of pyrethroids is 3-phenoxybenzoic acid (3-PBA) in mammals. Thus, in this study, we evaluated the immunotoxicity of β-CYP and 3-PBA in mouse macrophages, RAW 264.7 cells. MTT assays showed that both β-CYP and 3-PBA reduced cell viability in a concentration- and time-dependent manner. Flow cytometry with Annexin-V/PI staining demonstrated that both β-CYP and 3-PBA induced RAW 264.7 cell apoptosis. Furthermore, our results also showed that N-acetylcysteine partially blocked β-CYP- and 3-PBA-induced cytotoxicity and apoptosis. Intrinsic apoptotic pathway was stimulated by both β-CYP and 3-PBA exposure. In addition, we found that β-CYP and 3-PBA inhibited mRNA levels of pro-inflammatory cytokines with or without LPS stimulation. Phagocytosis assay showed that both β-CYP and 3-PBA inhibited phagocytic ability of macrophages. Moreover, it was also found that both β-CYP and 3-PBA increased reactive oxygen species (ROS) levels in RAW 264.7 cells. Accordingly, both β-CYP and 3-PBA were found to regulate the mRNA levels of oxidative stress-related genes in RAW 264.7 cells. Taken together, the results obtained in this study demonstrated that β-CYP and 3-PBA may have immunotoxic effect on macrophages and that elevated ROS may underlie the mechanism. The present study will help to understand the health risks caused by β-CYP and other pyrethroids.
Toxicological effects of ciprofloxacin exposure to Drosophila melanogaster

Abstract:The abuse of ciprofloxacin (CIP) may cause serious side effects and the mechanisms underlying these effects remain unclear. Here, we determinate the 48 h, 72 h and 96 h LC50 values of CIP to Drosophila melanogaster and demonstrate a series of adverse effects after D. melanogaster was exposed to CIP at a sublethal concentration (3.2 mg mL-1). Treated individuals showed shorter lifespan, delayed development and many of the treated larvae failed to pupate or hatch. Smaller body size was observed at every life stage when exposed to CIP and the size of pupae, the weight of third-instar larvae exhibited a perfectly dose-response relationship that the larger concentration exposed to, the smaller body size or lighter weight is. Moreover, reduction in fat body cell viability, elevated oxidative stress markers (SOD and CAT) and down-regulation of diap1, ex, two target genes of Yorkie (Yki), was observed in response to CIP exposure. Most importantly, we found two types of black spot in Drosophila and the proportion of larvae with a black spot was positively related to the treatment dose, which is new in the field. This study provides a scientific basis for the potential harm caused by abuse of quinolones with the goal of urging cautious use of antibiotics.

Abstract:Insect toxicology and chemical ecology are inherently interconnected disciplines, both dedicated to unraveling the intricate relationships between insects and the diverse array of chemical compounds that pervade their surroundings. Drosophila melanogaster , owing to its genetic and physiological similarities to other insects, serves as a robust model system in the study of insect toxicology. Moreover, state-of-the-art techniques in Drosophila neurobiology have extensively probed the chemosensory system of insects, providing significant insights into their adaptation to chemical environments. In this review, we emphasize the advancements achieved through the application of Drosophila genetics in investigations spanning both of these fields, significantly enhancing our understanding of the mode of action and resistance mechanisms of insecticides, as well as unraveling the molecular and cellular mechanisms underlying insect chemosensation and associated behaviors. The profound insights derived through this tiny fly not only enrich our understanding of the broader world of insects but also hold the potential to develop more effective and sustainable strategies for pest management.

Abstract:Beta-cypermethrin (beta-CYP) is a widely used pyrethroid pesticide, the extensive application of which may potentially cause damage to non-target organisms. To investigate the effect of beta-CYP on the antioxidant system of aquatic animals, adult zebrafish were exposed to environmentally relevant dosages (0.01, 0.1 and 1.0 mu g/L) of beta-CYP. The activities of four antioxidant enzymes in zebrafish liver and brain tissue were tested after 7, 15 and 30days of exposure. Our results showed that exposure of beta-CYP could induce different levels of increase in hepatic superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR) and glutathione peroxidase (GPx) activities at 7 and 15days post exposure (dpe), but caused apparent inhibition of hepatic SOD, GR and GPx activities at 30dpe. Unlike in liver tissue, SOD and CAT activities in zebrafish brain did not show any apparent response to beta-CYP during the whole treatment period. In addition, increased brain GPx activities were observed at 7 and 30dpe.

