Abstract:An orthogonal dispersive spectral-domain optical coherence tomography (SDOCT) system based on a spectrometer consisting of a high spectral resolution virtually-imaged phased array (VIPA) and a low resolution diffraction grating is developed. Two-dimensional (2D) dispersion generated by the combination of the VIPA and the grating in conjunction with a 2D CCD leads to an improved performance of the spectrometer. Ultrahigh spectral resolution of 0.002 nm within a free spectrum range of 50 nm is realized, providing the spectrometer with a spectral sampling rate up to similar to 10(5). The developed SDOCT realizes an imaging depth over 80 mm, which is the longest depth range ever achieved by SDOCT. The increased spectral sampling rate also results in a high signal-to-noise ratio of the SDOCT system. The application of the developed system is further illustrated by quantitative phase imaging of a glass plate and an optical lens. (C) 2012 Optical Society of America
An Absolute Wavenumber Calibration Method Based on Characteristic Spectral Line and Constrained Fitting Phase

Abstract:Spectral-domain Optical Coherence Tomography (SD-OCT) systems generally have nonlinear sampling problems in wavenumber domain. In order to realize the precise positioning of the discrete interfaces and the high-quality reconstruction of OCT images under conventional fast Fourier transform, it is necessary to solve the precise calibration problem of the absolute wavenumber of the discrete sampling points in the spectrometer. In this paper, an absolute wavenumber calibration method based on the absolute phase of the characteristic spectral line and the constraint polynomial fitting phase under precise optical path difference was proposed. In the sample arm of the SD-OCT system, metal gauges with precise thickness difference are used to obtain the absolute phase value of the characteristic spectral line, and the phase wrapping times corresponding to the characteristic spectral line are further accurately solved. Thus, this method overcomes the 2π ambiguity of spectral phase in conventional interferometric phase methods. At the same time, combined with the polynomial fitting phase of the high signal-to-ratio region under window constraint, the accurate calibration of the absolute wavenumber of each sampling point is realized. Finally, comprehensive comparisons between the proposed method and the traditional resampling method in terms of discrete interface positioning, axial resolution and image reconstruction quality verifies the significant advantages of this method.

Abstract:Abstract Ultra-deep three-dimensional (3D) optical coherence tomography (OCT) in turbid media is reported. Parallel detection along depth combined with scanning detection across lateral field is exploited in a swept-source OCT system to obtain the depth-resolved reflection matrices. Scanning detection enables the use of a highspeed swept source and a matched detector providing the required depth range for ultra-deep imaging. Disturbances intrinsic to scanning detection are compensated with negligible intra-frame phase variations to ensure satisfactory reconstruction by the inter-frame phase insensitive algorithm. 3D OCT imaging with an ultra-deep imaging depth of 11.5 times the scattering mean-free-path and a triple extension of the imaging depth achieved by a conventional SS-OCT is realized. The measurement time for 3D imaging (125μm×125μm×4mm ) is ~5 min, a significant leap over the time-domain matrix OCT where 6 min is taken for only one specified depth.
High-speed Spectral-Domain Optical Coherence Tomography at 830nm Wavelength

Abstract:We present a high-speed spectral domain optical coherence tomography (SD-OCT) system at 830 nm wavelength which is consisted of a fiber based Michelson interferometer and a custom-built spectrometer. The designed resolution of the spectrometer is about 67.4pm which limits the maximum detection depth 2.56mm in air. And the 35us exposure time of the high speed line scan CCD makes real-time imaging possible. Furthermore, a novel method of spectrometer calibration is put forward. The method can remove the influence of dispersion mismatch, thus accurately determine the distribution of wavelength on the line scan CCD, which leads to a precise interpolation and a subsequent better contrast image.

Abstract:The Czerny-Turner layout, which is most frequently used in miniature spectrometers, should follow Shafer's coma-free condition and Fastie's flat-field principal to eliminate the central wave's primary coma and maximize its resolution. However, the design process does not take the comas and resolutions at non-central waves into consideration. Based on the theory of primary coma in reflection optical system, the present paper points out that in the crossed beam design, the resolutions at wide 'spectral region present a "V" shape, while in the M design, the resolutions change little over the whole region, and present an approximately straight line shape, so the latter kind of spectrometer maintains a far more consistent resolution than the former one. Accordingly, this paper designs two kinds of spectrometers with spectrum regions from 400 to 600 nm, and carries out theoretical simulation and contrast experiment. The result demonstrates that for the two designs the resolutions at the fringe wavelength are 3.8 times and 1.5 times respectively that at the central wavelength, which accords with the conclusion of the theoretical simulation.

