Abstract:Improving the sensitivity of the fluorescence method is crucial in biosensor fields. In this study, an ultrasensitive and excellent selectivity method was developed for the detection of trace Cu2+ ions using 3-mercaptopropionic acid capped CdSSe/CdS/ZnS quantum dots (MPA-CdSSe/CdS/ZnS QDs) as a fluorescent probe. Through energy band engineering to optimize the interior inorganic structure and passivate the surface defects, the photoluminescence quantum yield of the MPA-CdSSe/CdS/ZnS QDs in aqueous solution had as high as 70 %. The X-ray diffraction (XRD) and Transmission Electron Microscope (TEM) analysis revealed the prepared CdSSe/CdS/ZnS QDs were cubic zinc blende structure with average diameter 7.2 nm. Further, the fluorescence of the MPA-CdSSe/CdS/ZnS QDs was significantly quenched by the Cu2+ with fast response time through an efficient photo-induced electron transfer (PET) progress. By optimizing the thickness of CdS shell, the MPA-CdSSe/CdS/ZnS QDs probe with appropriate CdS shell thickness exhibited excellent linear linearity and sensitivity for the detection of Cu2+ ions with limit of detection as low as 0.05 nM. Additionally, ions interference experiments demonstrated the good selectivity of the probe towards Cu2+. Furthermore, this sensing system could also be applied to the measurement of Cu2+ ions in real water samples with satisfactory recoveries. This work provided new insights to design and synthesize high-performance QDs-based fluorescence probes with controllable exciton emission behavior for their applications in biosensor and biological fields.
Highly sensitive determination of copper in HeLa cell using capillary electrophoresis combined with a simple cell extraction treatment.

Abstract:A new separation system of capillary electrophoresis (CE1) for the highly sensitive determination of copper was established by using ethylenediaminetetraacetic acid (EDTA) as a complexing agent and employing cetyltrimethylammonium chloride (CTAC) as a capillary inner wall modifier. Benefitted from the combination of field-enhanced sample injection (FESI) method, a limit of detection (LOD) of 2.7nM was obtained, which was much lower than that of the conventional methods. This made it possible to determine trace copper in HeLa cell only by a simple cell extraction (CE2) treatment. Two copper-extraction methods—acid-hydrolysis and freeze–thaw—were compared. Limited by the requirement of low ion strength in FESI, only the extract using freeze–thaw could be successfully applied in the determination. The effectiveness assessment of this CE2–FESI method was adopted by inductively coupled plasma-atomic emission spectrometry (ICP-AES) as a gold standard.

Abstract:A rapid and novel two-step dispersive liquid-liquid microextraction and dispersive micro-solid phase extraction method was established for the separation and enrichment of trace cadmium, nickel, and copper in food and environmental water prior to determination by inductively coupled plasma-mass spectrometry (ICP-MS). In the first microextraction step, carbon tetrachloride was employed to extract metal-diethyldithiocarbamate chelates from aqueous solution with ultrasound. In the following step, Fe3O4 magnetic nanoparticles were added and used to collect the analytes in the organic solvent. The sample pH, type and volume of extraction solvent, mass of magnetic nanoparticles, concentration of the chelating agent, concentration of sodium chloride, and sonication time were optimized. The linear dynamic range was from 0.01 to 20 micrograms per liter with correlation coefficients between 0.9990 and 0.9992. Enrichment factors were 78, 79, and 81 for cadmium, nickel, and copper, respectively. The limits of detection for cadmium, nickel, and copper were 0.007, 0.009, and 0.017 micrograms per liter, with relative standard deviations from 1.1 to 2.6 percent. The developed method was validated by the determination of cadmium, nickel, and copper in rice and mussel certified reference materials, food, and environmental water with satisfactory results.

Abstract:Here, we present a simple platform for the use of the enhanced emission of 16-mercaptohexadecanoic acid (16-MHA) capped CdSe quantum dots (QDs) as a probe for ultrasensitive copper(II) detection. In this study, the photoluminescence (PL) of the QDs was first enhanced by Ag nanoprisms which were self-assembled on Si surfaces and then further increased by photobrightening. Using this approach, the control and different analytes could be readily probed all on a single platform using fluorescence microscopy. The enhanced PL intensity of CdSe QDs was selectively quenched in the presence of Cu2+, accompanied by the emergence of a new red-shifted luminescence band. The quenching mechanism was found to be due to a cation exchange mechanism as confirmed by X-ray photoelectron spectroscopy (XPS) measurements. Herein, we have demonstrated that this simple methodology can offer a rapid and reliable detection of Cu2+ with a detection limit as low as 5 nM and a dynamic range up to 100, mu M in a fixed fast reaction time of 5 min. The potential applications of this technique were tested in two ways, for mixed-ion solutions and in physiological fluids, and both experiments exhibited good selectivity toward Cu2+.