Abstract:Ionotropic γ-aminobutyric acid (GABA) receptors in insects, specifically those composed of the RDL (resistant to dieldrin) subunit, serve as important targets for commonly used synthetic insecticides. These insecticides belong to various chemical classes, such as phenylpyrazoles, cyclodienes, meta-diamides, and isoxazolines, with the latter two potentially binding to the transmembrane inter-subunit pocket. However, the specific amino acid residues that contribute to the high sensitivity of insect RDL receptors to these novel insecticides remain elusive. In this study, we investigated the susceptibility of seven distinct Drosophila melanogaster Rdl point mutants against four meta-diamide and isoxazoline insecticides: isocycloseram, fluxametamide, fluralaner, and broflanilide. Our findings indicate that, despite exhibiting increased sensitivity to fluralaner in vitro, the RdlI276C mutant showed resistance to isocycloseram and fluxametamide. Similarly, the double-points mutant RdlI276F+G279S also showed decreased sensitivity to the tested isoxazolines. On the other hand, the RdlG335M mutant displayed high levels of resistance to all tested insecticides. Molecular modeling and docking simulations further supported these findings, highlighting similar binding poses for these insecticides. In summary, our research provides robust in vivo evidence supporting the idea that the inter-subunit amino acids within transmembrane M1 and M3 domains form the binding site crucial for meta-diamide and isoxazoline insecticide interactions. This study highlights the complex interplay between mutations and insecticide susceptibility, paving the way for more targeted pest control strategies.

Abstract:Diptera and Lepidoptera species have the highest levels of insecticide resistance, and the mechanism of drug resistance has been studied in detoxification metabolism genes such as P450, GST, EST, and ABC. Since Bactrocera dorsalis are resistant to a variety of chemicals, the pattern and mechanism of resistance in Bactrocera dorsalis have been investigated from a variety of aspects such as detoxification metabolism genes, detoxification enzymes, intestinal symbiotic bacteria, and synergists in the world. In this study, 51 species and 149 detoxification metabolism genes were annotated in the Suppression Subtractive Hybridization (SSH) library, and 12 candidate genes related to beta-cypermethrin resistance were screened and quantitatively expressed in this library. Two genes were found to be upregulated in the egg stage, three genes in the larval stage, one gene in the pupal stage, and five genes in the adult stage, and four genes were found to be upregulated in the midgut and the malacca ducts in the midgut. The expression of cyp6g1, cyp6a22, GST-Epsilon9, and Trypsin-4 genes was upregulated in resistant strains, with the most obvious upregulation occurring in the midgut and the Malpighian tubules. These results provide new insights into the study of pesticide resistance in quarantine insects.
Effects of cadmium on fecundity and defence ability of Drosophila melanogaster

Abstract:Cadmium (chemical symbol, Cd) is an extremely common pollutant that poses a toxicity threat to organisms. Therefore, we tested Drosophila melanogaster fecundity, Cd accumulation, and activity of two enzymes following Cd stress and used quantitative real-time polymerase chain reaction (qPCR) to quantify the mRNA expression levels of several genes involved in fecundity and defence. D. melanogaster was placed in a medium containing different concentrations of Cd (13, 26, and 52 mg L-1), following which, inductively coupled plasma atomic emission spectroscopy showed that Cd accumulation in Drosophila increased with the increase in its dietary intake. We also observed that Cd at these concentrations significantly prolonged the mating latency in females and reduced the number of eggs laid. However, the same Cd concentrations did not affect male fecundity. Acetylcholinesterase activity was only detected at 52 mg L-1 Cd in both sexes, whereas glutathione S-transferase activity was inhibited at 26 and 52 mg L-1 Cd in females. The results of qPCR indicated that exposure to 13-52 mg L-1 Cd affected the expression of reproduction-related genes, including downregulation of enok and upregulation of dally and dpp. The same level of exposure also induced transcriptional responses from three defence-related genes (hsp70, gstd2, and gstd6). Taken together, the results revealed that Cd exposure might negatively affect the expression of genes associated with D. melanogaster reproduction and trigger the transcription of defence-related genes. We suggest that further analyses of fecundity and defence responses may help develop indicators of Cd toxicity and improve our understanding of antitoxin defences.
Biochemical Toxicity and Transcriptome Aberration Induced by Dinotefuran in Bombyx Mori.

