Abstract:OBJECTIVE:To evaluate the correlation of CD44v6 and sCD44v6 expression with the biological activity of gastric carcinoma. METHODS:Mucosa samples from 103 gastric carcinoma patients and 10 healthy persons (control) were examined using immunohistochemical SP method; sCD44v6 level in peripheral blood samples was detected with ELISA in 86 gastric cancer patients, 30 gastric ulcer patients and 30 healthy controls. The follow-up period was 3-91 months for 88 patients in CD44v6 group and 1-91 months for 55 patients in sCD44v6 group. RESULTS:(1) Positive expression of CD44v6 was found in 60.2% of gastric carcinoma, where as 0% in the normal gastric mucosa. CD44v6 protein expression was positively correlated with TNM stage, perigastric lymph node metastasis, presence of cancerous embolic angiopathy and Borrmann classification (P <0.05), but was not correlated with invasion depth, differentiation, metastasis and survival (P > 0.05); 77.8% of the patient with liver metastasis had a strong CD44v6 protein expression. (2) sCD44v6 level in the peripheral blood of gastric carcinoma patient was significantly higher than that in gastric ulcer patient or healthy control; Compared with palliative gastrectomy, sCD44v6 level significantly went down after radical operation. No relationship between sCD44v6 level and the pathological features was found, which included invasion depth, perigastric lymph node metastasis, presence of cancerous embolic angiopathy, differentiation and Borrmann classification; The survival in the high sCD44v6 level group was longer than that in low sCD44v6 level group ( P = 0.0281), but no significant difference was observed by Cox Regression analysis (P = 0.415). (3) No apparent correlation was observed between CD44v6 expression in gastric cancer and sCD44v6 concentration in peripheral blood (P>0.05). CONCLUSION:Detection of CD44v6 expression in the gastric cancer may be helpful in evaluating the biological features and the survival in gastric carcinoma. The level of sCD44v6 in the blood may be also helpful in differential diagnosis, evaluation of surgical treatment and biological activity for gastric cancer. No apparent correlation is observed between CD44v6 expression in the cancer and that of sCD44v6 in the blood.
CD44 Expression is Predictive of Poor Prognosis in Pharyngolaryngeal Cancer: Systematic Review and Meta-Analysis

Abstract:Pharyngolaryngeal cancer is one of the most common head and neck cancer worldwide, and the early diagnosis and prognosis prediction are still difficult because of lacking in reliable cell markers. Although the expression of CD44 has been reported to correlate with poor prognosis of pharyngolaryngeal cancer in most literatures, some controversies still exist. Since the limited patient numbers within independent studies, here we performed a meta-analysis to clarify the correlations between CD44 expression and clinicopathological features and prognosis in pharyngolaryngeal cancer. A search of PubMed, ISI Web of Science and China National Knowledge Infrastructure databases (up to June 2013) was performed. Nineteen studies with 1,405 patients met the inclusion criteria. The expression of pan-CD44, including all variant isoforms, was detected in 58.0% (14.1-79.2%) specimens, while CD44-v6 (variant isoform 6 of CD44) was expressed in 54.8% (12-79.2%). In pooled analysis, CD44 expression was significantly associated with larger tumor size (T category, RR (relative risk) = 1.21, 95% CI: 1.01-1.46), lymph nodes metastasis (N category, RR = 1.94, 95% CI: 1.38-2.73) and poor prognosis [3-year overall survival (OS): RR = 0.70, 95% CI: 0.53-0.91; 5-year OS: RR = 0.66, 95% CI: 0.66-0.94]. In the stratified analysis of CD44 isoforms, high expression of CD44-v6 was related with a poor 5-year OS rate (RR = 0.53, 95% CI: 0.37077). We propose that CD44 expression is associated with tumor size, lymph node metastasis, and poor prognosis in pharyngolaryngeal cancer patients.

