Abstract:A well-defined amphiphilic centipede-like copolymer of styrene and methacrylic acid (PS-PS-PMAA) was synthesized by the combination of living anionic polymerization and atom transfer radical polymerization (ATRP). The synthetic approach involves the first coupling reaction of polystyrene (PS) backbone bearing 1,1-diphenylethene (DPE) pendant groups with living polystyryllithium (PSLi), and sequential anionic polymerization of t-butyl methacrylate (tBMA) initiated by resulting 1,1-diphenylmethyl anion, and final hydrolysis of obtained PS-PS-PtBMA. The centipede-like copolymer PS-PS-PMAA was characterized by 1H NMR, IR, GPC, and SLS measurements. The critical micelle concentration (CMC) of PS-PS-PMAA in water was determined by fluorescence probe technique. The self-assembly behavior of PS-PS-PMAA in water-THF mixture was observed by TEM. The results showed that the micellar morphology can be varied, such as vesicle, sphere, and agglomerate, depending on the THF content. These phenomena are worthy of further research in polymer physics field.
Synthesis and Self-Assembly of Amphiphilic ABA Type Triblock Copolymer with Well-Defined Glycopolymer Segments

Abstract:A novel amphiphilic ABA type triblock copolymer with well-defined glycopolymer segments was successfully synthesized via the atom transfer radical polymerization (ATRP) technique, using a bromo-terminated difunctional polysulfone as macroinitiator. The difunctional polysulfone macroinitiator was prepared by esterifying the phenolic end groups of polysulfone to α-haloesters. This macroinitiator was then used to initiate the polymerization of a glucose-carrying monomer, 3-O-methacryloyl-1,2:5,6-di-O-isopropylidene-D-glucofuranose (MAIpG), resulting an ABA type triblock copolymer. After acidolysis treatment, the isopropylidenyl groups of the protected sugar residues were removed, thus the amphiphilic ABA type triblock copolymer with well-defined glycopolymer segments was obtained. The polymers obtained were identified by FT-IR, 1H-NMR, GPC, and TGA. The self-assembly behavior of the amphiphilic glycopolymer in selective solvent (e.g. DMF/H2O) system was also preliminarily explored. The resultant amphiphilic glycopolymer shows potential applications in the fields of controlled release and delivery of drugs, micro-reactors, nano-materials, medical devices, and so on.

Abstract:Two pH-responsive amphiphilic diblock copolymers, namely polyisobutylene-block-poly[2-(N,N-dimethylamino) ethyl methacrylate] (PIB-b-PDMAEMA) and polyisobutylene-block-poly(metharylic acid) (PIB-b-PMAA), were synthesized via oxyanion-initiated polymerization, and their multiple self-assembly behaviors have been studied. An exo-olefin-terminated highly reactive polyisobutylene (HRPIB) was first changed to hydroxyl-terminated PIB (PIB-OH) via hydroboration-oxidation of C=C double bond in the chain end, and then reacted with KH to yield a potassium alcoholate of PIB (PIB-O−K+). PIB-O−K+ was immediately used as a macroinitiator to polymerize DMAEMA monomer, resulting in a cationic diblock copolymer PIB-b-PDMAEMA. With the similar synthesis procedure, the anionic diblock copolymer PIB-b-PMAA could be prepared via a combination of oxyanion-initiated polymerization of tert-butyl methacrylate (tBMA) and subsequent hydrolysis of tert-butyl ester groups in PtBMA block. The functional PIB and block copolymers have been fully characterized by 1H-NMR, FT-IR spectroscopy, and gel permeation chromatography (GPC). These samples allowed us to systematically investigate the effects of block composition on the pH responsivity and various self-assembled morphologies of the copolymers in THF/water mixed solvent. Transmission electron microscopy (TEM) images revealed that these diblock copolymers containing small amount of original PIB without exo-olefin-terminated group are able to self-assemble into micelles, vesicles with different particle sizes and cylindrical aggregates, depending on various factors including block copolymer composition, solvent polarity and pH value.

