Abstract:Nonlinear optical vibrational spectroscopies are powerful experimental tools for inspecting material properties that are difficult to acquire otherwise. As ultrafast lasers used in such experiments are typically of much broader bandwidth than vibrational modes, narrowband filtering is usually essential, and the utility of laser energy is often highly inefficient. Here we introduce an experimental scheme to break this trade-off. A broadband beam is spatially chirped as it reaches the sample, and generates sum-frequency signals upon overlapping with another broadband, unchirped beam. A narrowband spectrum can then be retrieved from the spatially dispersed image of signals, with both broadband pulses fully utilized. The scheme is also readily employed as a spatially resolved spectroscopy technique without scanning, and can be easily extended to other wave-mixing experiments.

Abstract:Far field single molecule localization microscopy (SMLM) has been established as a powerful tool to study biological structures with resolution far below the diffraction limit of conventional light microscopy. In recent years, the applications of SMLM have reached beyond traditional cellular imaging. Nanostructured interfaces are enriched with information that determines their function, playing key roles in applications such as chemical catalysis and biological sensing. SMLM enables detailed study of interfaces at an individual molecular level, allowing measurements of reaction kinetics, and detection of rare events not accessible to ensemble measurements. This paper provides an update to the progress made to the use of SMLM in characterizing nanostructured biointerfaces, focusing on practical aspects, recent advances, and emerging opportunities from an analytical chemistry perspective.

Abstract:Label-free vibrational imaging by stimulated Raman scattering (SRS) provides unprecedented insight into real-time chemical distributions. Specifically, SRS in the fingerprint region (400–1800 cm −1 ) can resolve multiple chemicals in a complex bio-environment. However, due to the intrinsic weak Raman cross-sections and the lack of ultrafast spectral acquisition schemes with high spectral fidelity, SRS in the fingerprint region is not viable for studying living cells or large-scale tissue samples. Here, we report a fingerprint spectroscopic SRS platform that acquires a distortion-free SRS spectrum at 10 cm −1 spectral resolution within 20 µs using a polygon scanner. Meanwhile, we significantly improve the signal-to-noise ratio by employing a spatial-spectral residual learning network, reaching a level comparable to that with 100 times integration. Collectively, our system enables high-speed vibrational spectroscopic imaging of multiple biomolecules in samples ranging from a single live microbe to a tissue slice.

Abstract:Sum frequency generation (SFG) vibrational spectroscopy has been proved to be a powerful technique which substantially impacts on many research areas in surface and interfacial sciences. This paper reviews the recent progress of applying this nonlinear optical technique in the studies of polymer surfaces and interfaces. The theoretical background of SFG is introduced first. Current applications of SFG in polymer science are then described in more detail to demonstrate the significance of this technique. Finally, a short summary is presented on this relatively new but widely applicable spectroscopic technique.

Abstract:Nonlinear optical spectroscopies for the investigation of solid surfaces and interfaces are reviewed. Emphasis is on the surface-specific second-order nonlinear optical response. Following a brief introduction on the fundamental, a broad variety of applications of second-harmonic (SHG) and sum-frequency (SFG) spectroscopic techniques are discussed. An outlook on exciting new opportunities provided by these techniques is presented.

Abstract:Moving the detection wavelength in vibrational sum-frequency generation (VSFG) spectroscopy to the near-infrared (> 700 nm) can potentially enable the study of molecular interfaces absorbing in the visible and give access to buried bio-interfaces at minimal absorption, reduced scattering, and negligible autofluorescence. Here, we employ an ultra-narrow bandpass thin-film optical interference filter on 180-fs, 1.03-$\mu$m laser pulses to generate an upconversion beam yielding a spectral resolution of 5 cm$^{-1}$ and VSFG wavelengths between 890 and 980 nm for molecular vibrations in the fingerprint region. We demonstrate that the beam rejected by the filter can be utilized for driving a supercontinuum-seeded near-infrared optical parametric amplifier serving as the front-end of a broadband LiGaS$_{2}$-based mid-infrared amplifier. Benchmark data on a phospholipid monolayer at the air-water interface acquired using the resulting VSFG spectrometer show the possibility of achieving high resolution and signal-to-noise ratio at short acquisition times. The scheme can also be utilized in other types of vibrational spectroscopy that derive their spectral resolution from bandpass-filtering of femtosecond near-infrared laser pulses, such as stimulated Raman scattering (SRS) and coherent anti-Stokes Raman scattering (CARS) spectroscopy.