Abstract:Objective To investigate the relationship between standard CD44(CD44s)expression and tumor stage and prognosis in colorectal cancer.Methods CD44s expression was detected by immunohistochemistry in tumor tissues and normal mucosa specimens from 74 cases of primary colorectal cancer.Results Of 74 cases,the expression of CD44s in colorectal cancer tissue and normal mucosa specimens was documented in 42% and 16%,respectively.The expression of CD44s in primary tumors significantly increased in stage Ⅲ and Ⅳ than in stage Ⅰ and Ⅱ(39% vs.6%,x~2=8.46,P＜0.01).A statistically significant correlation was observed between the overall survival and CD44s expression(x~2=17.82,P＜0.01).Furthermore,a poor prognosis of CD44s-positive tumors was observed in patients with stage Ⅲ and Ⅳ colorectal cancer(x~2=16.23,P＜0.01),but not in patients with stage Ⅰ and Ⅱ colorectal cancer(x~2=1.34,P＞0.05).Multivariate analysis indicated that TNM stage and CD44s expression were independent predictors of overall survival in colorectal cancer.Conclusion CD44s overexpression is involved in the progress of colorectal cancer and predicting the prognosis.

Abstract:Objective:To study the expression of the CD44 variant exon 6 product in colorectal cancer and evaluate its clinical implication. CD44v6 was determined in reseeted tumors obtained from 32 colorectal cancer patients.Methods:The expression of CD44v6 was analyzed by using immunohistochemical technique.Results:There was no significant correlation between CD44v6 and histologic type of the tumor, differentiated degree of the tumor, TNM grading, Dukes grading, or serum CEA level.Conclusion:There was a significant correlation between CD44v6 immunoreactivity and both lymph node and hematogenous metastasis.

Abstract:Objective To research the comelation between expression of CD44v6 in colorectal cancer and pathological factors. Methods The expression of CD44v6 in 79 cases with colorectal cancer was examined by im-munohistochemical staining. Results Our data showed that the expression of CD44v6 in colorectal cancer was not correlated with tumor region,depth of invasion and histologic type. The frequency of positive caseas in patients with lymph node metastasis was significantly higher than that in patients without lymph node metastasis(P<0. 01) , therefore. The expression of CD44v6 was significantly associated with lymph node metastasis. . Conclusions CD44v6 may be a recurrence and prognostic indicator in patients with colorectal cancer. It is very useful for patients with colorectal cancer in higher expression of CD44v6 adopting adjuvant chemotherapy and radiotherapy.

Abstract:OBJECTIVE:To investigate the relation between protein expression of 4 genes [P53,c-erbB-2,vascular endothelial factors(VEGF) and CD44]and survival rate in stage II( colorectal cancer(CRC) patients without radiochemotherapy after radical resection.METHODS:One hundred and fifty-nine cases of stage II(CRC without radiochemotherapy were enrolled in this study. The clinicopathological date and 5-year follow-up data were reviewed. Streptavidin-peroxidase immunohistochemical technique was used to detect the expression of P53, c-erbB-2, VEGF and CD44 in formalin-fixed, paraffin embedded sections of CRC tissues from above 159 patients.RESULTS:The 5-year survival rate was 82.4%. The rates of positive expression of P53, c-erbB-2, VEGF and CD44 were 58.5%(93/159), 26.4%(42/159), 57.9%(92/159) and 40.0%(54/159) respectively. The 5-year survival rates of positive expression patients were not significantly different with those of negative expression. chi(2) analysis showed that the positive expressions of 4 genes had no relationships with the prognosis.CONCLUSION:The expression of 4 gene proteins has no relationship with the prognosis of stage II( CRC patients without radiochemotherapy after radical resection.
Expression of CD44 Mrna in Human Colorectal Tumors