Abstract:pH-Responsive polymers have attracted much attention for biotechnology applications as carriers or matrix to facilitate intracellular or extracellular therapeutic drug delivery and release. In this paper, we report the development of new pH-responsive and hyperbranched copolymers with potential for such applications. These pH-responsive hyperbranched copolymers were synthesized via one pot reversible addition-fragmentation chain transfer (RAFT) copolymerization of propylacrylic acid (PAA) and a branching co-monomer poly(ethylene glycol diacrylate) (PEGDA) (Mn=258 Da) at the monomer feed molar ratios [PAA]0/[PEGDA]0 = 99/1, 90/10 and 80/20. The resultant poly(PAA-PEGDA) copolymers were characterized by Proton Nuclear Magnetic Resonance (1H NMR) and Gel Permeation Chromatography (GPC) to obtain the molecular weight, copolymer composition and degree of acrylate functionality. The hydrodynamic dimensions of these copolymers at pH range between 5.0 and 7.4 were studied using Dynamic Light Scattering technique (DLS). Moreover, these hyperbranched copolymers demonstrated composition- and size-dependent membrane disruptive properties by red blood cell hemolysis assay. Poly(PAA-PEGDA) with the copolymer composition [PAA]/[PEGDA]= 68/32, obtained from the copolymerization at the monomer feed molar ratio [PAA]0/[PEGDA]0 = 99/1, demonstrated significant membrane disruptive activity.

Abstract:A novel amphiphilic copolymer poly(ethylene glycol)-block-poly(N,N-dimethylamino-2-ethylmethacrylate)-blockpoly[6-(4-methoxy-azobenzene-4′-oxy) hexyl methacrylate] (PEG-b-PDMAEMA-b-PMMAzo) was prepared by ATRP polymerization. The self-assembly and responsive behaviors were investigated by SEM, TEM, LLS and UV-Vis spectra. The results indicated that the copolymers can self-assemble into spherical structures in aqueous media. The aggregate size can be tuned by pH and temperature. The trans-cis isomerization behavior of the formed aggregates was also examined. Upon irradiation with a linear polarized light, the elongation degree of the aggregates was increased with the irradiation time.

Abstract:A multi-responsive hydrophilic ABCBA-type pentablock copolymer, consisting of poly(ethylene glycol), poly(N-isopropylacrylamide), and poly(2-(diethylamino) ethyl methacrylate), PDEA-b-PNIPAM-b-PEG-b-PNIPAM-b-PDEA, was synthesized by consecutive reversible addition-fragmentation chain transfer (RAFT) polymerization. The polymerization showed all of the expected features of living radical polymerization and allowed the synthesis of copolymers with controlled lengths of the PNIPAM and PDEA blocks. Due to the multi-component and multi-functional nature of the pentablock copolymer, the solution properties can be manipulated by the changing of parameters such as temperature, pH etc. For example, the copolymer could be dissolved in acidic aqueous solution at room temperature, since the pH was below the pK(a) of the PDEA block, and the temperature was below the lower critical solution temperature (LCST) of the PNIPAM block. While in alkaline solution at room temperature, the copolymer self-assembled into core-shell-corona micelles, with the hydrophobic PDEA block as the core, the thermoresponsive PNIPAM block as the shell and the hydrophilic PEG block as the corona. At elevated temperatures in acidic media, the copolymer self-assembled into PNIPAM-core micelles with mixed hydrophilic PEG and pH-responsive PDEA coronas. Moreover, the self-assembly behavior of the pentablock copolymers in aqueous solution was investigated by varying the length of PDEA blocks.