Abstract:Dinotefuran is a third-generation neonicotinoid pesticide and is increasingly used in agricultural production, which has adverse effects on nontarget organisms. However, the research on the impact of dinotefuran on nontarget organisms is still limited. Here the toxic effects of dinotefuran on an important economic species and a model lepidopteran insect, Bombyx mori , were investigated. Exposure to different doses of dinotefuran caused physiological disorders or death. Cytochrome P450, glutathione S-transferase, carboxylesterase, and UDP glycosyl-transferase activities were induced in the fat body at early stages after dinotefuran exposure. By contrast, only glutathione S-transferase activity was increased in the midgut. To overcome the lack of sensitivity of the biological assays at the individual organism level, RNA sequencing was performed to measure differential expressions of mRNA from silkworm larvae after dinotefuran exposure. Differential gene expression profiling revealed that various detoxification enzyme genes were significantly increased after dinotefuran exposure, which was consistent with the upregulation of the detoxifying enzyme. The global transcriptional pattern showed that the physiological responses induced by dinotefuran toxicity involved multiple cellular processes, including energy metabolism, oxidative stress, detoxification, and other fundamental physiological processes. Many metabolism processes, such as carbon metabolism, fatty acid biosynthesis, pyruvate metabolism, and the citrate cycle, were partially repressed in the midgut or fat body. Furthermore, dinotefuran significantly activated the MAPK/ CREB, CncC/Keap1, PI3K/Akt, and Toll/IMD pathways. The links between physiological, biochemical toxicity and comparative transcriptomic analysis facilitated the systematic understanding of the integrated biological toxicity of dinotefuran. This study provides a holistic view of the toxicity and detoxification metabolism of dinotefuran in silkworm and other organisms.
Exploring the multilevel effects of triclosan from development, reproduction to behavior using Drosophila melanogaster

Abstract:Triclosan (TCS) is widely used as an antibacterial agent, but its residue in the environment poses a great threat. In this study, Drosophila melanogaster were treated with series concentrations of TCS and the effects on development, behavior, reproduction, and oxidative stress indicators were investigated. The results showed that that high concentrations of TCS severely interfered with the metamorphosis, resulting in lower hatching rate and longer development time. The hatching rate was only 75.00% ± 4.08% in 0.80 mg/mL TCS group. TCS also showed dose-dependent damage to the fertility of flies, causing ovarian defects and decreased the number of offspring. Almost no offspring adults hatched when exposed to high concentrations of TCS (0.50 and 0.80 mg/mL), and the hatching rate was 0% in 0.80 mg/mL TCS group. Larvae crawling, adult climbing and anti-starvation ability were also affected to varying degrees and showed hormesis. TCS could damage larval intestinal cells in a dose-dependent manner, and injury was lightened with culture time prolonging to 30 h. It is noteworthy that TCS caused redox imbalance with an increase on catalase (CAT) activity and decrease on reactive oxygen species (ROS) level. Our results conclude that TCS elicits multiple impacts on Drosophila and its rational use should be strengthened.

Autophagy Protects Murine Macrophages from Β-Cypermethrin-induced Mitochondrial Dysfunction and Cytotoxicity Via the Reduction of Oxidation Stress.