Abstract:Copper is an essential element for life and constitutes copper proteins, and attends life activities in human body. However, an excess of copper is poisonous, and can cause hepatitis, gastroenteritis and pneumonia. copper enters into biosphere from ores by weathering and erosion and exists in water in the form of a divalent copper ion. Therefore, it is necessary to detect copper ions in an ultrasensitive and quantitative way. Herein, a dual-functional DNAzymatic visual biosensor was developed, optimized and validated to detect divalent copper ions sensitively and selectively in tap water. In the method, dual DNAzymes with different functions were designed within one biosensor. One DNAzyme was Cu2+-dependent cleaving DNAzyme, which could be cleaved by Cu2+ for signal recognition. The other one was G-quadruplex DNAzyme, which was induced by terminal deoxynucleotidyl transferase (TdT) for "one-to-multiplex" signal amplification and output. In the middle of two steps, the exponential amplification reaction (EXPAR) was applied to amplify the recognitive signal exponentially. Under optimized conditions, Cu(2+ c)ould be selectively observed with the naked eyes with the detection limit (LOD) of 10 pmol/L and with the microplate reader with LOD of 7.6 pmol/L in about 2 h. Furthermore, Cu2+ in the concentration range of 10 pmol/L-200 pmol/L in tap water samples could be discriminated by naked eyes. The platform provides an ultrasensitive and colorimetric detective tool for the analysis of trace Cu2+ in tap water with high specificity, and will have broad application prospect in many fields.
Stacking and Simultaneous Determination of Estrogens in Water Samples by Ce with Electrochemical Detection

Abstract:A rapid and sensitive method based on transient ITP and field enhancement in CE with electrochemical detection at copper disk electrode was developed for the simultaneous separation and determination of three estrogens: estrone, 17β‐estradiol, and estriol. The effects of several important factors that influence the separation and detection were investigated. Under the optimum conditions, the estrogens could be separated in 0.06 mol/L sodium hydroxide solution within 14 min. With transient ITP by addition of 0.5% NaCl, a good linear response was obtained for three estrogens from 0.2 to 10 μmol/L, with correlation coefficients higher than 0.9993. The detection limits were 8.9 × 10−8, 6.7 × 10−8, and 1.1 × 10−7 mol/L (S/N = 3) for estriol, 17β‐estradiol, and estrone, respectively. This method was successfully employed to analyze different water samples from waterworks, tap water, fishpond, and river samples with recoveries in the range of 90.8–108.9%, and RSDs < 4.69%. The satisfied results demonstrated that this method was of convenient preparation, high sensitivity, and good repeatability, which could be applied to the rapid determination of environmental water samples.

Abstract:Field-amplified sample injection of copper(II) was investigated using capillary electrophoresis with chemiluminescence detection. The sensitivity of copper(II) has been improved markedly by the field-amplified sample injection technique and the detection limit reaches 2 x 10(-11) m. By injection of a short plug of water before sample introduction, the sensitivity can be further improved 5-fold and the detection limit reaches 4 x 10(-12) m. The relative standard deviations (n = 6) of the migration time and the peak height are 0.61% and 4.7% at 1.0 x 10(-9) M Cu(II), respectively. Parameters affecting the field-amplified sample injection, such as separation voltage and concentration of electrophoretic buffer, have been investigated.

Abstract:This paper reports a fluorescent turn-off assay for sensitive detection of Cu(2+) in an aqueous solution by using a copper-specific ssDNA Cu100 and Sybr Green I. By monitoring the fluorescence changes arose from different interactions of Sybr Green I with Cu100 and Cu100/Cu(2+) complex, the Cu(2+) could be linearly detected from 5.57 to 250 ppb, with a detection limit of 5.57 ppb. The feasibility of this assay was demonstrated by detecting Cu(2+) in certified reference materials and spiked water samples with satisfactory results.