Abstract:Abstract Unlike the single grating Czerny–Turner configuration spectrometers, a super-high spectral resolution optical spectrometer with zero coma aberration is first experimentally demonstrated by using a compound integrated diffraction grating module consisting of 44 high dispersion sub-gratings and a two-dimensional backside-illuminated charge-coupled device array photodetector. The demonstrated super-high resolution spectrometer gives 0.005 nm (5 pm) spectral resolution in ultra-violet range and 0.01 nm spectral resolution in the visible range, as well as a uniform efficiency of diffraction in a broad 200 nm to 1000 nm wavelength region. Our new zero-off-axis spectrometer configuration has the unique merit that enables it to be used for a wide range of spectral sensing and measurement applications.

Abstract:We propose a novel spectral-domain optical coherence tomography (SD-OCT) equipped with a Fresnel spectrometer, which utilizes a Fresnel zone plate (FZP) as both dispersion and focusing optics and thus spreads the spectral interferogram evenly in wavenumber domain because of the proportional relation between the focal length of the FZP and the wavenumber. With no need of the conversion calculation from wavelength to wavenumber in conventional SD-OCT, this new design is favorable for fast imaging with high resolution. As only a FZP and CCD are used, the Fresnel spectrometer is simple and compact. It is experimentally shown that its performance is as good as that of numerical interpolation in conventional SD-OCT. Imaging of bio-tissue by Fresnel SD-OCT is also demonstrated.

Abstract:The parameters of the spectrometer were analyzed in order to optimize the performance of spectral-domain optical coherence tomography (SD-OCT). Because probing depth is proportional to spectral resolution, it can be increased with the choice of a higher resolution spectrometer, which implies greater pixel numbers and lower imaging speed, or by sacrificing part of the spectrum, which compromises the axial resolution and side lobes. With a dynamic range of 8 bits or more, the signal-to-noise ratio remains constant for different noise levels. The results were verified experimentally with in vivo retinal SD-OCT imaging. Abstract:We develop a high speed spectral domain optical coherence tomography (SD-OCT) system based on a custom-built spectrometer and non-uniform discrete Fourier transform (NDFT) to realize minimized depth dependent sensitivity fall-off. After precise spectral calibration of the spectrometer, NDFT of the acquired spectral data is adopted for image reconstruction. The spectrometer is able to measure a wavelength range of about 138 nm with a spectral resolution of 0.0674 nm at central wavelength of 835 nm, corresponding to an axial imaging range of 2.56 mm in air. Zemax simulations and sensitivity fall-off measurements under two alignment states of the spectrometer are given. Both theoretical simulations and experiments are done to study the depth dependent sensitivity of the developed system based on NDFT in contrast to those based on conventional discrete Fourier transform (DFT) with and without interpolation. In vivo imaging on human finger from volunteer is conducted at A-scan rate of 29 kHz and reconstruction is done based on different methods. The comparing results confirm that reconstruction method based on NDFT indeed improves sensitivity especially at large depth while maintaining the coherence-function-limited depth resolution.

Abstract:Since many industrial materials have micro or submicro structures on the surface or subsurface, utrahigh-resolution is required in the inspection of these materials. Ultrahigh-resolution optical coherence tomography uses broadband light sources to achieve axial image resolutions on the scale of a few microns. We have been investigating an ultrahigh-resolution spectral-domain optical coherence tomography (SD-OCT) system using supercontinuum sources (SC) in free space. The effective SC spectrum has a full width at half maximum of 230 nm centered around 665 nm, and the imaging setup has an ultrahigh axial resolution of 0.9 μm in air, and a lateral resolution of 3.9 μm, with the system measurement range being 0.6 mm in axial direction. At a 50 μm axial position, the sensitivity can be 63 dB with 28600 axial scans per second at 2048 pixels per axial scan. Images of polystyrene microspheres solution with an average diameter of 5 μm and different sizes of industrial abrasive papers are presented to illustrate the performance of the system.