Abstract:The immunotoxicity of synthetic pyrethroid (SPs) has garnered much attention, and our previous research demonstrated that β-CYP causes immunotoxicity and oxidative stress in macrophages. Nevertheless, the underlying mechanism remains largely unknown. In this study, the murine macrophage RAW 264.7 cells and murine peritoneal macrophages (PMs) were exposed to β-CYP. The results showed that β-CYP elevated intracellular ROS levels in both RAW 264.7 cells and PMs. Exposure to β-CYP also caused mitochondrial dysfunction with reduced mitochondrial membrane potential (MMP), intracellular ATP level and mitochondrial DNA (mtDNA) content in the two cell types. In addition, exposure of RAW 264.7 cells to β-CYP for 12 h and 24 h enhanced autophagy, with elevated Beclin1, Rab7, Lamp1 and LC3-II expression levels, while 48 h of exposure attenuated autophagy. In contrast, exposure of PMs to β-CYP for 12 h promoted autophagy, whereas exposure for 24 h and 48 h impaired autophagy. Cotreatment with an antioxidant, N-acetyl-L-cysteine (NAC), partially blocked the reduced MMP, intracellular ATP level and autophagy disturbance. Moreover, cotreatment with an autophagy agonist, rapamycin (RAPA), partially blocked mitochondrial dysfunction and oxidative stress in the two cell types, whereas cotreatment with an autophagy inhibitor, 3-methyladenine (3-MA), augmented the abovementioned toxic effects. Furthermore, mitochondrial ROS levels in both RAW 264.7 cells and PMs were elevated by exposure to β-CYP, and molecular docking showed that β-CYP docked with mouse respiratory chain complex I by binding to the ND2, ND4, and ND5 subunits of the protein complex. Taken together, the data obtained in the present study demonstrate that oxidative stress partially mediates mitochondrial dysfunction and autophagy disturbance upon exposure to β-CYP in macrophages, and autophagy plays a protective role against the toxic effects.

Abstract:Tetrabromoethylcyclohexane (TBECH), as one emerging brominated flame retardants, is ubiquitous in the environment, including water and aquatic organisms. TBECH was found to exhibit endocrine-disrupting effects in different models, whereas a survey of comprehensive toxic effects of TBECH on zebrafish is limited. In the present study, zebrafish (Danio rerio) were waterborne exposed continuously to TBECH from embryonic stage (3 h post-fertilization (hpf)) to the time when the respective parameters were evaluated. Exposure to TBECH reduced hatchability of zebrafish embryos at 72 and 96 hpf, diminished heart rate of zebrafish larvae at 48 hpf, and increased malformation in zebrafish larvae at 96 hpf. In addition, exposure to TBECH diminished free swimming distance both in the light and under a photoperiod of 10 min light/10 min dark cycles in zebrafish larvae at 6 days post-fertilization (dpf). Moreover, exposure to TBECH elevated activities of superoxide dismutase (SOD) and catalase (CAT), malondialdehyde (MDA) content, whereas it reduced glutathione (GSH) content, in zebrafish larvae at 6 dpf. Accordingly, RT-qPCR analysis demonstrated that TBECH exposure increased the mRNA levels of sod1, sod2, cat, and gpx1 in zebrafish larvae at 6 dpf. With respect to the immune aspect, the mRNA levels of pro-inflammatory genes, including il-1b, il-6, il-8, and tnfa, in larval zebrafish at 6 dpf were increased by exposure to TBECH, while pretreatment with TBECH inhibited 24 h of exposure to LPS-stimulated elevation in the mRNA levels of the abovementioned four pro-inflammatory genes in zebrafish larvae at 6 dpf. Furthermore, TBECH treatment increased caspase-3 enzyme activities and regulated apoptosis-related genes in larval zebrafish at 6 dpf. Taken together, the data obtained in this study demonstrated that TBECH caused developmental and locomotor behavioral toxicity, immunotoxicity, oxidative stress and proapoptotic effects in early life zebrafish. The present study will help to understand the comprehensive toxicity of TBECH in zebrafish.
Cis-bifenthrin Causes Immunotoxicity in Murine Macrophages.

Abstract:Synthetic pyrethroids (SPs) are commonly used insecticides that have been detected in mammals, including humans, indicating a potential threat to human health. Bifenthrin (BF), as well as other pyrethroids, has been shown to possess neurotoxic, reproductive, hepatotoxic and nephrotoxic potential in mammals. However, studies regarding the immunotoxicity of BF and its mechanism are limited. In this study, we aim to exam the immunotoxicity of cis-BF on the murine macrophage cell line, RAW 264.7. MTT assay results demonstrated that cis-BF exposure induced apoptosis in RAW 264.7 cells in a concentration-dependent manner. We found that the expression of p53 and caspase-3 was up-regulated, while the expression of Bcl-2 was down-regulated during cis-BF-induced apoptosis. In addition, we also found that cis-BF exposure caused oxidative stress in RAW 264.7 cells in a dose-dependent manner. Interestingly, cis-BF exposure was found to inhibit the increase in transcription levels of IL-1β, IL-6 and TNF-α responding to LPS stimulation. We also found that the induced increase in IFN-β mRNA levels upon Sendai virus infection was blocked with cis-BF exposure. Finally, we found that cis-BF exposure increased ROS levels and dysregulated mRNA levels of oxidative stress-related genes in RAW 264.7 cells. The present study elucidates the immunotoxicity effect of cis-BF on macrophages and its possible underlying mechanism. The results from this study support the necessity to evaluate immune dysfunction in the risk assessment of cis-BF exposure.
Effects of cadmium on oxidative stress and cell apoptosis in Drosophila melanogaster larvae