Abstract:Two poly(2-(dimethylamino)ethyl methacrylate)-b-poly(acrylic acid) diblock copolymers, PDMAEMA(84)-b-PAA(18) and PDMAEMA(50)-b-PAA(18), were synthesized by the atom transfer radical polymerization (ATRP) and their dual-stimuli responsive behaviors to the changes in temperature and pH in aqueous solutions were investigated by UV-vis spectroscopy, dynamic light scattering (DLS), (1)H NMR spectroscopy and surface tension measurement. Different from PDMAEMA(84)-b-PAA(18) solutions where no aggregation is observed between pH 7.0 and 9.5, the PDMAEMA(50)-b-PAA(18) aggregates can exist in this broad pH range due to the hydrophobic interactions among the charge-balanced polyampholyte chains. At high pH, e.g., 11.0, the DMAEMA segments collapse to form the core of micelles due to the hydrophobic property of the de-protonized DMAEMA stabilized with the highly ionized AA segments on the surface of the micelles upon heating. At pH around the IEP, e.g., 9.5, large micelles can be formed in PDMAEMA(84)-b-PAA(18) solution upon heating, just like that at pH 11.0, while PDMAEMA(50)-b-PAA(18) first formed the micelles due to the electrostatic attraction between ionized AA segments and protonated DMAEMA segments, but the aggregation of the micelles was hardly happened upon heating due to the smaller DMAEMA segment. Moreover, LCST can be exactly estimated by surface tension experiment.

Abstract:In this article, the synthesis and self-assembly of a novel well-defined biocompatible amphiphilic POEGMA-PDMS-POEGMA triblock copolymer were studied. The copolymer was synthesized by atom transfer radical polymerization of oligo(ethylene glycol) methyl ether methacrylate (OEGMA) using alpha,omega-dibromo polydimethylsiloxane macroinitiator (Br-PDMS-Br). Br-PDMS-Br was synthesized through the esterification of alpha,omega-hydroxypropyl polydimethylsiloxane and 2-bromoisobutyryl bromide. The structures of the copolymers were confirmed by proton nuclear magnetic resonance spectroscopy, and gel permeation chromatography. The copolymers showed reversible aggregation in response to temperature cycles with a lower critical solution temperature (LCST) between 61 and 66 degrees C, as determined by ultraviolet-visible spectrophotometry and dynamic light scattering. The LCST values increased in proportion to the length of the hydrophilic block and were lower than that of the POEGMA homopolymer. The self-assembly behavior of the copolymers in aqueous solution was investigated by fluorescence spectroscopy and transmission electron microscopy. The critical micelle concentration value (1.08-0.26 10(-6) mol L-1) decreased as the length of the POEGMA chain increased. The POEGMA-PDMS-POEGMA copolymers can easily self-assemble into spherical micelles in aqueous solution. Such biocompatible block copolymers may be attractive candidates as "smart" thermo-responsive drug delivery systems. (C) 2014 Wiley Periodicals, Inc.

Abstract:A series of ABA triblock copolymers, consisting of double-bond-containing poly(phenoxyallene) (PPOA), poly(methyl methacrylate) (PMMA), or poly(butyl acrylate) (PBA) segments, were synthesized by sequential free radical polymerization and atom transfer radical polymerization (ATRP). A new bifunctional initiator bearing azo and halogen-containing ATRP initiating groups was first prepared followed by initiating conventional free radical homopolymerization of phenoxyallene with cumulated double bond to give a PPOA-based macroinitiator with ATRP initiating groups at both ends. Next, PMMA-b-PPOA-b-PMMA and PBA-b-PPOA-b-PBA triblock copolymers were synthesized by ATRP of methyl methacrylate and n-butyl acrylate initiated by the PPOA-based macroinitiator through the site transformation strategy. These double-bond-containing triblock copolymers are stable under UV irradiation and free radical circumstances.

