Abstract:In order to develop stimuli-responsive hydrogel, chitosan graft copolymer with chitosan back-bone and poly(N-isopropylacrylamide)-block-poly(acrylic acid) (PNIPAAm-b-PAA) branch chains was prepared by reversible addition-fragmentation chain transfer (RAFT) polymerization in DMF. The chain transfer agent was obtained by modification of chitosan with 3-benzylsulfanyl thiocarbonylsulfanyl propionic acid (BPATT) with 68% the degree of substitute. The graft polymerizations possessed controlled/living characteristics. The behavior of the graft copolymer in aqueous solution was investigated by dynamic light scattering, transmission electron microscopy, and UV-visible spectrophotometer. N-Phthaloylchitosan-graft-(poly(N-isopropylacrylamide)-block-poly(acrylic acid)) copolymer (N-phthaloylchitosan-g-(PNIPAAm-b-PAA)) could assemble to micelles in aqueous solution in range of 200-300 nm with narrow size distribution, and the hydrodynamic diameter could be controlled dependent on length of branch chains and temperature. The LCST values of micelle could be modulated from 34 to 40 °C by controlling the constitution of branch chains, pH, and concentration.

Abstract:A new reversible addition−fragmentation chain transfer (RAFT) agent, biotinylated trithiocarbonate, was successfully synthesized. Temperature-responsive biotinylated polymer, poly(N-isopropylacrylamide) (PNIPAAm) and water-soluble poly(N-(2-hydroxypropyl)methacrylamide) (PHPMA), have been prepared in one-step via RAFT polymerization using biotinylated trithiocarbonate as the RAFT agent. The results show that the polymerizations of N-isopropylacrylamide (NIPAAm) and N-(2-hydroxypropyl)methacrylamide (HPMA) are of living characters, and the corresponding polymers produced have ending biotin group. Biotin-ended diblock copolymer of PHPMA and PNIPAAm was also prepared by RAFT polymerization of NIPAAm using biotinylated PHPMA as macro chain transfer agent. The biotinylated PHPMA-b-PNIPAAm can form a new kind of core−shell nanostructure with biotin groups on the surface reversibly induced by temperature.

Abstract:The pH-responsive amphiphilic block copolymer micelles based on poly (2-(dimethylamino) ethyl methacrylate) (PDMAEMA) as a flexible corona and poly(hexafluorobutyl acrylate) (PHFBA) as a core have been synthesized by reversible addition-fragmentation chain transfer polymerization induced self-assembly (RAFT-PISA). This core-shell type amphiphilic copolymer micelles possessed the characteristics of pH-response from pendent tertiary amine groups in PDMAEMA chain. The morphology and size of amphiphilic block copolymer micelles were measured by dynamic light scattering (DLS) and transmission electron microscopy (TEM) analyses. The results indicated that the diameter of micelles narrowed down from 160 nm to 20 nm along with the increased pH value from 1.32 to 2.90. However, the diameter of the amphiphilic block copolymer micelles ranged broadly from 20 nm to more than 100 nm at pH >= 5.89. The amphiphilic block copolymer micelles were later used as reactors to synthesize cationic fluorinated polyacrylate emulsifier-free emulsion at pH 2.90, using fluorinated monomers and acrylate monomers as mixed monomers. The latex particles were presented as core-shell morphology with diameter of about 220 nm and narrow distribution, as characterized by TEM and DLS. The obtained emulsion with high zeta potential (+57.8 mV) had excellent stability.

