Abstract:We describe the direct synthesis of organophosphorus compounds from ubiquitous aryl and vinyl carboxylic acids via decarbonylative palladium catalysis. The catalytic system shows excellent scope and tolerates a wide range of functional groups (>50 examples). The utility of this powerful methodology is highlighted in the late-stage derivatization directly exploiting the presence of the prevalent carboxylic acid functional group. DFT studies provided insight into the origin of high bond activation selectivity and P(O)-H isomerization pathway.

Abstract:Aryl carboxylic acids are among the most abundant substrates in chemical synthesis and represent a perfect example of a traceless directing group that is central to many processes in the preparation of pharmaceuticals, natural products and polymers. Herein, we describe a highly selective method for the direct step-down reduction of carboxylic acids to arenes, proceeding via well-defined Pd(0)/(ii) catalytic cycle. The method shows a remarkably broad substrate scope, enabling to direct the classical acyl reduction towards selective decarbonylation by a redox-neutral mechanism. The utility of this reaction is highlighted in the direct defunctionalization of pharmaceuticals and natural products, and further emphasized in a range of traceless processes using removable carboxylic acids under mild, redox-neutral conditions orthogonal to protodecarboxylation. Extensive DFT computations were conducted to demonstrate preferred selectivity for the reversible oxidative addition and indicated that a versatile hydrogen atom transfer (HAT) pathway is operable.

Abstract:Decarbonylative borylation of carboxylic acids is reported. Carbon electrophiles are generated directly after reagent-enabled decarbonylation of the in situ accessible sterically-hindered acyl derivative of a carboxylic acid under catalyst controlled conditions. The scope and the potential impact of this method are demonstrated in the selective borylation of a variety of aromatics (>50 examples). This strategy was used in the late-stage derivatization of pharmaceuticals and natural products. Computations reveal the mechanistic details of the unprecedented C-O bond activation of carboxylic acids. By circumventing the challenging decarboxylation, this strategy provides a general synthetic platform to access arylpalladium species for a wide array of bond formations from abundant carboxylic acids. The study shows a powerful combination of experiment and computation to predict decarbonylation selectivity.

Abstract:Enabled by merging iridium photoredox catalysis and palladium catalysis, α-oxocarboxylate salts can be decarboxylatively coupled with aryl halides to generate aromatic ketones and amides at room temperature. DFT calculations suggest that this reaction proceeds through a Pd(0) -Pd(II) -Pd(III) pathway, in which the Pd(III) intermediate is responsible for reoxidizing Ir(II) to complete the Ir(III) -*Ir(III) -Ir(II) photoredox cycle.

Abstract:Cooperative bimetallic catalysis is a fundamental approach in modern synthetic chemistry. We report bimetallic cooperative catalysis for the direct decarbonylative heteroarylation of ubiquitous carboxylic acids via acyl C-O/C-H coupling. This novel catalytic system exploits the cooperative action of a copper catalyst and a palladium catalyst in decarbonylation, which enables highly chemoselective synthesis of important heterobiaryl motifs through the coupling of carboxylic acids with heteroarenes in the absence of prefunctionalization or directing groups. This cooperative decarbonylative method uses common carboxylic acids and shows a remarkably broad substrate scope (>70 examples), including late-stage modification of pharmaceuticals and streamlined synthesis of bioactive agents. Extensive mechanistic and computational studies were conducted to gain insight into the mechanism of the reaction. The key step involves intersection of the two catalytic cycles via transmetallation of the copper-aryl species with the palladium(II) intermediate generated by oxidative addition/decarbonylation.

Abstract:Formic acid is efficiently used as a C1 source to directly carboxylate allylic alcohols in the presence of a low loading of palladium catalyst and acetic anhydride as additive to afford β,γ‐unsaturated carboxylic acids with excellent chemo‐, regio‐, and stereoselectivity. The reaction proceeds through a carbonylation process with in situ‐generated carbon monoxide under mild conditions, avoiding the use of high‐pressure gaseous CO. A bisphosphine ligand with a large bite angle (4,5‐bis{diphenylphosphino}‐9,9‐dimethylxanthene, Xantphos) was found to be uniquely effective for this transformation. The regio‐ and stereoconvergence of this reaction is ascribed to the thermodynamically favored isomerization of the allylic electrophile in the presence of the palladium catalyst.