Abstract:A highly sensitive and accurate dual-signal strategy is developed for trace Cu2+ detection based on the inner filter effect (IFE) between Tween 20-gold nanoparticles (AuNPs) and CdSe/ZnS quantum dots (QDs). Tween 20-AuNPs are utilized as colorimetric probes and excellent fluorescent absorbers. The fluorescence of CdSe/ZnS QDs can be quenched efficiently by Tween 20-AuNPs via IFE. In the presence of D-penicillamine, D-penicillamine induces the aggregation of Tween 20-AuNPs and the fluorescent recovery of CdSe/ZnS QDs at high ionic strength. Upon addition of Cu2+, D-penicillamine tends to selectively chelate with Cu2+ and then forms the mixed-valence complexes, which consequently inhibits the aggregation of Tween 20-AuNPs and the fluorescent recovery. The dual-signal method is used to quantitatively detect trace Cu2+, with low detection limits of 0.57 μg/L and 0.36 μg/L for colorimetry and fluorescence, respectively. In addition, the proposed method using a portable spectrometer is applied to the detection of Cu2+ in water. This sensitive, accurate and miniature sensing system has potential in environmental evaluations.

Abstract:Abstract The extensive usage and production of copper may lead to toxic effects in organisms due to its accumulation in the environment. Traditional methods for copper detection are time consuming and infeasible for field usage. It is necessary to discover a real-time, rapid and economical method for detecting copper to ensure human health and environmental safety. Here we developed a colorimetric paper strip method and optimized spectrum method for rapid detection of copper ion based on the specific copper chelator bathocuproinedisulfonic acid disodium salt (BCS). Both biological assays and chemical methods verified the specificity of BCS for copper. The optimized reaction conditions were 50 mM Tris–HCl pH 7.4, 200 μM BCS, 1 mM ascorbate and less than 50 μM copper. The detection limit of the copper paper strip test was 0.5 mg/L by direct visual observation and the detection time was less than 1 min. The detection results of grape, peach, apple, spinach and cabbage by the optimized spectrum method were 0.91 μg/g, 0.87 μg/g, 0.19 μg/g, 1.37 μg/g and 0.39 μg/g, respectively. The paper strip assays showed that the copper contents of grape, peach, apple, spinach and cabbage were 0.8 mg/L, 0.9 mg/L, 0.2 mg/L, 1.3 mg/L and 0.5 mg/L, respectively. These results correlated well with those determined by inductively coupled plasma-mass spectrometry (ICP-MS). The visual detection limit of the paper strip based on Cu-BCS-AgNPs was 0.06 mg/L. Our study demonstrates the potential for on-site, rapid and cost-effective copper monitoring of foods and the environment.

Abstract:The authors describe a silicon nanoparticle-based fluorometric method for sensitive and selective detection of Cu2+. It is based on the catalytic action of Cu2+ on the oxidation of cysteine (Cys) by oxygen to form cystine and the by-product H2O2. The generated H2O2 is catalytically decomposed by Cu2+ to generate hydroxyl radicals which oxidize and destroy the surface of SiNPs. As a result, the blue fluorescence of the SiNPs is quenched. The method has excellent selectivity due to the dual catalytic effects of Cu2+, which is much better than most previously reported nanomaterial-based assays for Cu2+. Under the optimal conditions, the method has low detection limit (29 nM) and a linear response in a concentration range from 0.05 μM to 15 μM. The method has been successfully applied to the determination of Cu2+ in spiked real water samples, and the results agreed well with those obtained by the Chinese National Standard method (GB/T 7475-1987; AAS).

Abstract:In this study, we designed a system of integrating gold nanoparticles and cysteamine to detect Cu (2+) based on a colorimetric strategy. The quantified limit of detection towards Cu2+ is 0.4 mu M, which is similar to 50 times lower than the maximum contamination level (similar to 20 mu M) in drinking water permitted by US environmental protection agency. The practicality of this system was further validated by detecting Cu2+ in different water samples.

Abstract:A fluorescent probe was synthesized and demonstrated to be highly selective and sensitive in the reaction with copper (II) ion, generating a large variation of the fluorescence intensity in a dose–response manner. The probe contains a dansyl moiety as fluorophore and a multidentate ligand for copper (II) ion recognition. The reaction of the molecular probe with copper (II) ion proceeds rapidly and irreversibly in a 1 to 1 stoichiometric way, leading to the production of stable copper (II) complex, which subsequently results in the quenching of fluorescence. The detection limit for copper (II) ion was measured to be about 2ppb. It was also shown that the probe has high selectivity for copper (II) ion and good anti-interference ability against other transition metal ions. The herein reported very simple and reliable fluorescence probe could be employed for copper (II) ion detection in many aspects.