Abstract:Reversible addition–fragmentation chain transfer polymerization with a low-temperature initiator can be used to synthesize multifunctional block copolymers for polyplex formation. Utilizing three methacrylate monomers (triethylene glycol methyl ether methacrylate (MEO3MA), pyridyldisulfideethyl methacrylate (PDSM), N,N-dimethylethylenediamine methacrylate (DMAEMA)) and an azide bearing chain transfer agent for polymerization, the resulting triblock-copolymers possess two orthogonally reactive functionalities (a block with reactive disulfide side chains and an azide end-group) and two additional functional blocks for i) solubilization and ii) polyplex formation. Thereby, the MEO3MA block provides solubility in aqueous media and the DMAEMA block, having a tertiary amine in the side chain, allows polyplex formation with polyanionic biomacromolecules like DNA or RNA. Due to the reactive disulfide block the polyplexes can be (reversibly) crosslinked with dithiols. Because of the manifold of possibilities to modify the pDNApolyplexes, this polymers system is an interesting candidate for active-targeting and codelivery approaches.

Abstract:pH-Responsive polymers have attracted much attention for biotechnology applications as carriers or matrix to facilitate intracellular or extracellular therapeutic drug delivery and release. In this paper, we report the development of new pH-responsive and hyperbranched copolymers with potential for such applications. These pH-responsive hyperbranched copolymers were synthesized via one pot reversible addition-fragmentation chain transfer (RAFT) copolymerization of propylacrylic acid (PAA) and a branching co-monomer poly(ethylene glycol diacrylate) (PEGDA) (Mn=258 Da) at the monomer feed molar ratios [PAA]0/[PEGDA]0 = 99/1, 90/10 and 80/20. The resultant poly(PAA-PEGDA) copolymers were characterized by Proton Nuclear Magnetic Resonance (1H NMR) and Gel Permeation Chromatography (GPC) to obtain the molecular weight, copolymer composition and degree of acrylate functionality. The hydrodynamic dimensions of these copolymers at pH range between 5.0 and 7.4 were studied using Dynamic Light Scattering technique (DLS). Moreover, these hyperbranched copolymers demonstrated composition- and size-dependent membrane disruptive properties by red blood cell hemolysis assay. Poly(PAA-PEGDA) with the copolymer composition [PAA]/[PEGDA]= 68/32, obtained from the copolymerization at the monomer feed molar ratio [PAA]0/[PEGDA]0 = 99/1, demonstrated significant membrane disruptive activity.

Abstract:Reversible addition fragmentation chain transfer (RAFT) ab initio emulsion polymerization of styrene using poly(acrylic acid)-b-polystyrene trithiocarbonate (AA(x)-b-St(y) macroRAFT) as both mediator and surfactant was investigated with systematically changed y from 0 to 7 and x from 15 to 30. A well-controlled polymerization in terms of little coagulum, well predicted molecular weight and low polydispersity (PDI) (approximate to 1.2) could only be achieved by using the macroRAFTs of hydrophile-lipophile balance (HLB) values between 13 and 20. The final latex particle size could be tuned from 80 to 172 nm in diameter simply by changing x and y values while the molecular weight was targeted the same (30 K).

Abstract:A novel triblock copolymer containing azobenzene (AZO) and N-isopropylacrylamide (NIPAM) was synthesized by atom transfer radical polymerization (ATRP) using a poly(ethylene oxide) bifunctional macroinitiator (Br-PEO-Br). The triblock copolymer was characterized by H-1 NMR and gel permeation chromatography (GPC). Uniformly distributed polymeric micelles were obtained in aqueous solution by self-assembly method. Transmission electron microscope (TEM), fluorescence spectroscopy and dynamic light scattering (DLS) were used for the investigation of the self-assemblies and their stimuli-responsive behavior. Photoinduced release behavior was achieved with the as-prepared triblock copolymer micelles, which has not been observed in our previously-reported diblock copolymer micelles: the fluorescence intensity of Nile Red trapped in the triblock copolymer micelles dropped by approximately 40% after UV and Vis light alternative irradiation, indicating that a portion of the Nile Red was released to the aqueous media. Besides, the micelles shrank during the heating process.