Abstract:A new ABA amphiphilic triblock copolymer, comprising double-bond-containing poly(phenoxyallene) (PPOA) and poly(oligo(ethylene glycol) methyl ether methacrylate) (POEGMM) segments, was synthesized by a combination of conventional free radical polymerization, atom transfer radical polymerization (ATRP), and the site transformation strategy. A bifunctional initiator containing azo and Br-containing ATRP initiating groups was employed to initiate conventional free radical homopolymerization of phenoxyallene with a cumulated double bond to give a PPOA-based macroinitiator (Br-PPOA-Br) bearing ATRP initiating groups at both ends. Next, ATRP of OEGMM monomer was initiated by Br-PPOA-Br to provide the desired POEGMM-b-PPOA-b-POEGMM triblock copolymer. The critical micelle concentration of POEGMM-b-PPOA-b-POEGMM amphiphilic copolymer in aqueous solution was determined by fluorescence spectroscopy. The amphiphilic triblock copolymer could self-assemble into wormlike and spherical micelles in aqueous media. POEGMM-b-PPOA-b-POEGMM triblock copolymer can also be used as the stabilizer for the aqueous polymerization of styrene and methyl methacrylate.

Abstract:A series of perfluorocyclobutyl aryl ether-based amphiphilic ABA triblock copolymers consisting of hydrophilic poly[poly(ethylene glycol) methyl ether methacrylate] (PPEGMEMA) and hydrophobic poly(2,2'-bis(4-trifluorovinyl-oxyphenyl)propane) (PBTFVPP) blocks were synthesized via the site transformation strategy. A semi-fluorinated PBTFVPP segment was first prepared via thermal step-growth cycloaddition polymerization of BTFVPP trifluorovinyl aryl ether monomer followed by end functionalization to be transformed into Br-PBTFVBP-Br macroinitiator bearing one ATRP initiating group at each end. The target PPEGMEMA-b-PBTFVPP-b-PPEGMEMA triblock copolymers with relatively narrow molecular weight distributions (M-w/M-n <= 1.32) were synthesized via ATRP of PEGMEMA macromonomer initiated by Br-PBTFVBP-Br macroinitiator. A fluorescent probe technique was used to determine the critical micelle concentrations (cmc) of the obtained amphiphilic copolymers in aqueous solution. The morphologies of the micelles formed by the copolymers were investigated by transmission electron microscopy. It was shown that such triblock copolymers could self-assemble into ovals, well-defined cylinders, and spheres in aqueous solution with different initial water contents and compositions.

Abstract:A novel pH-sensitive fluorinated triphilic block cocopolymer P (EHMA-co-MAA)-b-PFOEA-b-P(EHMA-co-MAA)was synthesized by using the reversible addition fragmentation chain transfer (RAFT )method,incorporating methacrylic acid,2-ethylhexyl methacrylate,and 2-perfluorooctyl ethyl acrylate blocks used as hydrophilic,lipophilic,and fluorophilic units respectively.The chemical structure of block copolymer was characterized by infrared spectroscopy (IR),nuclear magnetic resonance (NMR),gel permeation chromatography (GPC),and fluorine element analysis(F-EA).The self-assembly of fluorinated block copolymer in dimethylformamide/water dispersions was investigated.Spherical micelles with silkworm chrysalis-shaped surface formed by copolymer were verified by scanning electron microscopy,and the diameter of micelles increased significantly along with the increase of the content of fluorinated block.The morphology transition from spheres to bowl-shaped vesicles was obtained by adding aqueous solution of HCl into the system,and as the concentration of HCl increases,vesicles will merge together.

Abstract:A novel POSS-containing methacrylate monomer (HEMAPOSS) was fabricated by extending the side chain between polyhedral oligomeric silsesquioxane (POSS) unit and methacrylate group, which can efficiently decrease the steric hindrance in free-radical polymerization of POSS-methacrylate monomer. POSS-containing homopolymers (PHEMAPOSS) with a higher degree of polymerization (DP) can be prepared using HEMAPOSS monomer via reversible addition-fragmentation chain transfer (RAFT) polymerization. PHEMAPOSS was further used as the macro-RAFT agent to construct a series of amphiphilic POSS-containing poly(N,N-dimethylaminoethyl methacrylate) diblock copolymers, PHEMAPOSS-b-PDMAEMA. PHEMAPOSS-b-PDMAEMA block copolymers can self-assemble into a plethora of morphologies ranging from irregular assembled aggregates to core-shell spheres and further from complex spheres (pearl-necklace-liked structure) to large compound vesicles. The thermo-and pH-responsive behaviors of the micelles were also investigated by dynamic laser scattering, UV spectroscopy, SEM, and TEM. The results reveal the reversible transition of the assembled morphologies from spherical micelles to complex micelles was realized through acid-base control. (C) 2014 Wiley Periodicals, Inc.