Abstract:Although a variety of whole-cell biosensors and biosorbents have been developed for detection and removal of heavy metal contaminants, few whole cells can be applied to both monitoring and remediation of copper pollution in water. In this study, a modified plasmid was constructed by incorporating a copper-sensing element and a copper-adsorbing element into a temperature-inducible plasmid, pBV220. This plasmid was subsequently transformed into an engineered Escherichia coli strain lacking copA and cueO. This dual-functional E. coli cell selectively responded to copper ions with a linear detection range of 0.01–25 μM at 37 °C and could express surface-displayed CueR when treated at 42 °C without any costly chemical inducers. The display of CueR on the cell surface specifically enhanced its copper adsorption capacity and rapidly removed copper ions from aqueous solutions. In addition, the CueR surface-displayed cells could be regenerated by adsorption-desorption cycles via pH regulation. Moreover, by simply using two different temperatures, the detection or adsorption of copper using this dual-functional whole cell was achieved without any cross-interference. Most importantly, it provided highly sensitive, accurate quantification, and effective removal of copper in real environmental water samples. Thus, this E. coli cell can be used for large-scale detection and remediation of copper pollutants.
Sensitive detection and intracellular imaging of free copper ions based on DNA-templated silver nanoclusters aggregation-inducing fluorescence enhancement effect

Abstract:Free copper ions (Cu+ and Cu2+) have critical toxicity to cells, although copper is an essential element for human body. Hence, sensitive monitoring is crucial to avoid over intake of Cu+/Cu2+. We herein designed a ssDNA sequence (A31) for synthetizing A31-templated silver nanoclusters (AgNCs), and demonstrated that Cu+/Cu2+ can induce the aggregation of A31-templated AgNCs and thus greatly enhanced the fluorescence emission of A31-templated AgNCs. Based on Cu+/Cu2+-induced fluorescence enhancement effect of A31-templated AgNCs, a label-free and signal-on fluorescent sensing platform was developed for the specific and sensitive detection of Cu+/Cu2+ in biological samples and intracellular imaging of Cu+/Cu2+ in cells. The signal-on fluorescent sensing platform could be used to rapidly detect Cu+ and Cu2+ with a detection limit of 0.1 µM within 30 min., and to perform the intracellular imaging of Cu+ and Cu2+ in cells with good cell permeability and biocompatibility. By using the signal-on fluorescent sensing platform, we have successfully detected Cu+ and Cu2+ in cells fluids and human serum with a recovery of 90-104% and a RSD (n = 5) < 5%, and performed the imaging of Cu+/Cu2+ in Hela cells. The developed fluorescent sensing platform has obvious analytical and imaging advantages such as signal-on, simple operation, short analysis time, both Cu+ and Cu2+ detection, similar or higher sensitivity, good cell permeability and biocompatibility, which promising a reliable approach for the rapid and on-site detection or imaging of free copper ions in biological samples in clinical diagnosis.
Determination of Trace Copper Ions with Ultrahigh Sensitivity and Selectivity Utilizing Cdte Quantum Dots Coupled with Enzyme Inhibition

Abstract:A fluorescent transducer with the combination of the unique property of CdTe quantum dots (QDs) and enzymatic inhibition assays was successfully constructed for the purpose of ultrasensitive determination of Cu2+ ions. Alcohol oxidase (AO) catalyzed the oxidation of methanol to produce hydrogen peroxide (H2O2), inducing the quenching of QDs fluorescence. In the presence of Cu2+ ions, the activity of AO was inhibited and therefore, the quenching of QDs fluorescence was decreased. Other metal ions showed no intensive inhibition to the AO activity even at 10 or 100 times Cu2+ ions concentration, presenting a high selectivity of this fluorescent sensor. Using this QDs–enzyme hybrid system, the detection limit for Cu2+ ions was found to be as low as 0.176ng/mL (2.75nM) due to the superior fluorescence property of QDs. Practical application of the QDs–enzyme hybrid system has been demonstrated by domestic waste water, agricultural irrigation water and lake water analysis. Results of Cu2+ determinations were in good agreement with those obtained by inductively coupled plasma analytical method. The coupling of efficient quenching of QDs photoluminescence by H2O2 generated from oxidase-catalyzed reaction and the effective enzymatic inhibition make this a simple and sensitive method for heavy metal ions detection.