Abstract:An in-depth investigation of the ultrasonic response of mechanophores containing weak covalent bonds within nanosized aggregates is vital for ultrasound imaging, drug delivery, and sensors. In this work, well-defined amphiphilic block SP-[poly(methyl methacrylate- b -N-isopropylacrylamide)] 2 SP-[p(MMA- b -NIPAM)] 2 ( P1 ) and amphiphilic random, SP-[poly(methyl methacrylate- r -N-isopropylacrylamide)] 2 SP-[p(MMA- r -NIPAM)] 2 ( P2 ) copolymers with SP mechanophore embedded at midpoint polymer chain are prepared by RAFT (reversible addition-fragmentation chain transfer) polymerization using a pre-synthesized novel functional bi (trithiocarbonate) and spiropyran (SP)-based RAFT reagent. Both P1 and P2 copolymers self-assemble in spherical micelles in AcCN/water mixed solvents, and several assemblies with different water contents are fabricated. The structural differentiation between block P1 and random P2 dominates the inverse influence of water content on the aggregate sizes of micelles. Under ultrasound irradiation, the SP mechanophore is activated within both micelles, and the amount of activated SP mechanophore increases progressively with prolonged sonication time. For P1 , the mechanochemical reaction (mechanochromism) kinetics in an aggregated state and a single chain state are different, while that of P2 micelles and liner are similar, inferred to be associated with the structure difference between both micelles. The mechanoactivation of SP mechanophore within micelles in AcCN/water mixed solvents is higher than in dissolved state due to the micellization of P1 and P2 , entangling the hydrophobic PMMA segments and partially swelling in the micellar core. Moreover, an increase in the medium's dielectric constant around the core's SP is also observed, promoting the SP mechanoactivation. This approach may provide a new strategy for preparing novel mechanophore-centered amphiphilic copolymers with colorful structures and high hydrophilicity. These can self-aggregate into nanoparticles in aqueous or mixed solutions, exhibiting effective effects of ultrasonic responsiveness. We expect this work to push forward the in-depth studies on the ultrasonic responsive mechanism of assembly and help open new research avenues into the mechanochemical reactions within nanosized aggregates.

Abstract:ABSTRACTWell‐defined tertiary amine‐based pH‐responsive homopolymers and block copolymers were synthesized via reversible addition‐fragmentation chain transfer (RAFT) polymerization using 4‐cyanopentanoic acid dithiobenzoate (CPAD) as the RAFT agent for homopolymers and a poly(ethylene glycol) (PEG) macro‐RAFT agent for the block copolymers. 1H NMR and gel permeation chromatography results confirmed the successful synthesis of these homopolymers and block copolymers. Kinetics studies indicated that the formation of both the homopolymers and the block copolymers were well defined. The pKa titration experiments suggested that the homopolymers and the related block copolymers have a similar pKa. The dynamic light scattering investigation showed that all of the block copolymers underwent a sharp transition from unimers to micelles around their pKa and the hydrodynamic diameter (Dh) was not only dependent on the molecular weight but also on the composition of the block copolymers. The polymer solution of PEG‐b‐PPPDEMA formed the largest micelle compare to the PEG‐b‐PDPAEMA and PEG‐b‐PDBAEMA with a similar molecular weight. © 2015 Wiley Periodicals, Inc. J. Polym. Temperature-Responsive Lactic Acid-Based Nanoparticles by RAFT-Mediated Polymerization-Induced Self-Assembly in Water

