Abstract:Polyglycerol nanogels are three-dimensional polymeric networks with a few hundred nanometer sizes and the ability to encapsulate and deliver cargos for a wide range of biomedical applications. However, time-consuming and multistep synthetic routes as well as milligram-scale production have hindered further development of these nanomaterials. In this work, we report on a straightforward synthetic method for the production of polyglycerol nanoarchitectures. Enzymatic ring-opening copolymerization of a mixture of glycidol and succinic anhydride resulted in polyglycerol nanogels with succinic acid segments in their backbone. Novozyme 435 was used as a dual catalytic agent to support ring-opening polymerization of the above-mentioned cyclic monomers as well as esterification of the produced oligomers to obtain nanogels. While succinic acid segments improved the biodegradability and loading capacity of nanogels, polyglycerol caused water solubility, high functionality, and biocompatibility. Nanogels were loaded with tacrolimus and photosensitizer 5,10,15,20-tetrakis(3-hydroxyphenyl)porphyrin (mTHPP)-a close congener of the approved photosensitizer temoporfin (mTHPC)-and their ability to improve the skin penetration of these therapeutic agents was investigated. mTHPP delivery experiments on human skin, which were quantified by fluorescence microscopy, showed that these nanogels deposit in the stratum corneum and release the loaded drug to viable epidermis of skin efficiently in comparison with commercially available base cream. Taking advantage of the straightforward synthesis as well as biodegradability, biocompatibility, high loading capacity, and efficient skin penetration, the synthesized nanogels could be used as future topical delivery systems.

Abstract:The synthesis of a new kind of amphiphilic poly(vinyl-co-ester)s via simple one-pot radical ring-opening polymerization (RROP) is reported. A series of biodegradable, biocompatible, and cross-linkable copolymers P(MDO-co-PEGMA-co-CMA) with different compositions were synthesized by copolymerization of 2-methylene-1,3-dioxepane (MDO), poly(ethylene glycol) methyl ether methacrylate (PEGMA) and 7-(2-methacryloyloxyethoxy)-4-methylcoumarin methacrylate (CMA). All the copolymers were well characterized by H-1 NMR, C-13 NMR, and gel permeation chromatography (GPC). The hydrolytic degradation and enzymatic degradation of the copolymers were investigated. The amphiphilic P(MDO-co-PEGMA-co-CMA) can self-assemble into micelles in aqueous solution. Polymeric micelles with different sizes were obtained through tuning the compositions of the copolymers, i.e., the hydrophilic/hydrophobic ratio. The size and morphology of the micelles were measured by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Because of the coumarin side groups in the micellar core, the micelles can be cross-linked upon exposure to long-wavelength UV light (lambda = 365 nm). These micelles also showed excellent biocompatibility by the in vitro cytotoxicity experiments. Furthermore, the biodegradable micelles were applied for the delivery of an anticancer drug doxorubicin (DOX). The cell viability of DOX-loading micelles compared to free DOX was discussed. The effects of cross-linking and the copolymer composition on the drug release behavior were well studied. The results indicate that this new kind of amphiphilic poly(vinyl-co-ester) s could serve as promising nanocarriers for the delivery of anticancer drugs.

Abstract:The synthesis of a new PEG based hyperbranched copolymer of poly(ethylene glycol) methyl ether methacrylate-co-ethylene glycol dimethacrylate (PEGMEMA-co-EGDMA) was achieved via a one-step in situ deactivation enhanced atom transfer radical polymerization (DE-ATRP). Then, hollow PEG based nanospheres were fabricated from this polymer using a solvent evaporation method and post-stabilisation strategy. Furthermore, the analysis using a cellular metabolic activity assay proved that the copolymer did not affect cellular metabolism, indicating that this PEG based polymeric nanosphere has potential for use in drug delivery applications.