Abstract:Spectral domain optical coherence tomography (SD-OCT) with ultrahigh resolution can be used to measure precise structures in the context of ophthalmic imaging. We designed an ultrahigh resolution SD-OCT system based on broadband superluminescent diode (SLD) as the light source. An axial resolution of 2.2 μm in tissue, a scan depth of 1.48 mm, and a high sensitivity of 93 dB were achieved by the spectrometer designed. The ultrahigh-resolution SD-OCT system was employed to image the human cornea and retina with a cross-section image of 2048 × 2048 pixels. Our research demonstrated that ultrahigh -resolution SD-OCT can be achieved using broadband SLD in a simple way.

Abstract:We develop a high speed multi-functional spectral domain (SD-OCT) system, using a broadband light source centered at 835nm and a custom-built spectrometer with a spectral resolution of 0.0674nm. After precise spectral calibration of the spectrometer, non-uniform discrete Fourier transform (NDFT) of the acquired spectral data is adopted for image reconstruction. In vivo imaging of human finger from volunteer at A-scan rate of 29 kHz with different reconstruction methods reveal that with the NDFT method, improved sensitivity fall-off especially at large depth is achieved in contrast to the conventional discrete Fourier transform (DFT) with interpolation method. And the phase information of the spectral data is used for functional extension to velocity mapping, Doppler images of rats' cerebral arteries at A-scan rate of 20 kHz are demonstrated. Furthermore, based on the spatial sinusoidal modulation in the reference arm, the complex spectral interferogram can be retrieved by harmonics analysis and digital synchronous demodulation which is free of mirror image and the coherence noise. Double imaging depth range on fresh shrimp at A-scan rate of 10 kHz is achieved and the resulting complex conjugate rejection ratio is up to 41dB.

Abstract:Optical coherence tomography (OCT), as a noninvasive imaging technique, has been widely applied in biological and medical fields. The light sources commonly used in OCT are the partially coherent light sources which usual exhibits typically the resolution of 15 to 20 μm. In another approach for an extremely broadband light source is a thermal light source (white light), which may have the potential to supply a compact and low-cost OCT system. In addition, current the majority of OCT measurements are executed in the time domain systems, which are limited in the image acquisition rate and sensitivity. However, a spectral domain optical coherence tomography system (SD-OCT) outperforms the conventional TD-OCT system in imaging acquisition rate and sensitivity. In order to improve both image acquisition rate and axial resolution economically, a white light source spectral-domain optical coherence tomography (SD-OCT) system is constructed, where a tungsten halogen lamp is chosen as the illumination source and a Michelson fiber interferometer is employed to generate interference pattern of object collected by a spectrometer. The object inner structure is obtained from the interference pattern by a Fourier transform. It is theoretically stated and experimentally verified that the white light source SD-OCT system is feasible and enables to realize near real time three dimensional imaging reconstructions of internal structure of engineering and biological materials.

Abstract:A high resolution spectral-domain optical coherence tomography (SD-OCT) system based on a thermal light source was presented. A novel normalized method was introduced to remove the background noise and the DC noise of the interference spectrum. A Gaussian spectral calibration procedure was performed to improve axial resolution and image quality before reconstructing OCT image. With the proposed method, the quality of the obtained images was greatly improved. Two-dimensional (2D) cross-sectional images with axial resolution of 1.2 μm were obtained. Furthermore, the film thickness of single-layer film sample was obtained. The experimental result demonstrates the SD-OCT system has potential for film thickness measurement and surface topography.

Abstract:Exploring the corresponding relation between structural and physical properties of materials at the atomic scale remains the fundamental problem in science. With the development of the aberration-corrected transmission electron microscopy (AC-TEM) and the ultrafast optical spectroscopy technique, sub-angstrom-scale spatial resolution and femtosecond-scale temporal resolution can be achieved, respectively. However, the attempt to combine both their advantages is still a great challenge. Here, we develop in situ optical spectroscopy with high temporal resolution in AC-TEM by utilizing a self-designed and manufactured TEM specimen holder, which has the capacity of sub-angstrom-scale spatial resolution and femtosecond-scale temporal resolution. The key and unique design of our apparatus is the use of the fiber bundle, which enables the delivery of focused pulse beams into TEM and collection of optical response simultaneously. The generated focused spot has a size less than 2 µm and can be scanned in plane with an area larger than 75 × 75 µm2. Most importantly, the positive group-velocity dispersion caused by glass fiber is compensated by a pair of diffraction gratings, thus resulting in the generation of pulse beams with a pulse width of about 300 fs (@ 3 mW) in TEM. The in situ experiment, observing the atomic structure of CdSe/ZnS quantum dots in AC-TEM and obtaining the photoluminescence lifetime (∼4.3 ns) in the meantime, has been realized. Further ultrafast optical spectroscopy with femtosecond-scale temporal resolution could be performed in TEM by utilizing this apparatus.
Study On Near-Far Ultraviolet Imaging Spectrometer With High Resolution