Abstract:This work demonstrates for the first-time biobased, temperature-responsive diblock copolymer nanoparticles synthesized by reversible addition-fragmentation chain-transfer (RAFT) aqueous emulsion polymerization-induced self-assembly (PISA). Here, monomers derived from green solvents of the lactic acid portfolio, N,N-dimethyl lactamide acrylate (DMLA) and ethyl lactate acrylate (ELA), were used. First, DMLA was polymerized by RAFT aqueous solution polymerization to produce a hydrophilic PDMLA macromolecular chain transfer agent (macro-CTA), which was chain extended with ELA in water to form amphiphilic PDMLA-b-PELA diblock copolymer nanoparticles by RAFT aqueous emulsion polymerization. PDMLAx homopolymers were synthesized targeting degrees of polymerization, DPx from 25 to 400, with relatively narrow molecular weight dispersities (Đ < 1.30). The PDMLA64-b-PELAy diblock copolymers (DPy = 10-400) achieved dispersities, Đ, between 1.18 and 1.54 with two distinct glass transition temperatures (Tg) identified by differential scanning calorimetry (DSC). Tg(1) (7.4 to 15.7 °C) representative of PELA and Tg(2) (69.1 to 79.7 °C) of PDMLA. Dynamic light scattering (DLS) studies gave particle z-average diameters between 11 and 74 nm (PDI = 0.04 to 0.20). Atomic force microscopy (AFM) showed evidence of spherical particles when dispersions were dried at ∼5 °C and film formation when dried at room temperature. Many of these polymers exhibited a reversible lower critical solution temperature (LCST) in water with a concomitant increase in z-average diameter for the PDMLA-b-PELA diblock copolymer nanoparticles.

Abstract:Polymerization-induced self-assembly via reversible addition–fragmentation transfer (RAFT)-mediated emulsion polymerization has served as a versatile platform for scalable preparation of well-defined block copolymer nanoparticles. However, kinetically trapped spherical micelles rather than higher-order morphologies are typically prepared by RAFT-mediated emulsion polymerization. Moreover, the mechanism for morphological evolution under RAFT-mediated emulsion polymerization conditions is still unclear. Herein, we report a redox-initiated RAFT-mediated emulsion polymerization of methacrylic monomers at relatively low temperatures. A diverse set of morphologies have been prepared by using glycidyl methacrylate (GlyMA) as the core-forming monomer. Effects of reaction temperature, molecular weight of macro-RAFT agent, degree of polymerization of PGlyMA, and monomer concentration on RAFT-mediated emulsion polymerization have been studied in detail. To give more insights into the mechanism of morphological evolution, other methacrylic monomers were also employed for RAFT-mediated emulsion polymerization. Finally, taking advantage of the intrinsic mechanism of RAFT-mediated emulsion polymerization, cross-linking has been explored for the preparation of higher-order morphologies. The current study not only expands the scope of RAFT-mediated emulsion polymerization for preparing various block copolymer nano-objects but also provides important insights into the morphological evolution under RAFT-mediated emulsion polymerization conditions.

Abstract:The influence of the macroRAFT agent architecture on the morphology of the self-assemblies obtained by aqueous RAFT dispersion polymerization in PISA is studied by comparing amphiphilic AC, A(B- co -C) diblocks and ABC triblock copolymers, constituted of N,N -dimethylacrylamide (DMAm = A), acrylic acid (AA = B) and N -cyanomethylacrylamide (CMAm = C) monomer units. The aim of this study is to better understand the impact of the presence of pH-sensitive acrylic acid units and their spatial distribution in block copolymers. Generally, the presence of a fully protonated intermediate block of PAA between the two blocks (based on PDMAm and PCMAm) favors the formation of higher-order morphologies. These ABC triblock copolymers assemblies are pH-responsive and can undergo morphological transitions through the controlled deprotonation or protonation of the PAA segment, but do not completely dissociate at high pH ( i.e. when AA units are fully ionized) unlike their A(B- co -C) diblock counterparts, where the pH-responsive monomer unit is homogeneously distributed within the solvophobic block.