Abstract:A new class of non-ionic amphiphiles have been synthesised using a combination of polyethylene glycol (PEG) and oligoglycerol dendrons as hydrophilic units and an alkoxy aryl moiety as hydrophobic unit. The resulting amphiphiles were found to aggregate in aqueous medium. Their aggregation behaviour was studied using dynamic light scattering (DLS), fluorescence spectroscopy, and cryogenic electron microscopy (cryo-TEM). The inner hydrophobic core of these aggregates in aqueous medium is capable of encapsulating lipophilic guest molecules. The encapsulation behaviour was studied using Nile red as a hydrophobic dye as well as Curcumin and Dexamethasone as hydrophobic drug candidates. Furthermore, for biological evaluation, cytotoxicity and cellular uptake was studied using different cancer cell lines. The biomedical application of synthesised amphiphiles was further investigated for dermal drug delivery on excised human skin using Nile red encapsulated in the nanocarrier. The release profile of drug/dye encapsulated amphiphiles was studied under physiochemical conditions in the presence of immobilized lipase Novozym 435.

Abstract:Enzymatic polymerization of glycerol‐based polyesters has emerged as a selective and sustainable strategy for the production of degradable, amphiphilic, PEG‐free polymeric materials. In the present work, we exploited the ability to produce copolyester variants of poly(glycerol adipate) (PGA), by adding a second functionalized diol in a one‐pot fashion. The produced glycerol‐based copolyesters, with a variety of amphiphilic balances and chemical functionalities along the backbone, have been screened as nanocarriers for drug delivery. The stability of the produced nanoparticles (NPs) was tested in relevant biological fluids and after freeze‐drying treatment. The new variants outperformed PGA homopolymers in the penetration through a gel‐like barrier as well as in their efficiency to encapsulate a poorly water‐soluble model drug. All the tested polymeric NPs formulations have demonstrated biocompatibility in both in vitro and in vivo experiments, that utilize Caenorhabditis elegans nematodes. Taken together, these preliminary results highlight the importance of introducing new chemistry along polyester backbones while tuning the amphiphilic nature of the final polymers to improve the performance of NP systems.

Abstract:Nowadays, the use of lipid-based nanocarriers for the targeted and controlled delivery of a variety of hydrophobic and hydrophilic bioactive-compounds and drugs has increased significantly. However, challenges such as thermodynamic instability, oxidation, and degradation of lipid membranes, as well as the unintended release of loaded compounds, have limited the use of these systems in the food and pharmaceutical industries. Therefore, the present study reviews the latest achievements in evaluating the characteristics, production methods, challenges, functional, and biological stabilization strategies of lipid-based carriers (including changes in formulation composition, structural modification, membrane-rigidity, and finally monolayer or multilayer coating with biopolymers) in different conditions, as well as molecular dynamics simulations. The scientists' findings indicate the effect of natural biopolymers (such as chitosan, calcium alginate, pectin, dextran, xanthan, caseins, gelatin, whey-proteins, zein, and etc.) in modifying the external structure of lipid-based carriers, improving thermodynamic stability and resistance of membranes to physicochemical and mechanical tensions. However, depending on the type of bioactive compound as well as the design and production goals of the delivery-system, selecting the appropriate biopolymer has a significant impact on the stability of vesicles and maintaining the bioaccessibility of the loaded-compounds due to the stresses caused by the storage-conditions, formulation, processing and gastrointestinal tract.
Supramolecular nanogels fabricated via host-guest molecular recognition as penetration enhancer for dermal drug delivery

Abstract:Nanogels that are assembled by supramolecular interactions as compared to covalent crosslinked nanogels, exhibit new functionalities with potential for easy processability, recycling and self-healing due to the nature of dynamic and reversible non-covalent interactions. Here we design a supramolecular polymer nanogel that utilize host-guest interactions between the groups pillar [5] arene and alkyl chains on hyperbranched polyglycerol backbone as crosslinking agents for a new dermal drug delivery system. The anti-inflammatory drug Dexamethasone (Dexa) can be efficiently loaded into the nanogels and released from the assemblies. Besides, the supramolecular polymer nanogels exhibit better drug loading capacity and skin penetration enhancement than the individual host polymer and guest polymer. In vitro skin permeation studies show that supramolecular polymer nanogels can improve the Nile red penetration through the skin by up to 9 fold, compared to the individual polymers or a conventional cream formulation on a barrier deficient skin model.
Polyglycerol‐Functionalized β‐Cyclodextrins as Crosslinkers in Thermoresponsive Nanogels for the Enhanced Dermal Penetration of Hydrophobic Drugs

