Abstract:Bis(difluoroboron)-1,2-bis[(1H-pyrrol-2-yl)methylene]hydrazine (BOPHY) is a new class of highly fluorescent organoboron compound. The new symmetric structural motif, the unusual absorption and emission properties, and better photo stability have attracted considerable attentions for the development of various functional chromophores. In the present work, a novel BOPHY derivative (BOPHY-NNBOH) for the selective sensing of Cu2+ was synthesized. BOPHY-NNBOH shows very weak fluorescence. However, it displays significant fluorescence enhancement in the presence of Cu2+. The method is sensitive and selective, and sensing of Cu2+ ions in HeLa cells is also demonstrated. To our knowledge, the present work is the first application of BOPHY derivatives in bioanalysis. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:Two N-alkylated diketopyrrolopyrrole derivatives, namely, FDPP-C8 and TDPP-C8, were prepared and applied in the ratiometric and fluorescent detection of Cu2+ in acetonitrile. The distinct optical changes of two probes were attributed to the fast radical process of diketopyrrolopyrrole triggered by Cu2+. This reaction mechanism was investigated by NMR, MS and EPR experiments and DFT theoretical calculations. The detection of Cu2+ with FDPP-C8 and TDPP-C8 were completed instantaneously, making them convenient for practical real-time operation. The limits of detection of FDPP-C8 and TDPP-C8 for Cu2+ were calculated as 65 and 127 nM, respectively. Relevant competitive ions exerted no effect. In addition, the intrinsic two-photon absorption properties of FDPP-C8 were also investigated and successfully used in the detection of Cu2+. Furthermore, given that Cu2+/Cu+-involved radical process is solvent dependent, TDPP-C8 was used to construct a INHIBIT logic gate at the molecular level with DMF and Cu2+ as inputs.

Abstract:Based on boron-dipyrromethene (BODIPY), taking 2-hydroxy-N-(2-hydroxyphenyl)benzamide as recognition site, a new fluorescent probe HHPBA-BODIPY aimed at sensitively detecting Cu ions was designed, synthesized and characterized.The emission spectra of HHPBA-BODIPY exhibited an intensive green fluorescence around 510 nm, with a maximum absorption near 500 nm. When Cu2+ ions are present, the fluorescence at 510 nm can be quenched with a good linearity between the copper ion concentrationand the fluorescence intensity and the detection limit is 0.35 μM. HHPBA-BODIPY is also selective toward Cu2+, while other metal ions show no interfere except Fe3+ and Cr3+ ions. In addition, HHPBA-BODIPY also proved efficient to detect Cu2+ in water samples which offers the possibility to detect trace amount of Cu2+ for environmental monitoring. Abstract:A BODIPY-based turn-on fluorescent probe 1 has been designed and synthesized for fluorescence detection of Cu2+ in aqueous media and imaging in living cells. The CN bonds are selectively hydrolyzed by the reaction with Cu2+ to generate formyl-BODIPY. The favorable features of probe 1 towards Cu2+ include significant fluorescence enhancement (over 290-fold), high selectivity and low detection limit (0.1 μM).

Abstract:A novel dual-function fluorescent probe PPH was conveniently synthesized by linking pyrazinehydrazide unit and pyrenyl fluorophore. The present probe PPH could be used for simultaneous detection of ClO− and Cu2+ through a fluorescence turn-on response. The probe PPH displayed excellent selectivity and sensitivity towards ClO− and Cu2+ over other anions and metal ions. The detection limits of PPH for ClO− and Cu2+ were calculated to be as low as 4.02 nM and 157 nM, respectively. The probe PPH had good pH stability and was highly responsive to ClO− and Cu2+ in a wide pH range. Furthermore, this probe could be readily applied to visualize ClO− and Cu2+ on the test paper. Additionally, this probe was successfully utilized to the fluorescence imaging of ClO− and Cu2+ in HeLa cells.