Abstract:Various poly(2-hydroxyethyl methacrylate-b-benzyl methacrylate) (PHEMA n –PBzMA m ) and poly(2-hydroxypropyl methacrylate-b-benzyl methacrylate) (PHPMA n –PBzMA m ) nano-objects have been prepared via reversible addition–fragmentation chain transfer (RAFT) alcoholic dispersion polymerization. Using either a PHPMA or PHEMA macro-CTA as a steric stabilizer, chain extension with BzMA was conducted in methanol, ethanol, or isopropanol. In each case, in situ self-assembly is driven by the growing PBzMA chains, which become insoluble in lower alcohols above a certain critical chain length. Empirically, PHPMA macro-CTA proved to be much more effective than PHEMA macro-CTA in such syntheses, since the former conferred higher colloidal stability in alcohol. By constructing two detailed phase diagrams, the final nanoparticle morphology is shown to be sensitive to the DP of the core-forming block (PBzMA), the total solids content, and also the mean DP of the stabilizer block (PHPMA). The latter effect is readily demonstrated for PHPMA macro-CTAs possessing mean DPs of 48 and 63. Using PHPMA48 as a steric stabilizer, a range of nano-objects (spheres, worms or vesicles) can be accessed simply by tuning the DP of the core-forming PBzMA block. In contrast, using the PHPMA63 stabilizer only produces spherical morphologies. Presumably this is because the latter confers more effective steric stabilization, which prevents the efficient fusion of spheres to form worms. Nevertheless the PHPMA63–PBzMA n formulation may still be useful, since it allows access to spherical nanoparticles with tunable mean diameters of 29–100 nm. Such phase diagrams are essential for the reproducible targeting of copolymer morphologies, since they enable mixed phase regions to be avoided and allow the predictable synthesis of pure spheres, worms, or vesicles at a given concentration. Finally, a block copolymer “jellyfish” was observed during these PISA syntheses, which suggests that such intermediates are most likely a generic feature of the in situ conversion of worms into vesicles.

Abstract:Water-soluble macromolecular chain transfer agents (Macro-CTAs) were developed for the microwave-assisted precipitation polymerization of N-isopropylacrylamide. Two types of Macro-CTAs, amphiphilic (Macro-CTA1) and hydrophilic (Macro-CTA2), were investigated regarding their activity for the facile formation of nanoparticles and double hydrophilic block copolymers by RAFT processes. While both Macro-CTAs functioned as steric stabilization agents, the variation in their surface activity afforded different levels of control over the resulting nanoparticles in the presence of cross-linkers. The cross-linked nanoparticles produced using the amphiphilic Macro-CTA1 were less uniform than those produced using the fully hydrophilic Macro-CTA2. The nanoparticles spontaneously formed core-shell structures with surface functionalities derived from those of the Macro-CTAs. In the absence of cross-linkers, both types of Macro-CTAs showed excellent control over the RAFT precipitation polymerization process with well-defined, double hydrophilic block copolymers being obtained. The power of combining microwave irradiation with RAFT procedures was evident in the high efficiency and high solids content of the polymerization systems. In addition, the "living" nature of the nanoparticles allowed for further copolymerization leading to multiresponsive nanostructured hydrogels containing surface functional groups, which were used for surface bioconjugation.
Responsive Microgels through RAFT-HDA Dynamic Covalent Bonding Chemistry

Abstract:This paper developed a method for preparing ultrasound-responsive microgels based on reversible addition fragmentation chain transfer-hetero Diels-Alder (RAFT-HAD) dynamic covalent bonding. First, a styrene cross-linked network was successfully prepared by a Diels-Alder (DA) reaction between phosphoryl dithioester and furan using double-ended diethoxyphosphoryl dithiocarbonate (BDEPDF) for RAFT reagent-mediated styrene (St) polymerization, with a double-ended dienophile linker and copolymer of furfuryl methacrylate (FMA) and St as the dienophile. Subsequently, the microgel system was constructed by the HDA reaction between phosphoryl disulfide and furan groups using the copolymer of polyethylene glycol monomethyl ether acrylate (OEGMA) and FMA as the dienophore building block and hydrophilic segment and the polystyrene pro-dienophile linker as the cross-linker and hydrophobic segment. The number of furans in the dienophile chain and the length of the dienophile linker were regulated by RAFT polymerization to investigate the effects of the single-molecule chain functional group degree, furan/dithioester ratio, and hydrophobic cross-linker length on the microgel system. The prepared microgels can achieve the reversible transformation of materials under force responsiveness, and their preparation steps are simple and adaptive to various potential applications in biomedical materials and adaptive electrical materials.
From RAFT Emulsion Polymerization to RAFT Dispersion Polymerization: A Facile Approach to tune Dispersities and Self-assembled Behaviors of Block Copolymers