Abstract:We present the synthesis and self-assembly behavior of a new semi-fluorinated amphiphilic triblock copolymer. A series of perfluorocyclobutyl aryl ether-based amphiphilic ABA triblock copolymer containing hydrophilic poly(ethylene glycol) segment as the middle block were synthesized by atom transfer radical polymerization (ATRP). ATRP of 4-(4'-p-tolyloxyperfluorocyclobutoxy)benzyl methacrylate was initiated by PEG-based bifunctional macroinitiators with different molecular weights to obtain the desired copolymers with narrow molecular weight distributions (M-w/M-n <= 1.30) and the number of perfluorocyclobutyl linkage can be tuned by the feed ratio and the conversion of the fluorine-containing methacrylic monomer. The critical micelle concentrations of these amphiphilic ABA triblock copolymers in aqueous media were determined by fluorescence probe technique. Abstract:Diblock copolymer poly[(N,N-dimethylamino)ethyl methacrylate]-block-poly(acrylic acid)(PDMA-b-PAA) with a low polydispersity(PDI=1.23) has been synthesized by atom transfer radical polymerization(ATRP).The zwitterionic properties of PDMA-b-PAA solutions and the effect of ionic strength of small molecule salt on the solution behavior have been investigated.It is shown that PDMA-b-PAA has both pH-and temperature-responsive assembly behavior with the IEP of 6.9 and the LCST of 37.8 ℃ and has anti-polyelectrolyte effect.Phase separation happened in the pH range of 5.4-7.3 and the perfect reversible thermal-responsive behaviors were clearly observed.

Abstract:In this work, a triblock copolymer poly(acrylic acid)-block-poly[6-(4-methoxy-azobenzene-4'-oxy) hexyl-methacrylate]- block-poly(N-isopropylacrylamide) (PAA-b-PAzoMA-b-PNIPAM) containing pH, photo and temperature sensitive groups was synthesized through three-step RAFT polymerization. By tuning the solvents, the copolymer could self-assemble into spherical micelles or vesicles with multi-stimuli responsiveness. Either the micelles or the vesicles underwent obvious size decreases when the temperature was above the LCST of PNIPAM, and size increases when the pH value was below the pK(a) of PAA without obvious morphology changes. In addition, under UV irradiation, the micelles disrupted into much smaller particles whereas the vesicles transformed into micelles with an apparent size decrease on account of trans-to-cis isomerization of the azobenzene moieties. Upon irradiation by linear polarized light, the micelles were elongated into spindles and short rods. Furthermore, the photo-isomerization rate of the azobenzene moiety could be modulated by pH and temperature. The multi-stimuli responsive properties of PAA-b-PAzoMA-b-PNIPAM will further expand the potential application of azobenzene copolymers in smart materials.
PDEGMA-b-PDMAEMA-b-PLMA triblock terpolymers and their cationic analogues: Synthesis, stimuli responsive self-assembly and micelleplex formation