Abstract:With the increase of human activities, cadmium (Cd) pollution has become a global environmental problem affecting biological metabolism in ecosystem. Cd has a very long half-life in humans and is excreted slowly in organs, which poses a serious threat to human health. In order to better understand the toxicity effects of cadmium, third instar larvae of Drosophila melanogaster (Canton-S strain) were exposed to different concentrations (1.125 mg/kg, 2.25 mg/kg, 4.5 mg/kg, and 9 mg/kg) of cadmium. Trypan blue staining showed that intestinal cell damage of Drosophila larvae increased and the comet assay indicated significantly more DNA damage in larvae exposed to high Cd concentrations. The nitroblue tetrazolium (NBT) experiments proved that content of reactive oxygen species (ROS) increased, which indicated Cd exposure could induce oxidative stress. In addition, the expression of mitochondrial adenine nucleotide transferase coding gene (sesB and Ant2) and apoptosis related genes (Debcl, hid, rpr, p53, Sce and Diap1) changed, which may lead to increased apoptosis. These findings confirmed the toxicity effects on oxidative stress and cell apoptosis in Drosophila larvae after early cadmium exposure, providing insights into understanding the effects of heavy metal stress in animal development.

Abstract:Cypermethrin (CYP), a widely used Type II pyrethroid pesticide, is one of the most common contaminants in the freshwater aquatic system. We studied the effects of CYP exposure on the induction of hepatic oxidative stress, DNA damage and the alteration of gene expression related to apoptosis in adult zebrafish. Hepatic mRNA levels for the genes encoding antioxidant proteins, such as Cu/Zn-Sod, Mn-Sod, Cat, and Gpx, were significantly upregulated when zebrafish were exposed to various concentrations of CYP for 4 or 8days. In addition, the main genes related to fatty acid β-oxidation and the mitochondrial genes related to respiration and ATP synthesis were also significantly upregulated after exposure to high concentrations (1 and 3μgL−1) of CYP for 4 or 8days. Moreover, in a comet assay of zebrafish hepatocytes, tail DNA, tail length, tail moment and Olive tail moment increased in a concentration-dependent manner. The significant induction (p<0.01) of all four parameters observed with CYP concentrations of 0.3μgL−1 or higher suggests that heavy DNA damage was induced even at low levels. Furthermore, several apoptosis- related genes, such as p53, Apaf1 and Cas3, were significantly upregulated after CYP exposure, and Bcl2/Bax expression ratio decreased, especially in groups treated with 1 and 3μgL−1 CYP for 8days. Taken together, our results suggested that CYP has the potential to induce hepatic oxidative stress, DNA damage and apoptosis in zebrafish. This information will be helpful in fully understanding the mechanism of aquatic toxicology induced by CYP in fish.
Digital gene expression profiling (DGE) of cadmium-treated Drosophila melanogaster

Abstract:Cadmium is highly toxic and can cause oxidative damage, metabolic disorders, and reduced lifespan and fertility in animals. In this study, we investigated the effects of cadmium in Drosophila melanogaster, performing transcriptome analysis by using tag-based digital gene expression (DGE) profiling. Among 1970 candidate genes, 1443 were up-regulated and 527 were down-regulated following cadmium exposure. Using Gene Ontology analysis, we found that cadmium stress affects three processes: transferase activity, stress response, and the cell cycle. Furthermore, we identified five differentially expressed genes (confirmed by real-time PCR) involved in all three processes: Ald, Cdc2, skpA, tefu, and Pvr. Pathway analysis revealed that these genes were involved in the cell cycle pathway and fat digestion and absorption pathway. This study reveals the gene expression response to cadmium stress in Drosophila, it provides insights into the mechanisms of this response, and it could contribute to our understanding of cadmium toxicity in humans.
Antioxidants improve β-cypermethrin degradation by alleviating oxidative damage and increasing bioavailability by Bacillus cereus GW-01