Abstract:Heterogeneous reversible addition-fragmentation chain transfer (RAFT) polymerization coupled with polymerization-induced self-assembly (PISA) can be performed through either RAFT emulsion polymerization or RAFT dispersion polymerization, providing opportunities for the synthesis of block copolymers with low dispersities and polymer nanoparticles with various morphologies. In this study, we have demonstrated the successful transition from photoinitiated RAFT emulsion polymerization to photoinitiated RAFT dispersion polymerization by solely changing the solvent composition from water to water/ethanol. Kinetic studies indicated that a slower polymerization was observed at a higher ethanol content. Size exclusion chromatography (SEC) analysis demonstrated that narrower molecular weight distributions were achieved as increasing the ethanol content due to the faster radical diffusion within the micellar core. The dispersity of block copolymer can be changed by varying the ethanol content used in photoinitiated heterogeneous RAFT polymerization. Moreover, reaction temperature has also been explored as a parameter to control the diffusion of radicals within the micellar core and therefore dispersities of block copolymers. Finally, the morphological evolution of block copolymer nanoparticles during the transition from RAFT emulsion polymerization to RAFT dispersion polymerization was also investigated. The present study is anticipated to significantly expand the scope of heterogeneous RAFT polymerization by allowing access to a wide range of block copolymers with different dispersities and polymer nanoparticles with diverse morphologies.

Abstract:Spiropyran (SP), which can respond to acid, ultraviolet irradiation, and mechanical force, has received great attention as a classic molecule for the construction of stimuli-responsive materials. However, the self-assembly behavior of SP with a tunable morphology has rarely been investigated. In the present work, a SP derivative bearing two carboxyl groups and one hydroxyl group was prepared and used as a model to investigate the optical properties, molecular self-assembly, and morphology of SP. This pH-responsive derivative could undergo a large morphology variation from a sphere- to a rodlike structure by adjusting the acidity. The indoline and pyridopyran fragments contributed to π-π stacking, whereas the carboxyl groups contributed to hydrogen bonding. Both π-π stacking and hydrogen bonding contributed to the pH-responsive self-assembly. The morphologies of spiropyran aggregates formed under different conditions were characterized by SEM, TEM, and FTIR spectroscopy. Control of the morphology of the spiropyran nanostructure was achieved.

Abstract:A novel multi stimuli-responsive silica nanocomposite with a core-shell structure is synthesized using reversible addition-fragmentation chain transfer (RAFT) polymerization. The SiO2 nanoparticles are chemically modified via 3-(trimethoxylsilyl) propyl methacrylate (MPS) which they are used in the core of nanocomposite. The macro-RAFT agent is prepared by RAFT polymerization of N-isopropylacrylamide (NIPAM), spirooxazine acryloyl monomer (SOM), and 2-(dodecylthiocarbonothioylthio)-2-methylpropionic acid (DDMAT as a RAFT agent). Finally, the smart nanocomposite is prepared by reaction between the macro-RAFT agent and 2-(Dimethylamino) ethyl methacrylate (DMAEMA) as a monomer. SOM as a photochromic monomer, DMAEMA and NIPAM as temperature and pH sensitive were used in the synthesized nanocomposite. The finally smart nanocomposite was characterized by using FT-IR, 1H-NMR, UV-Vis, GPC, DLS, and TEM techniques. The responsiveness of synthesized smart nanocomposite in different conditions (UV-Vis light, pH, temperature and the combination of pH and temperature) was investigated. The results show that the fabricated smart nanocomposite are good candidate nanomaterials for sensors application.