Abstract:Availability of highly sensitive assays for metal ions can help monitor and manage the environmental and food contamination. In the present study, a monoclonal antibody against Copper(II)–ethylenediaminetetraacetic acid was used to develop two sensitive ELISAs for Cu(II) analysis. Cobalt(II)–EDTA–BSA was the coating antigen in a heterologous indirect competitive ELISA (hicELISA), whereas Co(II)–EDTA–BSA–horseradish peroxidase (HRP) was the enzyme tracer in a heterologous direct competitive ELISA (hdcELISA). Both ELISAs were validated for detecting the content of Cu(II) in environmental waters. The ELISA data agreed well with those from graphite furnace atomic absorption spectroscopy. The methods of developing the Cu(II) hicELISA and hdcELISA are potentially applicable for developing ELISAs for other metals. The chelator–protein complexes such as EDTA–BSA and EDTA–BSA–HRP can form a suite of metal complexes having the consistent hapten density, location and orientation on the conjugates except the difference of the metal core, which can be used as ideal reagents to investigate the relationship between assay sensitivity and antibody affinities for the haptens and the analytes. The strategy of conjugating a haptenated protein directly with HRP can reduce the loss of HRP activity during the conjugation reaction and thus can be applicable for the development of ELISAs for small molecules.
Simultaneous Determination of Copper(II) and Cobalt(II) by Ion Chromatography Coupled with Chemiluminescent Detection

Abstract:In the absence of any special luminescent reagents, a weakly chemiluminescent emission was observed during the decomposition of hydrogen peroxide, catalyzed by transition-metal ions, such as Cu(II) and Co(II), in basic aqueous solution. The chemiluminescent intensity was significantly enhanced by the addition of ethyldimethylcetylammonium bromide and uranine. The signal-to-noise ratio ( S / N ) was proportional to the concentrations of Cu(II) and Co(II). Based on these phenomena, a flow-injection chemiluminescent method for the simultaneous separation and determination of Cu(II) and Co(II) was developed. The detection limits of the present chemiluminescent method for Cu(II) and Co(II) were 7. 5 and 0. 01 ng/ml, respectively. After ion chromatographic separation of Cu(II) and Co(II) by an IonPac CS5A column with oxalic acid and lithium hydroxide monohydrate as the eluent, the present chemiluminescent system was used as a post-column detector for these two transition metal ions in natural water samples.
An array of femtoliter wells for sensitive detection of copper using click chemistry

Abstract:Sensitive detection of copper ion (Cu2+), which is of great importance for environmental pollution and human health, is crucial. In this study, we present a highly sensitive method for measuring Cu2+ in an array of femtoliter wells. In brief, magnetic beads (MBs) modified with alkyne groups were bound to the azide groups of biotin-PEG3-azide (bio-PEG-N3) via Cu+-catalyzed click chemistry. Cu+ in the click chemistry reaction was generated by reducing Cu2+ with sodium ascorbate. Following the ligation, the surface of the MBs was modified with biotin, which could be labeled with streptavidin-β-galactosidase (SβG). The MBs complex was then suspended in β-galactosidase substrate fluorescein-di-β-d-galactopyranoside (FDG), and loaded into the array of femtoliter wells. The MBs sank into the wells due to gravity, and the resulting fluorescent product, generated from the reaction between SβG on the surface of the MBs and FDG, was confined within the wells. The number of fluorescent wells increased with higher Cu2+ concentrations. The bright-field and fluorescent images of the wells were acquired using an inverted fluorescent microscope. The detection limit of this assay for Cu2+ was 1 nM without signal amplification, which was 103 times lower than that of traditional fluorescence detection assays.

Abstract:In this work, ultrasound-assisted cloud point extraction (UA-CPE) and ultrasound-assisted dispersive liquid liquid microextraction (UA-DLLME) were investigated and compared firstly as ultrasound-assisted liquid phase microextraction methods, which were coupled with spectrophotometer for copper preconcentration and detection. Compared to conventional CPE and DLLME, the extraction patterns were changed and improved by the effect of ultrasound. As novel methods, their applications were expanded and the analytical performance of spectrophotometric determination for copper was considerably improved. The influence factors of UA-CPE and UA-DLLME were studied in detail. Under the optimal conditions, the limits of detection (LODs) for copper were 0.7 mu g L-1 of UA-CPE and 0.8 mu g L-1 of UA-DLLME with sensitivity enhancement factors (EFs) of 17 and 16. The developed methods were applied to the determination of trace copper in real water samples with satisfactory analytical results. (C) 2015 Elsevier B.V. All rights reserved.