Abstract:Microbial degradation of pesticide residues has the potential to reduce their hazards to human and environmental health. However, in some cases, degradation can activate pesticides, making them more toxic to microbes. Here we report on the β-cypermethrin (β-CY) toxicity to Bacillus cereus GW-01, a recently described β-CY degrader, and effects of antioxidants on β-CY degradation. GW-01 exposed to β-CY negatively affected the growth rate. The highest maximum specific growth rate (μm) appeared at 25 mg/L β-CY. β-CY induced the oxidative stress in GW-01. The activities of superoxide dismutase (SOD), catalyse (CAT), and glutathione-S-transferase (GST) were significantly higher than that in control (p < 0.01); but they are decreased as growth phase pronged, which is contrary to the β-CY degradation by GW-01 cells obtaining from various growth phase. Ascorbic acid (Vc), tea polyphenols (TP), and adenosine monophosphate (AMP) improved the degradation through changing the physiological property of GW-01. TP and AMP prompted the expression of gene encoding β-CY degradation in GW-01, while Vc does the opposite. Biofilm formation was significantly inhibited by β-CY, while was significantly enhanced by certain concentrations of TP and AMP (p < 0.05); while cell surface hydrophobicity (CSH) was negatively associated with β-CY concentrations from 25 to 100 mg/L, and these 4 antioxidants all boosted the CSH. Cells grown with β-CY had lower levels of saturated fatty acids but increased levels of some unsaturated and branched fatty acids, and these antioxidants alleviated the FA composition changes and gene expression related with FA metabolism. We also mined transcriptome analyses at lag, logarithmic, and stationary phases, and found that β-CY induced oxidative stress. The objective of this study was to elaborate characteristics in relation to the microbial resistance of pesticide poisoning and the efficiency of pesticide degradation, and to provide a promising method for improving pesticide degradation by microbes.

Abstract:Heavy metal pollutants are important abiotic environmental factors affecting pest habitats. In this study, Cd pre-exposure significantly increased the tolerance of Lymantria dispar larvae to β-cypermethrin, but did not significantly alter their tolerance to λ-cyhalothrin and bifenthrin. The activation of P450 by Cd exposure is the key mechanism that induces insecticide cross-tolerance in L. dispar larvae. Both before and after β-cypermethrin treatment, Cd exposure significantly increased the expression of CYP6AB224 and CYP6AB226 in L. dispar larvae. Silencing CYP6AB224 and CYP6AB226 reduced the tolerance of Cd-treated L. dispar larvae to β-cypermethrin. Transgenic CYP6AB224 and CYP6AB226 genes significantly increased the tolerance of Drosophila and Sf 9 cells to β-cypermethrin, and the recombinant proteins of both genes could significantly metabolise β-cypermethrin. Cd exposure significantly increased the expression of CnCC and Maf . CnCC was found to be a key transcription factor regulating CYP6AB224 - and CYP6AB226 -activated insecticide cross-tolerance in Cd-treated larvae. Decreasing reactive oxygen species (ROS) levels in the Cd-treated larvae or increasing ROS levels in the untreated larvae reduced or enhanced the expression of CnCC , CYP6AB224 and CYP6AB226 and β-cypermethrin tolerance in L. dispar larvae, respectively. Collectively, Cd exposure confers β-cypermethrin tolerance in L. dispar larvae through the ROS/ CnCC signalling pathway-mediated P450 detoxification.