Abstract:Objective To test the expression of CD44 gene in colorectal polyps, cancers in contrast to that of normal mucosae, and investigate its importance in the progression and metastasis of colorectal tumors. Methods The expression of CD44 mRNA was examined in cancerous tissues and adjacent noncancerous mucosae from 26 patients with colorectal cancer and in colorectal polyps from 7 patients by reverse transcription polymerase chain reaction and Southern blot hybridization. Results Strong expression of the epithelial form and variant forms containing variant exon 6 or 10 of the CD44 gene was detected in polyps and cancerous tissues, whereas it was very weak or not detectable in normal colonic mucosae. Significant correlations were not observed between the epithelial variant/hematopoietic variant (E/H) ratio and clinical features( t test, P 0 05). The E/H ratio was 1 4±1 0 in Dukes' stages A and B compared with 1 4±0 9 in stages C and D. The E/H ratio was 1 6±1 0 in cancers infiltrating to muscular layer compared with 1 3±0 9 in cancers infiltrating through the colonic wall. The E/H ratio was 1 4±1 0 in cancers with metastasis compared with 1 40±1 05 in cancers without metastasis. Conclusions Expression of CD44 epithelial forms was significantly higher in colorectal tumor tissue than in matched normal mucosa. CD44 variant forms were expressed in an early stage of colorectal tumorgenesis. Variant forms sharing exon V6 or V10 may play a role in colorectal tumorgenesis and progression. The clinical value of E/H ratio in colorectal cancer needs further elucidation.
The Clinical Significance of the CD44v6 Expression in Peripheral Blood Mononuclear Cell and Cancerous Tissue of Patients with Gastric Cancer

Abstract:Objective To investigate the clinical significance of cell surface differentiation antigen 44 (CD44) expression that shared spliced variant 6 (CD44v6)in peripheral blood mononuclear cell(PBMC)and cancerous tissue of patients with gastric carcinoma.Methods Flow cytometry was used to analyze the CD44v6 expression in the primary tumor tissue and these corresponding paracancer normal tissue after gastrectomy and in PBMC at pre-operation and post-operation one week later in 50 patients with gastric cancer and 20 normal controls.Results CD44v6 expression in PBMC was significantly higher in pre-operation of gastric cancer (9.72%±6.65%) than that in both normal controls(3.11%±1.15%) and post-operation one week later (4.44%±2.33%,P0.001 and P0.01 respectively). The significant increase (P0.01) of CD44v6 expression was found in cancerous tissue (3.67%±0.63%) when compared with corresponding paracancer normal tissue of gastric cancer(1.33%±0.51%). The level of CD44v6 expressions in PBMC and cancerous tissue in patients with gastric cancer of Ⅲ ± Ⅳ stage(12.56%±6.97% and 3.91%±0.87% respectively) was significantly higher than that of Ⅰ ±Ⅱ stage(6.89%±3.37% and 2.86%±0.65% respectively,both P0.05). CD44v6 expressions in PBMC and cancerous tissue in patients with lymph node metastasis (11.29%±5.36% and 4.41%±0.79% respectively)were significantly higher than that of patients without lymph node metastasis(7.32%±4.28% and 2.61%±0.78% respectively,both P0.05).Conclusions The level of CD44v6 expression in PBMC may be a marker of monitoring the changes of CD44v6 in gastric cancerous tissue and of important clinical value in the diagnosis,judgment of tumor development and metastasis and therapeutic evaluation for the patients with gastric cancer by flow cytometry.
Cd44v6 Overexpression Related to Metastasis and Poor Prognosis of Colorectal Cancer: A Meta-Analysis

Abstract:CD44v6 has recently been reported as a biomarker for colorectal cancer. However, the clinical and prognostic significance of CD44v6 in colorectal cancer remains controversial. Therefore, we performed a meta-analysis to clarify this issue. A comprehensive literature search was performed using Medline, Embase and Web of Science, and the statistical analysis was conducted using Stata software. A total of twenty-one studies including 3918 colorectal cancer cases were included. The pooled analysis showed that CD44v6 overexpression in colorectal cancer was an independent prognostic marker correlating with lower 5-year overall survival rate (OR=0.78, 95% CI=0.67-0.91, p=0.001). CD44v6 overexpression was also associated with more lymph node invasion (OR=1.48, 95%CI=1.02-2.15, p=0.04), and advanced Dukes stage (OR= 2.47, 95% CI=1.29-4.73, p=0.01). In addition, while excluding Zolbec's study, CD44v6 overexpression was associated with distance metastasis (OR=1.65, 95% CI= 1.13-2.40, p=0.01). Taken together, this meta-analysis suggested that CD44v6 is an efficient prognostic factor in colorectal cancer.