Abstract:A new probe PQ comprising quinoline as a fluorophore and 2-picolinic group as a recognition unit is designed and synthesized with high sensitivity and selectivity toward copper ion. The probe PQ shows almost no fluorescence in HEPES buffer (20 mM, pH 7.0), but displays obvious fluorescence enhancement after the addition of Cu2+, this response is considered to be attributed to the Cu2+-promoted hydrolysis of PQ to yield a highly fluorescent product, 2-methyl-7-hydroxy-quinoline with fluorescence signal greatly amplified. The probe PQ exhibits remarkable selectivity and sensitivity toward Cu2+ and the detection limit is as low as 0.15 p,M. These results reveal that PQ has the potential to discriminate Cu2+ over other cations in neat aqueous solution by fluorescence turn-on response. Abstract:Fluorescent probes are intriguing material for ion detection. In this study, 4,4-difluoro-4-bora3a,4a-diaza-s-indacene (BODIPY) containing a dipicolylethylenediamine unit was developed as a colorimetric and fluorescence "turn-off" probe for Cu 2+ . The probe exhibited higher selectivity for Cu 2+ than other common metal ions with a detection limit of 8.49 μM. With increasing Cu 2+ concentration, the probe showed a red-shift in the absorption spectrum as well as fluorescence quenching, possibly due to the intramolecular charge transfer effect of the probe–Cu(II) complex. Furthermore, the probe was used for imaging Cu 2+ in living cells based on confocal fluorescence imaging. The results show that the probe is an effective tool for detection copper ions.

Abstract:Here, we report a new Cu2+-selective fluorescent turn-on probe BODIPY-EP, in which the 2-pyridinecarboxylic acid is connected to a 6-hydroxyindole-based BODIPY platform through an ester linkage. The ester bond of BODIPY-EP is selectively hydrolyzed by the reaction with Cu2+ under mild and neutral conditions to generate BODIPY-OH, showing strong characteristic fluorescence of BODIPY-OH. The favorable features of BODIPY-EP towards Cu2+ include fast response, large fluorescence enhancement and high selectivity. We further demonstrated that the membrane-permeable probe reacts with intracellular Cu2+ and exhibits bright fluorescence in living cells.

Abstract:A turn-on fluorescent probe 1 has been developed for Cu2+ by the incorporation of picolinate onto a tricyanofuran-based fluorophore. The probe was weakly fluorescent due to the masked phenolic group and the resulted inhibition of the push-pull character of the fluorophore. The chemoselective reaction of probe I with Cu2+ leads to a dramatic fluorescence enhancement at 610 nm via the Cu2+-mediated hydrolysis of picolinate moiety and the subsequent release of the highly emissive fluorophore. This probe features high selectivity for Cu2+ over other cations, as well as high sensitivity with a low detection limit of 54 nM. Moreover, cell cytotoxicity and imaging tests indicate that probe 1 is biocompatible and can be utilized for imaging Cu2+ in live cells. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:A near-infrared distyryl boron dipyrromethene-based sensor bearing one bis(1,2,3-triazole)amino receptor has been synthesized. This probe selectively and quickly binds to Hg2+ and Cu2+ ions in CH3CN/H2O (5:1 v/v) and exhibits remarkably blue-shifted absorption and fluorescence bands due to the inhibition of the intramolecular charge transfer process. The fluorescence changes of this probe upon binding to Hg2+ or Cu2+ ion are totally different, undergoing a ratiometric fluorescence enhancement (for Hg2+) or a fluorescence quenching (for Cu2+) mechanism. The corresponding vivid color changes can be easily seen by the naked eye. This probe was further introduced into Hela cells for living cell imaging and found to discriminate Hg2+ and Cu2+ ions through two near-infrared fluorescence emission channels. These overall results indicate that this Click-derived near-infrared BODIPY-based probe is potentially useful for ratiometric and discriminative detection of Hg2+ and Cu2+ ions in solutions and living cells.

Abstract:Two fluorescent chemosensors for the detection of Cu2+ were designed, synthesized, and fully characterized. These sensor molecules bear a 9-cycloheptatrienyl fluorene (9-CHF) unit as the Cu2+ receptor and a boron-dipyrromethene (BODIPY) unit as the fluorophore. Both of these two sensors present high selectivity to Cu2+ and high sensitivity with acidity independence. Moreover, they exhibit turn-on fluorescence, which should be more practical in the design of the fluorescent chemosensor for detecting Cu2+.