Abstract:Enhancing the dermal penetration of insoluble drugs remains a challenge. To address this, β‐CD are functionalized with hyperbranched polyglycerol and are further used as crosslinkers to fabricate thermoresponsive nanogels based on N‐isopropylacrylamide (NIPAM). It is found that a higher NIPAM content and a moderate acrylation degree of the crosslinker benefitted the dermal penetration of a hydrophobic model cargo. Thermoresponsive nanogels (tNGs) are promising candidates for dermal drug delivery. However, poor incorporation of hydrophobic drugs into hydrophilic tNGs limits the therapeutic efficiency. To address this challenge, β‐cyclodextrins (β‐CD) are functionalized by hyperbranched polyglycerol serving as crosslinkers (hPG‐βCD) to fabricate βCD‐tNGs. This novel construct exhibits augmented encapsulation of hydrophobic drugs, shows the appropriate thermal response to dermal administration, and enhances the dermal penetration of payloads. The structural influences on the encapsulation capacity of βCD‐tNGs for hydrophobic drugs are analyzed, while concurrently retaining their efficacy as skin penetration enhancers. Various synthetic parameters are considered, encompassing the acrylation degree and molecular weight of hPG‐βCD, as well as the monomer composition of βCD‐tNGs. The outcome reveals that βCD‐tNGs substantially enhance the aqueous solubility of Nile Red elevating to 120 μg mL−1 and augmenting its dermal penetration up to 3.33 μg cm−2. Notably, the acrylation degree of hPG‐βCD plays a significant role in dermal drug penetration, primarily attributed to the impact on the rigidity and hydrophilicity of βCD‐tNGs. Taken together, the introduction of the functionalized β‐CD as the crosslinker in tNGs presents a novel avenue to enhance the efficacy of hydrophobic drugs in dermatological applications, thereby offering promising opportunities for boosted therapeutic outcomes.

Abstract:We here present the synthesis and characterization of a set of biodegradable core⁻multishell (CMS) nanocarriers. The CMS nanocarrier structure consists of hyperbranched polyglycerol (hPG) as core material, a hydrophobic (12, 15, 18, 19, and 36 C-atoms) inner and a polyethylene glycol monomethyl ether (mPEG) outer shell that were conjugated by ester bonds only to reduce the toxicity of metabolites. The loading capacities (LC) of the drugs, dexamethasone and tacrolimus, and the aggregate formation, phase transitions, and degradation kinetics were determined. The intermediate inner shell length (C15) system had the best overall performance with good LCs for both drugs as well as a promising degradation and release kinetics, which are of interest for dermal delivery.

Abstract:Due to the low cutaneous bioavailability of tacrolimus (TAC), penetration enhancers are used to improve its penetration into the skin. However, poor loading capacity, non-biodegradability, toxicity, and in some cases inefficient skin penetration are challenging issues that hamper their applications for the dermal TAC delivery. Here we present poly(lactide-co-glycerol) (PLG) as a water soluble, biodegradable, and biocompatible TAC-carrier with high loading capacity (14.5% w/w for TAC) and high drug delivery efficiencies into the skin. PLG was synthesized by cationic ring-opening copolymerization of a mixture of glycidol and lactide and showed 35 nm and 300 nm average sizes in aqueous solutions before and after loading of TAC, respectively. Delivery experiments on human skin, quantified by fluorescence microscopy and LC-MS/MS, showed a high ability for PLG to deposit Nile red and TAC into the stratum corneum and viable epidermis of skin in comparison with Protopic® (0.03% w/w, TAC ointment). The cutaneous distribution profile of delivered TAC proved that 80%, 16%, and 4% of the cutaneous drug level was deposited in the stratum corneum, viable epidermis, and upper dermis, respectively. TAC delivered by PLG was able to efficiently decrease the IL-2 and TSLP expressions in human skin models. Taking advantage of the excellent physicochemical and biological properties of PLG, it can be used for efficient dermal TAC delivery and potential treatment of inflammatory skin diseases.