Abstract:We aim to investigate the variation of CD44v6 expression in the normal-adenoma-primary carcinoma-liver metastasis sequence and its prognostic impact on colorectal carcinomas. The difference in CD44v6 expression between the tumor center and invasive front was also assessed. Immunohistochemistry was performed for CD44v6 on two cohorts. The first was tissue microarrays including 402 primary CRCs sampled from the tumor center and the invasive margin. The second was whole-tissue sections, consisting of 217 adenomas, 72 primary carcinomas, and the corresponding metastatic carcinomas. In the first cohort, we found that CD44v6 down-regulation was inclined to lymph node metastasis and perineural invasion, and had an unfavorable prognosis compared with CD44v6 up-regulation. In the second cohort, CD44v6 expression was predominant in adenoma over primary carcinoma and liver metastasis in multiple steps (normal < adenoma > primary carcinoma and liver metastasis). In addition, our analysis showed that CD44v6 expression was decreased at the invasion front of the CRC compared with the center of the tumor. In conclusion, the maximal expression of CD44v6 in adenoma plays a crucial role in colorectal carcinogenesis, while loss of CD44v6 expression on the cell surface of the tumor edge enhances the progression of metastasis. CD44v6 down-regulation is an independent prognostic factor for strikingly worse disease-specific survival.

Abstract:Objective To investigate the correlation of expression levels between CD 44s +,CD 44V 5 + as well as CD 44V 6 + cells in peripheral blood and tumor metastasis,prognosis of patients with colorectal carcinoma.Methods The expression rates of CD 44s +,CD 44V 5 + and CD 44V 6 + in peripheral blood cells from 62 patients with colorectal carcinoma were measured by the flow cytometry.The correlations of expression rates between CD 44s+ cells and CD 44V 5 + cells or between CD 44s + cells and CD 44V 6 + cells were analyzed.Results CD 44V 5 + and CD 44V 6+ cells percentages of patients with colorectal carcinoma were significantly higher,while CD 44s + cells percentage was marked lower,P0.05,than those of normal control,P0.01.CD 44V 5 + cells and CD 44V 6 + cells percentages of patients with tumor metastases were significantly higher than those of patients without tumor metastases,P0.01,while CD 44s + cells percentage was remarkable lower,P0.05.The correlationanalysis showed that CD 44s + cells expression rate was negatively correlated to CD 44V 5 + cells and CD 44V 6 + cells percentages,r 1=-0.84,P 10.05;r 2=-0.87,P 20.01.Conclusions CD 44V 5 + and CD 44V 6 + expressions of patients with colorectal carcinoma are closely related to tumor metastases,so the detection of CD 44 splicing variants expression rates and their transforming features can provide significant evidences for the development and prognosis of colorectal carcinoma.
Prognostic Value of CD44v6 Expression in Breast Cancer: a Meta-Analysis.

Abstract:PURPOSE:The prognostic value and clinical significance of CD44 variant isoform v6 (CD44v6) in breast cancer remains controversial. Our study aimed to generalize the correlation between CD44v6 expression and clinicopathological features and prognosis in breast cancer by using a meta-analysis.METHODS:We performed a comprehensive search of relevant literature from PubMed, Cochrane Database, and EMBASE database that were published before January 2018. The pooled ORs and HRs with 95% CIs were used to estimate the effects.RESULTS:Thirteen articles comprising 1,458 patients were included for analysis. The results revealed that CD44v6 expression was associated with histological grade (overall: OR=1.56, 95% CI [1.06, 2.29], P=0.023; Asian: OR=1.78, 95% CI [1.12, 2.85], P=0.016) and lymph node metastasis (overall: OR=1.96, 95% CI [1.01, 3.78], P=0.046; Asian: OR=2.11, 95% CI [1.00, 4.44], P=0.049). CD44v6 expression was significantly associated with poor prognosis in patients with breast cancer (overall survival: overall: HR=1.55, 95% CI [1.09, 2.22], P=0.015; Asian: HR=2.22, 95% CI [1.34, 3.68], P=0.002).CONCLUSION:Our meta-analysis demonstrates that CD44v6 is significantly associated with poor prognosis, histological grade, and lymph node metastasis in breast cancer patients, especially among Asian patients.