Abstract:A new thiophenyl pyrazoline probe for Cu2+ in aqueous solution was synthesized and characterized by IR, NMR, HRMS and X-ray analysis. Abstract:Based on a boron dipyrromethene (BODIPY) derivative containing an N, O and S tridentate ligand, a Cu2+ fluorescent probe BTCu was developed. The detection mechanism was verified as Cu2+-promoted oxidative dehydrogenation of an amine moiety, leading to a formation of a fluorescent Cu+–Schiff base complex. Free BTCu exhibited a maximum absorption wavelength at 496nm, and a very weak maximum emission at 511nm. Upon addition of various metals ions, it showed large fluorescence enhancement toward Cu2+ (417-fold in MeCN and 103-fold in MeCN/HEPES solution, respectively) with high selectivity. The detection limits are as low as 1.74×10−8M and 4.96×10−8M in the two different solutions, respectively. And BTCu could work in a wide pH range with an extraordinary low pKa of 1.21±0.06. Using fluorescence microscopy, the probe was shown to be capable of penetrating into living cells and imaging intracellular Cu2+ changes.

Abstract:A simple pyrazoline derivative BBD has been synthesized for detecting Cu2+ in EtOH/Hepes (V/V = 1:1, pH = 7). The probe has high selectivity for Cu2+ by quenching the fluorescence intensity, and was sensitive to pH. When excited at 398 nm, the fluorescence is emitted at 520 nm with a Stokes shift of 122 nm, which is larger than other reported pyrazoline skeleton probes. The detection limit (LOD) of the probe for Cu2+ is 0.862 μM which is lower than the value of WHO limit (31.5μM) in water. The 2:1 binding mode and probable recognition mechanism of the probe with Cu2+ were confirmed by Job's plot, IR, ESI-MS, and TD-DFT analysis. Moreover, the probe has been successfully applied to actual water samples as well as cell and zebrafish imaging.

The fluorescence intensity of DHP (10 μM) at 522 nm is well linearized with the Cu2+ concentration at 0-5 μM, and it has a detection limit of 0.29 nM (R2 = 0.9949). The complexation ratio of the probe DHP to Cu2+ is 2 : 1 and the complexation constant is 3.44 × 104 M-1 (R2 = 0.9974). In addition, the probe DHP can be recovered using EDTA and Cu2+ can be effectively monitored at pH 5-11, with good results in dipstick experiments and actual water samples. HepG-2 cells remained viable in excess of 90% after being exposed to DHP (50 μM) for 24 h, which demonstrates the extremely low toxicity of DHP, and it can be used for in vivo cell imaging.

Abstract:Herein, we report a highly selective fluorescent probe for the detection of Cu( II ). The detection mechanism relies on the Cu( II )-catalyzed oxidative hydroxylation of 2-(aminocarbonyl)phenylboronic acid into salicylamide, thus recovering the excited-state intramolecular proton transfer (ESIPT) effect and inducing more than 35-fold fluorescence enhancement. The simple structure and readily available fluorescent probe give a novel method for quantitatively detecting Cu( II ) in the linear range of 0–22 μM, with a limit of detection down to 68 nM, and exhibiting high selectivity for Cu( II ) over 16 other metal ions.

Abstract:A naphthalimide Schiff base fluorescent probe (BSS) was designed and synthesized from 4-bromo-1,8-naphthalic anhydride, and its structure was characterized by 1 HNMR, 13 CNMR, FTIR, and MS. Fluorescence emission spectra showed that probe BSS could realize the "turn-off" detection of Cu 2+ in acetonitrile solution, detection process with strong specificity and excellent anti-interference of other metal ions. In the fluorescence titration experiments, fluorescence intensity of BSS showed a good linear relationship with the Cu 2+ concentration (0–10 μmol/L), and the detection limit was up to 7.0 × 10 − 8 mol/L. Meanwhile, BSS and Cu 2+ could form a 1:1 complex (BSS-Cu 2+ ) during the reaction process. Under the same detection conditions, complex BSS-Cu 2+ had specific fluorescence recovery properties for H 2 PO 4 − and the whole process was not only fast (6 s) but also free of interference from other anions, with a detection limit was as low as 5.7 × 10 − 8 mol/L. In addition, complex BSS-Cu 2+ could be successfully applied to the detection of H 2 PO 4 − in actual water samples, which with excellent application prospects.