Abstract:In a century when environmental pollution is a major issue, polymers issued from bio-based monomers have gained important interest, as they are expected to be environment-friendly, and biocompatible, with non-toxic degradation products. In parallel, hyperbranched polymers have emerged as an easily accessible alternative to dendrimers with numerous potential applications. Glycerol (Gly) is a natural, low-cost, trifunctional monomer, with a production expected to grow significantly, and thus an excellent candidate for the synthesis of hyperbranched polyesters for pharmaceutical and biomedical applications. In the present article, we review the synthesis, properties, and applications of glycerol polyesters of aliphatic dicarboxylic acids (from succinic to sebacic acids) as well as the copolymers of glycerol or hyperbranched polyglycerol with poly(lactic acid) and poly(ε-caprolactone). Emphasis was given to summarize the synthetic procedures (monomer molar ratio, used catalysts, temperatures, etc.,) and their effect on the molecular weight, solubility, and thermal and mechanical properties of the prepared hyperbranched polymers. Their applications in pharmaceutical technology as drug carries and in biomedical applications focusing on regenerative medicine are highlighted.

Abstract:Biocompatible amphiphilic nanoparticles (NPs) with tunable particle morphology and surface property are important for their applications as functional materials. However, previously developed methods to prepare amphiphilic NPs generally involve several steps, especially an additional step for surface modification, greatly hindering their largescale production and widespread applications. Here, a versatile one-step strategy is developed to prepare biocompatible amphiphilic dimer NPs with tunable particle morphology and surface property. The amphiphilic dimer NPs, which consist of a hydrophobic shellac bulb and a hydrophilic poly(lactic acid) (PLA) bulb with PLA-poly(ethylene glycol) (PEG) on the bulb surface, are prepared in a single step by controlled co-precipitation and self-assembly. Amphiphilic PLA-PEG/shellac dimer NPs demonstrate excellent tunability in particle morphology, thus showing good performances in controlling the interfacial curvature and emulsion type. In addition, temperature-responsive PLA-poly(N-isopropyl acrylamide) (PNIPAM)/shellac dimer NPs are prepared following the same method and emulsions stabilized by them show temperature-triggered response. The applications of PLA-PEG-folic acid (FA)/shellac dimer NPs for drug delivery have also been demonstrated, which show a very good performance. The strategy of preparing the dimer NPs is green, scalable, facile and versatile, which provides a good platform for the design of dimer NPs with tunable particle morphology and surface property for diverse applications.

Abstract:This research is planned to synthesize and characterize interpenetrating polymer network (IPN) nanogels by combining natural (β-cyclodextrin) and synthetic (Polyvinyl alcohol) polymers by free radical polymerization utilizing N,N-methylenebisacrylic acid (MBA) and 2-acrylamido-2-methylpropane sulfonic acid (AMPS). FTIR, DSC, and TGA were employed to characterize the thermal stability of components and the conception of cross-linked networks. The porosity and amorphousness of the structure were shown using SEM and XRD. The entrapment efficiency of prepared nanogels was found to be more than 70%. The enormous increase in rosuvastatin solubility compared to the usual formulation led to a significant development in the drug's release profile and released more than 90% of the drug within 5–30 minutes. As compared to the commercially available tablet (Rovista®), triglyceride and total cholesterol levels were significantly lower in hyperlipidemic rats treated with RST-loaded nanogel. Therefore, the atherosclerosis index of nanogels demonstrated a considerable improvement in pharmacodynamic effectiveness in animals. GRAPHICAL