Abstract:Objective: To study the relationship between expressions of CD44v6 and PCNA proteins and clinical pathological factors of colorectal carcinoma (CRC). Methods: Expressions of CD44v6 and PCNA proteins were detected with immunohistochemistry in 65 cases of CRC. Results: The positive rate of CD44v6 in the metastasis group was significantly higher than that in the non-metastasis group(P < 0.05), and the high expression frequency of PCNA in patients with metastasis was also higher than that in patients with non-metastasis (P < 0.05). There was no positive correlation between the two protein expressions(rs′ = 0. 529, P > 0.05). In 31 cases of CRC with lymph node (LN) metastasis, CD44v6 expression in LN and primary carcinoma had significant correlationship(P< 0.05) and so did PCNA. Conclusion: CD44v6 may be an important marker in estimating neoplasm metastasis. PCNA may be a significant marker in estimating neoplasm malignant degree. CD44v6 and PCNA in CRC have no correlationship.

Abstract:Objective To investigate the expression of CD44 protein in hepatocellular carcinoma and clinical significances. Methods Immunohistochemistry was performed to examined 42 cases with hepatocellular carcinoma (HCC) and adjacent tissues and 10 cases of normal liver tissues for expression of CD44s and its isoforms CD44v6 proteins. Results Expression of CD44s and CD44v6 were all significantly higher in HCC tissues than it's surrounding liver and normal control, furthermore, be significantly correlated with the portal vein tumorous emboli, histological capsular infiltration and differentiation degree(P < 0.01, P < 0.05, P < 0.05, respectively). The positive expression of CD44s and CD44v6 were 17.6 %, 5.8 % respectively in groups that survival time was longer than or equal to two years, while 52.6 %, 42.2 % respectively less than two years(P < 0.05). Conclusion Expression of CD44s and CD44v6 is significantly correlated with differentiation, invasion and metastasis of HCC, and it is helpful to judge prognosis.

Abstract:Purpose:To study the relationship between the expression of CD44v6 with the clinicopathological charac- teristics and the prognosis in primary gastric carcinoma patients.Methods:A total of 188 paraffin-embedded gastric carcino- mas and 42 non-carcinomatous gastric mucosae was stained with the monoclonal antibodies CD44v6 using the EnVision~(TM) method.Results:The expression level of CD44v6 were significantly higher in the tumors (67.6%) than in the non-carcino- matous gastric mucosae (9.5%) (P0.01).The expression level of CD44v6 protein was related to lymph node metastasis (P0.05).Kaplan-Meier analysis revealed that patients with CD44v6-negative cancers had prolongation of overall survival (OS) and disease-free survival (DFS) (P0.01).By multivariate analysis,it demonstrated that vessel invasion,TNM stage and CD44v6 expression played important roles in OS or DFS (P0.05).Conclusions:The results suggest that CD44v6 is frequently expressed in primary gastric carcinoma,and CD44v6 might be used as one of the markers of poor prognosis in patients with gastric carcinoma.

Abstract:Objective To investigate the expression of CD44 variant 6 (CD44 v6) mRNA in human gastric cancer. Methods CD44v6 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR) in specimens of cancer tissue and normal gastric mucosa from 64 patients with gastric cancer. Results The expression of CD44 v6 mRNA was significantly higher in gastric cancer tissue than that in normal mucosa. The expression of CD44v6 mRNA was higher in cancer patients with lymph node and distant metastasis compared with those without metastasis. Conclusion The CD44v6 expression is significantly higher in gastric cancer than in normal tissue and is associated to lymph node involvement and distant metastasis.