Abstract:A variety of coordinated host-cell responses are activated as defense mechanisms against pore-forming toxins (PFTs). Bacillus thuringiensis (Bt) is a worldwide used biopesticide whose efficacy and precise application methods limits its use to replace synthetic pesticides in agricultural settings. Here, we analyzed the intestinal defense mechanisms of two lepidopteran insect pests after intoxication with sublethal dose of Bt PFTs to find out potential functional genes. We show that larval intestinal epithelium was initially damaged by the PFTs and that larval survival was observed after intestinal epithelium regeneration. Further analyses showed that the intestinal regeneration caused by Cry9A protein is regulated through c-Jun NH (2) terminal kinase (JNK) and Janus tyrosine kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathways. JAK/STAT signaling regulates intestinal regeneration through proliferation and differentiation of intestinal stem cells to defend three different Bt proteins including Cry9A, Cry1F or Vip3A in both insect pests, Chilo suppressalis and Spodoptera frugiperda . Consequently, a nano-biopesticide was designed to improve pesticidal efficacy based on the combination of Stat double stranded RNA (dsRNA)-nanoparticles and Bt strain. This formulation controlled insect pests with better effect suggesting its potential use to reduce the use of synthetic pesticides in agricultural settings for pest control. Bacillus thuringiensis (Bt) is the most successful bacteria to control insect pests through microbial pesticides and genetical modified crops. Its insecticidal proteins form pores on the midgut leading to target larvae death. However, the defense response of target insect pests is still unclear during this process. Here, we found that larvae intestine undergoes remodeling after intoxication with sublethal dose of Bt toxins. During the remodeling, the intestinal regeneration regulated by JNK and JAK/STAT signaling was induced after epithelial cells damaged by toxins. Surprisingly, this regulation is independent of EGFR pathway and unpaired gene that usually function in defense response in Drosophila . Finally, based on the findings, a mixture of dsRNA-nanoparticles and Bt bacterial strains was designed to improve pesticidal efficacy.

Effects and Mechanisms of Beta-cypermethrin on the Activity of Glutamine Synthetase in Mice Brain

Abstract:Objective To explore the effects and mechanisms of pyrethroids on excitatory neurotoxicity in mammal. Methods According to their body weights, healthy adult mice were randomly divided into five groups with 10 in each (5 males and 5 females). Four groups of mice were administrated by gastro-gavage with 0, 20, 40 and 80 mg/kg of beta-cypermethrin, diluted with salad oil respectively. Another group of mice was immediately given 300 mg/kg of reduced glutathione(GSH)by intraperitoneal injection after administration with 80 mg/kg of beta-cypermethrin- Three hours after administration, the activity and the mRNA expression of glutamine synthetase(GS) in brains were determined by colorimetric assay and RT-PCR, respectively. Results Two hours after administration, some excitotoxic symptoms were observed in the group of 40 and 80 mg/kg with or without GSH intervention, and the toxic symptoms were alleviated in mice by GSH intervention. There were no obvious symptoms in the group of 20 mg/kg. With the increasing of administrated dosage, the activity of GS in mice brain decreased. Statistical treatment showed that the GS activity of mice brain in groups of 40 mg/kg and 80 mg/kg were remarkably different with the control group (P<0. 001, P<0.01). The GS activity of mice brain in the intervention group was higher than that in the group of 80 rng/kg, however, the difference was not significant(P>0. 05). In addition, GS mRNA levels had no remarkable difference (P > 0. 05 ) among each group. Conclnsions The suppression of GS activity in mice brain induced by beta-cypermethrin was not mediated by GS gene transcriptional regulation. Toxic symptoms were alleviated obviously in mice, but the GS activity was not significantly improved by exogenetic GSH treatment.

Abstract:Dysdercus koenigii is a major global pest of cotton that causes severe economic loss. Among several control measures, pyrethroids are frequently used toxicants because of high efficacy at low dosages and relative safety. Since haemocytes are biomarkers of the physiological response and immunity of insect which determine the insecticide efficacy, the current study assessed the effect of a pyrethroid, β-cyfluthrin, on the total and differential haemocyte counts of D. koenigii. Haemolymph was collected from the fifth instars after the topical application of β-cyfluthrin (0.8, 1.6, 3.2, 6.4 and 12.8 mg/L) on the thoracic tergum. The haemolymph of control nymphs revealed 5270 haemocytes/mm3 which decreased instantly by 1.4-3.1-fold on β-cyfluthrin exposure; more reduction observed at lower dosages. Increase in exposure duration and β-cyfluthrin dosages fluctuated the count considerably, eventually raising them at lower dosages and diminishing at higher dosages. Among five kinds of haemocytes recorded in the haemolymph, the β-cyfluthrin exposure increased %prohaemocytes count; diminished %granulocytes and %plasmatocytes count while spherulocyte and oenocyte counts were inconsistent. The alterations in haemocyte counts indicate the immunity response trigger in D. koenigii due to β-cyfluthrin-induced stress. Further investigations may decipher the mechanisms involved and help to formulate the strategies for its management in fields.