Abstract:In this study, the synthesis, chemical modification and self-assembly in aqueous media of novel temperature-responsive and pH-responsive triblock terpolymers of the type (poly(diethylene glycol methyl ether methacrylate)-b-poly(2-(dimethylamino)ethyl methacrylate)-b-poly (lauryl methacrylate) (PDEGMA-b-PDMAEMA-b-PLMA) are reported. The triblock terpolymers were synthesized via RAFT polymerization and the molecular characterization was achieved by SEC, 1H-NMR and FTIR spectroscopy. Methyl iodide (CH3I) was utilized for chemical modification of the PDMAEMA blocks to increase the overall solubility of the triblock terpolymers in aqueous media and to obtain permanent positive charges, converting them to temperature-responsive block polyelectrolytes. PDEGMA-b-PDMAEMA-b-PLMA triblock terpolymers and their quaternized analogs self-assemble in aqueous media forming spherical micellar structures consisting of a hydrophobic PLMA core and stimuli-responsive PDMAEMA and PDEGMA corona, as evidenced by light scattering measurements. The dimensions of the formed micelles were influenced noticeably by variations in the solution temperature and pH. Moreover, we examine the formation of micelleplexes between quaternized PDEGMA-b-QPDMAEMA-b-PLMA thermoresponsive micelles and DNA molecules. The complexation capability of the formed micelleplexes was investigated by ethidium bromide quenching assays utilizing fluorescence spectroscopy techniques, and UV-vis spectroscopy. Light scattering techniques are utilized in order to determine the structural characteristics of the formed micelleplexes. The quaternized terpolymer micelles effectively interact with nucleic acid molecules with their surface charge and their size depending on the composition of block terpolymers, the N/P ratio, as well as the increase of temperature above the LCST value of the temperature responsive PDEGMA block.

Abstract:A new triblock copolymer poly(ethylene oxide)monomethyl ether-b-poly(acrylic aid)-b-poly[6-4-methoxy-azobenzene-4′-oxy)hexyl methylacrylate]was synthesized by the atom transfer radical polymerization (ATRP)method.The self-assembly behaviors in aqueous media,pH sensitive and photo-sensitive properties of this copolymer were studied by the means of transmis-sion electron microscopy (TEM),scanning electron microscopy (SEM),laser light scattering (LLS)and UV-Vis spectropho-tometer.It is found that the polymer can self-assemble into vesicle structure.With decreasing the pH value,the particle size of the aggregates increases sharply obviously bigger when pH<3.4.However,there is not significant change for the particle size of micelles when pH>3.4.Upon irradiation with polarized light,the aggregates are elongated with increasing the irradiation time.

Abstract:The pH-responsive amphiphilic poly(ε-caprolactone)-block-poly(acrylic acid) (PCL-b-PAA) copolymer was prepared by selective hydrolysis of one novel poly(ε-caprolactone)-block-poly(methoxymethyl acrylate) (PCL-b-PMOMA) block copolymer, which was synthesized by combining ring-opening polymerization (ROP) of ε-caprolactone (ε-CL) and atom transfer radical polymerization (ATRP) of methoxymethyl acrylate (MOMA). Selective hydrolysis of the hemiketal ester groups on the PMOMA block gave 100% deprotection without the cleavage of the PCL block. The self-assembly behavior of PCL-b-PAA was investigated by fluorescence spectroscopy, DLS and TEM. The spherical micelles were formed with the hydrophobic PCL block as the core and the hydrophilic PAA as the shell by a co-solvent evaporation method. Moreover, the size and size distribution of the micelles varied with pH value and ionic strength in aqueous solution. The cytotoxicity of the PCL-b-PAA was lower, which was confirmed by MTT assay.

Abstract:A novel fluorinated triblock copolymer incorporating 2-ethylhexyl methacrylate (EHMA), tert-butyl methacrylate (tBMA) and 1 H ,1 H ,2 H ,2 H -perfluorodecyl acrylate (FA) (PEHMA- b -P t BMA- b -PFA) was first synthesized using three successive reversible addition fragmentation chain transfer (RAFT) polymerization and the subsequent hydrolyzing at acidic condition. The as-fabricated triblock copolymer exhibited an interesting morphology evolution from the multi-compartment rod-like structure to spherical structure along with the addition of a selective solution. At the same time, a visible phase separation domain could be seen in the core area due to the existence of fluorocarbon segments. Furthermore, the self-assembly behavior of the triphilic copolymer at different pH was also verified by transmission electron microscopy, as well as the dynamic light scattering. These stimuli-responsive multi-compartment nanostructures may have potential applications in drug delivery.