Abstract:A near-far ultraviolet imaging spectrometer with high resolution is developed, which can be applied to the upper atmosphere observation in 115nm-295nm. The application demands of the corresponding remote sensing are analyzed. Because the imaging spectrum technology has been rarely utilized for the ionosphere observation in our country, we study some foreign advanced loads to apply the standard to our design. According to the analysis, the optical system of the imaging spectrometer is designed. The system includes an off-axis parabolic mirror as the telescope and Czerny-Turner structure as the imaging spectrum system. The photon counting detector is adopted to satisfy the observation for that the radiation is weak in near-far ultraviolet waveband. The receiving surface of the detector should be coated by different photocathode for better quantum efficiency in the near and far ultraviolet wavebands. The traditional Czerny-Turner system is not suit for the broadband imaging spectrometer because aberrations in the structure can't be corrected homogeneously. The geometric analysis and 1(st) order differential calculation method are introduced to improve disadvantages based on the aberration theory. We designed an example for the advanced imaging spectrometer. The results demonstrate that the modulation transfer function (MTF) in total field of view and the waveband is more than 0.6. The design has high spatial resolution and high spectral resolution. It certificates that the design theory can be applied to the near-far ultraviolet imaging spectrometer especial for upper atmosphere observation.

Abstract:Aiming at the requirements of real-time inspection in material industry, an ultrawide-field parallel SD-OCT system capable of visualizing cross-sectional image of internal structures by a single shot of a 2-D CMOS camera is developed. To achieve ultrawide-field parallel detection, one meniscus lens with negative dioptric is introduced as an additional relay lens and imaging optics including sample arm, relay lenses and spectrometer are optimized as a whole for desirable imaging quality. The developed system has an ultrawide-field of 35mm and an axial range of 8mm in air. The maximum on-axis sensitivity and the axial resolution are experimentally determined to be 65dB and 17.7μm, respectively. The on-axis lateral resolution is measured to be 44μm in parallel direction and 22μm in scanning direction. The system is applied to obtain 3-D volume images of a set of glass-samples covering an area of about 28mm by 15mm with thicknesses ranging between 2.1mm and 4mm, from which defects such as solid inclusion, micro-deformation and bubble are identified either on the surfaces or inside the samples.

Abstract:We investigated clinical imaging of the human retina with visible-light optical coherence tomography (vis-OCT) with a 1 µm axial resolution. Imaging was performed with rectangular modulation of the scanning beam that reduced image speckle noise and preserved fine anatomical features. B-scans acquired with scan modulation revealed new anatomical features and demonstrated increased contrast to noise ratio (CNR) over regularly acquired B-scans.

Abstract:Optical spectrometers play a key role in acquiring rich photonic information in both scientific research and a wide variety of applications. In this work, we present a new spectrometer with an ultrahigh resolution of better than 0.012 nm/pixel in the 170–600 nm spectral region using a grating-integrated module that consists of 19 subgratings without any moving parts. By using two-dimensional (2D) backsideilluminated complementary metal-oxide-semiconductor (BSI-CMOS) array detector technology with 2048 × 2048 pixels, a high data acquisition speed of approximately 25 spectra per second is achieved. The physical photon-sensing size of the detector along the one-dimensional wavelength direction is enhanced by a factor of 19 to approximately 428 mm, or 38912 pixels, to satisfy the requirement of seamless connection between two neighboring subspectral regions without any missing wavelengths throughout the entire spectral region. As tested with a mercury lamp, the system has advanced performance capabilities characterized by the highest k parameter reported to date, being approximately 3.58 × 104, where k = (working wavelength region)/(pixel resolution). Data calibration and analysis as well as a method of reducing background noise more efficiently are also discussed. The results presented in this work will stimulate further research on precision spectrometers based on advanced BSI-CMOS array detectors in the future.

Abstract:A miniature, broadband, astigmatism-corrected Czerny-Turner spectrometer is proposed. Theories of the broadband astigmatism correction using freeform cylindrical lens are thoroughly analyzed. Comparisons of the freeform cylindrical lens method with those of titling cylindrical lens or wedge cylindrical lens methods are also described. Results show the better spectrometer performance with the new optical design over a broadband spectral range from 300 to 800 nm.