Abstract:Poly (DL-lactic acid-co-glycolic acid)-co-poly-(ethylene glycol) was synthesized by bulk ring-opening polymerization of DL-lactide/glycolide/poly(ethylene glycol) using stannous chloride as an initiator. The molecular structure of the copolymer was analyzed by IR, H-1 NMR, and DSC. The degradation behavior of copolymer was assayed by the reduction of molecular weight, the loss-in-mass, and the changes of pH value for degradation medium. The different contents of PGA and PEG in the molecules of the copolymer could control the degradation rate of polymer. Human Serum Albumin (HSA) was chosen as the model hydrophilic drug and encapsulated in the copolymer. Abstract:Biodegradable polycarbonate-based ABA triblock copolymers were synthesized via organocatalyzed ring-opening polymerization and successfully formulated into chemically cross-linked hydrogels by strain-promoted alkyne-azide cycloaddition (SPAAC). The synthesis and cross-linking of these polymers are copper-free, thereby eliminating the concern over metallic contaminants for biomedical applications. Gelation occurs rapidly within a span of 60 s by simple mixing of the azide- and cyclooctyne-functionalized polymer solutions. The resultant hydrogels exhibited pronounced shear-thinning behavior and could be easily dispensed through a 22G hypodermic needle. To demonstrate the usefulness of these gels as a drug delivery matrix, doxorubicin (DOX)-loaded micelles prepared using catechol-functionalized polycarbonate copolymers were incorporated into the polymer solutions to eventually form micelle/hydrogel composites. Notably, the drug release rate from the hydrogels was significantly more gradual compared to the solution formulation. DOX release from the micelle/hydrogel composites could be sustained for 1 week, while the release from the micelle solution was completed rapidly within 6 h of incubation. Cellular uptake of the released DOX from the micelle/hydrogel composites was observed at 3 h of incubation of human breast cancer MDA-MB-231 cells. A blank hydrogel containing PEG-(Cat)12 micelles showed almost negligible toxicity on MDA-MB-231cells where cell viability remained high at >80% after treatment. When the cells were treated with the DOX-loaded micelle/hydrogel composites, there was a drastic reduction in cell viability with only 25% of cells surviving the treatment. In all, this study introduces a simple method of formulating hydrogel materials with incorporated micelles for drug delivery applications.

Abstract:In the present study, semi-interpenetrating polymer networks (semi-IPNs) were synthesized based on crosslinked acrylic acid (AA)/xanthan gum (XG) biopolymer in the presence of N, N'-hexane-1, 6-dilbisprop-2-enamide (MS) or 1,4-butandioldimethacrylate (BDOD) as the cross-linking agent. MS is a novel acrylic-urethane diene monomer prepared through the condensation reaction between AA and hexamethylene diisocyanate (HDI). Scanning electron microscopy (SEM), Fourier transform infrared (FT-IR), proton nuclear magnetic resonance spectroscopy (1H NMR), X-ray diffraction (XRD) and thermogravimetric (TGA) analyses were used to study the morphology, structure and thermal stability of MS and semi-IPNs. The effect of crosslinking agent type on different behaviors such as morphology, stability, swelling, and water-retention capabilities of the synthesized hydrogels were investigated. XG-PAA semi-IPNs exhibited a very high adsorption potential and stability. Hydrogel biocompatibility was confirmed by the outcomes of MTT assay and cell staining. We recommend XG-PAA semi-IPNs as an environmentally benign and readily non-toxic material with an excellent adsorption capacity for application in drug delivery systems, wound healing and dye removal.

Abstract:Application of degradable polymers for pharmaceutical formulations is an appropriate strategy to increase the efficacy and reduce the side effects of drugs. Methylprednisolone is a glucocorticoid used in the treatment of a wide range of inflammatory and immune disorders. Due to high rate of clearance of methylprednisolone from body, frequent administration is required which can increase its undesired effects. Therefore, the aim of this study was to prepare methylprednisolone loaded polymeric nanoparticles incorporating fatty acid based polyamide, to improve its efficacy and reduce the side effects. In this study, a new fatty acid based polyamide with ideal characteristics for application in pharmaceutical dosage forms was synthesized. Palmitic acid, a saturated fatty acid, a derivative of palm oil, was the main constituent of the polymer, which improved its degradability, flexibility, and hydrophobicity. Palmitic acid was functionalized by tris(2-aminoethyl)amine, and then polymerized by application of sebacoyl chloride as a cross linker. The structure of the polymer was confirmed by FTIR and 1HNMR spectroscopy. Cytotoxicity of the polymer was tested on WRL68cell line using cell proliferation (MTT) assay, which showed low toxicity profile of the polymer. To study the type of erosion, polymeric devices were prepared by melt casting method by using aluminium mold. The polymer had about 55% and 40% wet and dry weight loss respectively within 7 days. The scanning electron micrographs showed surface erosion of the polymer. Drug loaded polymeric nanoparticles (PNP) were prepared by using thin film hydration method. Drug loaded PNPs with a particles size of 60–300 nm and entrapment efficiency of 76% were achieved.