Abstract:The direct transformation of aryl carboxylic acids to aryl nitrile compounds is an interesting topic because carboxylic acids are not only abundant in nature but are also inexpensive and stable. Here, the synthesis of a series of aryl nitriles by palladium-catalyzed decarbonylative cyanation of carboxylic acids without base has been achieved. The successful decarbonylative cyanation of drug molecules and Gram-scale reaction to verify the practicality and operability of this method are analyzed.

Abstract:In this study, we demonstrate that phosphinoacridines are efficient bidentate ligands for palladium‐catalyzed carboxylative C‐N coupling reactions under blue LED irradiation. This method facilitates the direct synthesis of arylcarbamates using a range of non‐activated aryl halides, such as iodides and bromides, with various amines under atmospheric pressure of CO2. The optimized conditions exhibit high tolerance to sensitive functional groups, resulting in very good to excellent yields of the desired products. This approach expands the scope of palladium‐catalyzed carboxylation reactions and offers an environmentally friendly route to arylcarbamates.

Formate-Mediated Aryl Halide Reductive Cross-Coupling Catalyzed by Iodide-Bridged Palladium(I) Dimers: Experi-mental and Computational Studies

Abstract:The first efficient reductive cross-couplings of aryl halides mediated by an abundant feedstock, sodium formate, are de-scribed. These processes, which exploit air-stable Pd(I) iodide dimers, are especially effective for challenging 2-pyridyl systems due to intervention of chelated intermediates. Furthermore, orthogonality with respect to Suzuki and Buchwald-Hartwig coupling processes is displayed, as pinacol boronates and anilines are tolerated. Although palladium-catalyzed transfer hydrogenolyses of aryl halides mediated by formate are longstanding, it is not a major competing pathway under the present conditions. Experimental and computational studies corroborate a novel catalytic cycle for cross-coupling where the Pd(I) precatalyst, [Pd(I)(PtBu3)]2, is converted to the active dianionic catalyst, [Pd2I4][NBu4]2, from which aryl halide oxi-dative addition is more facile. Rapid, reversible Pd-to-Pd transmetalation delivers iodide-bridged diarylpalladium dimers. The hetero-diarylpalladium dimers are more stable than the homodimers and have lower barriers to reductive elimination, resulting in high cross-selectivity.

Abstract:A decarboxylative allylic benzylation cocatalyzed by Pd/photoredox in a regio- and enantioselective manner has been achieved. Readily available aryl acetic acids are used as benzylic nucleophile equivalents without preactivation. This mild and atom-economical protocol expands the scope of coupling partners of allylic electrophiles. Vinyl epoxides could also go through this transformation smoothly, affording various chiral homoallylic alcohols bearing all-carbon quaternary stereocenters.

Abstract:Aryl aldehydes are key synthetic intermediates in the manufacturing of active pharmaceutical ingredients. They are generated on scale (>1000 kg) through the palladium-catalysed formylation of aryl bromides using syngas (CO:H2). The best-in-class catalyst system for this reaction employs di-1-adamantyl-n-butylphosphine (cataCXium A), palladium(II) acetate, and tetramethylethylenediamine. Despite nearly 20 years since its initial report, a mechanistic understanding of this system remains incomplete. Here we use automation, kinetic analysis, and DFT calculations to develop a new mechanistic model for this best-in-class catalyst. We show that a combination of the migratory insertion step and dihydrogen activation step are likely involved in the turnover-limiting sequence. The reaction kinetics are responsive to the nature of the substrate, with electron-rich aryl bromides reacting faster and more selectively than their electron-poor counterparts due to the influence of electronics in the migratory insertion step. Our findings overturn the current paradigm and provide new mechanistic insight into palladium-catalysed formylation of aryl bromides.

Abstract:An efficient palladium-catalyzed decarboxylative ortho-acylation of 2-aryloxypyridines with -oxocarboxylic acids is described. In this new transformation, the aromatic C(sp2)H bond was successfully acylated to give diverse aromatic ketones regioselectively in moderate to good yields. The pyridine group can be removed easily after the acylation to give the corresponding 2-hydroxy aromatic ketones.

Abstract:A practical, efficient Pd(OAc)(2)/Cu(II)/PPh3 cocatalytic system has been developed. With only 1 mol% Pd(OAc)(2), 20 mol% copper(II) salt, and 0.5 equiv of inexpensive PPh3 as ligand, the direct arylation of benzoazoles with aryl bromides could be performed smoothly in mild condition, affording the desired arylated benzoazoles in good yields (75-93%).