Light on the interactions between nanoparticles and lipid membranes by interface-sensitive vibrational spectroscopy

Abstract:Nanoparticles are produced in natural phenomena or synthesized artificially for technological applications. Their frequent contact with humans has been judged potentially harmful for health, and numerous studies are ongoing to understand the mechanisms of the toxicity of nanoparticles. At the macroscopic level, the toxicity can be established in vitro or in vivo by measuring the survival of cells. At the sub-microscopic level, scientists want to unveil the molecular mechanisms of the first interactions of nanoparticles with cells via the cell membrane, before the toxicity cascades within the whole cell. Unveiling a molecular understanding of the nanoparticle-membrane interface is a tricky challenge, because of the chemical complexity of this system and its nanosized dimensions buried within bulk macroscopic environments. In this review, we highlight how, in the last 10 years, second-order nonlinear optical (NLO) spectroscopy, and specifically vibrational sum frequency generation (SFG), has provided a new understanding of the structural, physicochemical, and dynamic properties of these biological interfaces, with molecular sensitivity. We will show how the intrinsic interfacial sensitivity of second-order NLO and the chemical information of vibrational SFG spectroscopy have revealed new knowledge of the molecular mechanisms that drive nanoparticles to interact with cell membranes, from both sides, the nanoparticles and the membrane properties.

Abstract:The importance of interfaces for material science and electronic devices has stimulated great interest in the development of surface analytical tools. Among them, modern optical techniques using lasers have attracted the most attention in recent years. They have the advantage of being applicable to all interfaces accessible by light, and the high temporal, spatial, and spectral resolutions offer unique opportunities for studying ultrafast molecular dynamics and other transient phenomena at interfaces. Optical second harmonic generation (SHG) and sum-frequency generation (SFG) are particularly being noticed because of the many recent successful demonstrations of their versatility. This article briefly introduces these newly developed surface probes, first outlining the basic principles behind surface SHG and SFG, and then illustrating the power of the techniques with selected examples. A more complete treatment of the theory can be found in References 4–6. An overview of the earlier applications can be found in Reference 3.SHG arises from the nonlinear polarization P(2)(2ω) induced in a medium by an incident laser field E(ω). In the electric dipole approximation, P is given by:where is a second-order nonlinear susceptibility. For a medium with inversion symmetry, it follows directly from Eq. 1 that = 0. However, at an interface the surface nonlinear susceptibility is nonvanishing because there the inversion symmetry is necessarily broken.

Abstract:We demonstrate nonlinear focal modulation microscopy (NFOMM) to achieve superresolution imaging. Traditional approaches to superresolution that utilize point scanning often rely on spatially reducing the size of the emission pattern by directly narrowing (e.g., through minimizing the detection pinhole in Airyscan, Zeiss) or indirectly peeling its outer profiles [e.g., through depleting the outer emission region in stimulated emission depletion (STED) microscopy]. We show that an alternative conceptualization that focuses on maximizing the optical system's frequency shifting ability offers advantages in further improving resolution while reducing system complexity. In NFOMM, a spatial light modulator and a suitably intense laser illumination are used to implement nonlinear focal-field modulation to achieve a transverse spatial resolution of ∼60 nm (∼λ/10). We show that NFOMM is comparable with STED microscopy and suitable for fundamental biology studies, as evidenced in imaging nuclear pore complexes, tubulin and vimentin in Vero cells. Since NFOMM is readily implemented as an add-on module to a laser-scanning microscope, we anticipate wide utility of this new imaging technique.

Abstract:A sum frequency generation (SFG) vibrational micro-spectroscopy system was developed to examine buried heterogeneous biointerfaces. A compact optical microscope was constructed with total-internal reflection (TIR) SFG geometry to monitor the tightly focused SFG laser spots on interfaces, providing the capability of selectively probing different regions on heterogeneous biointerfaces. The TIR configuration ensures and enhances the SFG signal generated only from the sample/substrate interfacial area. As an example for possible applications in biointerfaces studies, the system was used to probe and compare buried interfacial structures of different biological samples attached to underwater surfaces. We studied the interface of a single mouse oocyte on a silica prism to demonstrate the feasibility of tracing and studying a single live cell and substrate interface using SFG. We also examined the interface between a marine mussel adhesive plaque and a CaF2 substrate, showing the removal of interface-bonded water molecules. This work also paves the way for future integration of other microscopic techniques such as TIR-fluorescence microscopy or nonlinear optical imaging with SFG spectroscopy for multimodal surface or interface studies.

Abstract:Optical second harmonic generation (SHG) and infrared-visible sum-frequency generation (SFG) as surface probes have recently attracted a great deal of attention because of their seemingly unlimited possible applications to all areas of surface science. We review here the recent progress in this field. We first discuss how surface-specific these techniques really are. This is important because the bulk contribution to the signal sets a limit to the sensitivity of such surface probes. We then give a number of examples to illustrate the power of the techniques. Most of them are chosen to show that SHG and SFG are unique in providing specific information about the particular surface or interfacial systems. The following problems will be discussed in some detail. 1. Measurements of molecular nonlinearities and the environmental effect on soluble and insoluble molecular monolayers at air/water interfaces. 2. Studies of molecular orientations of liquid surfaces. 3. Creation and detection of monolayer gratings on crystalline surfaces and measurements of anisotropic surface diffusion. Abstract:Over the last decade, infrared-visible sum-frequency generation has been developed into a powerful vibrational spectroscopic technique, especially for surface studies. We give here a brief review of the technique and a few recent applications of the technique to liquid and polymer surfaces. We also describe how the technique has found unique applications in our studies of surface melting of ice, ferroelectric ice films, and molecular chirality in chiral liquids. Doubly resonant sum-frequency generation as surface-specific two-dimensional spectroscopy is discussed.