Abstract:New biobased hyperbranched polymers were designed and synthesized from transgenic soybean oil by using dicumyl peroxide (DCP) or sulfur as reaction reagents. The structures of the samples and the reaction mechanisms were confirmed and investigated by Fourier transform infrared spectroscopy and nuclear magnetic resonance methods. The gel permeation chromatography (GPC) results showed that the molecular weight of the fully soybean-oil-based polymers could be easily adjusted by controlling the amounts of the reaction reagents rather than vary the reaction time. The oligomers and decomposition products of DCP in the DCP-initiated polymers could be completely removed by using a solution–precipitation method, and finally, several purified fully soybean-oil-based hyperbranched polymers with high molecular weights were obtained. This work makes a great contribution to the sustainability of hyperbranched polymers.
Amphiphilic Co-polypeptides Self-Assembled into Spherical Nanoparticles for Dermal Drug Delivery

Abstract:Poly(l-glutamic acid)-b-poly(Nγ-acetyl-l-2,4-diaminobutyric acid), P(Glu-b-NADA), amphiphilic block-co-polypeptides with different hydrophobic/hydrophilic ratios were synthesized as drug carriers for dermal delivery. The block-co-polypeptides were prepared using ring-opening polymerization (ROP) of γ-benzyl l-glutamate N-carboxyanhydride and polycondensation of the activated urethane derivative of Nγ-acetyl-l-2,4-diaminobutyric acid. Structures and conjugations of two blocks were successfully confirmed by 1H NMR, FTIR, DOSY NMR, GPC, and TGA. The nanocarriers were loaded with 5,10,15,20-tetrakis(3-hydroxyphenyl)porphyrin (m-THPP), a close congener of the approved photosensitizer 5,10,15,20-tetrakis(3-hydroxyphenyl)chlorin (m-THPC, temoporfin), and the loading capacity was found to be dependent on the composition of the block-co-polypeptides. Among the synthesized polymers, P(Glu55-b-NADA20) with moderate hydrophobic content showed the highest drug loading capacity of 4 wt % and self-aggregated into spherical nanoparticles with a size of 180–200 nm, which was confirmed by TEM and DLS. In an in vitro drug release study, P(Glu55-b-NADA20) could release m-THPP in a controllable manner. Furthermore, the synthesized polymer P(Glu55-b-NADA20) did not show high toxicity against HaCaT and HeLa cells up to 1000 and 500 μg mL–1, respectively, in an in vitro cell viability assay. Finally, it was shown in an ex vivo skin penetration study that the ionic amphiphilic block co-polypeptide enhanced the m-THPP penetration into human skin compared to base cream up to a factor of 12. m-THPP was released from P(Glu55-b-NADA20) to the viable epidermis while the polymer was deposited in the skin’s stratum corneum. Taking advantage of its excellent biodegradability, the low cytotoxcity, and efficient skin penetration, the synthesized block-co-polypeptide has the potential for future topical delivery systems.

Abstract:Biodegradable in situ thermo-sensitive hydrogels have potential applications in ophthalmic pharmaceutical preparations. Herein, a thermo-gel of PNICL-PEG-PNICL loaded with the natural compound epigallocatechin gallate (EGCG) used as a drug sustained release carrier was prepared by block bending strategy. Compared to mono polymers that cannot form a gel, mixing two block copolymers through a block blending strategy allows for the preparation of thermo-sensitive hydrogels. The temperature sensitive behavior of the system can be easily regulated by varying the blending ratios of the two polymers. Furthermore, the internal structure of the gel was investigated that indicated the system formed a permeable network through micelle aggregation. The biosafety of the system and its ability to sustainably release EGCG at 37 o C for 240 h were preliminarily demonstrated in vitro . This study has revealed the internal structural changes of block blending thermo-gels and tentatively justified their potential as drug carriers.