Abstract:Metal-catalyzed alkene arylalkoxylation is a powerful complexity-building strategy for the synthesis of oxygen heterocycles from simple γ-unsaturated alcohols, but only a few examples of catalytic enantioselective methods exist. Herein, an efficient palladium-catalyzed enantioselective arylalkoxylation of γ-hydroxyalkenes with aryl halides is reported. The salient features of this transformation include a remarkable broad substrate scope, mild reaction conditions, and good functional group tolerance, delivering a series of chiral tetrahydrofurans containing a tertiary or quaternary stereocenter in good yields with up to 95% ee. The Xu10 ligand with a suitable side-arm was responsible for the high reactivity and good enantioselectivity of this transformation.

Abstract:Palladium-catalyzed cyanation of aryl halides and pseudo halides with potassium hexacyanoferrate is described employing the hemilabile, bulky, and electron-rich MOP-type ligands. When the mixture of t-BuOH and H2O was used as the solvent and K2CO3 as the base, the MOP-type ligands showed high efficiency for the palladium-catalyzed cyanation. The effect of ligand structure was studied in detail, and 2-di-tert-butylphosphino-2-isopropoxy-1,1-binaphthyl was the more effective for the cyanation. The catalyst system allows the cyanation of unactivated aryl chlorides, and even aryl mesylates to occur in good yields. Furthermore, the reactivity of different arylated reagents in the catalytic system was found to be: ArBr > ArCl >> ArOMs > ArOSO(2)Im > ArOSO2NMe2.

Abstract:Generation of alkenes through decarboxyolefination of alkane carboxylates has significant synthetic value in view of the easy availability of a variety of carboxylic acids and the synthetic versatility of alkenes. Herein we report that palladium catalysts under irradiation with blue LEDs (440 nm) catalyze decarboxylative desaturation of a variety of aliphatic carboxylates to generate aliphatic alkenes, styrenes, enol ethers, enamides, and peptide enamides under mild conditions. The selection of a dual phosphine ligand system is the key enabler for the successful development of this reaction. The Pd-catalyzed decarboxylative desaturation is utilized to achieve a three-step divergent synthesis of Chondriamide A and Chondriamide C in overall 68% yield from simple starting materials. Mechanistic studies suggest that, distinct from palladium catalysis under thermal condition, irradiation-induced palladium catalysis involves irradiation-induced single-electron transfer and dynamic ligand-dissociation/association process to allow two phosphine ligand to work synergistically.

Abstract:Although nucleophilic benzylation-type reaction to introduce various aromatic systems into molecules has been widely explored, the related pyrrolylmethylation version remains to be disclosed. Reported herein is a palladium-catalysed multiple auto-tandem reaction between N-Ts propargylamines, allyl carbonates and aldimines in the presence of an acid, proceeding through sequential allylic amination, cycloisomerisation, vinylogous addition and aromatisation steps. A diversity of formal pyrrolylmethylated amine products were finally furnished efficiently. In addition, switchable regiodivergent 3-pyrrolylmethylation and 4-pyrrolylmethylation were realised by tuning catalytic conditions. Moreover, remote chirality transfer with readily available enantioenriched starting materials was well achieved with an achiral ligand, relying on diastereoselective generation of η2-Pd(0) complexes between Pd(0) and chiral 1,3-diene intermediates in the key vinylogous addition step. A few control experiments was conducted to elucidate the palladium-involved tandem reaction and regiodivergent synthesis.

Abstract:Palladium-catalyzed carbonylation reactions are efficient methods for synthesizing valuable molecules. However, realizing a carbonylation with excellent yield and chemo-, regio-, and enantioselectivities by classical low-valent palladium catalysis is highly challenging. Herein, we describe an enantioselective carbonylation reaction using a high-valent palladium catalysis strategy and employing a chiral sulfoxide phosphine (SOP) ligand. This double aminocarbonylation reaction begins with the formation of a carbamoylpalladium(II) species, which undergoes enantioselective oxidative addition with a cyclic diaryliodonium salt to generate a palladium(IV) intermediate, followed by a second CO insertion and reductive elimination. The mechanism has been illustrated with experimental and computational studies.