Abstract:Vibrational spectroscopy has been extensively applied to the study of molecules in gas phase, in condensed phase, and at interfaces. The transition from spectroscopy to spectroscopic imaging of living systems, which allows the spectrum of biomolecules to act as natural contrast, is opening new opportunities to reveal cellular machinery and to enable molecule-based diagnosis. Such a transition, however, involves more than a simple combination of spectrometry and microscopy. We review recent efforts that have pushed the boundary of the vibrational spectroscopic imaging field in terms of spectral acquisition speed, detection sensitivity, spatial resolution, and imaging depth. We further highlight recent applications in functional analysis of single cells and in label-free detection of diseases.

Abstract:Sum-frequency generation vibrational spectroscopy (SFG-VS) can provide detailed information and understanding of the molecular composition, interactions, and orientational and conformational structure of surfaces and interfaces through quantitative measurement and analysis. In this review, we present the current status of and discuss important recent developments in the measurement of intrinsic SFG spectral lineshapes and formulations for polarization measurements and orientational analysis of SFG-VS spectra. The focus of this review is to present a coherent description of SFG-VS and discuss the main concepts and issues that can help advance this technique as a quantitative analytical research tool for revealing the chemistry and physics of complex molecular surfaces and interfaces.

Abstract:Sum-frequency generation vibrational spectroscopy (SFG-VS) was first developed in the 1980s and it has been proven a uniquely sensitive and surface/interface selective spectroscopic probe for characterization of the structure, conformation and dynamics of molecular surfaces and interfaces. In recent years, there have been many progresses in the development of methodology and instrumentation in the SFG-VS toolbox that have significantly broadened the application to complex molecular surfaces and interfaces. In this review, after presenting a unified view on the theory and methodology focusing on the SFG-VS spectral lineshape, as well as the new opportunities in SFG-VS applications with such developments, some of the controversial issues that have been puzzling the community are discussed. The aim of this review is to present to the researchers and students interested in molecular surfaces and interfacial sciences up-to-date perspectives complementary to the existing textbooks and reviews on SFG-VS.

Abstract:The ability to achieve sub-wavenumber resolution (0.6 cm(-1)) and a large signal-to-noise ratio in high-resolution broadband sum-frequency generation vibrational spectroscopy (HR-BB-SFG-VS) allows for the detailed SFG spectral lineshapes to be used in the unambiguous determination of fine spectral features. Changes in the structural spectroscopic phase in SFG-VS as a function of beam polarization and experimental geometry proved to be instrumental in the identification of an unexpected 2.78 +/- 0.07 cm(-1) spectral splitting for the two methyl groups at the vapor/dimethyl sulfoxide (DMSO, (CH3)(2)SO) liquid interface as well as in the determination of their orientational angles. Abstract:The difficulty in achieving high spectral resolution and accurate line shape in sum-frequency generation vibrational spectroscopy (SFG-VS) has restricted its use in applications requiring precise detection and quantitative analysis. Recently, the development of high-resolution broadband sum-frequency generation vibrational spectroscopy (HR-BB-SFG-VS) with sub-wavenumber resolution generated by synchronizing two independent amplifier lasers have opened new opportunities for probing an intrinsic SFG response. Here, we present a new flexible approach to achieve HR-BB-SFG-VS. In this system, two regeneration amplifiers shared the same oscillator laser as the seed, and a time-asymmetric visible pulse with a nearly Lorentzian line shape filtered by an etalon was used to overlap with a femtosecond broadband infrared pulse. This Lorentzian line shape of the visible pulse can greatly simplify the spectral fitting and analysis. We also demonstrated that the single-sided long visible pulse provided both high spectral resolution (1.4 cm-1) and effective suppression of the non-resonant background by detuning the time delay between visible and infrared pulses in SFG-VS measurements. With this new SFG setup, a pair of spectral splittings by 3.1 ± 0.7 and 3 ± 0.2 cm-1 for the symmetric and antisymmetric stretching of the CH3 group was resolved at the CH3CN/TiO2(110) surface, which are tentatively attributed to two different orientational methyl groups. These technological advancements can help broaden the applications of HR-BB-SFG-VS and provide solid ground for a better understanding of complex molecular structures and dynamics at interfaces.

Abstract:The quest for ultrahigh detection sensitivity with spectroscopic contrasts other than fluorescence has led to various novel approaches to optical microscopy of biological systems. Coherent nonlinear optical imaging, especially the recently developed nonlinear dissipation microscopy (including stimulated Raman scattering and two-photon absorption) and pump-probe microscopy (including excited-state absorption, stimulated emission, and ground-state depletion), provides new image contrasts for nonfluorescent species. Thanks to the high-frequency modulation transfer scheme, these imaging techniques exhibit superb detection sensitivity. By directly interrogating vibrational and/or electronic energy levels of molecules, they offer high molecular specificity. Here we review the underlying principles and excitation and detection schemes, as well as exemplary biomedical applications of this emerging class of molecular imaging techniques.