The aim of this study was to evaluate the prognostic value of CD44v6 and investigate its functional roles.METHODS:The expression of CD44v6 in 208 primary gastric adenocarcinoma patient samples was examined using immunohistochemistry and its correlation with clinicopathological parameters, and 5-year patient survival was assessed. Two pairs of MGC-803 stable cells with either CD44v6 overexpression or knockdown were created. The effect of CD44v6 on cell proliferation, colony formation, migration and apoptosis was investigated using these two pairs of cells.RESULTS:Overexpression of CD44v6 was observed in all cancer cell lines. The 5-year survival rate of patients with positive CD44v6 expression is significantly worse compared to those with negative expression (38.8 vs. 73.6 %). CD44v6 and TNM stage are two independent prognostic factors of primary gastric adenocarcinoma. The risk factors for the positive CD44v6 expression are location of tumor, depth of invasion, lymph node metastasis, Lauren classification and TNM stage. In MGC-803 cells, CD44 stimulated proliferation and colony formation, antagonized oxaliplatin-induced apoptosis, but did not affect migration.CONCLUSION:CD44v6 is an important prognosis marker in gastric cancer. Tissue specificity may affect the functions of CD44v6, and further work is needed to elucidate its regulation.
[Expression and Clinicopathological Significance of CD44V6 Expression in Non-Small Cell Lung Cancer].

Abstract:BACKGROUNDTo study the expression and clinicopathological significance of CD44V6 in non-small cell lung cancer (NSCLC).METHODSCD44V6 expression in lung cancer tissues was detected in 90 patients with lung cancer by immunohistohemistry SABC method.RESULTSThe positive rate of CD44V6 expression in NSCLC was 51.1% (46/90), which was related to TNM stage, but not to cell differention, age and sex of the patients (P > 0.05). The positive rate of CD44V6 expression was 32.2% (10/31) in stage I+II patients and 61.0% (36/59) in stage III+IV disease. A highly significant difference was existed between the two groups (P < 0.01). The positive rate of CD44V6 expression was significantly higher in NSCLC with lymph node metastasis (61.0%, 36/59) than that without lymph node metastasis (32.2%, 10/31) (P < 0.01). The 3-year survival rate (65.5%, 19/29) in patients with low CD44V6 expression was significantly higher than that (35.0%, 7/20) with high CD44V6 expression (P < 0.05).CONCLUSIONSOverexpression of CD44V6 may be involved in the progression and metastasis of the cancer and prognosis of the patients with lung cancer. It might be helpful to evaluate the progression of the cancer, and to predict the metastasis and progression of lung cancer.
Prognostic Significance of CD44 in Human Colon Cancer and Gastric Cancer: Evidence from Bioinformatic Analyses

Abstract:CD44 is a well-recognized stem cell biomarker expressed in colon and gastric cancer. In order to identify whether CD44 mRNA could be used as a prognostic marker in colon and gastric cancer, bioinformatic analyses were used in this study. cBioPortal analysis and COSMIC analysis were used to explore the CD44 mutation. CD44 mRNA levels were evaluated by using SAGE Genie tools and Oncomine analysis. Kaplan-Meier Plotter was performed to identify the prognostic roles of CD44 mRNA in these two cancers. In this study, first, we found that low alteration frequency of CD44 mRNA in colon and gastric cancer. Second, the high CD44 mRNA level was found in colon and gastric cancer, and it correlated with a benign survival rate in gastric cancer. Third, CD4 and CD74 may be used as markers to predict the prognosis of colon and gastric cancer. However, the deep mechanism(s) of these results remains unclear, further studies have to be performed in the future.
Relation of NM23-H1 and CD44v6 Expression to Patient's Prognosis in the Non-Small Cell Lung Cancer Without Lymph Node Metastasis

Abstract:[Objective] To research the expressions of NM23-H1、CD44v6 in the non-small cell lung cancer without lymph node metastasis(N0NSCLC)and their clinical significances.[Methods] The immunohistochemistry was used to detect the expressions of NM23-H1 and CD44v6 in 60 N0NSCLC from operatal patients and the relationship between the expressions and 5-years prognosis of patients after operation was analysed.[Results] Both of NM23-H1 and CD44v6 expressions were no significant differences between squamous cell carcinoma and adenocarcinoma.However,the expression of NM23-H1 related positively to 5-year survival rate of the patient(r=0.887),but that of CD44v6 negatively(r=-0.784).[Conclusion] Detection of NM23-H1 and CD44v6 expressions might be useful to predict the prognosis of patients with N0 NSCLC.
